Transcriptional regulation of alcohol induced liver fibrosis in a translational porcine hepatocellular carcinoma model

Hepatocellular carcinoma (HCC) is the 5th most common and 2nd deadliest cancer worldwide. HCC risk factors include alcohol induced liver cirrhosis, which prompts hepatic inflammation, cell necrosis, and fibrosis deposition. As 25% of HCC cases are associated with alcohol induced liver disease, understanding the effects of the cirrhotic liver microenvironment on HCC tumor biology and therapeutic responses are critical. This study utilized the Oncopig Cancer Model-a transgenic pig model that recapitulates human HCC through induced expression of KRAS(G12D) and TP53(R167H )driver mutations-to investigate the molecular mechanisms underlying alcohol induced liver disease. Oncopigs (n = 5) underwent fibrosis induction via infusion of ethanol and ethiodized oil (1:3 v/v dosed at 0.75 mL/kg) into the hepatic arterial circulation. Eight-weeks post induction, liver tissue samples from fibrotic and age-matched control (n = 5) Oncopigs were collected for histological evaluation and transcriptional profiling. Increased hepatic inflammation and fibrosis was observed in fibrotic Oncopigs via pathological assessment. Transcriptional profiling (RNA-seq) resulted in the identification of 4387 differentially expressed genes between Oncopig fibrotic and control livers. GO term enrichment analysis identified pathway alterations associated with cirrhosis progression in humans, including cell proliferation, angiogenesis, extracellular matrix deposition, and oxidation-reduction. Key alterations include activation of hepatic stellate cells, increased matrix metalloproteinase production, and altered expression of ABC and SLC transporter genes involved in transport of anticancer drugs.These results demonstrate Oncopig liver fibrosis recapitulates transcriptional hallmarks of human cirrhosis, making the Oncopig an ideal model for studying the effects of the cirrhotic liver microenvironment on HCC tumor biology and therapeutic response. Technical application of a ternary alternative jet fuel blend – Chemical characterization and impact on jet engine particle emission

The use of alternative fuels is an essential element in future aviation. There are currently different approaches with regard to fuel production processes and feedstock materials. This requires flexible handling of available fuel quantities. The production of fuel mixtures (blends) is inevitable for an effective usage of commercially available fuel components. Within the framework of the project DEMO-SPK approx. 600 t of a ternary mixture (multiblend) of alternative jet fuels was produced and used in a real airport infrastructure at the airport Leipzig/Halle. Production and application were accompanied by extensive R&D activities to show that on-spec, semi-synthetic multiblends can be produced from several different synthetic fuels. One important aspect is the emission of soot particles from aircraft engines. The Multiblend Jet A-1 has been analyzed in a flow reactor setup regarding the formation of soot precursor compounds in comparison to a reference Jet A-1. The experiment revealed a lower formation of relevant soot precursors which also corresponds to the higher hydrogen content of the Multiblend Jet A-1 compared to the reference fuel. The lower soot formation in a real aircraft engine has been proven during ground runs of an A300-600 aircraft with PW4158 engines. The Multiblend Jet A-1 showed lower particle number and particle mass emission than the reference fuel. The difference to the reference fuel decreased with increasing power settings. In summary, the emission of particle mass is reduced by similar to 29 % and the number of emitted particles is reduced by similar to 37 % if the ICAO landing-and-take-off cycle is used for evaluation.

Na-containing manganese-based cathode materials synthesized by sol-gel method for zinc-based rechargeable aqueous battery

Zinc-based rechargeable aqueous batteries are regarded as one of the most safe and inexpensive energy storage systems compared to the conventional rechargeable batteries with organic electrolytes. Herein, the inexpensive novel cathode materials of Na-containing manganese-based composite of Na0.44MnO2 /Mn2O3 and Na2/3Fe1/2Mn1/2O2 are designed and synthesized correspondingly by sol-gel methods combined with freeze-drying technique, and are applied in the zinc-based aqueous batteries for the first time. The secondary particles of Na0.44MnO2/Mn2O3 composite with a cubic box and the molecular sieve configurations are composed of nano primary particles with about 150-300 nm, and the obtained O3-Na2/3Fe1 /2Mn1/2O2 at 850 degrees C (O3-NFM-850) appears to homogenous distribution with 3D solid block structure and platelet-shaped particles, while the spherical core-shell structure of P2 -Na2/3Fe1/2Mn1/2O2 at 850 degrees C (P3-NFM-850-PVA) after dispersed by poly (vinyl alcohol) are linked to each other by amorphous carbon. Finally, the cathode of Na0.44MnO2/Mn2O3 and P3-NFM-850-PVA deliver the reversible capacities of 68.3 mAh g(-1) and 42 mAh g(-1) even after 200 cycles at 0.4 A/g respectively without capacity fading. Inhibition effect of methanol addition on carbon deposition in propane pyrolysis

Methanol-mixed fuels consisting of methanol, propane, and a small amount of auxiliary solvent are widely used as inexpensive industrial fuels, but the carbon deposition in the pyrolysis of this fuel seriously restrains its popularization. This paper studied the characteristics of gaseous products and carbon deposition in the pure propane and methanol-propane mixture pyrolysis experimentally, analyzed the effect of methanol addition on the reaction path of propane pyrolysis and production rate of key species by using detailed reaction mechanism. Results show that methanol addition can restrain C2H4 converting to C2H2 in the later stage of pyrolysis. The carbon deposition rate and particle diameter increase exponentially with the increase of reaction temperature in both pure propane and methanol-propane blend pyrolysis. Methanol addition can significantly reduce the carbon deposition rate and particle diameter. The reaction path analysis indicates that the main reaction paths forming benzene are C3H3 -> A1 and C4H4 -> I-C6H6 -> A1, and C3H3 is a key specie to form benzene. The pyrolysis of methanol-propane mixture generates more H-2 and H than that of pure propane pyrolysis, which makes most C3H6 convert to C2H4, reduces the amount of C3H3 and suppresses the conversion of C2H4 to C2H2. And then the amount of benzene is reduced significantly and the generation of subsequent PAHs and carbon deposition is inhibited. Lots of H-2 also restrains the hydrogen abstraction from benzene resulting in the delay of the formation of PAHs and carbon deposition. This study is of great significance to research the soot formation in hydrocarbon combustion with alcohol fuel.

Multifunctional nucleating agents with simultaneous plasticizing, solubilizing, nucleating and their effect on polyvinyl alcohol foams

Currently, preparing microcellular polymer foams by supercritical CO2(Sc-CO2) is a newly trend and challenge. The main limitation of Sc-CO2 used in microcellular foaming industry is its solubility. In this paper, four kinds of amines (formamide, ethylenediamine, polyamide dendrimer (PAMAM) and polyhedral oligomeric silsesquioxane (POSS)) were introduced into polyvinyl alcohol (PVA) to improve the foaming behaviors. The results of differential scanning calorimetry showed that formamide and POSS could plasticize PVA to a certain extent. Through SEM analysis, formamide and ethylenediamine has a nucleating effect and improves the foaming effect. Furthermore, the content of formamide has a great influence on the foaming behavior of PVA. TG-IR analysis showed that -NH2 can react with CO2 to form -HNCOO- at normal temperature and pressure, which contributed to the diffusion of CO2 and improved the uniformity of cell size. With increasing formamide content, the cell density increased from 3.22 x 10(9) cm(-3) to 1.79 x 10(10) cm(-3).

BACKGROUND Near-infrared (NIR) spectroscopy coupled with principal component analysis (PCA) and partial least squares (PLS) regression was used to analyse a series of different Irish Whiskey samples in order to define their spectral profile and to assess the capability of the NIR method to identify samples based on their origin and storage (e.g. distiller, method of maturation). The ability of NIR spectroscopy to quantify the level of potential chemical adulterants was also investigated. Samples were spiked with 0.1%, 0.5%, 1.0%, 1.5% and 2.0% v/v of each adulterant (e.g. methanol, ethyl acetate, etc.) prior to NIR analysis. RESULTS The results of this study demonstrated the capability of NIR spectroscopy combined with PLS regression to classify the whiskey samples and to determine the level of adulteration. Moreover, the potential of NIR coupled with chemometric analysis as a rapid, portable, and non-destructive screening tool for quality control, traceability, and food/beverage adulteration for customs and other regulatory agencies, to mitigate beverage fraud was illustrated. CONCLUSION Given the non-specificity of the NIR technique, these positive preliminary results indicated that this method of analysis has the potential to be applied to identify the level of adulteration in distilled spirits. The rapid nature of the technique and lack of consumables or sample preparation required allows for a far more time and cost-effective analysis per sample. (c) 2021 Society of Chemical Industry.

Prior drinking motives predict alcohol consumption during the COVID-19 lockdown: A cross-sectional online survey among Belgian college students

The global outbreak of coronavirus disease 2019 (COVID-19) and the resulting lockdown measures have raised concerns regarding their effect on alcohol consumption. We investigated alcohol use during lockdown in a population of college students, usually characterized by social and heavy drinking. We also tested the predictive role of pre-lockdown drinking motives on alcohol consumption during lockdown. We collected data from 1951 French-speaking Belgian students during the lockdown period (April 1st – May 3rd, 2020) through a crosssectional online survey. Participants self-reported their daily alcohol consumption (1) during a typical week in normal circumstances (i.e., before lockdown), and (2) since lockdown onset. We also assessed drinking motives and severity of alcohol use before lockdown. Our findings showed that 68.2% of the sample reported a lower alcohol consumption during lockdown compared to before lockdown, 17.2% conversely reporting a higher consumption. Enhancement, social and coping motives were all associated with heavy drinking before lockdown. Enhancement and social motives predicted lower alcohol consumption during lockdown among heavy drinkers. Conversely, coping motives, as well as social motives among low drinkers, predicted higher consumption during lockdown. Conformity motives, as well as enhancement motives among low and moderate drinkers, did not predict alcohol consumption before or during lockdown. Overall, several pre-lockdown drinking motives reliably predicted alcohol consumption during lockdown and could thus be used to identify at-risk populations and to tailor intervention programs on alcohol misuse during sanitary crises.
