Herein, the first demonstrations of [2 + 2 + 1] cycloaddition reactions was reported that allow the facile synthesis of isoxazolines I [R1 = cyclopropyl, Ph, 2-naphthyl, etc.; R2 = CH2OH, COOEt, Ph, etc.], employing N-tosylhydrazones, tert-Bu nitrite (TBN) and alkenes as reactants. This process represented a new type of cycloaddition reaction with a distinct mechanism that does not involve the participation of nitrile oxides. This approach was both general and practical and exhibits a wide substrate scope, nearly universal functional group compatibility, tolerance of moisture and air, the potential for functionalization of complex bioactive mols. and is readily scaled up. Both control experiments and theor. calculations indicated that this transformation proceeds via the in situ generation of a nitronate from the coupling of N-tosylhydrazone and TBN, followed by cycloaddition with an alkene and subsequent elimination of a tert-butyloxy group to give the desired isoxazoline.

(MLSA) using 16S rRNA, rpoB, gyrB and rpoD concatenated sequences revealed that the closest relatives of FIT28T are Pseudomonas zeae OE48.2T, ‘Pseudomonas crudilactis’ UCMA 17988, Pseudomonas tensinigenes ZA5.3T, Pseudomonas helmanticensis OHA11T , Pseudomonas baetica a390T, Pseudomonas iridis P42T, Pseudomonas atagonensis PS14T and Pseudomonas koreensis Ps 9-14T, within the Pseudomonas koreensis subgroup of the Pseudomonas fluorescens lineage. The genome size of FIT28T is about 6.7Mb with 59.09 mol% DNA G+C content. Average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values calculated from the genomic sequences of FIT28T, and the closely related P. zeae OE48.2T are 95.23 and 63.4%, resp. Biochem., metabolic and chemotaxonomic studies further support our proposal that Pseudomonas germanica sp. nov., should be considered a novel species of the genus Pseudomonas. Hence, the type strain FIT28T (=LMG 32353T =DSM 112698T) has been deposited in public cell-type culture centers.

TP0446131, developed as an antidepressant agent, was found to cause lenticular opacity in a 13-week repeated-dose study in dogs. Histopathologically, the lenticular opacity was observed as a degeneration of the lens fibers, characterized by irregularity in the ordered arrangement of the fibers which is necessary to maintain the transparency of the lens, and was considered to manifest clinically as cataract. To evaluate the development mechanism of the lenticular opacity, the chemical constituents of the lens, which is known to be associated with the development of cataract, were examined. The results of liquid chromatography-tandem mass spectrometry analysis revealed an increase in the amplitudes of 3 unknown peaks in a dose- and time-dependent manner in the lens, with no remarkable changes in the other chemical components tested. In addition, the content of cholesterol, alterations of which have been reported to be associated with cataract, remained unchanged. The mass spectral data and chromatographic behavior of the 3 peaks indicated that these peaks corresponded to sterol-related substances, and that one of them was 7-dehydrocholesterol, a precursor of cholesterol biosynthesis. This finding suggested that TP0446131 exerts some effects on the cholesterol biosynthesis pathway, which could be involved in the development of the cataracts. Furthermore, increases in the levels of these sterol-related substances were also detected in the serum, and were, in fact, noted prior to the onset of the cataract, suggesting the possibility that these substances in the serum could be used as potential safety biomarkers for predicting the onset of cataract induced by TP0446131.

Addnl., we have identified and optimized multiple potent new series of SC4MOL and HSD17B7 inhibitors and shown that these small mols. enhance oligodendrocyte formation. SC4MOL inhibitor CW4142 induced accumulation of SC4MOLs sterol substrates in mouse brain and represents an in vivo probe of SC4MOL activity. Mechanistically, the cellular accumulation of these 8,9-unsaturated sterols represents a central driver of enhanced oligodendrocyte formation, as exogenous addition of purified SC4MOL and HSD17B7 substrates but not their 8,9-satd analogs promotes OPC differentiation. Our work validates SC4MOL and HSD17B7 as novel targets for promoting oligodendrocyte formation, underlines a broad role for 8,9-unsaturated sterols as enhancers of oligodendrocyte formation, and establishes the first high-quality small mols. targeting SC4MOL and HSD17B7 as novel tools for probing diverse areas of biol.

Recently, we opened a synthetic access to antitumoral platinum complexes of the type cis-[(NHC)1(NHC)2PtIICl(L)] which interact with DNA in a way correlated to the complex charge and to the sterical accessibility of the leaving chlorido ligand. We now identified mitochondria rather than nuclei as the cellular target of the neutral dichlorido complex 1 (L = Cl) and the delocalized lipophilic cationic phosphine complex 2 (L = PPh3), both carrying the same cis-bis(1,3-dibenzylimidazol-2-ylidene) ligands. Their uptake into 518A2 melanoma cells was concentration-dependent and distinctly greater for complex 2 which was also more cytotoxic against sensitive cancer cell lines with submicromolar IC50 values. Both complexes interfered strongly with various forms of DNA in vitro, but only complex 2 caused a melanoma cell cycle arrest in G1-phase, setting both apart from the S-phase arresting drug cisplatin. Studies of the intracellular localisation of 1 and 2 were carried out with their alkyne-tagged analogs 6 and 7, which showed identical patterns of cancer cell cytotoxicity, cell cycle interference and effects on mitochondria. Click reactions with 7-hydroxycoumarin azide, colocalisation with Mitotracker and confocal microscopy, proved complexes 6 and 7 to accumulate mainly in the mitochondria rather than the nuclei of melanoma cells. Complex 1 and even more so complex 2 reduced the mitochondrial membrane potential and also increased the cellular ROS levels. As a consequence, both complexes caused stress fiber formation in the F-actin cytoskeleton of melanoma cells, most distinctly so complex 2 which also activated the apoptotic cascade mediated by capases-3 and -7.

Objectives: This study aims to investigate the prevalence of vitamin D deficiency in Chinese centenarians and to identify the factors associated with vitamin D deficie. Methods: Details on sociodemographics and lifestyle characteristics were collected using a structured questionnaire. Anthropometrics and blood samples were obtained. Vitamin D deficiency was defined as serum 25-hydroxyvitamin D concentration < 20ng/mL (50nmol/L). The overall prevalence of vitamin D deficiency among these 943 centenarians was 39.9% (376 subjects) and the mean serum vitamin D concentrations were 22.7±9.5 (ng/mL). Female centenarians had higher prevalence of vitamin D deficiency than male (44.0% vs. 21.7%, P < 0.001). Multivariate logistic regression analyses showed that being female, urban residency, lower body mass index, higher serum parathyroid hormone levels, no fish consumption, and less sun exposure time were all significant and independent determinants of vitamin D deficiency. No significant associations of vitamin D deficiency with ethnic, education, geog. location, tea drinking, alc. use, or smoking were found in this study. Conclusion: Vitamin D deficiency was common in Chinese centenarians, especially in women. Given that vitamin D deficiency is linked to numerous adverse health outcomes, dietary, outdoor activities and other intervention measures are needed to correct vitamin D deficiency in this population. This is a retrospective study of 23 patients from 2002-2020 over 18 years. The patients were evaluated on the basis of their age, obstetric history and the treatment received. All the patients were scored according to Modified World Health Organization (WHO) prognostic scoring system as adapted by FIGO Year 2000. Low risk group patients were given injection Methotrexate + Calcium Leucovorin and high risk group were given EMACO regimen. After treatment, the patient follow up was till date or min. up to 2 years. The response to treatment was evaluated by regular clin. and radiol. examination and serum β-hCG level estimation Three out of 23 patients who relapsed after primary chemotherapy were given second line chemotherapy. All three patients achieved complete remission (CR) after salvage chemotherapy. Thus overall response rate is 100%. Proper diagnosis, close monitoring and follow up with β-hCG value is of utmost importance in the management of GTN, WHO/FIGO scoring should be done and managed with chemotherapy treatment according to the risk assessment.
