Two polysaccharides, DGS1 and DGS2, were obtained by solid-state fermentation (SSF) of Fusarium solani DO7, an endophytic fungus isolated from the orchid Dendrobium officinale. Structural characterizations revealed that DGS1 consisted of arabinose, glucose, mannose and galactose with a molar ratio of 2.9:13.4:3.0:1, resp., and contained (1 → 5)-Araf, (1 → 4)-Glcp, (1 → 6)-Glcp, (1 → 3)-Manp, (1 → 2,6)-Manp and (1 → 6)-Galp glycosidic linkages, while DGS2 was composed of arabinose, glucose, mannose and galactose in a molar ratio of 3.5:8.1:2.1:1, resp., and contained (1 → 5)-Araf, (1 → 4)-Glcp, (1 → 6)-Glcp, (1 → 3)-Manp and (1 → 6)-Galp glycosidic linkages. Neither polysaccharide was toxic to human embryonic kidney cells or mouse RAW 264.7 macrophage cells. An immunomodulatory activity assay indicated that both polysaccharides could significantly enhance the levels of TNF-α, IL-6 and NO by activating TNF-α, IL-6 and iNOs gene expression, resp., especially DGS2. Interestingly, DGS2 also possessed relatively high antioxidant activity. These results illustrate that, due to its cost-effectiveness and environmentally friendly features, SSF has significant potential as a com. competitive source of natural products, including fungal polysaccharides, with immunomodulatory activity.

The solubility of borneol, camphor, and isoborneol in four solvents (i.e., acetone, ethanol, p-cymene, and p-xylene) was measured by the static equilibrium method within the temperature range of 296.65-348.85 K under the atm. The fusion points and fusion enthalpies of borneol and isoborneol were determined by differential scanning calorimetry. The Apelblat equation, Wilson model, and NRTL model were employed to correlate the measured solubility data, and the NRTL model fits the measured data better. Mixing properties of each solute-solvent pair are calculated by the NRTL model.

Bicontinuous microemulsions gelled with a low mol. weight gelator have been shown to be an orthogonally self-assembled system. With the mech. stability provided by the gel network, gelled non-toxic bicontinuous microemulsions have the potential to be an efficient transdermal drug delivery carrier. However, up to now no suitable system has been formulated for transdermal drug delivery. To fill this gap, we formulated and characterized a gelled non-toxic bicontinuous microemulsion suitable for the mentioned application. Starting from a previously studied scouting system, namely, H2O-n-octane-n-octyl β-D-glucopyranoside (β-C8G1)-1-octanol, the co-surfactant and the oil were replaced by non-toxic components. Subsequently, the expensive pure surfactant was replaced by cheap tech.-grade surfactants (Plantacare series) to make the system economical. Having formulated the non-toxic microemulsion H2O-IPM-Plantacare 1200 UP-1,2-octanediol, three low mol. weight gelators were studied with regard to the gelation of both the scouting system and the non-toxic system. The chosen gelators were 12-hydroxyoctadecanoic acid (12-HOA), 1,3:2,4-dibenzylidene-D-sorbitol (DBS), and N,N’-dibenzoyl-L-cystine (DBC). We found that only DBS gels the non-toxic microemulsion. The gelled non-toxic bicontinuous microemulsion H2O-IPM-Plantacare 1200 UP-1,2-octanediol was characterized with oscillatory shear rheometry and small-angle neutron scattering (SANS) at a DBS concentration of 0.3 wt% to verify that the system is indeed a gel and that the microstructure of the microemulsion is not altered by the gel network.

A water-soluble acidic polysaccharide containing 3-O-Me galactose, named YB-PS4, was isolated from the twigs and leaves of Juniperus tibetica Kom. Its structure was characterized by monosaccharide composition anal., methylation, and NMR spectroscopy. It was concluded that YB-PS4 had a backbone composed of (2,4)-a-Rhap-(1, 3,5)-a-Araf-(1, 2,4)-a-Galp-(1 and 4)-a-GalpA-(1, with branches of 2)-a-Rhap-(1,3)-a-Araf-(1 and 2)-3-O-Me-a-Galp-(1). The possible repetitive units were speculated and further analyzed by oligosaccharide anal. YB-PS4 showed inhibitory effects on complement activation through the classical pathway (CH50 = 94.23 ± 8.90 mug/mL) and alternative pathway (AP50 = 194.76 ± 9.20 mug/mL). Preliminary mechanism study indicated that it interacted with C1q, C2, C3, C4 and C5. These studies pointed a way to understand the active constituents of J. tibetica and provided scientific bases for YB-PS4 as a complement inhibitor.

Click chem. is an effective tool to fabricate three-dimensional gel network in an accurate way. In this work, Pluronic F127 gels are prepared by thiol-ene click chem., utilizing double-bond end-capped Pluronic F127 (F127DA) and pentaerythritol tetras-mercaptopropionate (PETMP) as the starting materials and triethylamine as the catalyst. Both the mass fraction of F127DA and the initial concentration of PETMP have effects on the properties of the resulting F127 gels. The optimized F127 gel prepared under a F127DA mass fraction of 15 wt% and a PETMP concentration of 17.62 mmol/L exhibits the highest swelling ratio (10) and has the largest elongation at break (194%). When the F127DA mass fraction and PETMP concentration are fixed at 15 wt% and 48.26 mmol/L, resp., the resulting F127 gel has a high stress of 0.25 MPa. The present example has presented a method to fabricate three-dimensional gel network in a controlled way.

Shape-anisotropic polymeric colloids having chem. distinct compartments are promising materials, however, introducing site-sp. surface functionality to block copolymer particles has not yet been actively investigated. The current contribution demonstrates the selective surface functionalization of nanostructured, ellipsoidal polystyrene-b-polybutadiene (PS-b-PB) particle and investigate their effects on the particle shape. Photo-induced thiol-ene click reaction was used as a selective functionalization chem. for modifying the PB block, which was achieved by controlling the feed ratio of functional thiols to the double bonds in PB. Importantly, the controlled particle elongation was observed as a function of the degree of PB functionalization. Such an increase in the aspect ratio is attributed to the (i) increased incompatibility of the PS and modified PB block and (ii) the reduced surface tension between the particles and surrounding aqueous medium, both of which contributes to the further elongation of ellipsoids. Further tunability of the elongation behavior of ellipsoids was further demonstrated by controlling the particle size and chem. structure of functional thiols, showing the versatility of this approach for controlling the particle shape. Finally, the utility of surface functionality was demonstrated by the facile complexation of fluorescent dye on the modified surface of the particle via favorable interaction, which showed stable fluorescence and colloidal dispersity.
