Glutathione-s-transferase is believed to be involved in the resistance to chemotherapeutic drugs, which depends on the interaction with the cell membranes. In this study, we employed Langmuir monolayers of a mixture of phospholipids and cholesterol (MIX) as models for tumor cell membranes and investigated their interaction with the anticancer drugs cisplatin (CDDP) and doxorubicin (DOX). We found that both DOX and CDDP expand and affect the elasticity of MIX monolayers, but these effects are hindered when glutathione-s-transferase (GST) and its cofactor glutathione (GSH) are incorporated. Changes are induced by DOX or CDDP on the polarization-modulated IR reflection absorption spectroscopy (PM-IRRAS) data for MIX/GST/GSH monolayers, thus denoting some degree of interaction that is not sufficient to alter the monolayer mech. properties. Overall, the results presented here give support to the hypothesis of the inactivation of DOX and CDDP by GST and point to possible directions to detect and fight drug resistance. Alzheimer’s disease (AD) is characterized by multiple pathol. hallmarks, including β-amyloid aggregation, oxidative stress, and metal dys-homeostasis. In the absence of treatments addressing its multi-factorial pathol., the authors designed novel multifunctional adamantane-based semicarbazones and hydrazones (1-12) targeting AD hallmarks. Of these, 2-pyridinecarboxaldehyde (N-adamantan-1-yl)benzoyl-4-amidohydrazone (10) was identified as the lead compound, which demonstrated: pronounced iron chelation efficacy; attenuation of CuII-mediated β-amyloid aggregation; low cytotoxicity; inhibition of oxidative stress; and favorable characteristics for effective blood-brain barrier permeation. Structure-activity relationships revealed that pyridine-derived hydrazones represent a promising pharmacophore for future design strategies due to their ability to bind critical FeII pools. Collectively, the unique multifunctional activity of these agents provides a novel therapeutic strategy for AD treatment. Engineering microbes to produce isoprenoids can be limited by the competition between product formation and cell growth because biomass and isoprenoids are naturally derived from central metabolism Recently, a two-step synthetic pathway was developed to partially decouple isoprenoid formation from central carbon metabolism The pathway used exogenously added isopentenols as substrates. In the present study, we systematically optimized this isopentenol utilization pathway in Escherichia coli by comparing enzyme variants from different species, tuning enzyme expression levels, and using a two-stage process. Under the optimal conditions found in this study, ~300 mg/L lycopene was synthesized from 2 g/L isopentenol in 24 h. The strain could be easily modified to synthesize two other isoprenoid mols. efficiently (248 mg/L β-carotene or 364 mg/L R-(-)-linalool produced from 2 g/L isopentenol). In recent years, the increasing demand for clean and potable water has created a need for efficient, cost-effective wastewater treatment. One of the most promising methods is heterogeneous photocatalysis due to its ability to mineralize organic mols. This paper describes the development of a flow-through packed bed reactor system based on a packed column comprising glass beads coated with com. available TiO2 (P25). The resulting deposited films and corresponding powders were characterized using thermal anal. (TGA-DSC-MS), X-ray diffraction (XRD), scanning/transmission electron microscopy (SEM/TEM), sp. surface area (BET) and XPS measurements. The photocatalytic efficiency of the reactor was tested by observing the degradation rate of Plasmocorinth B, an organic dye, and 18 bisphenols co-dissolved in deionized water and simulated wastewater under UV light. The developed photocatalytic reactor effectively removed organic dye and bisphenols from the aqueous medium using a combination of adsorption and photocatalysis. The goal of this study was to perform in silico identification of bioinsecticidal potential of 42 monoterpenes against Drosophila melanogaster and Reticulitermes chinensis Snyder. Quant. structure-activity relationship (QSAR) modeling was performed for both organisms, while docking and mol. dynamics were used only for Drosophila melanogaster. Neryl acetate has the lowest interaction energy (-87 kcal/mol) against active site of acetylcholinesterase, which is comparable to the ones of methiocarb and pirimicarb (-90 kcal/mol) and reported PDB binder 9-(3-iodobenzylamino)-1,2,3,4-tetrahydroacridine (-112.67 kcal/mol). Interaction stability was verified by mol. dynamics simulations and showed that the stability of ACHE active site complexes with three selected terpenes is comparable to the one of the pirimicarb and methiocarb. A more accurate HPLC-MS screening method combining functional enzyme assay and affinity ultrafiltration screening assay was developed and applied for the screening of natural product inhibitors of α-glucosidase from Cercis chinensis Bunge fruits. The enzyme assay was conducted to prescreen botanical extracts, in which maltose was used as the substrate and detection object. That showed the Cercis chinensis Bunge fruits demonstrated higher α-glucosidase inhibitory activity (IC50 = 11.94 ± 1.23μg/mL) than acarbose (IC50 = 44.03 ± 4.37μg/mL) (n = 3, p < 0.05). Subsequently, twelve bioactive components targeting α-glucosidase were screened out and identified using affinity ultrafiltration coupled to liquid chromatog.-mass spectrometry. The known inhibitor, acarbose, was used as a pos. control and competitive ligand to eliminate false positives. Untargeted metabolomics was performed to study the profiles of 101 chems. in lotus seeds using ultrahigh-performance liquid chromatog.-photodiode array detection-high-resolution tandem mass spectrometry. Among them, 16 dimeric, 18 trimeric, and 4 tetrameric proanthocyanidins were theor. identified based on the d.p., and the number of linkages and the presence of two dihydroflavonols and three glycosylated alkaloids were determined for the first time. The proanthocyanidin, flavonoid, amino acid, and total compound contents were quantified, revealing decreases in their levels during maturation as well as a polymerization process formation of polymers from monomers during seed maturation. Interestingly, glycosylated alkaloids were only detected in seed cotyledons being highest at green-brown stage, whereas proanthocyanidins were present at a concentration of 8,226.19 ± 249.96μg/g (dry weight) in green-brown stage of seed coats. New VLE data are presented for CO2 + 3,7-dimethyl-1-octanol (37DM1O) + 1-decanol (C10OH) at 35, 55 and 75°C and pressures between 68 and 237 bar. Phase compositions were analyzed using a static analytic app with dual sampling and online chromatog. The mixture displays virtually no enhanced solubility, liq curves remain convex throughout, and coexisting detached two-phase regions were not detected. CO2 can fractionate all measured 37DM1O:C10OH ratios though with limited efficacy, as highlighted by relative solubilities ranging from 1.1 to 1.5. RK-ASPEN, SR-POLAR, PR-BM and PC-SAFT correlations failed at 35°C, the optimum fractionation temperature At 55 and 75°C, RK-ASPEN and SR-POLAR produced the smallest percentage av abs deviations in equil pressure of 4.0%, and RK-ASPEN was marginally superior at correlating vapor and liq phase curves. The aim of this study is to determine the phenolic composition of Hypericum linarioides Bosse, to assess its various biol. properties, and finally to confirm the exptl. data of enzyme inhibitions via docking studies. LC-MS/MS anal. performed over methanol extract (ME) revealed the presence of 27 phytochems. Antioxidant activity results indicated that ME had the strongest DPPH radical scavenging and ferric ion reducing capacity than water and hexane extracts XTT assays revealed the strongest anti-proliferative activity of ME against MCF-7 and HT-29 cancer cells. Flow cytometry results also confirmed the notable apoptotic effect of ME on MCF-7 cells through mitochondria-dependent pathway. Enzyme activity results exhibited the most promising acetylcholinesterase and α-glycosidase inhibitory activities of the ME. Mol. docking calculations of α-glycosidase with the major compounds (1, 15, 17 and 19) indicated that 17 (isoquercitrin) had the best docking binding energy (-10.92 kcal/mol) with appropriate binding interactions.
