In the present study, germinated brown rice (GBR) from three Japonica and three Indica rice cultivars were subjected to metabolomics anal. and volatile profiling. The statistical assessment and pathway anal. of the metabolomics data demonstrated that in spite of significant metabolic changes in response to the germination treatment, the Japonica rice cultivars consistently expressed higher levels of several health-promoting compounds, such as essential amino acids and γ-aminobutyric acid (GABA), than the Indica cultivars. No clear discriminations of the volatile profiles were observed in light of the subspecies, and the concentrations of the volatile organic compounds (VOCs), including alkenes, aldehydes, furans, ketones, and alcs., all exhibited significant reductions ranging from 26.8% to 64.1% after the germination. The results suggest that the Japonica cultivars might be desirable as the raw materials for generating and selecting GBR food products for health-conscious consumers.

It is still uncertain whether the consumption of Lachnum sp. polysaccharides (LEP) alleviates colorectal cancer (CRC) through the gut microbiota. In this study, our efforts are focused on the influence of LEP on CRC, intestinal barrier and inflammation, and fecal microbiota and the metabolites, in azoxymethane (AOM)/dextran sodium sulfate (DSS)-induced CRC mice. Results showed that LEP inhibited CRC mouse colon shortening and weight loss, decreased tumor incidence, restored intestinal barrier integrity, and reduced excessive inflammation. LEP consumption significantly altered microbiota overall structure and community, with reduced pernicious bacteria (such as Parabacteroides, Escherichia_Shigella, Desulfovibrio and Helicobacter), and increased beneficial bacterium (such as Alistipes, Alloprevotella and Ruminiclostridium). Fecal-metabolome profile indicated that a total of 43 metabolites were clearly changed, with 10 down-regulated and 33 up-regulated metabolites. In addition, short-chain fatty acids (SCFAs), including acetic acid, propionic acid and n-butyric acid, were significantly increased after LEP administration. Moreover, a strong correlation between the fluctuant gut microbiota and metabolites was found. These findings provided not only deeper insights into the responsibility of LEP for CRC alleviation, and but also the potential of LEP as a promising candidate for CRC prevention and treatment.

Scrophulariae Radix (SR) is one of the oldest and most frequently used Chinese herbs for oriental medicine in China. In order to investigate the difference in chem. constituents using different processing methods, the two-dimensional (2D) 1H-13C heteronuclear single quantum correlation (1H-13C HSQC)-based metabolomics approach was applied to extensively characterize the difference in the chem. components in the extracts of SR processed using different processing methods. In total, 20 compounds were identified as potential chem. markers that changed significantly with different steaming durations. Seven compounds can be used as potential chem. markers to differentiate processing by sweating, hot-air drying, and steaming for 4 h. These findings could elucidate the change of chem. constituents of the processed SR and provide a guide for the processing. In addition, our protocol may represent a general approach to characterizing chem. compounds of traditional Chinese medicine (TCM) and therefore might be considered as a promising approach to exploring the scientific basis of traditional processing of TCM.

Intracellular bacteria (ICBs) are among the most life-threatening causes of drug resistance. Challenges remains in the intracellular drug release specific to ICB-infected cells and efficient uptake into ICBs. In this study, mannose-grafted polymers containing enzymes-responsive and tetraphenylethylene segments (mPET) are assembled into nanoparticles with loading complexes of deferoxamine-ciprofloxacin conjugates with Fe3+ (DFeC). The aggregation-induced emission (AIE) of tetraphenylethylene segments is overlapped with DFeC absorptions, leading to fluorescence resonance energy transfer (FRET)-caused quenching of mPET@DFeC nanoparticles. Nanoparticles are efficiently acquired by infected macrophages via mannose mediation, and the DFeC release is triggered by intracellular lipase and alk. phosphatase specific to ICB-engulfed macrophages, followed by deferoxamine-mediated ingestion of ciprofloxacin into ICBs. The gradual alleviation of FRET effect and the concurrent restoration of AIE activity demonstrate capabilities of dynamically tracking the drug release and ICB treatment outcome. The mPET@DFeC treatment inhibits the hematol., hepatic and nephric toxicities caused by ICB infections, and all the infected mice survive with dramatic reductions of bacterial levels in livers (over 430 folds), spleens (over 240 folds) and kidneys (5.6 × 104 folds). Thus, this study has provided a feasible strategy to achieve intracellular enzymes-responsive and traceable release of antibiotics and then deferoxamine-mediated bacterial ingestion for ICB destruction.

Stimuli-responsive polymers are of great interest due to their ability to translate changing environmental conditions into responses in defined materials. One possibility to impart such behavior is the incorporation of optically active mols. into a polymer host. Here, we describe how sensor mols. that consist of a π-extended benzothiadiazole emitter and a naphthalene diimide quencher can be exploited in this context. The two optically active entities were connected via different spacers and, thanks to attractive intramol. interactions between them, the new sensor mols. assembled into cyclic structures in which the fluorescence was quenched by up to 43% when compared to solutions of the individual dyes. Detailed spectroscopic investigations of the sensor mols. in solution show that the extent of donor/acceptor interactions is influenced by various factors, including solvent polarity and ion concentration The new sensor mol. was covalently incorporated into a polyurethane; the investigation of the optical characteristics in both the solid and solvent-swollen states indicates that a stimulus-induced formation of associated dye pairs is possible in polymeric materials. Indeed, a solvatochromic quenching effect similar to the behavior in solution was observed for solvent-swollen polymer samples, leading to an effective change of the green emission color of the dye to a yellow color.

Keemun, Assam, Darjeeling and Ceylon black teas are honored as the world’s four most famous black teas, and their excellent aroma qualities are well received by people around the world. In this study, aroma components in these four types of teas were analyzed by comprehensive two-dimensional gas chromatog. time-of-flight mass spectrometry (GC × GC-TOFMS) and gas chromatog.-olfactometry (GC-O) technologies. A total of 42 aroma-active compounds were ultimately identified, especially benzeneacetaldehyde, geraniol, (Z)-3-hexen-1-yl hexanoate, trans-β-ionone, cis-linalool oxide (pyranoid), hotrienol, and Me salicylate presented the strongest aroma strengths with pleasant scents in all tested teas. The quantification results indicated that 19 compounds including (Z)-3-hexenol, 1-octen-3-ol, linalool, phenylethyl alc., hexanal, benzeneacetaldehyde, limonene, heptanoic acid, (Z)-3-hexen-1-ol, acetate, benzyl alc., trans-linalool oxide (furanoid), hotrienol, 1-octen-3-one, 2-nonanone, (E)-2-octenal, nonanal, β-myrcene, 2-pentylfuran, and methylpyrazine were identified as the key compounds with odor activity values (OAVs) higher than 1.0 in the world’s four most famous black teas. Notably, the comparison of GC-O and OAV calculation results showed that Me salicylate (Ceylon), (E)-2-octenal (Assam), benzeneacetaldehyde (Keemun) and linalool and trans-linalool oxide (furanoid) (Darjeeling) might be the most definitive odorants in the corresponding tea categories.

In this research work, a water-soluble polysaccharide (LAP) isolated from the fruits of Lycium arabicum was investigated. LAP contains carbohydrates (82.45±1.23%), protein (1.56±0.21%), and uronic acids (3.56±0.34%). The anal. of the monosaccharide composition revealed the presence of rhamnose, arabinose, galactose, glucose and mannose in a molar ratio of 4.7 : 1.5 : 1 : 8.7 : 16.4 : 5.6. The extracted polysaccharide (PS) was considered as heterogeneous and highly branched by interpreting its GC/MS, FT-IR and NMR data. Crystallinity of LAP was inferred from its X-ray diffractometry (XRD) and SEM (SEM) anal. LAP exhibited an interesting stability at high temperatures (∼254°C) and in a wide range of pH (3-9) deduced, resp., from its DSC and zeta potential anal. LAP displayed a strong antioxidant activity at low concentrations evaluated by the 2,2-diphenyl-1-picrylhydrazyl (DPPH)-radical scavenging, ferric reducing activity power (FRAP), free radical scavenging ability, superoxide radical-scavenging and hydroxyl radical-scavenging abilities. Inhibition of erythrocyte hemolysis and lipid peroxidation was also assessed. In 5 h, LAP treatment allowed the protection of the damaged erythrocytes caused by AAPH (2,2-azobis(2-amidinopropane) dihydrochloride), to reduce the level of malondialdehyde (MDA) as well as to increase the reduced glutathione (GSH) level.

In the search for highly reactive oxidants we have identified high-valent metal-fluorides as a potential potent oxidant. The high-valent Ni-F complex [NiIII(F)(L)] (2, L=N,N’-(2,6-dimethylphenyl)-2,6-pyridinedicarboxamidate) was prepared from [NiII(F)(L)]- (1) by oxidation with selectfluor. Complexes 1 and 2 were characterized by using 1H/19F NMR, UV-vis, and EPR spectroscopies, mass spectrometry, and X-ray crystallog. Complex 2 was found to be a highly reactive oxidant in the oxidation of hydrocarbons. Kinetic data and products anal. demonstrate a hydrogen atom transfer mechanism of oxidation The rate constant determined for the oxidation of 9,10-dihydroanthracene (k2=29 M-1 s-1) compared favorably with the most reactive high-valent metallo-oxidants. Complex 2 displayed reaction rates 2000-4500-fold enhanced with respect to [NiIII(Cl)(L)] and also displayed high kinetic isotope effect values. Oxidative hydrocarbon and phosphine fluorination was achieved. Our results provide an interesting direction in designing catalysts for hydrocarbon oxidation and fluorination.
