Effects of post-harvest fungal infection of apples on chemical characteristics of cider

The impact of fungal post-harvest infection of apple on the chemical composition of cider was investigated through a comparative analysis of ciders obtained from apples (Gala variety) separately infected by five fungal species (Alternaria alternata, Botrytis cinerea, Diplodia seriata, Monilinia fructigena, and Penicillium expansum) and cider from sound apples. The content of several flavor-active molecules belonging to different chemical groups, such as alcohols, esters, acids, aldehydes, phenols, and lactones, significantly varied among ciders. Particularly, cider from sound apples had a higher concentration of ethyl ester acetate, fatty acid ethyl esters and fatty acids, molecules that contribute to fruity and sweet scent. Principal component analysis well discriminated ciders, evidencing species-specific fungal effect. Differences in precursor availability in juices and biosynthesis pathways in fungi could explain changes in aroma profile of ciders. This study provides information on the potential risk to produce cider from infected apples due to the possible quality depreciation.

Fuel cells (FC) produce electricity in a continuous mode through a catalytic reaction and have many possible applications, as for example, in the transportation sector substituting the combustion engines. These devices can be regarded as a free emission technology if the fuel used in them is obtained in a renewable mode, such as hydrogen from the reforming of light alcohols obtained from biomass fermentation or gasification. In fact, proton exchange membrane fuel cells (PEMFC) use hydrogen as fuel that, in turn, has to be free of carbon monoxide (CO) since the later chemical species poisons the platinum based catalyst applied in the electrochemical process. This review aims at clarifying how multicomponent catalysts can be used in the hydrogen production from light alcohols reforming to overcome the limitations of current catalysts. Specifically, their low thermal stability, the CO formation that is not suitable for FC use, the carbon (coke) production that poisons the reforming catalyst, or byproducts (i.e. CH4) generation that reduces the hydrogen amount produced. Enhanced flux and fouling resistance forward osmosis membrane based on a hydrogel/MOF hybrid selective layer

Although forward osmosis (FO) technology has been widely adopted for water treatment, it still faces great challenges, including low permeability and fouling problem. Herein, a novel forward osmosis membrane was developed with a stable, uniform and defect-free polyvinyl alcohol/polydopamine coated zeolitic imidazolate framework (PVA/PDA@ZIF-8) hybrid selective layer to simultaneously enhance its transport property and fouling resistance. The prepared FO membrane exhibits improved water flux without losing the selectivity, since the PDA@ZIF-8 provide additional preferential passageway for water transporting while hinder the diffusion of salt. The optimized membrane shows a higher water flux than pristine PVA membrane (14.2 vs. 8.7 L m(-2) h(-1) in FO mode, 24.3 vs. 14.8 L m(-2) h(-1) in PRO mode) with a reasonable selectivity considering as J(s)/J(w) value (0.44 vs. 0.4 g L-1 in FO mode, 0.38 vs. 0.33 g L-1). Moreover, the dynamic fouling experiments with organic foulants (protein and polysaccharide) indicate that the prepared membrane exhibits strong antifouling property and excellent permeation recovery ability (>95.0%) due to the optimized surface property. The chemical composition and potential role of epicuticular and intracuticular wax in four cultivars of table grapes

Plant cuticular wax is the first barrier to resist biotic and abiotic stress. However, little is known about the compositional differences between the epicuticular and intracuticular wax in grape berry. The compositional, morphological and functional features of cuticular wax in grape berries of Vitis vinifera cv. ‘Kyoho’, ‘Muscat Hamburg’, ‘Redglobe’, and ‘Zuijinxiang’ were investigated. The total and epicuticular wax of four berries were mainly composed of terpenoids, hydrocarbons, alcohols, fatty acids and esters. Oleanolic acid was the most abundant terpenoid among the four cultivars. Scanning electron microscopy revealed the crystalline flakes structure of the cuticular wax. Additionally, the removal of epicuticular wax accelerated the weight loss, browning, and softening of grape berries, indicating the plastic-wrap-like effect of the cuticular wax on postharvest quality. This study provided the theoretical basis for further application of the fruit wax or waxy analogue.

Separate cohorts of humanized Val66Met transgenic mice were assessed for either trait impulsivity in the 5-choice serial reaction time (5-CSRT) touchscreen task, or propensity towards obtaining ethanol in an operant paradigm. It was found that female hBDNE(val/val) mice exhibited both greater impulsivity compared to hBDNF(Met/Met) mice of the same sex as shown by a higher number of premature responses at one of three increased inter-trial intervals tested in the 5-CSRT task, and a greater propensity toward stable ethanol self-administration relative to male mice of the same genotype in the operant paradigm. By contrast, male mice showed no difference between genotypes in impulsivity or stable ethanol self-administration. The hBDNF(Met/Met) genotype appears to sex-specifically alter aspects of both impulsive behaviour and addiction propensity. These results suggest that impulse behaviour may be a possible predictor of addiction risk.

Live predator stress in adolescence results in distinct adult behavioral consequences and dorsal diencephalic brain activation patterns

Exposure to traumatic events during childhood increases the risk of adult psychopathology, including anxiety, depression, alcohol use disorders and their co-morbidity. Early life trauma also results in increased symptom complexity, treatment resistance and poor treatment outcomes. The purpose of this study was to establish a novel rodent model of adolescent stress, based on an ethologically relevant life-threatening event, live predator exposure. Rats were exposed to a live predator for 10 min. at three different time points (postnatal day (PND)31, 46 and 61). Adult depression-, anxiety-like behaviors and ethanol consumption were characterized well past the last acute stress event (two weeks). Behavioral profiles across assessments were developed to characterize individual response to adolescent stress. CNS activation patterns in separate groups of subjects were characterized after the early (PND31) and last predator exposure (PND61). Subjects exposed to live-predator adolescent stress generally exhibited less exploratory behavior, less propensity to venture into open spaces, a decreased preference for sweet solutions and decreased ethanol consumption in a two-bottle preference test. Additional studies demonstrated blunted cortisol response and CNS activation patterns suggestive of habenula, rostromedial tegmental (RMTg), dorsal raphe and central amygdala involvement in mediating the adult consequences of adolescent stress. A test of interaction effects between alcohol protective behavioral strategies and antecedents of alcohol-related consequences

The purpose of the present study was to examine interaction effects between alcohol protective behavioral strategies (PBS) and a variety of risk and protective factors for experiencing negative alcohol-related consequences using data from a large, multi-site sample of college students. We recruited 5090 college students (71.2% female; 77.4% White; Mean Age = 20.89, SD = 4.44) who reported any alcohol use in the past month from psychology department participant pools at 10 universities across the U.S. to complete an online survey. Alcohol PBS use was a statistically significant negative predictor of alcohol-related problems while controlling for each antecedent variable individually. In terms of moderation effects, 26 out of 35 interactions (74.3%) were statistically significant in the models controlling only for sex. When controlling for frequency and quantity of alcohol use, 24 of 33 interactions (72.7%) were statistically significant. The nature of these interaction effects was largely the same: more frequent alcohol PBS use reduced the positive associations between risk factors and alcohol-related problems. The lack of specificity of these interaction effects calls into question the limited contributions of tests of interaction effects between alcohol PBS and individual antecedent variables. Comprehensive tests of interaction effects between alcohol PBS and antecedent variables with large samples are needed to move the field forward, especially replication attempts of interactions effects reported in single studies.

A Randomized Trial of Albumin Infusions in Hospitalized Patients with Cirrhosis

BACKGROUND Infection and increased systemic inflammation cause organ dysfunction and death in patients with decompensated cirrhosis. Preclinical studies provide support for an antiinflammatory role of albumin, but confirmatory large-scale clinical trials are lacking. Whether targeting a serum albumin level of 30 g per liter or greater in these patients with repeated daily infusions of 20% human albumin solution, as compared with standard care, would reduce the incidences of infection, kidney dysfunction, and death is unknown. METHODS We conducted a randomized, multicenter, open-label, parallel-group trial involving hospitalized patients with decompensated cirrhosis who had a serum albumin level of less than 30 g per liter at enrollment. Patients were randomly assigned to receive either targeted 20% human albumin solution for up to 14 days or until discharge, whichever came first, or standard care. Treatment commenced within 3 days after admission. The composite primary end point was new infection, kidney dysfunction, or death between days 3 and 15 after the initiation of treatment. RESULTS A total of 777 patients underwent randomization, and alcohol was reported to be a cause of cirrhosis in most of these patients. A median total infusion of albumin of 200 g (interquartile range, 140 to 280) per patient was administered to the targeted albumin group (increasing the albumin level to =30 g per liter), as compared with a median of 20 g (interquartile range, 0 to 120) per patient administered to the standard-care group (adjusted mean difference, 143 g; 95% confidence interval [CI], 127 to 158.2). The percentage of patients with a primary end-point event did not differ significantly between the targeted albumin group (113 of 380 patients [29.7%]) and the standard-care group (120 of 397 patients [30.2%]) (adjusted odds ratio, 0.98; 95% CI, 0.71 to 1.33; P = 0.87). A time-to-event analysis in which data were censored at the time of discharge or at day 15 also showed no significant between-group difference (hazard ratio, 1.04; 95% CI, 0.81 to 1.35). More severe or life-threatening serious adverse events occurred in the albumin group than in the standard-care group. CONCLUSIONS In patients hospitalized with decompensated cirrhosis, albumin infusions to increase the albumin level to a target of 30 g per liter or more was not more beneficial than the current standard care in the United Kingdom. Assessment of Annual Cost of Substance Use Disorder in US Hospitals

IMPORTANCE A persistently high US drug overdose death toll and increasing health care use associated with substance use disorder (SUD) create urgency for comprehensive estimates of attributable direct costs, which can assist in identifying cost-effective ways to prevent SUD and help people to receive effective treatment. OBJECTIVE To estimate the annual attributable medical cost of SUD in US hospitals from the health care payer perspective. DESIGN, SETTING, AND PARTICIPANTS This economic evaluation of observational data used multivariable regression analysis and mathematical modeling of hospital encounter costs, controlling for patient demographic, clinical, and insurance characteristics, and compared encounters with and without secondary SUD diagnosis to statistically identify the total attributable cost of SUD. Nationally representative hospital emergency department (ED) and inpatient encounters from the 2017 Healthcare Cost and Utilization Project Nationwide Emergency Department Sample and National Inpatient Sample were studied. Statistical analysis was performed from March to June 2020. EXPOSURES International Statistical Classification of Diseases, Tenth Revision, Clinical Modification (ICD-10-CM) principal or secondary SUD diagnosis on the hospital discharge record according to the Clinical Classifications Software categories (disorders related to alcohol, cannabis, hallucinogens, inhalants, opioids, sedatives, stimulants, and other substances). MAIN OUTCOMES AND MEASURES Annual attributable SUD medical cost in hospitals overall and by substance type (eg, alcohol). The number of encounters (ED and inpatient) with SUD diagnosis (principal or secondary) and the mean cost attributable to SUD per encounter by substance type are also reported. RESULTS This study examined a total of 124 573 175 hospital ED encounters and 33 648 910 hospital inpatient encounters from the 2017 Healthcare Cost and Utilization Project Nationwide Emergency Department Sample and National Inpatient Sample. Total annual estimated attributable SUD medical cost in hospitals was $13.2 billion. By substance type, the cost ranged from $4 million for inhalantrelated disorders to $7.6 billion for alcohol-related disorders. CONCLUSIONS AND RELEVANCE This study’s results suggest that the cost of effective prevention and treatment may be substantially offset by a reduction in the high direct medical cost of SUD hospital care. The findings of this study may inform the treatment of patients with SUD during hospitalization, which presents a critical opportunity to engage patients who are at high risk for overdose. Aligning incentives such that prevention cost savings accrue to payers and practitioners that are otherwise responsible for SUD-related medical costs in hospitals and other health care settings may encourage prevention investment.
