To explore bioactive polysaccharides from medicinal fungi, a homogeneous heteropolysaccharide SB1-1 was extracted from the stroma of Shiraia bambusicola by enzyme assisted extraction, ethanol precipitation, anion-exchange and size-exclusion chromatogs. Chem. and spectroscopic analyses showed that SB1-1 was a neutral polysaccharide composed of mannose, glucose and galactose at a molar ratio of 1.1:1.9:1.0, and had a mol. weight of 18.3 kDa. SB1-1 was composed of a mannan core and glucose and galactose side chains. The mannan core was composed of (1 → 2)-α-Manp substituted by the side chains (1 → 6)-α-D-Manp, (1 → 4)- α-D-Glcp and (1 → 6)-α-D-Glcp with different ds.p. at the C-6. The galactose side chains had the backbone of (1 → 6)-β-D-Galf. There were two branch sites on every five sugars on average at the O-2 of →6)-β–Galf(1→, and the branches consisted of (→2)-β-D-Galf(1→ and terminal β-D-Galf) 1→. The potential immunomodulatory activities of SB1-1 on RAW264.7 macrophages were investigated. The results showed that SB1-1 could activate macrophage and significantly improve its phagocytic ability by neutral red uptake assay. Addnl., SB1-1 enhanced the productions of IL-1, IL-6, IL-12 and TNF-α in the RAW264.7 macrophages by stimulation of SB1-1, while significantly increased the production of inducible nitric oxide synthase (iNOS) production This study demonstrated that the galactofuranose-containing polysaccharide SB1-1 from S. bambusicola which is a novel mannogalactoglucan, could enhance the activation of macrophages.

In this research work, a water-soluble polysaccharide (LAP) isolated from the fruits of Lycium arabicum was investigated. LAP contains carbohydrates (82.45±1.23%), protein (1.56±0.21%), and uronic acids (3.56±0.34%). The anal. of the monosaccharide composition revealed the presence of rhamnose, arabinose, galactose, glucose and mannose in a molar ratio of 4.7 : 1.5 : 1 : 8.7 : 16.4 : 5.6. The extracted polysaccharide (PS) was considered as heterogeneous and highly branched by interpreting its GC/MS, FT-IR and NMR data. Crystallinity of LAP was inferred from its X-ray diffractometry (XRD) and SEM (SEM) anal. LAP exhibited an interesting stability at high temperatures (∼254°C) and in a wide range of pH (3-9) deduced, resp., from its DSC and zeta potential anal. LAP displayed a strong antioxidant activity at low concentrations evaluated by the 2,2-diphenyl-1-picrylhydrazyl (DPPH)-radical scavenging, ferric reducing activity power (FRAP), free radical scavenging ability, superoxide radical-scavenging and hydroxyl radical-scavenging abilities. Inhibition of erythrocyte hemolysis and lipid peroxidation was also assessed. In 5 h, LAP treatment allowed the protection of the damaged erythrocytes caused by AAPH (2,2-azobis(2-amidinopropane) dihydrochloride), to reduce the level of malondialdehyde (MDA) as well as to increase the reduced glutathione (GSH) level.

Keemun, Assam, Darjeeling and Ceylon black teas are honored as the world’s four most famous black teas, and their excellent aroma qualities are well received by people around the world. In this study, aroma components in these four types of teas were analyzed by comprehensive two-dimensional gas chromatog. time-of-flight mass spectrometry (GC × GC-TOFMS) and gas chromatog.-olfactometry (GC-O) technologies. A total of 42 aroma-active compounds were ultimately identified, especially benzeneacetaldehyde, geraniol, (Z)-3-hexen-1-yl hexanoate, trans-β-ionone, cis-linalool oxide (pyranoid), hotrienol, and Me salicylate presented the strongest aroma strengths with pleasant scents in all tested teas. The quantification results indicated that 19 compounds including (Z)-3-hexenol, 1-octen-3-ol, linalool, phenylethyl alc., hexanal, benzeneacetaldehyde, limonene, heptanoic acid, (Z)-3-hexen-1-ol, acetate, benzyl alc., trans-linalool oxide (furanoid), hotrienol, 1-octen-3-one, 2-nonanone, (E)-2-octenal, nonanal, β-myrcene, 2-pentylfuran, and methylpyrazine were identified as the key compounds with odor activity values (OAVs) higher than 1.0 in the world’s four most famous black teas. Notably, the comparison of GC-O and OAV calculation results showed that Me salicylate (Ceylon), (E)-2-octenal (Assam), benzeneacetaldehyde (Keemun) and linalool and trans-linalool oxide (furanoid) (Darjeeling) might be the most definitive odorants in the corresponding tea categories.

Scrophulariae Radix (SR) is one of the oldest and most frequently used Chinese herbs for oriental medicine in China. In total, 20 compounds were identified as potential chem. markers that changed significantly with different steaming durations. Seven compounds can be used as potential chem. markers to differentiate processing by sweating, hot-air drying, and steaming for 4 h. These findings could elucidate the change of chem. constituents of the processed SR and provide a guide for the processing. In addition, our protocol may represent a general approach to characterizing chem. compounds of traditional Chinese medicine (TCM) and therefore might be considered as a promising approach to exploring the scientific basis of traditional processing of TCM.

Plants in Amorphophallus genus are rich in soluble dietary fibers and have long been used as food and traditional medicine in Asian countries. In the present investigation, natural O-acetyl-glucomannans were extracted from tubers of Amorphophallus rivirei (konjac), A. albus and two local types of A. bulbifer (wild type M and wild type B), which were designated as KGM, AGM, MGM, and BGM, successively. Comparative studies were conducted on their physicochem. properties and primary structures. The four AcGMs had about 90% (weight/weight) neutral sugar, 11% (weight/weight) moisture, and 1%-2% (weight/weight) ash. Relatively high contents of acetyl groups (17%-24%, weight/weight) were detected. Mannose and glucose as the main components existed in molar ratios of 1.41, 1.41, 1.21, and 1.58 for KGM, AGM, MGM, and BGM, resp. Relative mol. weights of AcGMs increased in the order of: KGMName: (2S,3S,4R,5R)-2,3,4,5,6-Pentahydroxyhexanal.

The reaction begins with the homolytic fission of the C-I bond in iodoform induced by visible-light irradiation, produced active radicals. The following steps involved as radical trapping by DMF, nucleophilic substitution, Vilsmeier-type reagent formation, and the final iodination of the alc. The excellent performance of the model reaction in the flow photoreactor demonstrated the potential of this system for practical application.

In this study, the structural characterization and anti-inflammation effect of dilute alkali-soluble polysaccharides from purple sweet potato were investigated. Three fractions (F-1, F-2 and F-3) were obtained by purifying crude polysaccharides on DEAE-52 cellulose column. The main fraction (F-1) was further purified on Sephadex G-200 column to afford purified alkali-soluble sweet potato polysaccharide (ASPP). The chem. structure of ASPP was analyzed by gas chromatog., Fourier transform IR spectroscopy, methylation anal. and NMR spectroscopy. Monosaccharide compositional anal. showed ASPP was composed of rhamnose, arabinose, xylose, mannose and glucose in the molar ratio of 2.8:1.9:1.0:7.6:53.3. Moreover, the backbone of ASPP was composed of 1,4-linked Glcp with side chains attached to the O-6 position. The anti-inflammation effect of ASPP was further investigated by in vitro and in vivo experiments Results showed ASPP could inhibit the levels of nitric oxide, interleukin (IL)-6, IL-1β and TNF-α but increase the production of IL-10 in lipopolysaccharide (LPS)-treated RAW 264.7 macrophage cells. In addition ASPP could reduce the secretion of IL-6, IL-1β and TNF-α in LPS-treated mice. Our results suggest ASPP can be developed as a novel anti-inflammation agent.

We synthesized a series of novel alicyclic compounds by modifying 1,5-anhydro-D-glucitol with two to four palmitoyl chains, and we explored their self-assembly and gelation behaviors in paraffin. The obtained organogels were studied by field emission SEM, at. force microscopy, variable-temperature Fourier transform IR spectroscopy, X-ray diffraction anal., polarized optical microscopy, and transmission spectroscopy. While all the palmitoylated derivatives spontaneously formed fibrous networks and gelated the paraffin, an acetylated derivative of 1,5-anhydro-D-glucitol did not gelatinize the solvent, thus indicating the importance of aliphatic chains for gelation. Interestingly, α- and β- D-glucopyranose with five palmitoyl chains neither gelatinized the solvent nor formed fibrous networks, thus suggesting that the absence of C-1 substitution in 1,5-anhydro-D-glucitol is important for gelation. Fourier transform IR spectroscopy suggested that the formation of weak hydrogen bonds between the carbonyl groups and the C-H groups was the driving force for formation of the supramol. fibers and for gelation of the solvent.

Bladder cancer is one of the most malignant tumors closely associated with macrophage immune dysfunction. The Chinese medicine polyporus has shown excellent efficacy in treating bladder cancer, with minimal side effects. However, its material basis and mechanism of action remain unclear. A new water-soluble polysaccharide (HPP) with strong immunomodulatory activity was isolated from the fungus Polyporus umbellatus (Pers.) Fries. HPP had an average mol. weight of 6.88 kDa and was composed mainly of an <-(1→ 4)-linked D-galactan backbone. The immunomodulatory activity of HPP was determined in vitro, and the results revealed that it could obviously increase the secretion of immune factors by IFN-γ-stimulated macrophages, including nitric oxide (NO), interleukin-6 (IL-6), interleukin-1β (IL-1β), RANTES and interleukin-23 (IL-23), and the expression of the cell membrane mol. CD80. In addition, HPP was recognized by Toll-like receptor 2 (TLR2) and activated the signaling pathways of NF-κB and NLRP3 in a bladder cancer microenvironment model, indicating that HPP could enhance host immune system function. These findings demonstrated that HPP may be a potential immune modulator in the treatment of immunol. diseases or bladder cancer therapy.
