Comparative transcriptome analysis reveals the key regulatory genes for higher alcohol formation by yeast at different a-amino nitrogen concentrations

Higher alcohols are important flavor substance in alcoholic beverages. The content of alpha-amino nitrogen (alpha-AN) in the fermentation system affects the formation of higher alcohols by Saccharomyces cerevisiae. In this study, the effect of alpha-AN concentration on the higher alcohol productivity of yeast was explored, and the mechanism of this effect was investigated through metabolite and transcription sequence analyses. We screened 12 most likely genes and constructed the recombinant strain to evaluate the effect of each gene on high alcohol formation. Results showed that the AGP1, GDH1, and THR6 genes were important regulators of higher alcohol metabolism in S. cerevisiae. This study provided knowledge about the metabolic pathways of higher alcohols and gave an important reference for the breeding of S. cerevisiae with low-yield higher alcohols to deal with the fermentation system with different alpha-AN concentrations in the brewing industry.

A bionic PEEK composite structure with negatively charged surface adsorbing molecular brushes possessing improved biotribological properties for artificial joints

To improve the contact mode of artificial joints and their tribological performance, inspired by the ‘soft-on-hard’ structure of ‘cartilage-subchondral bone’ as well as negatively charged characteristic of the cartilage surface with brush-like molecules, a polyetheretherketone (PEEK)-hydrogel composite structure with negatively charged surface adsorbing cationic molecules through electrostatic interaction was designed and fabricated. Characterization results demonstrated that polyacrylic acid (PAA) hydrogel strengthened by polyvinyl alcohol (PVA) microcrystals was successfully fabricated on the PEEK substrate through ultra-violet (UV) radiation. Adsorption of cationic monomers was also confirmed. The PEEK-hydrogel composite structure had significantly reduced friction coefficient, similar to 80% lower than that of PEEK, owing to the synergistic effect of biphasic and boundary lubrication. This work proposes a novel route to develop the friction surface of artificial joints with advanced performance and contributes to the understanding of lubrication mechanism of artificial joints.

The association between reinforcement sensitivity and substance use is mediated by individual differences in dispositional affectivity in adolescents

Background: Adolescence marks the onset of substance use experimentation and adolescents are particularly vulnerable to certain negative effects of substances. Some evidence indicates reinforcement sensitivity is associated with substance use, though little is known about mechanisms underlying such association. Aims: in the current study were to examine, (1) associations between behavioral activation (BAS) and behavioral inhibition (BIS) system sensitivity, positive (PA) and negative affectivity (NA), and alcohol use and alcohol problems as well as tobacco, and marijuana use, and whether (2) associations are mediated by PA or NA. Methods: Participants were a community sample of N = 125 adolescents (M-age = 15.67 years; SD = 0.93; 52% boys) who completed self-report measures. Results: evinced associations, generally as expected, across variables (all ps < 0.05). In mediation analyses, an association emerged between BIS sensitivity and alcohol use, mediated by NA (95%CIs [0.034; 0.390]); greater BIS sensitivity was associated with greater NA and greater NA was associated with greater alcohol use. These findings were replicated with alcohol problems. An association also emerged between BAS sensitivity and marijuana use, mediated by PA (95%CIs [-0.296; -0.027]); greater BAS sensitivity was associated with greater PA and greater PA was associated with lower marijuana use. Finally, BIS sensitivity was associated with tobacco use through NA (95%CIs [0.023; 0.325]) and PA (95%CIs [0.004; 0.116]), with NA linked to greater, but PA linked to lower tobacco use. BAS sensitivity was also associated with tobacco use through PA (95%CIs [-0.395; -0.049]), with PA linked again to lower tobacco use. Conclusions: There are unique and shared effects of domains of reinforcement sensitivity on adolescent substance use and these vary with index of dispositional affectivity and type of substance considered. What happens when pesticides are solubilised in binary ionic/zwitterionic-nonionic mixed micelles?

Hypothesis: Surfactants have been widely used as adjuvants in agri-sprays to enhance the solubility of pesticides in foliar spray deposits and their mobility through leaf cuticles. Previously, we have characterised pesticide solubilisation in nonionic surfactant micelles, but what happens when pesticides become solubilised in anionic, cationic and zwitterionic and their mixtures with nonionic surfactants remain poorly characterised. Experiments: To facilitate characterisations by SANS and NMR, we used nonionic surfactant hexaethylene glycol monododecyl ether (C12E6), anionic sodium dodecylsulphate (SDS), cationic dodecyltrimethylammonium bromide (DTAB) and zwitterionic dodecylphosphocholine (C12PC) as model adjuvant systems to solubilise 3 pesticides, Cyprodinil (CP), Azoxystrobin (AZ) and Difenoconazole (DF), representing different structural features. The investigation focused on the influence of solubilisates in driving changes to the micellar nanostructures in the absence or presence of electrolytes. NMR and NOESY were applied to investigate the solubility and location of each pesticide in the micelles. SANS was used to reveal subtle changes to the micellar structures due to pesticide solubilisation with and without electrolytes. Findings: Unlike nonionic surfactants, the ionic and zwitterionic surfactant micellar structures remain unchanged upon pesticide solubilisation. Electrolytes slightly elongate the ionic surfactant micelles but have no effect on nonionic and zwitterionic surfactants. Pesticide solubilisation could alter the structures of the binary mixtures of ionic/zwitterionic and ionic/nonionic micelles by causing elongation, shell shrinkage and dehydration, with the exact alteration being determined by the molar ratio in the mixture. A novel B2O3/B4C-modified composite adhesive with wide operative temperature range for alumina fiber fabric bonding

A heat resistance adhesive with wide operative temperature range for bonding alumina fiber fabric was developed by organic-inorganic modification. The polyvinyl alcohol modified by B2O3 generated a complex cross-linked network connected by B-O-C bonds, which can enhance the bonding strength of adhesive after heat-treatment from RT to 400 degrees C. Moreover, the addition of B4C can enhance the bonding strength of adhesive after heat-treatment from 400 degrees C to 800 degrees C due to the formation of molten B2O3 and borosilicate glass. Significantly, the appropriate addition of B4C can make the adhesive form a denser structure without transforming the fracture mode of the bonding joints, which is conducive to enhance the strength performance of bonding joints after sintered at 800 degrees C. On the contrary, the excessive addition of B4C will transform the fracture mode of the bonding joints into brittle fracture, which will degrade the strength performance of bonding joints.

One-step complexed preparation of nitrogen and Cu co-doped oxidative active carbon catalysts Cu-N/OAC for furfural selective hydrogenation with high yield

A facile procedure for preparing copper and nitrogen co-doped active carbon (Cu-N/OAC) by one-step complexed was reported and applied in liquid-phase hydrogenation of furfural (FAL). The facile procedure resulted in high Cu nanoparticles dispersion on OAC with Cu-0 and Cu+ sites and apparently promoted the catalytic activities during furfural hydrogenation reaction. The obtained Cu-N/OAC-800 shows 99.5% FAL conversion with 98.4% selectivity to furfuryl alcohols (FOL) under reaction condition of 150 degrees C, 2 MPa and 6 h. These results indicated that the excellent catalytic performance of the catalyst was due to the synergic effects of nitrogen doping and Cu metal active sites.

Using surfactants for controlling rotifer contamination in mass cultivation of Chlorella pyrenoidosa

Frequent microzooplankton contamination such as rotifers in Chlorella mass culture often causes devastating effects on biomass production. In this study, five surfactants were used to control contamination by a rotifer, Brachionus calyciflorus, in Chlorella pyrenoidosa XQ-20044 cultures. Laboratory results showed that the minimal effective concentrations of surfactants for complete control of B. calyciflorus were 7.5, 10, 10, 10, and 15 mg L-1 for primary alcohol ethoxylate, sodium dodecylbenzene sulfonate, sodium alcohol ether sulfate, coconut diethanolamide, and sodium dodecyl sulfate, respectively. However, only 10 mg L-1 sodium dodecylbenzene sulfonate and 7.5-10 mg L-1 primary alcohol ethoxylate had no negative effects on algal growth. Moreover, in a 5 m(2) open raceway pond, 10 mg L-1 sodium dodecylbenzene sulfonate ensured swift and total elimination of rotifers, while the final dry weight of the algae reached >0.74 g L-1 whereas the cost of using sodium dodecylbenzene sulfonate was only $ 0.014. The results of this study indicated that sodium dodecylbenzene sulfonate approach is fast-acting, highly effective, and a low-cost treatment. Therefore, sodium dodecylbenzene sulfonate has great potential for biological contamination control in microalgae mass culture without a negative impact on algal growth.

Rational design of a porous nanofibrous actuator with highly sensitive, ultrafast, and large deformation driven by humidity

Humidity-responsive bilayered actuators which can dynamically change their shapes under humidity gradients, have recently motivated significant interest in a wide variety of emerging fields, including artificial muscles, intelligent sensors, and smart devices. However, this kind of actuators frequently suffer from slow responsiveness, prolonged actuating period, and small deformation, which is due to the dense structure hindering the diffusion of water molecules. Herein, we employ a simple programmable electrospinning approach to fabricate a highly porous polyvinyl alcohol/polyvinyl butyral (PVA/PVB) bilayered actuator for achieving excellent humidity-triggered deformation. Benefiting from the differential affinity of the two layers to water and the elaborately structural design, the porous bilayered actuator exhibits superior actuating performance in response to humidity gradient. Briefly, only 283 ms is needed for driving the shape change, and the whole deformation merely requires 10 s; besides, large bending amplitude with the curvature of 11.2 cm-1 is achieved, and the shape deformation process can be repeated at least 30 times without fatigue. Finally, we demonstrate the porous nanofibrous actuator can be used as artificial plants to perform opening and closing motions and a self-driven detector for monitoring humidity in real-time.

Tailoring Multistimuli Responsive Micropatterns Activated by Various Mechanical Modes

A variety of applications are demonstrated based on this system, including 1) a moisture erasable highly sensitive pressure responsive device and pattern replicator with a high fidelity; 2) a moisture erasable scratch/magneto-mechanical re-writable tablet; 3) an electro-mechanical controllable smart window with an ultra-sensitive strain responsive transmittance modulation and a low operating voltage; 4) various types of strain responsive, moisture erasable, and laser writable information recording/encryption devices. This work provides new routes for designing innovative wrinkled systems triggered by diverse mechanical fashions and can decode multiple environmental stimuli into optical signals for widespread application.

Gelatin/Poly (vinyl alcohol)/Inorganic filler composites for phantom breasts

The effect of the various inorganic filler (Zn, ZnO, CuO, Co(NO3)(2), Fe2O3) on the structural, electronic, atomic, and absorption properties to the performance of composite phantom breast with polyvinyl alcohol (PVA) as a binder has been studied. The bonding characteristic analyzed from the Fourier transforms infrared (FTIR) spectra and the structural properties in the form of the crystallite size, porosity, and dislocation density determined from the X-ray diffraction (XRD) spectra. The atomic, electronic, and absorption ability of composite were analyzed from the irradiation intensity after the X-rays for the energies from 60 keV to 85 keV at 10 mAs passing through the samples. The functional group of C=O, C-H, and C-O show covalent bonding as a cross-linking between atoms from the various filler and the atom from the matrix materials. The crystallite size shows a direct correlation between the linear attenuation coefficient and the mass attenuation coefficient. The linear attenuation coefficient of composite gelatin/PVA/ZnO is 0.255 cm(-1), for composite gelatin/PVA/CuO is 0.233 cm(-1) shows good for breast phantom, and for composite gelatin/PVA/Zn is 0.343 cm(-1 )shows excellent characteristics for breast phantom. This study provided a new composite with excellent characteristics as a guide to produce breast phantom.
