Alzheimer′s disease is a neurodegenerative disorder whose pathol. mechanisms, despite recent advances, are not fully understood. However, the deposition of beta amyloid -peptide and neuroinflammation, which is probably aggravated by dysbiotic microbiota, seem to play a key role. Anti-Gal are the most abundant xenoreactive natural antibodies. They are supposed to stem from immunization against the gut microbiota and have been implicated in the pathogenesis of several diseases, including multiple sclerosis. These antibodies target the alpha-Gal epitope, expressed on the terminal sugar units of glycoprotein or glycolipid of all mammals except apes, Old World monkeys and humans. The alpha-Gal is constitutively expressed in several bacteria constituting the brain microbiota, and alpha-Gal-like epitopes have been detected in gray matter, amyloid plaque, neurofibrillary tangles and corpora amylacea of the human brain, suggesting a potential link between anti-Gal and Alzheimer′s disease etiopathogenesis. For the first time, our study searched for possible alterations of anti-Gal Ig levels in Alzheimer′s disease patients. IgG and IgM blood levels were significantly lower, and IgA significantly higher in patients than in healthy subjects. These results suggest that such Igs might be implicated in Alzheimer′s disease pathogenesis and open new scenarios in the research for new biomarkers and therapeutic strategies.

Optical diffusers with high transmission, high haze and excellent diffusing ability are widely needed in backlight unit (BLU) of liquid crystal displays (LCDs). However, there are still drawbacks in the particles-doped optical diffusers, including the distribution of the scattered particles due to agglomeration of particles and complex coating process. In this paper, polymer/liquid crystal composite films which are based on the epoxy resins, polyamine (PA) and varied thiol monomers were firstly prepared by thermally initiated polymerization The morphologies and optical properties of the composite films were systematically investigated. By controlling the functionality of thiol in the system of epoxy resins/thiol/PA/LCs, as the light scattering of anisotropic LCs and isotropic polymer framework is both achieved in the optical diffusers, good diffusing ability and high visible transmission (total transmission >96.0% and transmission haze >95.0%) are obtained.

It is well known in industrial applications involving powders and granular materials that the presence of electrostatic charges influences drastically the material flowing properties. The triboelec. charges are produced during flow at the contacts between the grains and at the contacts between the grains and the container. Unfortunately, the triboelec. effect is still poorly understood, even at the fundamental level. Therefore, the approach to solve practical problems is mostly empirical. Moreover, reproducible electrostatic measurements are difficult to perform. In the present study, the ability of a set of excipients and active pharmaceutical ingredients (APIs) to produce electrostatic charges during flow in contact with different materials is analyzed with a recently developed instrument called GranuCharge. While different excipients have almost the same triboelec. behavior and a low chargability, APIs show complex triboelec. properties. Some APIs charge a lot while other APIs charge less. Afterward, the electrostatic behavior of API/excipient blends is considered. We show that the net charge of the blend is a complex function of the relative quantity of API in the mixture Moreover, both the quantity and the sign of the charge are found to depend on the material in contact with the powder during the flow.

Isomalt, a sugar alc., was melt-extruded prior to compression in order to improve its tabletting properties. After fusion, crystalline isomalt was transformed into an amorphous form as shown by x-ray diffraction and DSC. The tabletting properties of amorphous isomalt were dramatically improved. Mixtures formulated with paracetamol (50%) and extruded isomalt yielded hard tablets. However, extruded isomalt powder showed agglomeration problems due to recrystallization of the amorphous phase into a stable crystalline form in the presence of atm. moisture. The evolution of the moisture content correlated well with the compressibility data. The tablets made of extruded isomalt powder had a lower friability in comparison to the tablets formulated with non-extruded isomalt powder. Their disintegration was fast and a rapid dissolution rate was recorded. Extruded isomalt displayed excellent tabletting properties; however, further experiments should be conducted to delay or even prevent recrystallization of amorphous isomalt.

The aim of the present study is to evaluate the effect of a new formulation parameter on the characteristics of dosage form and manufacturing process. In the authors’ previous work the effects of isomalt and compression force on compaction and tablet properties were investigated. In this research paper the particles were agglomerated with purified water without using other binders. To set up the 33 type factorial design a new independent variable – mixing speed (X2) – was added to paracetamol:isomalt ratio in granules (X1) and to compression force during the tabletting process (X3). Each of these variables was examined at 3 levels to characterize their power and interactions. As granule’s properties mass flow, densities and particle size were investigated. From the aspect of compression process lubrication coefficient and friction work were the evaluated response variables. As tablet characteristics tablet resistance to crushing and tensile strength were studied. The results indicated that the agglomeration process did not need any other binders to obtain granules with good quality. Moreover, the addition of mixing speed as a new independent variable was appropriate, since it had significant effect among others on the particle size distribution of the granules and on tablet characteristics. According to the results of the tablet quality tests the optimal mixing speed at granulation is needed which offers not only sufficient homogeneity of the granules but also appropriate tablet characteristics. Furthermore, the enhancing effect of isomalt on the compaction of granules and on the tablet characteristics is observed

Poly(urea-formaldehyde) (PUF) microcapsules filled with dicyclopentadiene (DCPD) were prepared by in situ polymerization and the effect of synthesis parameters, such as pH of the solution and agitation rate, on microcapsules size and shell thickness was evaluated. SEM (SEM) and Fourier transform infra-red spectroscopy (FTIR) were performed. Adjusted pH conditions (pH = 3.5) and agitation rate (1350 RPM) were found using a design of experiments (DOE). SEM results indicated that microcapsule size was directly affected by agitation rate, whereas shell thickness was mostly affected by pH. After obtaining adjusted synthesis conditions, microcapsules presenting mean size of 60μm and mean shell thickness of 4μm were embedded in an epoxy matrix for evaluating the self-healing effect. FTIR and SEM analyses in damaged samples suggested that a healing agent was delivered to the crack location.

The aim of this work is to develop a new generation of repellent products with a long-lasting protection based on a natural component, para-menthane-3,8-diol (PMD). The active is first rendered soluble in a surfactantless microemulsion (H2O/iPrOH/PMD) and then in classical microemulsions. The presence of self-associated nanostructures is detected by dynamic light scattering (DLS). A synergetic system of surfactants (Cremophor RH40 and Texapon N70) is used. Addnl., 2-ethylhexane-1,3-diol and Et (-)-(S)-lactate are incorporated. The final product contains, as main components, 46% of H2O, 25% of iPrOH, 20% of non-H2O-soluble PMD, and only 4% of surfactants. Investigations of lasting protection on human volunteers are carried out using a cage test bioassay protocol and Aedes aegypti mosquitoes. A complete protection of 315 min is found on the test persons using the surfactantless microemulsion. An extension is observed with the final formulation to reach a mean of complete protection of 385 min. This study demonstrates that alternative formulations using a natural active instead of synthetic chems. like N,N-diethyl-m-methylbenzamide (DEET) can be efficient for human protection against mosquitoes.

Characterization of mosquito repellents using arm-in-cage tests are performed by assessing the 95% ED (ED95), half-life and complete protection time (CPT). This study fully characterizes these properties for p-menthane-3,8-diol (PMD), which has not been widely studied, and a long-acting formulation containing a PMD-vanillin composite. A series of arm-in-cage tests against Aedes aegypti (Diptera: Culicidae) mosquitoes were devised using 6 volunteers to estimate CPT or 10 to estimate the ED95 and half-lives for three repellents: 20% N,N-diethyl-3-methylbenzamide (DEET), 30% PMD and a novel 30% PMD-vanillin formulation. Non-linear regression anal. was used to characterize the relationship between applied dose and CPT. PMD and DEET showed a very similar log dose relationship to CPT; however, the PMD-vanillin formulation exhibited a sigmoidal ′S-shaped′ relationship. This resulted in a 1.5-fold higher CPT for PMD-vanillin compared with that of 20% DEET when applied at a dose of 1.6 mg/cm2, but little difference was observed at lower doses of 0.8-1 mg/cm2. The ED95 value for the 30% PMD and PMD-vanillin formulations were 0.25 and 0.24 mg/cm2, resp., these being higher than that for 20% DEET (0.09 mg/cm2). The half-lives for 30% PMD and 20% DEET were similar (2.23 vs. 2.74 h), but longer for the PMD-vanillin formulations (3.8 h). A full characterization for other repellent formulations, particularly those claiming extended longevity, should be conducted in order to identify differences at various applied doses.

In addition to its primary function as an insect repellent, DEET has many “”off-label”” properties, including a deterrent effect on the attraction of gravid female mosquitoes. DEET neg. affects oviposition sites. While deorphanizing odorant receptors (ORs) using the Xenopus oocyte recording system, we have previously observed that DEET generated outward (inhibitory) currents on ORs sensitive to oviposition attractants. Here, we systematically investigated these inhibitory currents. We recorded dose-dependent outward currents elicited by DEET and other repellents on ORs from Culex quinquefasciatus, Aedes aegypti, and Anopheles gambiae. Similar responses were observed with other plant-derived and plant-inspired compounds, including Me jasmonate and Me dihydrojasmolate. Inward (regular) currents elicited by skatole upon activation of CquiOR21 were modulated when this oviposition attractant was coapplied with a repellent. Compounds that generate outward currents in ORs sensitive to oviposition attractants elicited inward currents in a DEET-sensitive receptor, CquiOR136. The best ligand for this receptor, Me dihydrojasmolate, showed repellency activity but was not as strong as DEET in our test protocol.

The essential oil of catmint, Nepeta cataria L., contains nepetalactones, that, on hydrogenation, yield the corresponding dihydronepetalactone (DHN) diastereomers. The DHN diastereomer {(4R,4aR,7S,7aS)-4,7-dimethylhexahydrocyclopenta[c]pyran-1(3H)-one, DHN 1} was evaluated as mosquito repellent, as was the mixture of diastereomers {mostly (4S,4aR,7S,7aR)-4,7-dimethylhexahydrocyclopenta[c]pyran-1(3H)-one, DHN 2} present after hydrogenation of catmint oil itself. The repellency of these materials to Aedes aegypti L. and Anopheles albimanus Wiedemann mosquitoes was tested in vitro and found to be comparable to that obtained with the well-known insect repellent active ingredient N,N-diethyl-3-methylbenzamide (DEET). DHN 1 and DHN 2 also repelled the stable fly, Stomoxys calcitrans L., in this study. DHN 1, DHN 2, and p-menthane-3,8-diol (PMD), another natural monoterpenoid repellent, gave comparable levels of repellency against An. albimanus and S. calcitrans. Laboratory testing of DHN 1 and DHN 2 using human subjects with An. albimanus mosquitoes was carried out. Both DHN 1 and DHN 2 at 10% (wt:vol) conferred complete protection from bites for significant periods of time (3.5 and 5 h, resp.), with DHN2 conferring protection statistically equivalent to DEET. The DHN 1 and DHN 2 diastereomers were also efficacious against black-legged tick (Ixodes scapularis Say) nymphs.
