With its origin estimated around Dec. 2019 in Wuhan, China, the ongoing SARS-CoV-2 pandemic is a major global health challenge. The demand for scalable, rapid and sensitive viral diagnostics is thus particularly pressing at present to help contain the rapid spread of infection and prevent overwhelming the capacity of health systems. While high-income countries have managed to rapidly expand diagnostic capacities, such is not the case in resource-limited settings of low- to medium-income countries. Aiming at developing cost-effective viral load detection systems for point-of-care COVID-19 diagnostics in resource-limited and resource-rich settings alike, we report the development of an integrated modular centrifugal microfluidic platform to perform loop-mediated isothermal amplification (LAMP) of viral RNA directly from heat-inactivated nasopharyngeal swab samples. The disks were pre-packed with dried n-benzyl-n-methylethanolamine modified agarose beads used to selectively remove primer dimers, inactivate the reaction post-amplification and allowing enhanced fluorescence detection via a smartphone camera. Sample-to-answer anal. within 1 h from sample collection and a detection limit of approx. 100 RNA copies in 10μL reaction volume were achieved. The platform was validated with a panel of 162 nasopharyngeal swab samples collected from patients with COVID-19 symptoms, providing a sensitivity of 96.6% (82.2-99.9%, 95% CI) for samples with Ct values below 26 and a specificity of 100% (90-100%, 95% CI), thus being fit-for-purpose to diagnose patients with a high risk of viral transmission. These results show significant promise towards bringing routine point-of-care COVID-19 diagnostics to resource-limited settings.

Vitamin D3 metabolites are capable of controlling gene expression in mammalian cells through two independent pathways: vitamin D receptor (VDR) and sterol regulatory element-binding protein (SREBP) pathways. In the present study, we dissect the complex biol. activity of vitamin D by designing synthetic vitamin D3 analogs specific for VDR or SREBP pathway, i.e., a VDR activator that lacks SREBP inhibitory activity, or an SREBP inhibitor devoid of VDR activity. These synthetic vitamin D probes permitted identification of one of the vitamin D-responsive genes, Soat1, as an SREBP-suppressed gene. The chem. probes developed in the present study may prove useful in dissecting the intricate interplay of vitamin D actions, thereby providing insights into how vitamin D target genes are regulated.

A new monomer, 2,4,6-tris(4-(1H-pyrrol-1-yl)phenoxy)-1,3,5-triazine (Tria-Py) was synthesized via the reaction of 4-(1H-pyrrol-1-yl)phenol with 2,4,6-trichloro-1,3,5-triazine. It was electrochem. polymerized in a dichloromethane/acetonitrile solvent mixture The polymer P(Tria-Py) was characterized via CV,FTIR,SEM and UV-vis spectroscopy. Spectroelectrochem. anal. revealed electronic transitions at 346, 508 and 665 nm, revealing π to π* transitions, and polaron and bipolaron band formations resp. We also made a dual-type, complementary-colored electrochromic device using P(Tria-Py)/poly(3,4-ethylenedioxythiophene) (PEDOT) in a sandwich configuration. The device switches between dark blue and red colors with a switching time of 2.2 s and an optical contrast (% ΔT) of 25%.

In this study, a new ferrocenyldithiophosphonate functionalized; [O-1-(4-phenyl)-1H-pyrrole ferrocenyldithiophosphonate (TPFc)] monomer and its conductive copolymers with pyrrole and 4-(2,5-di(thiophen-2-yl)-1H-pyrrol-1-yl)butane-1-amine were synthesized, characterized and their electrochromic properties were investigated. The spectroelectrochem. analyses demonstrated that the copolymer films reveal a reversible cycling with distinctive color changes in their reduced/oxidized forms. For the copolymer films P(TPFc-co-Py)/DCM, P(TPFc-co-Py)/CH3CN and P(TPFc-co-TPA)/DCM, the maximum optical contrasts (ΔT %) were measured as 28, 12 and 27% at 800, 900 and 900 nm, resp. Spectroelectrochem. anal. also revealed that all copolymers prepared with TPFc have lower band gaps compared to those of corresponding polymers.

A new method was developed for the in vitro sun protection factor (SPF) evaluation of sunscreen samples. A new type of substrate, a hydroxyalkyl cellulose-coated plate, was also prepared specifically for hydrophilic samples. This new substrate was required because hydrophilic samples would be unlikely to wet the surface of the standard cosmetic PMMA UV evaluation plate. A super-hydrophilic quartz plate was prepared by corona-discharge treatment before an aqueous solution of hydroxyalkyl cellulose was spread on it. A flat and uniform hydroxyalkyl cellulose film was subsequently formed through the evaporation of water. Special care was taken to inhibit the generation of spatial non-uniformity. Six hydrophilic sunscreen samples with in vivo SPF values of 56, 55, 52, 25, 15, and 4, were then applied to the prepared hydroxyalkyl cellulose-coated plate, as well as a super-hydrophilic quartz plate and a flat hydrophobic PMMA plate. The thicknesses of the applied layers were determined using a wheel-shaped wet film thickness gauge immediately after the application, and UV transmission was measured using an SPF analyzer. The value of in vitro SPF was calculated from the UV absorbance and the thickness of the layer. For two out of the six samples, PMMA plate could not be available, as the samples were unable to wet the PMMA surface. Relatively small differences were observed between the in vitro SPF values when the super-hydrophilic and hydroxyalkyl cellulose-coated plates were used. Samples exhibiting higher in vivo SPF were also associated with higher in vitro SPF values, although a linear relationship was not observed In contrast to the super-hydrophilic plate whose half-life of the super-hydrophilicity is only approx. five days, the hydrophilicity of the hydroxyalkyl cellulose-coated plate scarcely varied during six months of storage. Finally, a simplified evaluation method was also proposed. The validity of the method was verified through a ring test where three researchers employed this method in different laboratories at three independent organizations.

In this paper, we presented Focused Beam Reflectance Measurement (FBRM) studies applied in the case of the non-typical spherical crystallization methods of the active pharmaceutical ingredient ambroxol hydrochloride. Spherical agglomeration and controlled cooling crystallization methods were carried out based on our previous studies regarding these methods and the crystal formation processes were investigated with FBRM probe. Critical agglomeration periods and other mechanisms, such as the breakage of the agglomerates and the precipitation of small particles, were investigated. Chord length values were observed continuously and samples were taken out in certain points of the crystallization Particle size anal. was carried out in each case to obtain information on the particle size enlargement caused by the crystallization methods. With the application of light microscopic anal., aspect ratio and roundness values were also determined to obtain pieces of information on the sphericity-changes of the particles throughout the crystallization and large improvements were presented either for these parameters (aspect ratio from ∼ 1.7 to ∼ 1.3; roundness from ∼ 2.4 to ∼ 1.5) and the mean particle size (from ∼13μm to 120-400μm). Among others, it was revealed that after the spherical agglomeration method (SA), mostly crystal agglomerates were formed, while slow cooling crystallization with alternating temperature profile (ATP) yielded larger and probably harder individual crystals.

This study identifies the volatile organic compounds (VOCs) emitted by citrus when infested with California red scale (Aonidiella aurantii) and determines which of these elicit behavior responses in the predator Rhyzobius lophanthae. Headspace solid-phase micro extractions (HS-SPME) technique combined with gas chromatog.-mass spectrometry (GC-MS) was used to identify compounds and Y-tube olfactometer to determine R. lophanthae behavior responses. According to the results, 22 VOCs were detected in infested citrus plants and some of them were increased in lemon, orange and tangerine by A. aurantii infestation. R. lophanthae individuals were attracted to infested citrus saplings. According to bioassays with the olfactometer, they were attracted to Me salicylate and D-limonene at dosages of 1 and 10μL/mL by using a Y-tube olfactometer. These results explain how citrus volatiles can affect the response of the predator R. lophanthae.
