Abstract: Tea plant (Camellia sinensis L.) is capable of accumulating a large amount of fluorine (F) in leaves without showing toxicity symptoms and thus offers a good model for exploring F tolerance mechanisms. Here, gas chromatog. time-of-flight mass spectrometry (GC-TOF-MS) was used to investigate metabolic changes in leaves of tea seedlings under control (0 mM), low F (0.2 mM) and high F (0.8 mM) conditions. Differentially changed metabolites such as galacturonic acid, lactose, fructose, malic acid, alanine were identified by the comparison among the three F treatment groups. A pathway map depicted based on the KEGG database reflected the involvement of pectin biosynthesis metabolism in F stress response. The gene expression and enzyme activity of key enzymes involved in pectin biosynthesis pathway and the content of pectic polysaccharides were increased by exogenous F treatments, indicating the promotion effect of F on the pectin biosynthesis. Pectin was also immunochem. stained in vivo using monoclonal antibody (2F4), which confirmed the increment. The increased pectin might contribute to combining the exogenous F in tea leaves. This research provided some novel insights into further research on F detoxification of plants.

The monoterpene linalool is a major contributor to aroma and flavor in peach (Prunus persica) fruit. It accumulates during fruit ripening, where up to ∼40% of the compound is present in a non-volatile glycosylated form, which affects flavor quality and consumer perception by retronasal perception during tasting. Despite the importance of this sequestration to flavor, the UDP-glycosyltransferase (UGT) responsible for linalool glycosylation has not been identified in peach. UGT gene expression during peach fruit ripening and among different peach cultivars was analyzed using RNA sequencing, and transcripts correlated with linalyl-β-d-glucoside were selected as candidates for functional anal. Kinetic resolution of a racemic mixture of R,S-linalool was shown for PpUGT85A2, with a slight preference for S-(+)-linalool. PpUGT85A2 was shown to catalyze synthesis of linalyl-β-d-glucoside in vitro, although it did not exhibit the highest enzyme activity between tested substrates. Subcellular localization of PpUGT85A2 in the cytoplasm and nucleus was detected. Application of linalool to peach leaf disks promoted PpUGT85A2 expression and linalyl-β-d-glucoside generation. Transient expression in peach fruit and stable overexpression in tobacco and Arabidopsis resulted in significant accumulation of linalyl-β-d-glucoside in vivo. Taken together, the results indicate that PpUGT85A2 expression is a major control point predicting linalyl-β-d-glucoside content.

Eight carbon (C8) compounds are the key characteristic flavors of mushrooms. The quant. anal. of the volatiles in mushrooms is challenging especially with the unattended HS-SPME-GC-MS. An unattended HS-SPME-GC-MS/MS in combination with novel sample preparation of the complete control of volatile emissions was developed for the quantitation of the C8 volatiles in mushrooms. The sample preparation strategy was composed of freeze-drying, rehydration, and the addition of a 15% citric acid solution With this strategy, the volatile emission from mushroom was fully controlled at a certain time point. This method was found to be highly reliable, sensitive, precise, and accurate. This method was successfully applied to measure the contents of the C8 volatiles in the beech, button, and shiitake mushrooms. 1-Octene-3-ol was the most predominant compound in the mushrooms, representing 62.4, 69.0, and 89.2% of the total C8 volatiles in the beech, button, and shiitake mushrooms, resp.

In this study, we demonstrate that the com. available organoborane reagent 9-borabicyclo[3.3.1]nonane (H-B-9-BBN) can serve as a catalyst for the sequential double hydroboration of alkynes using pinacolborane (HBpin). This strategy, which is effective for a wide range of terminal alkynes, is predicated upon a key C(sp3)-B/B-H transborylation reaction. Elemene is a natural compound extracted from Zingiberaceae plants, and is used in various cancer. However, the efficacy and safety elemene combined with chemotherapy in advanced gastric cancer (GC) are lack of systematic assessment. We searched the PubMed, EMBASE, Web of Science, Cochrane Library, China Academic Journals (CNKI), Chinese Science and Technol. Journals (CQVIP) and Chinese Biomedical Literature databases. Randomized controlled trials (RCTs) comparing elemene plus chemotherapy with chemotherapy alone in participants with advanced GC and reporting at least one of the following outcomes were selected and assessed for inclusion. JADAD scale was used to assess the quality. Data was screened and extracted by two independent investigators. The primary clin. outcome was overall response rate (ORR); the secondary outcomes were quality of life (QOL) and adverse events (AEs). Anal. was performed using Review Manager 5.3. Sixteen RCTs matched the selection criteria, which reported on 969 subjects. Risk ratios (RR) and corresponding 95% confidence intervals (CIs) were pooled for ORR, life quality based on KPS, and risk of AEs. Compared to chemotherapy alone, elemene combined with chemotherapy in the treatment of GC may increase the efficiency of ORR(RR: 1.41; 95% CI: 1.23-1.60; P<.0001), improve their life quality based on KPS (RR: 1.84; 95% CI: 1.45-2.34; P<.00001), and reduce the adverse reactions, including leukopenia(RR: 0.73; 95% CI: 0.62-0.85; P<.00001), neutropenia (RR: 0.75; 95% CI: 0.60-0.95; P=.02), anemia (RR: 0.76; 95% CI: 0.60-0.95; P=.02), thrombocytopenia (RR: 0.56; 95% CI: 0.43-0.73; P<.00001). Nausea and vomiting (RR: 0.84; 95% CI: 0.84-1.07; P=.39), diarrhea (RR: 0.69; 95% CI: 0.41-1.15; P=.15), neurotoxicity (RR: 0.77; 95% CI: 0.59-1.00; P=.05) and hepatic dysfunction (RR: 0.95; 95% CI: 0.58-1.54; P=.83) were similar between two groups. Elemene may have the potential to improve the efficacy and reduce the AEs of chemotherapy for gastric cancer. However, the long-term, high-quality researches with a large sample size in different populations are required. Salix variegata Franch. is a riparian shrub species that can tolerate long-term complete submergence; however, the mol. mechanisms underlying this trait remain to be elucidated. In this study, we subjected S. variegata plants to complete submergence for 60 d and collected stems to perform transcriptomic and metabolomic analyses, as well as quant. reverse transcription-polymerase chain reaction (qRT-PCR) assays. Results revealed that photosynthesis and the response to light stimulus were inhibited during submergence and recovered after desubmergence. Ethylene and abscisic acid (ABA) signaling could be important for the long-term submergence tolerance of S. variegata. Jasmonic acid (JA) signaling also participated in the response to submergence. Raffinose family oligosaccharides, highly unsaturated fatty acids, and specific stress-related amino acids accumulated in response to submergence, indicating that they may protect plants from submergence damage, as they do in response to other abiotic stressors. Several organic acids were produced in S. variegata plants after submergence, which may facilitate coping with the toxicity induced by submergence. After long-term submergence, cell wall reorganization and phenylpropanoid metabolic processes (the synthesis of specific phenolics and flavonoids) were activated, which may contribute to long-term S. variegata submergence tolerance; however, the detailed mechanisms require further investigation. Several transcription factors (TFs), such as MYB, continuously responded to submergence, indicating that they may play important roles in the responses and adaptation to submergence. Genes related to oxidative stress tolerance were specifically expressed after desubmergence, potentially contributing to recovery of S. variegata plants within a short period of time.

Purpose: The aim of the study was to examine the effects of resveratrol upon hepatic endoplasmic reticulum stress (ERS) and insulin sensitivity in vivo and in vitro. Material and methods: C57BL/6J mice were fed a high-fat diet (HFD) for 8 wk, and insulin resistance was evaluated by the i.p. glucose tolerance test (IPGTT). Mice were then treated with resveratrol for 12 wk and blood and liver samples collected. Blood biochem. indicators were determined by kits, liver protein expression was determined by western blot, and morphol. changes were observed by histol. staining. Palmitic acid (PA)-induced insulin-resistant HepG2 cells were established. Cells were exposed to 100, 50 or 20 μM resveratrol for 24 h, and proliferation/cytotoxicity was determined Cells were divided into five groups: control, PA, PA + Rev (100 μM), PA + Rev (50 μM) and PA + Rev (20 μM) groups. After 24 h of treatment, cellular proteins were analyzed the same way as animal tissues. Results: The IPGTT confirmed that the insulin resistance model was established successfully. After resveratrol treatment, fasting blood glucose and cholesterol levels declined and the quant. insulin sensitivity check index increased. Western-blot results showed that resveratrol-treated HFD mice had reduced hepatic levels of p-PERK, ATF-4 and TRIB3, and increased the levels of ATF-6, p-AKT and p-GSK3β. In the cell model, resveratrol with 100 and 50 μM enhanced ERS and insulin resistance, whereas 20 μM had beneficial effects, similar to the animal model. Conclusion: Resveratrol reduced hepatic ERS, thereby improving insulin sensitivity and glucose levels. However, high doses of resveratrol had harmful effects on cells, elevating ERS and insulin resistance. The safe dose of resveratrol needs further investigation.

In this study, therapeutic-laden Konjac glucomannan (KGM) hydrogel was prepared by injecting polydopamine (PDA)/5-fluorouracil (5-FU)/calcium folinate (CF)/KGM alk. solution to the target area, for the synergistic tumor photothermal and chemotherapy. In alk. solution, dopamine can be synchronously polymerized to form PDA, while KGM can be hydrolyzed and transformed into a thermally irreversible KGM hydrogel. Besides, the alk. condition can increase the solubility of 5-FU. The hydrogel matrix can control the release of 5-FU and cf. PDA renders the composite hydrogel excellent tumor photothermal therapy ability. Moreover, CF can be used as a synergist to enhance the tumor chemotherapy efficiency of 5-FU. With convenient handling, excellent photothermal conversion ability and high biocompatibility, the multifunctional hydrogel system is anticipated to find a promising translational potential in clin. therapeutic applications.
