Cross-linked enzyme aggregates (CLEAs) are considered as an effective tool for the immobilization of enzyme. In this study, Pencillium notatum lipase (PNL) was immobilized as carrier free cross-linked enzyme aggregates using glutaraldehyde (GLA) and Ethylene glycol-bis [succinic acid N-hydroxysuccinimide] (EG-NHS) as crosslinking agents. The optimal conditions for the synthesis of an efficient lipase CLEAs such as precipitant type, the nature and amount of crosslinking reagent, and crosslinking time were optimized. The recovered activities of CLEAs were considerably dependent on the concentration of GLA; however, the activity recovery was not severely affected by EG-NHS as a mild cross-linker. The EG-NHS aggregates displayed superior hydrolytic (52.08 ± 2.52%) and esterification (64.42%) activities as compared to GLA aggregates which showed 23.8 ± 1.86 and 34.54% of hydrolytic and esterification activity, resp. Morphol. anal. by fluorescence and scanning electron microscope revealed that EG-NHS aggregates were smaller in size with larger surface area compared to GLA aggregates. The pH optima of both types of CLEAs were displaced to slightly alk. region and higher temperature as compared to native enzyme. Highest enzyme activity of CLEAs was achieved at the pH of 9.0 and 42 °C temperature Moreover, a significant improvement in the thermal resistance was also recorded after immobilization. After ten reusability cycles in aqueous medium, GLA and EG-NHS cross-linked lipase CLEAs preserved 63.62% and 70.9% of their original activities, resp. The results suggest that this novel CLEA-lipase is potentially usable in many industrial applications.

To investigate if topical administration of ambroxol promotes tear secretion and to compare with Diquas ophthalmic eye drop. Two consecutive studies were conducted using sixteen (32 eyes) New Zealand White rabbits. The first study compared the efficacy of ambroxol hydrochloride (0.05%, 0.2%, and 1.0%) on tear and mucin secretion when administered twice daily. Tear secretion was assessed by Schirmer test I and mucin production by conjunctival impression cytol. and PAS stain. The second study compared 0.2% ambroxol hydrochloride with Diquas. A human goblet cell line and human conjunctival tissue were used to test the effect of ambroxol hydrochloride on the expression of aquaporin 5 (AQP5) and MUC5AC, using reverse transcriptase-quant. polymerase chain reaction and immunoblotting. All three concentrations of ambroxol hydrochloride demonstrated significant efficacy on tear stimulation within 2 wk of treatment and total mucin component appeared increased. When administered topically twice daily, 0.2% ambroxol hydrochloride was more effective in augmenting tear secretion than Diquas. With 24 h of treatment, 5μM of ambroxol hydrochloride upregulated AQP5 and MU5AC mRNA and MUC5AC protein in a goblet cell line. When tested on preserved human conjunctiva tissue, a trend of increased production of MUC5AC protein was seen (P = 0.26). Ambroxol is effective in augmenting tear secretion at the ocular surface in rabbits. With actions desirable of a candidate dry eye drug, further investigation of ambroxol and related compounds is warranted to explore their value toward clin. application.

Medicaments essential for alleviation of diseases may sometime adversely affect dental health by eroding the enamel, owing to their acidic nature. It is therefore highly desirable to be able to detect these effects quickly and reliably. In this study, we evaluated the erosive capacity of four most commonly prescribed respiratory disease syrup medicaments on enamel using micro-energy-dispersive X-ray fluorescence spectrometry (μ-EDXRF) and attenuated total reflection Fourier transform IR spectroscopy (ATR-FTIR). Fifty-five enamel fragments obtained from 30 bovine teeth were treated with artificial saliva (S), acebrofilin hydrochloride (AC), ambroxol hydrochloride (AM), bromhexine hydrochloride (BR), and salbutamol sulfate (SS); by immersing in 3 mL of resp. solutions for 1 min, three times a day at intervals of 1 h, for 5 days. μ-EDXRF anal. of enamel surface did not reveal significant erosion caused by the medications. However, ATR-FTIR showed a detectable shift in the phosphate (PO4) antisym. stretching mode (ν3) at ~985 cm-1 for AM, BR, and SS, indicating erosion. Multivariate statistical anal. showed that AC, AM, SS, and BR could be classified with 70%, 80%, 100%, and 100% efficiency from S (control), further highlighting the ability of ATR-FTIR to identify degree of erosion. This suggests ATR-FTIR may be used to rapidly and nondestructively investigate erosive effects of medicaments.

The present study aims to evaluate the effect of brushing with fluoride dentifrice on teeth severely affected by erosion due to respiratory medicaments. Enamel (n = 50) and dentin (n = 50) bovine specimens were prepared and treated with artificial saliva (S-control), acebrofilin hydrochloride (AC), ambroxol hydrochloride (AM), bromhexine hydrochloride (BR), and salbutamol sulfate (SS) and subjected to cycles of demineralization (immersing in 3 mL, 1 min, three times a day at intervals of 1 h, for 5 days) followed by remineralization (saliva, 37°C, 1 h). Simulated brushing with fluoridated toothpaste was performed using 810 strokes in a reciprocal-action brushing simulator. SEM, micro energy dispersive X-ray fluorescence (μ-EDXRF) spectroscopy and attenuated total reflection Fourier transform IR (ATR FTIR) spectroscopy were then performed. μ-EDXRF images showed extensive erosion after treatment with all medicaments. SEM images showed enamel erosion in order SS > BR > AC = AM > S after brushing and fluoridation. FTIR results were in agreement. In case of dentin, μ-EDXRF measurements showed significant difference in mineral content (percent weight of calcium and phosphate) in SS + brushing + fluoridation treated enamel compared to control, while μ-EDXRF images showed erosive effects in the order SS > AM>BR > AC = S post brushing + fluoridation. SEM images showed erosion in the order SS > AM = BR > AC > S post brushing + fluoridation. Again, FTIR multivariate results were in agreement. Overall, our study shows that proper oral care is critical when taking certain medication. The study also demonstrates the possible use of FTIR for rapid clin. monitoring of tooth erosion in clinics.

A UV-spectrophotometric and RP-HPLC method was developed and validated for concurrent estimation of fexofenadine hydrochloride (FEX), montelukast sodium (MLK) and ambroxol hydrochloride (AMB). In RP-HPLC chromatog., the retention time for FEX, MLK and AMB was 2.775 min, 9.529 min and 5.261 min, resp. and detection at max 210 nm for all three of drugs (overlain spectra). Linear regression anal. shows a good linear relationship; in the concentration range of 10-30, 10-50 and 5-25μg/mL; for FEX, MLK and AMB with r2 > 0.999. The UV-spectrophotometric estimation was done by two methods; zero order and first derivative method. These methods were based on the measurement of absorbance at 220.20, 283.42 and 245.84 nm in zero order; 221, 287.52 and 262.25 nm in first order derivative method. In both UV methods, the drugs follow the Beer-Lambert’s law in the concentration range of 6-26, 4-24 and 8-28μg/mL, resp. showing correlation coefficient r2 > 0.999. The RP-HPLC and UV methods showed good reproducibility and recovery with the percent relative standard deviation (RSD) less than 5%. These methods were successfully applied for reliable quantification of FEX, MLK and AMB in a com. and routine anal. in combined dosage form.

Given the inexpensive, nontoxic, and recyclable nature of ChCl:MA, this protocol is environmental friendly.

Epipremnum aureum (Linden and Andre) G. S. Bunting (E. aureum) is an ornamental foliage rich in phytoconstituents and serves as a potential source of bioactive compounds possessing beneficial biol. activities. The present study was carried out to isolate the pyrrolidine alkaloids of leaves with in vitro antitermite and antirepellant efficacy on paper and soil by direct-choice and no-choice assay. Spectrometric anal. of alkaloid fractions via UV-visible spectroscopy (UV-VIS), Fourier transform IR spectroscopy (FTIR) and gas chromatog. mass spectrometry (GC-MS) helped in the characterization of pyrrolidine alkaloids. Superior antitermite activities were observed, associated with high levels of 5-Oxoproline as compare to Me 5-oxo-2-pyrrolidinecarboxylate. Presence of higher contents of 4-Morpholineethanol showed effective antitermite activity and even low levels of 3-Amino-4-Hydroxybutanoic acid and 1H-1,2,4-Triazol-3-amine, N-methyl- also exposed significant activities for repellence and mortality of termites. Repellence and percentage mortality of termites by these alkaloid fractions were similar in results when compared with the standard alkaloid Nicotine and chem. termiticide Monocrotophos. These findings were also supported by SEM studies. Pyrrolidine alkaloids present in E. aureum have exposed strong in vitro antitermite and antirepellant activities against subterranean termites Odontotermes obesus. These findings strongly support the use of E. aureum to bear potential termiticidal properties, however further details need to be carried out.

The voltage-gated sodium channel NaV1.7 is believed to be a critical mediator of pain sensation based on clin. genetic studies and pharmacol. results. Clin. utility of nonselective sodium channel blockers is limited due to serious adverse drug effects. Here, we present the optimization, structure-activity relationships, and in vitro and in vivo characterization of a novel series of NaV1.7 inhibitors based on the oxoisoindoline core. Extensive studies with focus on optimization of NaV1.7 potency, selectivity over NaV1.5, and metabolic stability properties produced several interesting oxoisoindoline carboxamides (16A, 26B, 28, 51, 60, and 62) that were further characterized. The oxoisoindoline carboxamides interacted with the local anesthetics binding site. In spite of this, several compounds showed functional selectivity vs. NaV1.5 of more than 100-fold. This appeared to be a combination of subtype and state-dependent selectivity. Compound 28 showed concentration-dependent inhibition of nerve injury-induced ectopic in an ex vivo DRG preparation from SNL rats. Compounds 16A and 26B demonstrated concentration-dependent efficacy in preclin. behavioral pain models. The oxoisoindoline carboxamides series described here may be valuable for further investigations for pain therapeutics.
