The inflammatory and cholesterol-depleting effects of CD were first investigated in low-d. lipoprotein receptor knockout (Ldlr-/) mice that were transplanted with Npc1nih or Npc1wt bone marrow and were fed either regular chow or a high-fat, high-cholesterol (HFC) diet for 12 wk, thereby creating an extreme model of lysosomal cholesterol-induced metabolic inflammation. In the final three weeks, these mice received daily injections of either control (saline) or CD s.c. Subsequently, the inflammatory properties of CD were investigated in vitro in two macrophage cell lines and in murine bone marrow-derived macrophages (BMDMs). While CD administration improved cholesterol mobilization outside lysosomes in BMDMs, an overall pro-inflammatory profile was observed after CD treatment, evidenced by increased hepatic inflammation in vivo and a strong increase in cytokine release and inflammatory gene expression in vitro in murine BMDMs and macrophages cell lines. Nevertheless, this CD-induced pro-inflammatory profile was time-dependent, as short term exposure to CD did not result in a pro-inflammatory response in BMDM. While CD exerts desired cholesterol-depleting effects, its inflammatory effect is dependent on the exposure time. As such, using CD in the clinic, especially in a metabolic inflammatory context, should be closely monitored as it may lead to undesired, pro-inflammatory side effects.

High-performance electromagnetic interference shielding (EMI) materials are desirable in aerospace, military and mobile electronics applications. However, fabrication of smart and ultra-lightweight EMI shielding materials with adjustable EMI shielding performance through an energy-efficient and environmental-friendly process is still challenging. Herein, we demonstrate using a conductive biomass-derived D-glucaric acid-chitosan/single-walled carbon nanotube (SWNT/GA-chitosan) polymer composite layer and a crystallized paraffin layer to fabricate ultra-lightweight polyurethane foams with shape memory and adjustable EMI shielding functions, of which the shape change via external stimulus enables the foams to adjust EMI shielding efficiency autonomously. The foam coated with synthesized conductive composite layers shows an exceptional EMI shielding effectiveness of 56 dB at an ultra-low d. of 0.03 g/cm3 with only 0.171 vol% SWNT. The foam also exhibits ultra-high durability over 2000 compression-recovery cycles. Furthermore, introduction of paraffin layer as a reversible network results in shape memory foams with high fixity ratio >95% and recovery ratio >90%. These foams exhibit a wide range of adjustable EMI shielding efficiency from 18 dB to 30 dB by controlling their macroscopic shape from compression. Such smart foams with shape memory and adjustable EMI shielding functions fabricated by an environmental-friendly strategy represent a promising material candidate for the wide applications of EMI shielding.

A green, novel and extremely efficient nanocatalyst was successfully synthesized by the immobilization of Ni as a transition metal on Fe3O4 nanoparticles coated with tryptophan. This nanostructured material was characterized using Fourier transform IR spectroscopy, transmission electron microscopy, SEM, energy-dispersive X-ray spectroscopy, thermogravimetric anal., inductively coupled plasma optical emission spectroscopy, vibrating sample magnetometry and X-ray diffraction. The prepared nanocatalyst was applied for the oxidation of sulfides, oxidative coupling of thiols and synthesis of 5-substituted 1H-tetrazoles. The use of non-toxic, green and inexpensive materials, easy separation of magnetic nanoparticles from a reaction mixture using a magnetic field, efficient and one-pot synthesis, and high yields of products are the most important advantages of this nanocatalyst.

Decaying cyanobacterial blooms carry a potential risk for submerged macrophyte and periphyton biofilms in aquatic environments. This study comprehensively studied the responses in growth, oxidative response, detoxification pathway, and ultrastructure characteristics of aquatic plants to Microcystis aeruginosa (M. aeruginosa) exudates and extracts released during the decline phase. Particular emphasis was placed on the variation of extracellular polymeric substances (EPS) and quorum-sensing signaling mols. The results showed that superoxide dismutase, peroxidase, and glutathione S-transferase were significantly induced as antioxidant response, and the malondialdehyde content increased. Increased content of MC-LR (1.129 μg L-1) and NH+4-N (1.35 mg L-1) were found in the decline phase of M. aeruginosa, which played a vital role in the damage to submerged plants. In addition, a change in the amount of osmophilic granules and a variation of organelles and membranes was observed A broad distribution of α-D-glucopyranose polysaccharides was dominant and aggregated into clusters in biofilm EPS in response to exposure to decaying M. aeruginosa. Furthermore, exposure to exudates and extracts changed the abundance and structure of the microbial biofilm community. Increased contents of N-acylated-L-homoserine lactone signal mol. might result in a variation of biofilm EPS production in response to decaying M. aeruginosa. These results expand the understanding of how submerged macrophyte and periphyton biofilms respond to environmental stress caused by exudates and extracts of decaying M. aeruginosa.

A novel method for the synthesis of epoxydibenzo[b,f][1,5]diazocines exhibiting a V-shaped mol. architecture is reported. The unique approach is based on unprecedented base-catalyzed, solvent-free autocondensation and cross-condensation of fluorinated o-aminophenones. The structure of the newly synthesized diazocines was confirmed independently by X-ray anal. and chiroptical methods. The rigidity of the diazocine scaffold allowed for the separation of the racemate into single enantiomers that proved to be thermally stable up to 140°C. Furthermore, the inertness of the diazocine scaffold was demonstrated by performing a series of typical transformations, including transition metal-catalyzed reactions, proceeding without affecting the bis-hemiaminal subunit.

In developing the new features within the CL-MS plug-in, we identified addnl. complexities associated with the application of CL reagents, arising primarily from digestion-induced bias in yield measurements and ambiguities in site localization. A strategy is presented involving the use of redundant site labeling data from overlapping peptides, the imputation of missing data, and a normalization routine to determine relative protection factors. These elements together provide for a robust structural interpretation of CL-MS/MS data while minimizing the over-reporting of labeling site resolution Finally, to minimize bias, we recommend that digestion strategies for the generation of useful overlapping peptides involve the application of complementary enzymes that drive digestion to completion.
