Genetic polymorphisms in pri-let-7a-2 are associated with ischemic stroke risk in a Chinese Han population from Liaoning, China: a case-control study

Ischemic stroke is a complicated disease, and its pathogenesis has been attributed to the occurrence of genetic polymorphisms. Evidence has suggested that the microRNA let-7a is involved in the pathogenesis of ischemic stroke. Pri-miRNA is the primary transcript, which undergoes several processing steps to generate pre-miRNA and, later, mature miRNAs. In this case-control study, we analyzed the distribution of pri-let-7a-2 variants in patients at a high risk for ischemic stroke and the interactions of pri-let-7a-2 variants and environmental factors. Blood samples and clinical information were collected from 1086 patients with ischemic stroke and 836 healthy controls between December 2013 and December 2015 at the First Affiliated Hospital of China Medical University. We found that the rs1143770 CC genotype and the C allele were associated with a decreased risk of ischemic stroke, whereas the rs629367 CC genotype was associated with an increased risk for ischemic stroke. Moreover, these two single-nucleotide polymorphisms were in linkage disequilibrium in this study sample. We analyzed gene-environment interactions and found that rs1143770 exerted a combined effect on the pathogenesis of ischemic stroke, together with alcohol use, smoking, and a history of hypertension. Therefore, the detection of pri-let-7a-2 polymorphisms may increase the awareness of ischemic stroke risk. This study was approved by the Institutional Ethics Committee of the First Affiliated Hospital of China Medical University, China (approval No. 2012-38-1) on February 20, 2012, and was registered with the Chinese Clinical Trial Registry (registration number: ChiCTR-COC-17013559) on December 27, 2017.

Comparative associations of problematic alcohol and cannabis use with suicidal behavior in US military veterans: A population-based study

Alcohol use disorder (AUD) and cannabis use disorder (CUD) are each associated with increased suicidal behavior, but it is unclear how their comorbidity relates to suicide risk. Understanding these associations in U.S. military veterans is especially important, given their heightened risk for suicide, high prevalence of AUD, and increasing access to cannabis. We compared associations of probable AUD, CUD, and AUD/CUD with suicide ideation, plan(s), and attempt(s) in a nationally representative sample of 4,069 veterans surveyed in 2019-2020 as part of the National Health and Resilience in Veterans Study. Among veterans who screened positive for current AUD, 8.7% also screened positive for current CUD. Among veterans who screened positive for current CUD, 33.3% screened positive for current AUD. Current and lifetime positive screens for AUD, CUD, and AUD/ CUD were each strongly and independently associated with past-year suicide ideation and lifetime suicide ideation, plan(s), and attempt(s) [odds ratios (ORs) = 1.6-8.7]. Relative to veterans who screened positive for AUD only, veterans who screened positive for AUD/CUD and CUD only had higher odds of past-year suicide ideation (AUD/CUD: OR = 3.3; CUD only: OR = 2.4), lifetime suicide ideation (AUD/CUD: OR = 1.9; CUD only: OR = 2.6) and lifetime suicide plan(s) (AUD/CUD: OR = 1.7; CUD only: OR = 6.1). Collectively, findings suggest that screening positive for CUD might be an especially strong indicator of suicide ideation and planning in veterans with and without AUD, independent of sociodemographic, military, trauma, and other psychiatric factors. These findings underscore the importance of routine screening for CUD in suicide prevention efforts, especially as cannabis becomes more widely available.

The compaction of chromatin fibers has been a topic of controversy since the discovery of chromosomes in the 19th century. Although chromatin fibers were first identified using electron microscopy, the chromatin fibers on the surface of chromosome structures in plants remain unclear due to shrinking and breaking caused by prior chromosome isolation or preparation with alcohol and acid fixation, and critical point drying occurred into dehydration and denatured chromosomal proteins. This study aimed to develop a high-quality procedure for the isolation and preparation of plant chromosomes, maintaining the native chromosome structure, to elucidate the organization of chromatin fibers on the surface of plant chromosomes by electron microscopy. A simple technique to isolate intact barley (Hordeum vulgare) chromosomes with a high yield was developed, allowing chromosomes to be observed with a high-resolution scanning ion microscopy and helium ion microscopy (HIM) imaging technology, based on a scanning helium ion beam. HIM images from the surface chromatin fibers were analyzed to determine the size and alignment of the chromatin fibers. The unit size of the chromatin fibers was 11.6 +/- 3.5 nm and was closely aligned to the chromatin network model. Our findings indicate that compacting the surface structure of barley via a chromatin network and observation via HIM are powerful tools for investigating the structure of chromatin.

A new biomimetic set -up to understand the role of the kinematic, mechanical, and surface characteristics of the tongue in food oral tribological studies

A biomimetic tribological set-up was developed to better integrate physiological characteristics in oral friction studies. Polyvinyl alcohol was used to design tongue-mimicking samples (TMSs) with controlled bulk rigidity and surface roughness. The set-up allowed normal stresses and sequences of shearing motion to be applied between a TMS and a rectangular aluminum plate mimicking the hard palate. Cottage cheese with and without suspended microcrystalline cellulose particles of two sizes (small: 15 mu m and large: 250 mu m) was used as the model food in the experiments, making it possible to introduce heterogeneities well above the perception threshold, which often get excluded in conventional tribology studies. The feasibility of the set-up was tested by measuring how friction coefficient values were affected by food properties (viscosity, particle presence), TMS properties (surface roughness, bulk rigidity), and operational parameters (normal stress, shearing velocity). The measurements with the set-up were found to be repeatable, confirming its reliability. Particle presence led to increases in friction coefficient values, while food viscosity showed less of an influence, comparatively. Increases in the surface roughness and bulk rigidity of the TMSs led to a pronounced augmentation of the friction coefficient values. The friction coefficient values showed dependence on normal stress, increase in normal stress led to significant decrease in friction. Moreover, friction notably increased between the lowest and highest shearing velocities. In conclusion, this set-up proved to be efficient and reliable in measuring friction coefficient values and variation, paving the way for a better understanding of oral tribology and its dynamic nature.

The proposed cation distribution based on saturation magnetization data established the amount of cadmium occupying the tetrahedral sites of spinel lattice, which was well supported by the variation of DC resistivity with cadmium. The unparalleled matching between the experimentally observed lattice constant and that provided by the proposed cation distribution as well as Rietveld refinement confirms the amounts of cadmium entering tetrahedral and octahedral sites of the spinel lattice. All the samples of cadmium substituted cobalt ferrite assure their suitability for high frequency application with the pertinent properties.

TEM study of chronic alcoholism effects on early carcinogenesis by probing the nanoscale structural alterations of cell nuclei

Nanoscale structural alteration in the nuclei of cells with the progression of carcinogenesis is due to the rearrangements of the basic building blocks in the cell such as DNA, RNA, lipids, etc. Although epigenetic modifications underlie the development of cancer, exposure to carcinogenic chemicals such as alcohol also enhances the development of cancer. We report the effects of chronic alcoholism on early-carcinogenesis based on changes in the degree of nanoscale structural alterations (L-d) in nuclei. For this, transmission electron microscopy (TEM) imaging of the nuclei of colonic cells is performed for the following four mouse models: control mice; chronic alcoholic mice treated with ethanol (i.e., EtOH mice); mice treated with colonic carcinogen azoxymethane (AOM) and dextran sulfate sodium (DSS) that induced colitis (i.e., AOM + DSS mice); and chronic alcoholic or EtOH treated mice, together with AOM and DSS treatment (i.e., AOM + DSS + EtOH mice). The disordered optical lattices are constructed from their respective TEM images of thin colonic cell nuclei and the L-d values are calculated using the inverse participation ratio (IPR) technique from the spatially localized eigenfunctions of these lattices. Results show no significant difference in the average L-d value of the colon cell nuclei of alcohol treated mice relative to its control [i.e., L-d(C) similar to L-d(EtOH)]; however, an increase in the L-d value of alcohol treated precancerous cells [i.e., L-d(AOM + DSS + EtOH) > L-d(AOM + DSS)], indicating that alcohol accelerates the early carcinogenic process.

Acid hydrolysis and enzymatic hydrolysis are the main methods for releasing glycosidically bound volatiles (GBV). However, acid hydrolysis yields a strong pungent odor, and enzymatic hydrolysis is time consuming. In the present study, a new method, ultrasound hydrolysis, is reported to release GBV. This method is simple, environmentally friendly, fast and effective. Large differences were observed in the released aglycones and glycosyls between ultrasound and enzymatic hydrolysis of GBV. More types of aglycones were released under ultrasound than enzymatic hydrolysis. Alcohols and esters were the main aglycones under enzymatic hydrolysis, and terpenoids, esters and aldehydes were the main aglycones under ultrasound hydrolysis. The glycosyls released under ultrasound hydrolysis were mannose, glucose and sucrose, and those released under enzymatic hydrolysis were galactose and sucrose. The present study gives a new insight into a hydrolytic method for GBV by using ultrasound hydrolysis and can provide a reference method for fruit juice aromatization.

Synthesis and electrochemical properties of V(2)O(5)(center dot)nH(2)O compound with reduced graphene oxide/polyvinyl alcohol film as the free-standing cathode for coin-typed aqueous Zn-ion batteries

Recently aqueous zinc-ion batteries (AZIBs) have been widely studied on the aspects of electrode materials and electrolytes, but it is still far from practical application. The limitation of AZIBs is that it is difficult to have both high specific capacity and good cycle stability. Herein, a free-standing cathode V(2)O(5)(center dot)nH(2)O/reduced graphene oxide-polyvinyl alcohol (denoted as V(2)O(5)(center dot)nH(2)O/rGO-PVA) film was synthesized by one-step hydrothermal method and vacuum filtration. As the cathode for the coin-typed AZIBs, the V(2)O(5)(center dot)nH(2)O/rGO-PVA film cathode delivers the ultrahigh capacity of 553 mA h g(-1) at the current density of 0.1 A g(-1). And the capacity is 194 mA h g(-1) at 0.5 A g(-1), after 300 cycles, it still remains 152 mA h g(-1). At the power density of 183 W.kg(-1), the high energy density is calculated as 354 Wh.kg(-1) based on the mass of the cathode. The energy storage mechanism was studied by some ex-situ characterizations as well. This work not only proposes a strategy for the synthesis of V(2)O(5)(center dot)nH(2)O/rGO-PVA film, but also demonstrates that the as-obtained free-standing cathode is beneficial to the assembly of coin-typed AZIBs and exhibits excellent electrochemical performance.

Photochemistry of biochar during ageing process: Reactive oxygen species generation and benzoic acid degradation

In this study, the photogeneration of center dot OH and O-1(2) and the degradation mechanism of organic pollutants in biochar suspension under the simulated solar light irradiations were investigated. Biochar derived from rice husk with 550 degrees C of charring temperature (R550) was selected to degrade benzoic acid. It was found that 10 g/L of R550 could degrade 78.7% of benzoic acid within 360 min at pH 3, and the degradation efficiency was promoted to 95.2% as ultraviolet (UV) presented. By checking the production of p-hydroxybenzoic acid, UV accelerated the production of center dot OH, which was confirmed by the enhanced degradation efficiency of 59.2% caused by the evaluated center dot OH as UV appeared. The furfuryl alcohol loss in the R550 suspension under light irradiations testified to the production of O-1(2), which contributed to 9.3% of benzoic acid degradation. Oxidization treatment using gradient concentrations of H2O2 was employed to enhance the ageing process of biochar. As the ageing processed, the biochar possessed a declined performance towards center dot OH production from O-2 activation and the radical degradation of organic pollutants. As a contrast, the evaluated content of O-1(2) and enhanced non-radical degradation of organic pollutants was reached as UV presented. The further study indicated that phenolic hydroxyl groups on biochar facilitated the production of center dot OH via the electron transfer, and quinone like structures (C=O) on biochar boosted the generation of O-1(2) via the energy transfer.
