Protein biomarkers play an important role in the diagnosis and treatment of disease. Herein, the authors report an ultrasensitive magnetic-bioluminescent-nanoliposome based technique using a portable ATP luminometer for the point-of-care testing (POCT) of protein biomarkers in blood. The DNA repair enzyme tyrosyl-DNA-phosphodiesterase 1 (TDP1) is a current inhibition target to improve the efficacy of cancer chemotherapy. Previous studies showed that compounds combining adamantane and monoterpenoid fragments are active against TDP1 enzyme. In order to prepare insulin-like growth factor 1 (IGF-1) more economically and efficiently, the structure prediction and molecular docking of three IGF-1 fusion proteins were performed by computer simulation. The most suitable expression form of IGF-1 fusion protein was screened out. A prokaryotic expression vector of IGF-1 fusion protein was constructed and transformed into Escherichia coli Origami B(DE3) strain to obtain the recombinant strain. After induction with IPTG, the target protein was purified from the soluble fractions of the bacteria cell lysate by affinity chromatography, desalination, thrombin digestion and affinity chromatography of the enzyme digested products. An activity evaluation system was established by 3T3 cell proliferation method and the activity of the obtained IGF-1 was measured. The results showed that the sequence of the IGF-1 fusion protein prokaryotic expression vector was correct and the fusion protein was soluble upon 0.05 mmol/L IPTG induction at 25 ℃ for 16 h. After preliminary purification, thrombin digestion and re-purification, IGF-1 target protein with purity over 90% was obtained. Using the established activity evaluation system, the specific activity of IGF-1 was 2.47×105 U/mg, which was close to the standard product available at the market. The preparation technology of IGF-1 developed in this study may facilitate the development and industrial production of IGF-1 drugs. In this study, the influence of grapefruit seed extract (GFSE) was studied on citric acid and zinc oxide incorporated cellulose-based hydrogel films. The effect of GFSE (0.25-1.0%, volume/volume) on hydrogel films was examined for their physico-chem., mech., thermal, antioxidant and antibacterial properties. The swelling ratio, tensile strength and thermal stability of films decreased with increase in GFSE concentrations On the other hand, elongation (%) and antioxidant activity of films were found to be 34-60% and 25-79%, resp. for increasing amounts of GFSE. FESEM and FETEM clearly showed that two different types of nanoparticles (50-90 nm and 5-10 nm) were present in GFSE incorporated films. The release of polyphenolic compounds and zinc from films was in sustained manner. Furthermore, the fabricated hydrogel films exhibited antibacterial activity against both E. coli and S. aureus. All these results indicate that the prepared nanocomposite hydrogel films could find their potential applications in wound healing.

With the development of power industry, the recycling of waste insulator silicone rubber (WSR) remains a significant challenge. Herein, an efficient alcoholysis strategy for the surface modification of WSR was explored by using nerol as alcoholysis reagent, and then the nerol modified WSR (Nerol-WSR) was utilized in polypropylene toughening. FTIR spectra indicate that nerol can served as nucleophilic reagent to cleave the Si-O bonds in WSR, resulting in an efficient surface grafting of nerol moieties on WSR. The grafting content of nerol moieties gradually increases with alcoholysis times. Morphol. investigation demonstrates that with the increase of alcoholysis time, the dispersion state of Nerol-WSR particles in PP matrix is significantly improved, along with a much smaller particle size. Accordingly, with the addition of 10 weight% of Nerol-WSR, the resulting PP composites can achieve a remarkable increase of approx. 200% in impact strength. This should be due to the grafted nerol moieties can substantially improve the interfacial adhesion of the composites and hence facilitate the dispersion of WSR particles in PP matrix. Therefore, the stress concentration of PP can also be effectively delivered to the adjacent Nerol-WSR and thus retard the propagation of cracks. In addition, the addition of Nerol-WSR can also increase the loss tangent values of the composites, which indicates its potential application in polymer damping.

Metal Ni and Co are widely used in the catalytic amination of alcs. and Ni is more favored due to its better catalytic performance. Here we compared their catalytic performance for the amination of ethylene glycol and unexpectedly found that Co is more active than Ni. DFT calculation results showed that the energy barriers for the dissociation of O-H and C-H bond in an ethylene glycol mol. are smaller on Co (0001) plane, accounting for higher ethylene glycol conversion and primary amine yield on metal Co catalyst. The C-C bond dissociation in a glycolaldehyde (dehydrogenation intermediate of ethylene glycol) mol. has significantly lower energy barrier on Ni (111) plane, being consistent with lower selectivity of primary amine on metal Ni catalyst. We also compared their catalytic performance for the amination of other diols and the results showed that Co is more active when short-chain diols are used as the substrate.
