The reaction was achieved via sequential functionalization of the β and α-sites of the initially formed N-heteroarenium salts followed by a C-N cleavage, proceeding with the striking features of broad substrate scope, excellent functional groups tolerance, high chemoselectivity and atom-efficiency, and applicable for streamline synthesis of some biomedical mols. The strategy utilized will pave the avenues for further development of catalytic transformations of inert organo-systems to functional frameworks.

Classical lipid transporters are suggested to modulate cellular vitamin D uptake. This study investigated the vitamin D levels in serum and tissues of mice deficient in SR-B1 (Srb1-/-), CD36 (Cd36-/-) and ABC-G5/G8 (Abcg5/g8-/-) and compared them with corresponding wild-type (WT) mice. All mice received triple-deuterated vitamin D3 (vitamin D3-d3) for six weeks. All knockout mice vs. WT mice showed specific alterations in their vitamin D concentrations Srb1-/- mice had higher levels of vitamin D3-d3 in the serum, adipose tissue, kidney and heart, whereas liver levels of vitamin D3-d3 remained unaffected. Addnl., Srb1-/- mice had lower levels of deuterated 25-hydroxyvitamin D3 (25(OH)D3-d3) in the serum, liver and kidney compared to WT mice. In contrast, Cd36-/- and WT mice did not differ in the serum and tissue levels of vitamin D3-d3, but Cd36-/- vs. WT mice were characterized by lower levels of 25(OH)D3-d3 in the serum, liver and kidney. Finally, Abcg5/g8-/- mice tended to have higher levels of vitamin D3-d3 in the serum and liver. Major alterations in Abcg5/g8-/- mice were notably higher levels of 25(OH)D3-d3 in the serum and kidney, accompanied by a higher hepatic mRNA abundance of Cyp27a1 hydroxylase. To conclude, the current data emphasize the significant role of lipid transporters in the uptake, tissue distribution and activation of vitamin D.

Three phloroglucinol based polyphenolic compounds have been synthesized and their potential efficacy against gastric ulceration in mice was investigated. Polyphenol (I, II, III) were synthesized through Vilsmeier-Haack formylation, Schiff-base condensation of diformyl- phloroglucinol with 4-amino salicylic acid and Duff formylation reactions resp. We administered indomethacin, a non-steroidal anti inflammatory drug (NSAID), to induce gastric ulceration in Balb/c mice and tested antiulcer efficacy of synthesized phloroglucinol derivatives The pathogenesis of NSAID-induced ulceration is known to be associated to upregulation of matrix metalloproteinase (MMP)-9. All derivatives of phloroglucinol were found to prevent gastric ulceration in mice through depletion of oxidative stress in gastric tissue. Phloroglucinol derivatives also possessed antioxidant activity and they suppressed antioxidant enzymes in gastric tissues to almost normalcy during gastroprotection. Compound III was found to be most efficient anti-ulcer agent among others that inhibited the MMP-9 activity near to control value during prevention of NSAID-induced gastric ulceration. Thus, new polyphenolic compounds derived from phloroglucinol have antioxidant as well as anti-ulcer activity. We conclude that phloroglucinol derivatives as potent anti-ulcer compound in prevention of indomethacin induced gastric ulceration via attenuation of MMP-9 activity.

The formation of and dynamic changes in aroma during white tea processing have not previously been systematically investigated. In this study, advanced comprehensive two-dimensional gas chromatog.-time-of-flight mass spectrometry was employed to investigate the mechanism of white tea aroma formation. A total of 172 volatiles were identified and mainly comprising endogenous volatiles, which displayed diverse change trends during the withering period. In this process, free aroma precursor amino acids and glycosidically bound volatiles (GBVs) were found to contribute to the formation of white tea aroma, with the differential expression of aroma-related key genes accounting for various accumulation of endogenous volatiles and GBVs. In addition, the drying was also shown to play an important role in the formation of white tea aroma. Our study provides the first characterization of white tea aroma formation and establishes a theor. basis for quality control during white tea processing operations.

The development of the central nervous system is associated with tremendous changes in the cell glycome that is tightly regulated. Herein, we have employed bioorthogonal Cu-free click chem. to image temporal distribution of different glycans in live mouse hippocampal neurons during their maturation in vitro. We show development-dependent glycan patterns with increased fucose and decreased mannose expression at the end of the maturation process. We also demonstrate that this approach is biocompatible and does not affect glycan transport although it relies on an administration of modified glycans. The applicability of this strategy to tissue sections unlocks new opportunities to study the glycan dynamics under more complex physiol. conditions.

The flower extracts of Saraca asoca were evaluated in their three different phenol. stages of flowering [bud (S1), mature (S2) and senescent (S3)] in terms of chem. composition and antioxidant activity. The GC/MS based fingerprinting led to identification of 85 metabolites, including 9 amino acids, 20 organic acids, 7 fatty acids, 20 sugar and sugar acids, 8 sugar alcs., 13 phenols and phenolic acids and 8 others compounds The three flowering stages showed prominent changes in their metabolite profile during the process of maturation of the flowering stages from bud to mature to senescence stages determined via GC/MS based metabolomics and chemometric approaches. The amounts and composition of metabolites in each stage showed statistically significant differences, which were reflected in their antioxidant capacities. The three phenol. stages showed antioxidant activities in a dose dependent manner, but the senescent stage showed highest superoxide radical scavenging activity (IC50 = 65.17 ± 2.647 mg/mL) and metal chelating effect (6.65 ± 0.331 mg/mL) in agreement with their high content of phenolic acids. These differences were strongly reproduced in the chemometric analyses (PCA, PLS-DA and s-PLS-DA), identifying the most distinctive features of the three flowering stages. This study might be beneficial to select the most potent flowering stage for incorporation in functional food.

The identification of safe and effective drugs that inhibit tumor invasion and metastasis is required to improve the clinical outcome of patients with colon cancer. The present study aimed to investigate the inhibitory effects and possible mechanisms of action of resveratrol against the invasion and metastasis of colon cancer. AKT1‑knockdown SW480 and SW620 colon cancer cells were used to detect the effects of resveratrol on cell invasion and metastasis, as well as changes in the expression of epithelial‑mesenchymal transition (EMT) markers and serine/threonine kinase (AKT)/glycogen synthase kinase (GSK)‑3β/Snail signaling pathway‑related molecules in vitro. Furthermore, nude mice were inoculated with SW480 cells in the tail vein to establish an in vivo lung metastasis model of colon cancer, to investigate the effects of resveratrol on lung metastasis in colon cancer. The results revealed that resveratrol treatment and AKT1 knockdown significantly inhibited cell migration and invasion in colon cancer, and markedly increased E‑cadherin expression and decreased that of N‑cadherin, phospho (p)‑AKT1, p‑GSK‑3β, and Snail in colon cancer both in vitro and in vivo. Furthermore, the effects of resveratrol were significantly weaker in the AKT1‑knockdown cells. In conclusion, resveratrol may suppress the invasion and metastasis of colon cancer through reversal of EMT via the AKT/GSK‑3β/Snail signaling pathway. AKT1 may therefore be a key regulator of EMT in colon cancer cells and a potential therapeutic target for this disease.

Interactions between bacteriophages (phages) and biofilms are poorly understood despite their broad ecol. and water quality implications. Here, we report that biofilm exposure to lytic polyvalent phages at low concentrations (i.e., 102-104 phages/mL) can counterintuitively promote biofilm growth and densification (corroborated by confocal laser scanning microscopy (CLSM)). Such exposure hormetically upregulated quorum sensing genes (by 4.1- to 24.9-fold), polysaccharide production genes (by 3.7- to 9.3-fold), and curli synthesis genes (by 4.5- to 6.5-fold) in the biofilm-dwelling bacterial hosts (i.e., Escherichia coli and Pseudomonas aeruginosa) relative to unexposed controls. Accordingly, the biofilm matrix increased its polysaccharide and extracellular DNA content relative to unexposed controls (by 41.8 ± 2.3 and 81.4 ± 2.2%, resp.), which decreased biofilm permeability and increased structural integrity. This contributed to enhanced resistance to disinfection with chlorine (bacteria half-lives were 6.08 ± 0.05 vs 3.91 ± 0.03 min for unexposed controls) and to subsequent phage infection (biomass removal was 18.2 ± 1.2 vs 32.3 ± 1.2% for unexposed controls), apparently by mitigating diffusion of these antibacterial agents through the biofilm. Overall, low concentrations of phages reaching a biofilm may result in unintended biofilm stimulation, which might accelerate biofouling, biocorrosion, or other biofilm-related water quality problems.
