Substance Use Disorder (SUD) is a chronic and relapsing disease characterized by craving, loss of control, tolerance and physical dependence. At present, the combination of pharmacotherapy and psychosocial intervention is the most effective management strategy in preventing relapse to reduce dropout rates and promote abstinence in SUD patients. However, only few effective medications are available. Transcranial Magnetic Stimulation (TMS) is a non-invasive brain stimulation technique that modulates the cellular activity of the cerebral cortex through a magnetic pulse applied on selected brain areas. Recently, the efficacy of TMS has been investigated in various categories of SUD patients. The present review analyzes the application of repetitive TMS in patients with alcohol, tobacco, and cocaine use disorder. Although the number of clinical studies is still limited, repetitive TMS yields encouraging results in these patients, suggesting a possible role of TMS in the treatment of SUD.

Solubility determination, model evaluation, Hansen solubility parameter and thermodynamic properties of benflumetol in pure alcohol and ester solvents

The equilibrium solubility and thermodynamic properties of benflumetol in methanol, ethanol, npropanol, n-butanol, n-pentanol, n-hexanol, n-heptanol, n-octanol, methyl acetate, ethyl acetate, n-propyl acetate, n-butyl acetate and n-pentyl acetate were reported. Solubility determinations were performed through the laser monitoring method at T = (278.15-323.15) K and p = 101.3 kPa. It is found that the mole fraction solubility of benflumetol increases apparently with the augmented experimental temperature. In addition, the solubility of benflumetol in all ester solvents is greater than that in alcohol solvents. The maximum mole fraction solubility of benflumetol is obtained in n-pentyl acetate (4.061 x 10(-2), T = 323.15 K), and the minimum mole fraction solubility of benflumetol is obtained in methanol (1.341 x 10(-5), T = 278.15 K). The Hansen solubility parameter for benflumetol as well as selected solvents was summarized to analyse the probabilities of miscibility between solute and solvents. The analysis consequence shows that the miscibility of benflumetol with selected solvents is caused by a combination of factors. All recorded solubility of benflumetol were regressed by 9.11 model, modified Apleblat model, two-Suffix Margules model, NRTL model and UNIQUAC model. By comparison, it can be found that the average ARD and 10 4 RMSD of the modified Apelblat model and NRTL model are smaller than the other three models, indicating that these two moedls are more suitable for correlating the solubility data. In addition, the apparent thermodynamic properties of benflumetol in all neat solvents were calculated and investigated by the Van’t Hoff equation. The results illustrated that the dissolution process of benflumetol is an entropy-driven endothermic process. (C) 2020 Elsevier Ltd.

Substance use and abuse, COVID-19-related distress, and disregard for social distancing: A network analysis

Research shows that there has been a substantial increase in substance use and abuse during the COVID-19 pandemic, and that substance use/abuse is a commonly reported way of coping with anxiety concerning COVID-19. Anxiety about COVID-19 is more than simply worry about infection. Research provides evidence of a COVID Stress Syndrome characterized by (1) worry about the dangers of COVID-19 and worry about coming into contact with coronavirus contaminated objects or surfaces, (2) worry about the personal socioeconomic impact of COVID-19, (3) xenophobic worries that foreigners are spreading COVID-19, (4) COVID-19-related traumatic stress symptoms (e.g., nightmares), and (5) COVID-19-related compulsive checking and reassurance-seeking. These form a network of interrelated nodes. Research also provides evidence of another constellation or syndrome, characterized by (1) belief that one has robust physical health against COVID-19, (2) belief that the threat of COVID-19 has been exaggerated, and (3) disregard for social distancing. These also form a network of nodes known as a COVID-19 Disregard Syndrome. The present study, based on a population-representative sample of 3075 American and Canadian adults, sought to investigate how these syndromes are related to substance use and abuse. We found substantial COVID-19-related increases in alcohol and drug use. Network analyses indicated that although the two syndromes are negatively correlated with one another, they both have positive links to alcohol and drug abuse. More specifically, COVID-19-related traumatic stress symptoms and the tendency to disregard social distancing were both linked to substance abuse. Clinical and public health implications are discussed.

Preparation of 3D Printed Chitosan/Polyvinyl Alcohol Double Network Hydrogel Scaffolds

In this work, a 3D printed double-network (DN) hydrogel scaffold is designed with chitosan (CS) and polyvinyl alcohol (PVA) as the framework matrix. The addition of PVA into the CS-based hydrogel clearly enhances the mechanical properties and lowers the swelling behaviors of the hydrogels. The crosslinking of CS with genipin can perform the pre-crosslinking to improve the viscosity and 3D printability of the hydrogel precursor, while increasing the PVA content results in lowering the viscosity and 3D printability of the pre-crosslinked hydrogel. The antibacterial property results of the DN hydrogel display that the hydrogel have favorable long-lasting antibacterial ability. The appropriate pre-crosslinked hydrogel with the CS/PVA mass ratio of 3:10 and pre-crosslinking time of 7 h is used for 3D printing to prepare the 3D printed porous DN hydrogels. Moreover, the anti-tumor drug doxorubicin (DOX) is loaded into the 3D printed porous DN hydrogels and the in vitro release study displays the sustainable drug release behavior. And the DOX release from hydrogel scaffold can be adjusted by the pH value of release environment. All of the results indicate that the porous DN CS/PVA hydrogel scaffolds have great application potential for tissue regeneration.

In vivo safety assessment, biodistribution and toxicology of polyvinyl alcohol microneedles with 160-day uninterruptedly applications in mice

Dissolving microneedles (DMNs) are widely used in drug delivery systems since they are based on one-step application, which is simple and convenient for patients, especially for the patients such as diabetes who need daily or long-term self-administration. In general, the matrix materials of DMNs are water-soluble materials that can release the encapsulated drugs gradually by dissolving in the skin without generating sharp needle waste. However, the matrix materials of DMNs will also leave in the skin after application. Thus, it is vital to evaluate whether the matrix material of DMNs dissolved in the skin will cause health risks such as toxicity to the body or some skin-related complications to patients who frequent or long-term administration. In this work, PVA, as one of the typical matrix materials of DMNs, was selected to prepare the DMNs to research the safety of PVA-based MNs to the body after being dissolved in the skin. Briefly, in a 160 – days trial, the healthy mice were daily administrated by PVA MNs. The results showed that PVA materials mainly accumulated in the skin tissues of mice after dissolving and the concentration of PVA in the insertion sites gradually decreased and was almost undetectable at 6 days after administration. The observation of general conditions, blood hematological analysis and histological examinations of the mice demonstrated that the PVA-based MNs do not cause appreciable toxicity to the healthy mice after daily insertion in a 160 – days trial. Altogether, these results encourage further studies of PVA MNs for biomedical applications and support translation of PVA-based DMNs from pre-clinical development into clinical trials.

Childhood psychopathology mediates associations between childhood adversities and multiple health risk behaviours in adolescence: analysis using the ALSPAC birth cohort

Background Childhood adversity strongly predicts adolescent multiple health risk behaviours (MRBs) such as alcohol/tobacco use, self-harm and physical inactivity, and both adversities and MRBs are associated with premature mortality and several chronic health conditions that are among the leading causes of death in adults. It is therefore important to understand the relationship between adversities and MRBs and what could mediate any association. The aim of this study was to explore whether childhood psychopathology mediates associations between adversities and MRBs. Methods Participants were young people in the Avon Longitudinal Study of Parents and Children (ALSPAC) (N = 5,799). Using structural equation modelling, we explored the associations between adversities before 9 years and MRBs at age 16 years. We also explored potential mediating pathways through dimensional psychopathology measured by the Strength and Difficulties Questionnaire subscales at age 12 years. Results There were strong positive associations between adversities and MRBs (beta .25, 95% CI 0.20, 0.31, p < .001) suggesting that each additional adversity is associated with a 0.25 increase in number of MRBs out of 13 total risk behaviours. We found robust evidence of mediating pathways from adversities through conduct problems (beta .05, 95% CI 0.03, 0.06, p < .001), hyperactivity/inattention (beta .02, 95% CI 0.01, 0.03, p < .001) and peer relationship problems (beta -.02, 95% CI -0.03, -0.02, p < .001) to MRBs. Conclusions Increased conduct problems and hyperactivity/inattention appear to partially explain the relationship between adversities and MRBs. Peer relationship problems also appear to reduce the association between adversities and MRBs, and further research is needed to understand how to encourage peer connectivity without increasing risk of MRBs. These results suggest that interventions aimed at reducing MRBs among those exposed to childhood adversities could focus on prevention of behavioural problems. Diabetes and liver cancer risk: A stronger effect in Whites than Blacks?

Background: Both diabetes and liver cancer are overrepresented among African Americans, but limited information is available on the interrelationship of these two diseases among African Americans. We examined the association of diabetes with the incidence of liver cancer and whether this varied by participant self-reported race/ ethnicity. Methods: Using the Southern Community Cohort Study, we conducted a cancer follow up (2002-2016) of a cohort of mostly low-income participants aged 40-79 with diabetes (n = 15,879) and without diabetes (n = 59,077) at study baseline. Cox regression was used to compute Hazard Ratios (HR) and 95% CIs for the risk of incident liver cancer. Results: With 790,132 person years of follow up, 320 incident cases of liver cancer were identified. In analyses controlling for age, sex, race, BMI, current and former smoking, total alcohol consumption, family history of liver cancer, any hepatitis infection, hyperlipidemia and socioeconomic factors, the association between diabetes and risk of liver cancer differed significantly (P-interaction = 0.0001) between participants identifying as Black/African American (AA) or White/European American (EA). Diabetes was associated with 5.3-fold increased cancer risk among EAs (HR 5.4, 95% CI 32-93) vs an 80% increase (HR 1.8, 95% CI 1.3-25) among AAs. Furthermore, controlling for diabetes greatly attenuated the higher risk of liver cancer among AAs; indeed, while the cancer risk among those without diabetes was twice as high among AAs than EAs (HR 2.0, 95% CI 1.4-2.9), no excess in Ms was observed among those with diabetes (HR = 0.7, 95% CI = 0.4-1.1). Conclusion: While liver cancer risk in general is greater in Ms than EAs and diabetes increases this risk in both racial/ethnic groups, diabetes appears to impact liver cancer to a much greater extent among EAs. The findings raise the possibility of racially different mechanisms and impacts of diabetes on this often fatal cancer among Ms and EAs. (C) 2020 Elsevier Inc. All rights reserved.

Rational design of a porous nanofibrous actuator with highly sensitive, ultrafast, and large deformation driven by humidity

Humidity-responsive bilayered actuators which can dynamically change their shapes under humidity gradients, have recently motivated significant interest in a wide variety of emerging fields, including artificial muscles, intelligent sensors, and smart devices. However, this kind of actuators frequently suffer from slow responsiveness, prolonged actuating period, and small deformation, which is due to the dense structure hindering the diffusion of water molecules. Herein, we employ a simple programmable electrospinning approach to fabricate a highly porous polyvinyl alcohol/polyvinyl butyral (PVA/PVB) bilayered actuator for achieving excellent humidity-triggered deformation. Benefiting from the differential affinity of the two layers to water and the elaborately structural design, the porous bilayered actuator exhibits superior actuating performance in response to humidity gradient. Briefly, only 283 ms is needed for driving the shape change, and the whole deformation merely requires 10 s; besides, large bending amplitude with the curvature of 11.2 cm-1 is achieved, and the shape deformation process can be repeated at least 30 times without fatigue. Finally, we demonstrate the porous nanofibrous actuator can be used as artificial plants to perform opening and closing motions and a self-driven detector for monitoring humidity in real-time.

Synthesis and Application of Platinum-Based Hollow Nanoframes for Direct Alcohol Fuel Cells

Although platinum (Pt)-based catalysts are suffering from high costs and limited reserves, they are still irreplaceable in a short period of time in terms of catalytic performance. Structural optimization, composition regulation and carrier modification are the common strategies to improve the activity and stability of Pt-based catalyst. Strikingly, the morphological evolution of Pt-based electrocatalyst into nanoframes (NFs) have attracted wide attention to reduce the Pt consumption and improve the electrocatalytic activity simultaneously. Contrary to Pt-based solid nanocrystalline materials, Pt-based NFs have many advantages in higher atomic utilization, open space structure and larger specific surface area, which facilitate electron transfer, mass transport and weaken surface adsorption by more unsaturated coordination sites. Here we introduce the detailed preparation strategies of Pt-based NFs with different etching methods (oxidative etching, chemical etching, galvanic replacement and carbon monoxide etching), crystal structure evolution and formation mechanism, efficient applications for oxygen reduction reaction (ORR), methanol oxidation reaction (MOR) and ethanol oxidation reaction (EOR) in direct alcohol fuel cells (DAFCs). Based on the high-efficiency atom utilization, open space structure and diverse alloy composition, Pt-based NFs exhibit superior activity, stability and anti-poisoning than commercial counterparts in the application of DAFCs. The current challenges and future development of Pt-based NFs are prospected on the type of NFs materials, synthesis and etching methods, crystal control and catalytic performance. We propose a series of improvement mechanisms of Pt-based NFs, such as small size effect, high-energy facets, Pt-skin construction and Pt-C integration, thereby weakening the molecule absorption, increasing the Pt utilization, strengthening the intrinsic stability, and alleviating the metal dissolution and support corrosion. Additionally, the scale-up synthesis of catalytic materials, membrane electrodes assembly, and development of the start-stop system and the circulation system design are essential for the commercial application of Pt-based NFs and industrial manufacturing of DAFCs. More importantly, the reaction mechanism, active site distribution and dynamic changes in the catalytic material during the catalytic reaction are crucial to further explain the maintenance and evolution of catalytic performance, which will open a window to elucidate the improvement mechanism of the catalyst in the fuel cell reactions. This review work would promote continuous upgradations and understandings on Pt-based NFs in the future development of DAFCs.
