A simple electrochem. mediated method for the conversion of alkyl carboxylic acids to their borylated congeners is presented. This protocol features an undivided cell setup with inexpensive carbon-based electrodes and exhibits a broad substrate scope and scalability in both flow and batch reactors. The use of this method in challenging contexts is exemplified with a modular formal synthesis of jawsamycin, a natural product harboring five cyclopropane rings.

Despite its widespread use, diclofenac has gastrointestinal liabilities common to nonsteroidal antiinflammatory drugs (NSAIDs) that might be reduced by concomitant administration of a gastrointestinal cytoprotectant such as nitric oxide (NO). A series of novel diclofenac esters containing a nitrosothiol (-S-NO) moiety as a NO donor functionality has been synthesized and evaluated in vivo for bioavailability, pharmacol. activity, and gastric irritation. All S-NO-diclofenac derivatives acted as orally bioavailable prodrugs, producing significant levels of diclofenac in plasma within 15 min after oral administration to mice. At equimolar oral doses, S-NO-diclofenac derivatives I [R = R’ = Me, Et, RR’ = (CH2)5] and II (n = 2, 3) displayed rat antiinflammatory and analgesic activities comparable to those of diclofenac in the carrageenan-induced paw edema test and the mouse phenylbenzoquinone-induced writhing test, resp. All tested S-NO-diclofenac derivatives I and II were gastric-sparing in that they elicited markedly fewer stomach lesions as compared to the stomach lesions caused by a high equimolar dose of diclofenac in the rat. Nitrosothiol esters of diclofenac comprise a novel class of NO-donating compounds having therapeutic potential as nonsteroidal antiinflammatory agents with an enhanced gastric safety profile.

A novel series of morpholinyl dendrimer phthalocyanines were synthesized. Their structures were characterized by 1H NMR, IR, elemental anal., ESI-MS as well as MALDI-TOF MS. The photophys. properties of these phthalocyanines were studied by UV/visible and fluorescence spectroscopic methods. The photophys. properties of these dendrimer phthalocyanines exhibited dependence on the number of morpholinyl groups and the central ion. The photoinduced intramol. electron transfer from the morpholinyl groups to the phthalocyanine ring was evidenced by the remarkably quenched fluorescence intensity and shortened lifetime of the silicon phthalocyanine. This difference in photoinduced intramol. electron transfer effect for axially and peripherally substituted morpholinyl dendrimer phthalocyanines was also studied. Besides, introduction of morpholinyl groups on the dendrimer structure could enhance water solubility as well as increase the singlet oxygen quantum yield.

Wu, Xiubing published the artcileSynthesis and surface properties of alkyl β-D-thioglucopyranoside, Related Products of alcohols-buliding-blocks, the publication is Journal of Molecular Liquids (2019), 282-289, database is CAplus.

Alkyl thioglycosides are a class of nonionic sugar-based sulfur-containing surfactants and bioreagents. The surfactants 1,2-trans alkyl β-D-thioglucopyranosides with different alkyl chain length (n = 6-12) were stereoselectively prepared by the Helferich method. Their properties including HLB number, logP value, water solubility, foam property, emulsifying property, surface property and thermotropic liquid crystal property were mainly investigated. The results showed that their HLB numbers and water-solubility decreased as the related logP values increased with increasing the alkyl chain length. Alkyl β-D-thioglucosides were already insoluble in water with n = 10. Both β-D-thioglucopyranosides (n = 8, 9) reduced the surface tension of the related aqueous solution to nearly 29 mN.m-1 at the critical micelle concentration (CMC), they also had excellent foaming ability and foam stability. Nonyl β-D-thioglucopyranoside had good emulsifying properties for both n-octane/water system and toluene/water system. Alkyl β-D-thioglucopyranosides (n = 6-12) were observed to have the thermotropic liquid crystal properties.

Despite the frequent occurrence of γ-branched amines in bioactive mols., the direct catalytic asym. synthesis of this structural motif containing a remote stereocenter remains an important synthetic challenge. Here, authors report an amide-directed, rhodium-catalyzed highly diastereo- and enantioselective hydroboration of unactivated internal alkenes. This method provided facile access to enantioenriched amines containing β,γ-vicinal stereocenters. The application of this strategy to the synthesis of bioactive mols. was demonstrated. Computational studies indicated that migratory insertion of the alkene into rhodium hydride controls the enantioselectivity.

In the two-step fermentative production of vitamin C, its precursor 2-keto-L-gulonic acid (2-KLG) was synthesized by Ketogulonicigenium vulgare through co-culture with Bacillus megaterium. The reconstruction of the amino acid metabolic pathway through completed genome sequence annotation demonstrated that K. vulgare was deficient in one or more key enzymes in the de novo biosynthesis pathways of eight different amino acids (L-histidine, L-glycine, L-lysine, L-proline, L-threonine, L-methionine, L-leucine, and L-isoleucine). Among them, L-glycine, L-proline, L-threonine, and L-isoleucine play vital roles in K. vulgare growth and 2-KLG production The addition of those amino acids increased the 2-KLG productivity by 20.4%, 17.2%, 17.2%, and 11.8%, resp. Furthermore, food grade gelatin was developed as a substitute for the amino acids to increase the cell concentration, 2-KLG productivity, and L-sorbose consumption rate by 10.2%, 23.4%, and 20.9%, resp. As a result, the fermentation period decreased to 43 h in a 7-L fermentor.

The nonmirror image relation between absorption and fluorescence spectra of conjugated polymers contrasts with most organic chromophores and is widely considered a signature of interchromopohore energy funneling. The authors apply broad-band ultrafast fluorescence spectroscopy to resolve the evolution of fluorescence spectra for dilute solutions of conjugated oligothiophenes, where no energy transfer is possible. Fluorescence spectra evolve from a mirror image of absorption, which lacks vibronic structure, toward a spectrally narrower and vibronically structured species on the hundreds of femtosecond to early picosecond time scale. The authors’ anal. of this fluorescence spectral evolution shows that a broad distribution of torsional conformers is driven to rapidly planarize in the excited state, including in solid films, which is supported by Raman spectroscopy and quantum chem. modeling. The authors’ data have important implications for understanding different energy-transfer regimes that are delineated by structural relaxation.

The master regulator of salicylic acid (SA)-mediated plant defense, NPR1 (NONEXPRESSER OF PR GENES 1) and its paralogs NPR3 and NPR4, act as SA receptors. After the perception of a pathogen, plant cells produce SA in the chloroplast. In the presence of SA, NPR1 protein is reduced from oligomers to monomers, and translocated into the nucleus. Several of these analogs can induce SA-mediated defense and inhibit growth of Pseudomonas syringae in Arabidopsis. These analogs, when sprayed on Arabidopsis, can induce the accumulation of the master regulator of plant defense NPR1. In a yeast two-hybrid system, these analogs can strengthen the interactions among NPR proteins. We demonstrated that these analogs can induce the expression of the defense marker gene PR1. Furthermore, we hypothesized that these SA analogs could be potent tools against the citrus greening pathogen Candidatus liberibacter spp. In fact, our results suggest that the SA analogs we tested using Arabidopsis may also be effective for inducing a defense response in citrus. Several SA analogs consistently strengthened the interactions between citrus NPR1 and NPR3 proteins in a yeast two-hybrid system. In future assays, we plan to test whether these analogs avoid degradation by SA hydroxylases from plant pathogens.
