Live predator stress in adolescence results in distinct adult behavioral consequences and dorsal diencephalic brain activation patterns

Exposure to traumatic events during childhood increases the risk of adult psychopathology, including anxiety, depression, alcohol use disorders and their co-morbidity. Early life trauma also results in increased symptom complexity, treatment resistance and poor treatment outcomes. The purpose of this study was to establish a novel rodent model of adolescent stress, based on an ethologically relevant life-threatening event, live predator exposure. Rats were exposed to a live predator for 10 min. at three different time points (postnatal day (PND)31, 46 and 61). Adult depression-, anxiety-like behaviors and ethanol consumption were characterized well past the last acute stress event (two weeks). Behavioral profiles across assessments were developed to characterize individual response to adolescent stress. CNS activation patterns in separate groups of subjects were characterized after the early (PND31) and last predator exposure (PND61). Subjects exposed to live-predator adolescent stress generally exhibited less exploratory behavior, less propensity to venture into open spaces, a decreased preference for sweet solutions and decreased ethanol consumption in a two-bottle preference test. Additional studies demonstrated blunted cortisol response and CNS activation patterns suggestive of habenula, rostromedial tegmental (RMTg), dorsal raphe and central amygdala involvement in mediating the adult consequences of adolescent stress. Thus, adolescent stress in the form of live-predator exposure results in significant adult behavioral and neurobiological disturbances. Childhood trauma, its impact on neurodevelopment and the subsequent development of mood disorders is a pervasive theme in mental illness. Release of catechin from Azivash gum-polyvinyl alcohol electrospun nanofibers in simulated food and digestion media

In this study, the electrospinning process was applied for the encapsulation of catechin in nanofibers from Azivash (Corchorus olitorius. L) gum-polyvinyl alcohol. By increasing catechin concentration (from 500 to 1000 mg L-1), encapsulation efficiency was lowered but loading capacity increased. A positive correlation was observed between catechin addition and diameter of nanofibers. FTIR spectroscopy highlighted the interactions between catechin and polymer solution which increased the thermal stability of nanofibers. Results of release trends demonstrated that approximately 50% and <70% of the total catechin was released in simulated gastric fluid and simulated intestinal fluid, respectively. Also, a different catechin release rate from nanofibers was observed in simulated low-fat and high-fat food media, respectively. After 12 days, about 90% of the total catechin was released. It was confirmed that designed nanofibers have the capacity to be applied in active packaging for improving the oxidative stability of food and pharmaceutical products. Childhood psychopathology mediates associations between childhood adversities and multiple health risk behaviours in adolescence: analysis using the ALSPAC birth cohort

Background Childhood adversity strongly predicts adolescent multiple health risk behaviours (MRBs) such as alcohol/tobacco use, self-harm and physical inactivity, and both adversities and MRBs are associated with premature mortality and several chronic health conditions that are among the leading causes of death in adults. It is therefore important to understand the relationship between adversities and MRBs and what could mediate any association. The aim of this study was to explore whether childhood psychopathology mediates associations between adversities and MRBs. Methods Participants were young people in the Avon Longitudinal Study of Parents and Children (ALSPAC) (N = 5,799). Using structural equation modelling, we explored the associations between adversities before 9 years and MRBs at age 16 years. We also explored potential mediating pathways through dimensional psychopathology measured by the Strength and Difficulties Questionnaire subscales at age 12 years. Results There were strong positive associations between adversities and MRBs (beta .25, 95% CI 0.20, 0.31, p < .001) suggesting that each additional adversity is associated with a 0.25 increase in number of MRBs out of 13 total risk behaviours. We found robust evidence of mediating pathways from adversities through conduct problems (beta .05, 95% CI 0.03, 0.06, p < .001), hyperactivity/inattention (beta .02, 95% CI 0.01, 0.03, p < .001) and peer relationship problems (beta -.02, 95% CI -0.03, -0.02, p < .001) to MRBs. Conclusions Increased conduct problems and hyperactivity/inattention appear to partially explain the relationship between adversities and MRBs. Peer relationship problems also appear to reduce the association between adversities and MRBs, and further research is needed to understand how to encourage peer connectivity without increasing risk of MRBs. These results suggest that interventions aimed at reducing MRBs among those exposed to childhood adversities could focus on prevention of behavioural problems. An efficacy trial of adaptive interventions for alcohol use disorder

Background: Adaptive interventions, sometimes referred to as stepped care, are those interventions in which the type or dosage of treatment offered to patients is tailored to baseline clinical presentation and then adjusted over time in response to patient progress or lack thereof. Currently, no adaptive brief interventions exist specifically for alcohol use disorder (AUD). Method: This study used a sequential multiple assignment randomized trial design with 160 individuals with AUD recruited both locally and nationally who had a goal to reduce or abstain from drinking. Participants received brief advice (BA) and then the study reassessed them three weeks later; the study randomized those who did not respond to BA, defined as reducing their drinking to low-risk guidelines, to two session of motivational interviewing (MI) or more BA. The study then reassessed participants at week 8. The study re-randomized non-responders to receive either MI alone or MI plus behavioral self-control therapy (BSCT), also referred to as coping skills therapy, and evaluated participants at week 13. Results: Overall, participants receiving any BSCT made the greatest reductions in drinking. Participants who received MI at week 4 and BSCT at week 8 outperformed all other groups. Conclusion: Findings reveal that prolonged treatment, more sessions, and/or a specific combination of MI and BSCT provided optimal outcomes. Future research should determine whether such an algorithm holds across heterogenous groups of individuals with AUD.

Osmotic dehydration and convective drying of kiwifruit (Actinidia deliciosa) – The influence of ultrasound on process kinetics and product quality

The aim of this study was to assess the impact of ultrasound on two subsequent processes – initial osmotic dehydration and convective drying of kiwifruit (Actinidia deliciosa). The effect of ultrasound (at a frequency of 25 kHz) was assessed both in terms of process kinetics and product quality. During the study, three different osmotic agents were used – erythritol, sorbitol, and sucrose – in an aqueous solution at a concentration of 50% (w/w). The essential kinetic parameters were analyzed – water loss and increase of dry matter during osmotic dehydration, and evolution of moisture content and temperature of samples during convective drying (drying curves). Product quality was assessed on the basis of color, water activity, and content of relevant bioactive components – polyphenols and carotenoids. It was found that the application of ultrasound during osmotic dehydration resulted in higher water loss and solid gain. This surely results from the phenomena occurring during the propagation of the elastic waves in the liquid medium (mainly related to cavitation) and enhancement of the mass transfer. The use of ultrasound during convective drying also had a positive effect on the kinetics of this process. In most cases, during the ultrasonically assisted drying operations, a significant increase in drying rate was observed, leading to a reduction in drying time. This may be due to the intensification of both heat and mass transfer as a result of the mechanical and thermal effects of ultrasound. The assessment of product quality showed that the use of sugar alcohols was a good alternative to sucrose, and ultrasound-assisted convective drying increased the retention of valuable carotenoids and polyphenols. Moreover, in all dried kiwifruit slices, water activity was below 0.6, which can guarantee the microbiological stability of the tested samples.

Effect of ethanol on the microstructure and rheological properties of whey proteins: Acid-induced cold gelation

The microstructure and mechanical properties of ethanol-denatured, acid-induced whey protein (WPI) cold-set gels have been monitored by confocal laser scanning microscopy and rheology. As the ethanol concentration increased, the porosity of the gels progressively decreased, with gels prepared at ethanol concentrations of 50 and 70 g/100 g lacking microscopically visible pores. Large aggregates were observed at ethanol concentrations of 70 g/100 g immediately after alcohol addition. A sharp increase in storage modulus was noted at ethanol concentrations between 30 and 50 g/100 g, while the gel strength at 50 g/100 g, was up to three times higher compared to that of heat-denatured gels. In general, the ethanol-denatured whey protein cold-set gels were prone to phase separation. Ethanol-denatured cold-set whey protein gels exhibited mechanical and microstructural properties that may offer alternative means of structuring protein-enriched food formulations and designing novel food products.

A Qualitative Investigation of the Experience of Mindfulness Training Among Police Officers

The primary objective of this study was to qualitatively assess the experience of mindfulness-based resilience training (MBRT) among police officers. MBRT is an 8-week intervention designed to enhance police officer resilience in the context of acute and chronic stressors inherent to policing. MBRT has demonstrated preliminary efficacy in decreasing aggression, burnout, alcohol use, sleep disturbance, and improving cortisol reactivity, psychological flexibility, and non-reactivity. Participants (n = 5) were police officers who completed an individual semi-structured interview post-MBRT. A coding schema was developed to identify and categorize participant responses, and then applied the final coding framework to all participant interviews. Results revealed perceived improvements in intrapersonal and interpersonal functioning, benefits of MBRT, and strategies for overcoming potential barriers to mindfulness practice. These preliminary qualitative results are consistent with quantitative psychological and physiological MBRT outcomes and provide further support for MBRT feasibility and acceptability.

Diabetes and liver cancer risk: A stronger effect in Whites than Blacks?

Background: Both diabetes and liver cancer are overrepresented among African Americans, but limited information is available on the interrelationship of these two diseases among African Americans. We examined the association of diabetes with the incidence of liver cancer and whether this varied by participant self-reported race/ ethnicity. Methods: Using the Southern Community Cohort Study, we conducted a cancer follow up (2002-2016) of a cohort of mostly low-income participants aged 40-79 with diabetes (n = 15,879) and without diabetes (n = 59,077) at study baseline. Cox regression was used to compute Hazard Ratios (HR) and 95% CIs for the risk of incident liver cancer. Results: With 790,132 person years of follow up, 320 incident cases of liver cancer were identified. In analyses controlling for age, sex, race, BMI, current and former smoking, total alcohol consumption, family history of liver cancer, any hepatitis infection, hyperlipidemia and socioeconomic factors, the association between diabetes and risk of liver cancer differed significantly (P-interaction = 0.0001) between participants identifying as Black/African American (AA) or White/European American (EA). Diabetes was associated with 5.3-fold increased cancer risk among EAs (HR 5.4, 95% CI 32-93) vs an 80% increase (HR 1.8, 95% CI 1.3-25) among AAs. Furthermore, controlling for diabetes greatly attenuated the higher risk of liver cancer among AAs; indeed, while the cancer risk among those without diabetes was twice as high among AAs than EAs (HR 2.0, 95% CI 1.4-2.9), no excess in Ms was observed among those with diabetes (HR = 0.7, 95% CI = 0.4-1.1). Conclusion: While liver cancer risk in general is greater in Ms than EAs and diabetes increases this risk in both racial/ethnic groups, diabetes appears to impact liver cancer to a much greater extent among EAs. The findings raise the possibility of racially different mechanisms and impacts of diabetes on this often fatal cancer among Ms and EAs. (C) 2020 Elsevier Inc. All rights reserved.
