Major depressive disorder (MDD) is a leading cause of disability, and there is an urgent need for new therapeutics. Stress-mediated induction of pro-inflammation in the periphery contributes to depression-like behaviors both in humans and in exptl. models. Inflammatory cytokine interleukin-6 (IL-6) has emerged as a potential therapeutic target. Our studies demonstrated that metabolism of flavanol rich cocoa preparation (FRP) led to the accumulation of select phenolic acids that may contribute to its anti-inflammatory activity. Using a repeated social defeat stress (RSDS) model of depression, we showed that oral administration of FRP attenuates susceptibility to RSDS-mediated depression, supporting the further development of FRP as a novel therapeutic for the treatment of stress disorders and anxiety in humans.

Com. production of 2-keto-L-gulonic acid (2-KLG), the immediate precursor of L-ascorbic acid, is by Ketogulonicigenium vulgare in co-culture with Bacillus megaterium. We used flux balance anal. (FBA) to study a genome-scale metabolic model (GSMM) of K. vulgare, iWZ663, and found that K. vulgare is deficient in nutrient biosynthetic pathways. Individually omitting L-glycine, L-cysteine, L-methionine, L-tryptophan, adenine, thymine, thiamine and pantothenate from complete chem. defined medium (CDM), caused biomass formation of K. vulgare to decrease to 1%, 21%, 16%, 1%, 26%, 57%, 73% and 24%, resp. Based on these results and FBA, a minimal chem. defined medium (MCDM) was developed that supported monoculture of K. vulgare (0.28 OD600) and 2-KLG production (3.59 g/L), which were similar to those in complete CDM or corn steep liquor powder (CSLP) medium. This study demonstrated the potential of using GSMM and FBA to characterize nutrient requirements, optimize CDM, and study interactions in co-culture.

Producing renewable biofuels from biomass is a promising way to meet future energy demand. Here, we demonstrated a lignin to diesel route via dimerization of the lignin oil followed by hydrodeoxygenation. The lignin oil undergoes C-C bond dehydrogenative coupling over Au/CdS photocatalyst under visible light irradiation, co-generating diesel precursors and hydrogen. The Au nanoparticles loaded on CdS can effectively restrain the recombination of photogenerated electrons and holes, thus improving the efficiency of the dimerization reaction. About 2.4 mmol gcatal-1 h-1 dimers and 1.6 mmol gcatal-1 h-1 H2 were generated over Au/CdS, which is about 12 and 6.5 times over CdS, resp. The diesel precursors are finally converted into C16-C18 cycloalkanes or aromatics via hydrodeoxygenation reaction using Pd/C or porous CoMoS catalyst, resp. The conversion of pine sawdust to diesel was performed to demonstrate the feasibility of the lignin-to-diesel route.

Aminoalcs. have been addressed as activating buffers for alk. phosphatase. However, there is no record on the buffer activation regarding organophosphorus hydrolase (OPH). Here we reported the activating effects of aminoalcs. on OPH-catalyzed hydrolysis of diisopropylfluorophosphate (DFP), an analog mol. of G-type warfare agents. The kinetic parametors kcat, Vmax and kcat/Km in the OPH reaction were remarkably increased in the buffers (pH 8.0, 25 °C) containing aminoalcs. with C2 between nitrogen (N) and oxygen (O) in their structures, including triethanolamine (TEA), diethanolamine, monoethanolamine, 1-amino-2-propanol, 2-amino-2-methyl-1-propanol, and triisopropanolamine. In contrast, much lower or no rate-enhancing effects were observed in the adding of amines, alcs., amine/alc. mixtures, or 3-amino-1-propanol (C3 between N and O). The 300 mM TEA further increased DFP-degrading activities of OPH mutants F132Y and L140Y, the previously reported OPH mutants with desirable activities towards DFP. However, the treatment of ethylenediaminetetraacetate (EDTA) markedly abolished the TEA-induced activation of OPH. The product fluoride effectively inhibited OPH-catalyzed hydrolysis of DFP by a linear mixed inhibition (inhibition constant Ki ∼ 3.21 mM), which was partially released by TEA adding at initial or later reaction stage. The obtained results indicate the activation of OPH by aminoalc. buffers could be attributed to the reduction of fluoride inhibition, which would be beneficial to the hydrolase-based detoxification of organophosphofluoridate.

In this paper, the latest new progresses of mixed cultures fermentation producing 2-KGA are reviewed from the aspects of key enzymes of Ketogulonicigenium vulgare, extracellular substances of co-helper, Omics and exogenous additives. Solid support SiO2 was found to possess favorable catalytic and dispersancy parameters for the condensation reaction. An efficient synthesis of 1,4-disubstituted 1,2,3-triazole derivatives I (R = C6H5, 2-ClC6H4, 3-O2NC6H4, etc.) and II was studied. 1,4-Disubstituted 1,2,3-triazoles containing isoxazole and thymidine structures were synthesized in 84-96% yields starting from various terminal isoxazole ether alkynes and β-thymidine azide derivatives via a 1,3-dipolar cycloaddition using copper acetate, sodium ascorbate as the catalyst under ultrasonic assisted condition. Furthermore, the quorum sensing inhibitory activities of synthesized compounds were evaluated with Chromobacterium violaceum (C. Violaceum CV026) based on their inhibition of violacein production, with compound C10-HSL as a pos. control. The compounds II (R = C6H5, 4-ClC6H4,and 4-C(CH3)3C6H4) exhibited considerable levels of inhibitory activity against violacein production, and IC50 values were 217 +/- 19, 223 +/- 20 and 42.8 +/- 4.5 μM, resp., which highlighted the potential of these compounds as lead structures for further research towards the development of novel QS inhibitors.

Summary : High-temperature fermented apple is a new fermented product with characteristic black appearance, which is prepared through natural fermentation of whole fresh apple at controlled high temperature (60-90°C) and humidity (relative humidity of 50-90%). In this work, melanoidins from high-temperature fermented apple were obtained by ultrafiltration and subsequent diafiltration. The structural characterization of melanoidins was investigated in the present study. Results showed that the mol. weight of the obtained melanoidins was about 179 kDa and the melanoidins were comprised primarily of furans by pyrolysis-gas chromatog.-mass spectrometry. Antioxidant activities of the high-temperature fermented apple and its melanoidins were evaluated by ABTS, DPPH, hydroxyl radicals, ferric reducing antioxidant power and Fe2+-chelating assays. Both high-temperature fermented apple and its melanoidins showed extremely high antioxidant activities in scavenging hydroxyl radicals. High correlation between the antioxidant activity assayed methods and the physicochem. characteristics of melanoidins was obtained. The contribution of melanoidins to the total antioxidant capacity of high-temperature fermented apple was determined
