Effects of algae subtype and extraction condition on extracted fucoxanthin antioxidant property: A 20-year meta-analysis

Algal fucoxanthin as a carotenoid pigment possesses various health benefits, among which, the antioxidant property is one of the most explored. Current research indicated that algal fucoxanthin is generally extracted from different subtypes (microand macro-algae) under varying extraction conditions. However, it lacks information whether algae subtypes and extraction conditions present a remarkable impact on the antioxidant property of the extracted fucoxanthin. In this study, the effects of algae subtype and extraction condition (i.e., extraction solvents, temperature, time, pressure and illumination condition) on fucoxanthin antioxidant property were investigated by performing a meta-analysis. The subtotal standard mean difference (SMD) of the microalgae and macroalgae subtypes were 12.59 (95% confidence interval (CI): 3.63-21.56) and 7.20 (3.44-10.96), respectively, presenting an overlapping range. This suggested that no statistically significant differences existed in the fucoxanthin antioxidant property extracted from two subtypes, which was consistent with the results from the subgroup analysis and meta-regression. Similar observations were found for algal fucoxanthin extracted by alcohols (SMD (CI):7.18 (3.36-11.00)) or alkanes (SMD (CI):11.88 (3.62-20.15)). Moreover, the employed extraction conditions including extraction time (SMD (CI) for >= 60 min vs <60 min: 8.03 (3.55-12.50) and 7.97 (2.49-13.45)), pressure (SMD (CI) for normal vs pressurized: 7.68 (4.00-11.35) and 10.64 (0.21-21.03)), and illumination (SMD (CI) for dark vs normal: 6.91 (2.31-11.50) and 9.45 (4.17-14.73)) showed no statistical influence on fucoxanthin antioxidant property. However, extraction at higher temperature produced stronger fucoxanthin antioxidant property (SMD (CI) for room temperature vs >= 40 degrees C: 31.43 (12.27-50.59) and 7.21 (3.69-10.74)). Additionally, the fucoxanthin antioxidant property exhibited a positive concentration-dependent correlation according to meta-regression analysis. Two-dimensional (2D) lamellar graphene oxide (GO) membranes represent promising porous materials in alcohol dehydration. Building evidence suggests that dihydromyricetin (DHM), a bioactive flavonoid isolated from Hovenia dulcis, provides hepatoprotection by enhancing ethanol metabolism in the liver by maintaining hepatocellular bioenergetics, reductions of oxidative stress, and activating lipid oxidation pathways. The present study investigates the utility of DHM on hepatic mitochondrial biogenesis via activation of the AMP-activated protein kinase (AMPK)/Sirtuin (Sirt)-1/PPARG coactivator 1 (PGC)-1 alpha signaling pathway. We utilized a forced drinking ad libitum study that chronically fed 30% ethanol to male C57BL/61 mice over 8 weeks and induced ALD pathology. We found that chronic ethanol feeding resulted in the suppression of AMPK activation and cytoplasmic Sirt-1 and mitochondrial Sirt-3 expression, effects that were reversed with daily DHM administration (5 mg/kg; intraperitoneally [i.p.]). Chronic ethanol feeding also resulted in hepatic hyperacetylation of PGC-1 alpha, which was improved with DHM administration and its mediated increase of Sirt-1 activity. Furthermore, ethanol-fed mice were found to have increased expression of mitochondrial transcription factor A (TFAM), reduced mitochondrial content as assessed by mitochondrial DNA to nuclear DNA ratios, and significantly lower levels of hepatic ATP. In contrast, DHM administration significantly increased TFAM expression, hepatic ATP concentrations, and induced mitochondrial expression of respiratory complex III and V. In total, this work demonstrates a novel mechanism of DHM that improves hepatic bioenergetics, metabolic signaling, and mitochondrial viability, thus adding to the evidence supporting the use of DHM for treatment of ALD and other metabolic disorders. Improving the performance of a photonic PCR system using TiO2 nanoparticles

Nucleic acid amplification using polymerase chain reaction (PCR) method has been widely used in different fields such as agricultural science, medicine, pathogen identification, and forensics to name a few. Today, it seems inevitable to have a robust, simple PCR system for diagnostics at the point-of-care (POC) level. Many photonic PCR systems have been proposed in the literature that benefit from plasmonic photothermal heating to achieve the common PCR thermal cycling. However, non-homogeneous temperature distribution is a challenge in some of them. In the present work, to achieve more efficient gene amplification, the effect of adding TiO2 nanoparticles has been investigated in a photonic PCR benefiting from plasmonic photothermal heating. The system enjoys higher heating and cooling rates than those of conventional PCR systems while having much lower energy consumption. The average heating and cooling rates are 4.44 degrees C/s and 2.65 degrees C/s, respectively. The system demonstrates acceptable temperature stability and accuracy with negligible deviation from the set points. Furthermore, the system is capable of amplifying eight gene samples simultaneously which makes it a viable choice for POC diagnostics. A part of the Alcohol Oxidase gene has been amplified using the plasmonic thermal cycler. It is demonstrated that adding TiO2 nanoparticles with a proper concentration to the sample provides a more specific band that can be ascribed to more homogeneous temperature distribution owing to enhanced thermal conductivity. Early life stress is known to impact vulnerability to psychopathological disorders in adulthood, including anxiety and alcohol use disorder (AUD), but the mechanisms underlying susceptibility to these outcomes are not fully understood. In the current study, we used adolescent social isolation (ASI) to determine whether Heterogeneous Stock (HS) rats, an outbred model used for genetic fine-mapping, could be used to study the genetics contributing to ASI-induced anxiety- and AUD-like behavior. We isolated (ASI) or group-housed (adolescent group-housed; AGH) 64 male HS rats at 4 weeks of age. After 5 weeks in these housing conditions, multiple anxiety and coping/despair-like behaviors were measured. All rats were then individually housed and assessed for voluntary ethanol self-administration. At euthanasia, synaptoneurosomes were isolated from a subset of brains to examine the expression of two proteins associated with alcohol drinking-related behaviors, GluA1 and SK2, in the dorsal (dHC) and ventral hippocampus (vHC). We found that ASI increased hyperactivity in the open field test relative to AGH, with no changes in other anxiety-like behaviors. Surprisingly, ASI rats demonstrated decreased immobility and increased climbing in the forced swim test relative to AGH. In contrast to prior studies by us and others, we found no difference in self-administration of 20% ethanol, with decreased ethanol self-administration in ASI relative to AGH rats at higher ethanol concentrations. Furthermore, while ASI in Long-Evans rats resulted in decreased SK2 expression in vHC synaptosomes, no differences were seen in vHC synaptosomes for SK2 or GluA1 in HS rats. These results demonstrate that HS rats are protected against many of the negative effects previously seen in response to ASI, namely anxiety-like behavior and increased ethanol self-administration. One-year Follow-Up Results with Hydrogel Implant in Therapy of Hallux Rigidus: Case Series with 44 Patients

Background The Cartiva implant (CI) is being increasingly used in the surgical therapy of hallux rigidus. Patients and Methods As part of a retrospective case series with prospective follow-up (average follow-up period: 12 months), a total of 44 patients (male/female = 16/28; mean age at the time of surgery: 55.4 years) with 44 CI were analysed (VAS, EFAS-, AOFAS-score). Using a correlation analysis and a machine learning algorithm, risk factors for therapy failure were investigated. Results The overall survival rate of the CI was 93% at 12 months. The VAS, EFAS and AOFAS scores showed a significant improvement in comparison to the preoperative condition. The mobility of the metatarsophalangeal joint showed no increase. Patients with a medium osteoarthritis grade and a medium level of clinical restraint showed the greatest improvement in relation to their preoperative condition. Conclusion The CI can be seen as an effective therapy for hallux rigidus. Nonetheless, realistic patient expectations must be communicated.

We use point of sales (POS) data from supermarkets, convenience stores, home centers, drug stores, and electronics retail stores for a nationwide analysis. Our analysis revealed the following. Second, they willingly stayed home during the semi-lockdown. Third, they continued to purchase infection prevention essentials during periods of both low and high levels of infection. Patterns of bi-directional relations across alcohol use, religiosity, and self-control in adolescent girls

Examining predictors of alcohol use among adolescent girls is increasingly important to enhance prevention efforts, given that the gender gap in alcohol use is steadily closing. While both religiosity and self-control have been independently associated with decreased alcohol use, little research has explored 1) whether religiosity and self-control are reciprocally related and 2) whether the reciprocal association between these constructs may indicate different patterns in the development of alcohol use. As such, this study examined whether there are multiple patterns of reciprocal relationships across religiosity, self-control, and alcohol use among adolescent girls. Latent variable mixture modeling was combined with an autoregressive cross-lagged panel model to identify discrete, prototypical patterns of longitudinal associations (i.e., subgroups) across religiosity, self-control, and alcohol use among 2,122 girls ages 13-17. Psychosocial covariates (e.g., conduct problems) were examined as predictors of each subgroup. Two subgroups were identified. Self-control was associated with reduced alcohol use in both the majority (87.56% of the sample) and minority (12.44% of the sample) subgroups, but only the majority subgroup also demonstrated associations between religiosity, self-control, and alcohol use. Religiosity may predict lower alcohol use in a majority of adolescent girls but this association may not be present among all girls, suggesting that there is a qualitative difference in how religiosity is associated with self-control and alcohol use between subgroups. Results also suggest that higher levels of conduct problems may predict which girls are more likely to demonstrate associations between only self-control and alcohol use, and demonstrate no significant associations with religiosity.
