In this work, we studied formation of free radicals during the decomposition of several water-soluble azo compounds by 50 kHz ultrasound in aqueous solutions Using the spin trp 3,5-dibromo-4-nitrosobenzene sulfonate (DBNBS) tertiary carbon-centered radicals from 2,2′-azobis (N,N’-bimethyleneisobutyramidine) dihydrochloride (VA-044), 2-(carbamoylazo)isobutyronitrile (V-30), and 2,2′-azobis(2-amidinopropane) dihydrochloride (AAPH) and •CH3 radicals from 1,1′-azobis(N,N’-dimethylformamide) (ADMF) were detected in argon-saturated solutions and the corresponding oxygen-centered radicals (alkoxyl and peroxyl) from VA-044, V-30, and AAPH were identified using the spin trap 5,5′-dimethyl-1-pyrroline-N-oxide (DMPO) in aerated sonicated solutions No free radicals from 4,4′-dihydroxyazobenzene-3,3′-dicarboxylic acid, disodium salt (DHAB) could be found in either system. While VA-044 and AAPH could also be readily decomposed by heat (42.5 °C and 80 °C), V-30 decomposition only occurred in the ultrasound-exposed solutions The most likely mechanism of decomposition of azo compounds by ultrasound is their thermolysis in the heated shell of the liquid surrounding cavitating bubbles driven by ultrasound and/or by pyrolysis inside these bubbles. Here, we show that D-mannose suppresses LPS-induced macrophage activation by impairing IL-1β production In vivo, mannose administration improves survival in a mouse model of LPS-induced endotoxemia as well as decreases progression in a mouse model of DSS-induced colitis. Phosphomannose isomerase controls response of LPS-activated macrophages to D-mannose, which impairs glucose metabolism by raising intracellular mannose-6-phosphate levels. Such alterations result in the suppression of succinate-mediated HIF-1α activation, imposing a consequent reduction of LPS-induced Il1b expression. Disclosing an unrecognized metabolic hijack of macrophage activation, our study points towards safe D-mannose utilization as an effective intervention against inflammatory conditions.

This study assessed PoIyPEPI1018 combined with first-line maintenance therapy in patients with MSS mCRC. Patients and Methods: Eleven patients with MSS mCRC received PolyPEPI1018 and Montanide ISA51VG adjuvant s.c., combined with tluoropyrimidine/biol. following first-line induction with chemotherapy and a biol. (NCT03391232). In Part A of the study, 5 patients received a single dose; in Part B, 6 patients received up to three doses of Poly- PEPI1018 every 12 wk. The primary objective was safety; secondary objectives were preliminary efficacy, immunogenicity at peripheral and tumor level, and immune correlates. PolyPEPI1018 vaccination was safe and well tolerated. No vaccine-related serious adverse event occurred. Eighty percent of patients had CD8+ T-cell responses against ≥3 TAAs. Increased d. of tumor-infiltrating lymphocytes were detected post-treatment for 3 of 4 patients’ liver biopsies, combined with increased expression of immune-related gene signatures. Three patients had objective response according to REClSTv1.1, and 2 patients qualified for curative surgery. Longer median progression-free survival for patients receiving multiple doses compared with a single dose (12.5 vs. 4.6 mo; P = 0.017) suggested a dose-efficacy correlation. The host HLA genotype predicted multi-antigen specific T-cell responses (P = 0.01) indicative of clin. outcome. PolyPEPI1018 added to maintenance chemotherapy for patients with unresectable, MSS mCRC was safe and associated with specific immune responses and antitumor activity warranting further confirmation in a randomized, controlled setting.

This study was carried out to study the effects of selenylation on physicochem. and biol. properties of polysaccharide (SPP) extracted from Sargassum pallidum. The selenized derivative of SPP (Se-SPP) with the selenium content of 2419 μg/g was synthesized by sodium selenite/dilute nitric acid method. Physicochem. characterization indicated that selenylation modification resulted in some changes in chem. composition, monosaccharide composition, mol. weight and surface morphol. of polysaccharides. FT-IR spectroscopy showed that a new absorption peak appeared at 675 cm-1 in Se-SPP probably due to the substitution of selenyl groups. Bioactivity assay showed that Se-SPP exhibited higher scavenging radical activities and ferrous ion chelating activities than native SPP. Compared with SPP and acarbose, Se-SPP showed more significantly inhibitory effect on α-glucosidase activity in a noncompetitive inhibition type. The IC50 values of SPP, Se-SPP and acarbose were determined as 1.579, 0.896 and 2.742 mg/mL, resp. These results suggest that Se-SPP can be used to develop a new selenium-complementary ingredient in functional foods.

Dehydrogenative coupling-based reactions have emerged as an efficient route toward the synthesis of a plethora of heterocyclic rings. Herein, we report an efficacious, nickel-catalyzed synthesis of two important heterocycles such as quinoline and quinoxaline. The catalyst is molecularly defined, is phosphine-free, and can operate at a mild reaction temperature of 80°C. Both the heterocycles can be easily assembled via double dehydrogenative coupling, starting from 2-aminobenzyl alc./1-phenylethanol and diamine/diol, resp., in a shorter span of reaction time. This environmentally benign synthetic protocol employing an inexpensive catalyst can rival many other transition-metal systems that have been developed for the fabrication of two putative heterocycles. Mechanistically, the dehydrogenation of secondary alc. follows clean pseudo-first-order kinetics and exhibits a sizable kinetic isotope effect. Intriguingly, this catalyst provides an example of storing the trapped hydrogen in the ligand backbone, avoiding metal-hydride formation. Easy regeneration of the oxidized form of the catalyst under aerobic/O2 oxidation makes this protocol eco-friendly and easy to handle.
