Summary : Yam is a common ‘medicine food homol.’ vegetable in Asia, and its peel is often considered a food residue during processing or cooking. In this work, the effects of hydrogen peroxide modification on the dietary fibers (DFs) from Chinese yam peel (CYP) were investigated. The structural characteristics of soluble dietary fiber (SDF), insoluble dietary fiber (IDF), modified soluble dietary fiber (MSDF) and modified insoluble dietary fiber (MIDF) were analyzed using Fourier-transform IR spectroscopy, X-ray diffraction, differential scanning calorimetry, granularity anal., SEM and GC-MS (monosaccharide composition). As results, after modification with hydrogen peroxide, the sizes of the DFs were reduced and MIDF exposed more cellulose. Experiments on the physicochem. and functional properties of DFs showed that MSDF and MIDF obtained a better water holding capacity, oil absorption capacity, swelling capacity and absorption abilities with altered structures, which is of great importance in food processing and development.

Trichoderma viride was utilized to ferment the defatted rice bran (DRB), and soluble dietary fiber was prepared before and after fermentation, named as B-SDF and F-SDF, resp. Optimum fermentation conditions for yield of F-SDF were determined using response surface methodol. (RSM). The yield of F-SDF was significantly increased from 10.5 (weight/weight, %) to 33.4 (weight/weight, %) under the optimal conditions (inoculum size = 10%, pH = 5.8 and time = 41 h). Fourier transform IR spectroscopy (FTIR) demonstrated that both B-SDF and F-SDF had typical characteristics of the IR spectrum of polysaccharides, and X-ray diffraction (XRD) revealed they were mainly substances of amorphous structure. In addition, F-SDF showed looser structure by SEM (SEM) anal., increased mol. weight by high-performance gel permeation chromatog. (HPGPC) anal., and more complex monosaccharide composition by high-performance anion exchange chromatog. (HPAEC) anal. Furthermore, functional properties anal. showed that F-SDF exhibited higher water-holding capacity (WHC), oil-holding capacity (OHC), water solubility (WS) and cholesterol absorption capacity (CAC) than B-SDF. In summary, the present study suggested that SDF from defatted rice bran obtained through Trichoderma viride fermentation has a higher potential to be used as a functional ingredient in foods.

Alzheimer’s disease (AD) is a neurodegenerative disorder leading to cognitive deficits and cognitive decline. Since no cure or preventing therapy is currently available to counteract AD, natural-derived compounds are investigated to find new potential neuroprotective agents for its treatment. In the present study, we tested the neuroprotective effect of lavender and coriander essential oils (EOs) and their main active constituent linalool, against the neurotoxicity elicited by Aβ1-42 oligomers, a key mol. factor in the neurodegeneration of AD. Importantly, our findings on neuronally differentiated PC12 cells exposed to Aβ1-42 oligomers are in accordance with previous in vivo studies reporting the neuroprotective potential of lavender and coriander EOs and linalool. We found that lavender and coriander EOs at the concentration of 10μg/mL as well as linalool at the same concentration were able to improve viability and to reduce nuclear morphol. abnormalities in cells treated with Aβ1-42 oligomers for 24 h. Lavender and coriander EOs and linalool also showed to counteract the increase of intracellular reactive oxygen species production and the activation of the pro-apoptotic enzyme caspase-3 induced by Aβ1-42 oligomers. Our findings provide further evidence that these EOs and their main constituent linalool could be natural agents of therapeutic interest against Aβ1-42-induced neurotoxicity.

We performed a NCI-sponsored, prospective study of neoadjuvant FOLFIRINOX followed by chemoradiation with carboplatin/paclitaxel followed by surgery in patients with locally advanced gastric or gastroesophageal cancer. Patients and Methods: The primary objective was to determine completion rate of neoadjuvant FOLFIRINOX x 8 followed by chemoradiation. Secondary endpoints were toxicity and pathol. complete response (pCR) rate. Exploratory anal. was performed of circulating tumor DNA (ctDNA) to treatment response. From Oct. 2017 to June 2018, 25 patients were enrolled. All patients started FOLFIRINOX, 92% completed all eight planned cycles, and 88% completed chemoradiation. Twenty (80%) patients underwent surgical resection, and 7 had a pCR (35% in resected cohort, 28% intention to treat). Tumor-specific mutations were identified in 21 (84%) patients, of whom 4 and 17 patients had undetectable and detectable ctDNA at baseline, resp. Presence of detectable post-chemoradiation ctDNA (P = 0.004) and/or postoperative ctDNA (P = 0.045) were associated with disease recurrence. Here we show neoadjuvant FOLFIRINOX followed by chemoradiation for locally advanced gastroesophageal cancer is feasible and yields a high rate of pCR. ctDNA appears to be a promising predictor of postoperative recurrence.

Potato peel was used for the extraction of three types of polysaccharides (PW, PAL, and PAC) using water, alk., and acid treatments, resp. The structure of the PP polysaccharides was examined by means of Fourier transform IR spectroscopy (FT-IR) anal. and Gas chromatog.-mass spectrometry (GC-MS). The results suggest that the extracted polysaccharides constituted essentially three functional groups: C=O, C-H, and O-H. The polysaccharides were comprised of low proportions of proteins, 17-23% uronic acids, and approx. 70% carbohydrates. PAL, PW, and PAC with mol. weights of 2.25 × 103, 2.18 × 103, and 1.92 × 103 kDa, resp., were composed of rhamnose, xylose, mannose, arabinose, glucose, and galactose. Functional properties (solubility, oil holding capacity, foaming, and emulsion properties) of these polysaccharides were evaluated. Among three, PAL showed the highest fat-binding capacity which was 7.50 g/g with the solubility of 95.06%. The three polysaccharides possessed appreciable in vitro anti-oxidant (scavenging DPPH and ABTS+ radicals, chelating ferrous ions, and reducing power) potential. PAL exhibited the strongest reducing power, scavenging activity on DPPH radicals and chelating capability on ferrous ions. PP polysaccharides can be used as promising natural antioxidants in food, pharmaceutical, and cosmetic preparations

An efficient and operationally simple gold(III)-catalyzed glycosylation protocol was developed using newly synthesized bench top stable phenylpropiolate glycosyl (PPG) donors. Gold(III)-catalyzed activation of PPGs proceeds well with various carbohydrate and noncarbohydrate-based glycosyl acceptors and leads to their corresponding O/N-glycosides in good to excellent yields with regeneration of reusable and easily separable phenylpropiolic acid. Differentially protected PPGs reacted well under the optimized reaction conditions. In particular, good anomeric selectivity was observed with mannosyl and rhamnosyl PPG donors. A preliminary mechanistic study reveals that the presence of a triple bond adjacent to the ester group is essential for activation, and PPG-based donor shows higher reactivity than analogous acetate and benzoate donors.
