A review. Neurol. disorders have been growing in recent years and are highly prevalent globally. Resveratrol (RES) is a natural product from plant sources such as grape skins. This compound has shown biol. activity in many diseases, in particular, those that act on the central nervous system. The mechanism of action and the key points in neurol. disorders were described and show the targeted mechanism of action. Due to the insolubility of this compound; the use of nanotechnol.-based systems has been proposed for the incorporation of RES and RES-loaded nanocarriers have been designed for intranasal administration, oral or parenteral routes to deliver it to the brain. In general, these nanosystems have shown to be effective in many studies, pharmacol. and pharmacokinetic assays, as well as some cell studies. The outcomes show that RES has been reported in human clin. trials for some neurol. diseases, although no studies were performed in humans using nanocarriers, animal and/or cellular models have been reported to show good results regarding therapeutics on neurol. diseases. Thus, the use of this nutraceutical has shown true for neurol. diseases and its loading into nanocarriers displaying good results on the stability, delivery and targeting to the brain.

A novel rod-shaped, Gram-stain-neg., strictly aerobic and alginate-degrading marine bacterium, designated CCB-QB4T, was isolated from a surface of algal turf collected from a coastal area of Penang, Malaysia. The cells showed motility by a lateral flagellum. The rod-shaped cells formed long chains end-to- end. Phylogenetic anal. based on the 16S rRNA gene sequence of strain CCB-QB4T showed 94.07, 92.69, 91.52 and 90.90% sequence similarity to Algibacillus agarilyticus RQJ05T, Catenovulum maritimum Q1T, Catenovulum agarivorans YM01T and Catenovulum sediminis D2T, resp. Strain CCB-QB4T formed a cluster with A. agarilyticus RQJ05T. Strain CCB-QB4T was catalase-neg., oxidase-pos., and degraded agar, alginate, and starch. Cell growth was observed at 15-40°C, at pH 7.0-10.0 and in the presence of 1-6% (w/v) NaCl and glucose. The major fatty acids were summed feature 3 (C16:1 ω7c/iso-C15:0 2-OH), C16:0 and C18:1 ω7c. The polar lipids were phosphatidylethanolamine, two unidentified aminolipids, two unidentified glycolipids, an unidentified phospholipid and unidentified lipid. The major respiratory quinone was ubiquinone-8. The genomic DNA G+C content was 46.7 mol%. Based on the phenotypic, chemotaxonomic and phylogenetic data, strain CCB-BQ4T represents a novel species in a new genus, for which the name Saccharobesus litoralis gen. nov., sp. nov. is proposed. The type strain is CCB-QB4T (=JCM 33513T=-MBL 5008T).

The metabolic oligosaccharide engineering (MOE) strategy using unnatural sialic acids has recently enabled the visualization of the sialome in living systems. However, MOE only reports on global sialylation and dissected information regarding subsets of sialosides is missing. Described here is the synthesis and utilization of sialic acids modified with a sydnone reporter for the metabolic labeling of sialoconjugates. The positioning of the reporter on the sugar significantly altered its metabolic fate. Further in vitro enzymic assays revealed that the 9-modified neuraminic acid is preferentially accepted by the sialyltransferase ST6Gal-I over ST3Gal-IV, leading to the favored incorporation of the reporter into linkage-specific α2,6-N-linked sialoproteins. This sydnone sugar presents the possibility of investigating the roles of specific sialosides.

Glucose electrooxidation to glucaric acid, a highly valuable platform chem., was conducted, for the 1st time, in two consecutive steps employing a bare Au electrode in a batch reactor. The 1st step, at low potential, enabled the effective formation of the intermediate, gluconic acid, promoted by the oxidation of the aldehyde group on C1. For this reaction step, except for the reaction time, all the operational parameters had a strong impact on the selectivity. At the optimal conditions of pH 11.3, 5° and 0.04M initial glucose, a maximum selectivity of 97.6% was obtained, the highest reported to date for the electrochem. process, with a conversion of 25%. At higher potential, gluconic acid was further converted to glucaric acid by the oxidation of the hydroxymethyl group on C6. For this oxidation step, the variation of the operational parameters (pH, gluconic acid initial concentration, reaction temperature and time) had no conspicuous effect on the selectivity, while the applied potential had a major role: at 1.1 VRHE, a maximum selectivity of 89.5% to glucaric acid was obtained, which is one of the highest values reported in literature so far. In all experiments, irresp. of conditions and reaction time, a maximum concentration of ∼1.2 mM of glucaric acid was achieved, and a drastic decrease of the c.d. was observed in the 1st hours of electrolysis. Addnl. control experiments revealed a poisoning process caused by glucaric acid that, once formed, remains strongly bonded to the catalyst’s active sites, thus deactivating it.

Stimulation by NaCl or KCl of the ABA-induced increase in proline was synergistically enhanced by CaCl2 or MgCl2 as well as by 1,3-bis[tris(hydroxymethyl)methylamino]propane chloride (BTP-Cl), N-methyl-D-glucamine chloride (NMG-Cl), or 2-amino-2-hydroxymethyl-1,3-propandiol chloride (TRIS-Cl). This enhancing effect did not depend on the osmolarity and occurred when Cl- was higher than K+ in the incubation medium, but not vice versa. When CaCl2 or MgCl2 or NMG-Cl were added, the higher the Cl-:K+ ratio in the external solution the higher was the increase in proline. When the excess of Cl- to K+ was obtained with BTP-Cl the highest enhancing effect resulted with a Cl-:K+ ratio of 3:1 while, at a 5:1 ratio, the KCl stimulation was completely suppressed. The inhibiting effect of proline accumulation by NH4+ and 4,4′-diisothiocyano-2,2′-disulfonicacid stilbene was reversed to varying degrees depending on the magnitude of the excess of Cl- on K+ concentration in the medium. Also, the inhibition of proline accumulation obtained by tetraethylammonium chloride, monensin, and D-mannose was similarly reverted. Thus, Cl- elicits an increase in ABA-induced proline which requires the simultaneous presence of K+ (or Na+).

The gray-headed flying fox (Pteropus poliocephalus) is an endemic Australian fruit bat, known to carry zoonotic pathogens. We recently showed they harbor bacterial pathogen Klebsiella pneumoniae and closely related species in the K. pneumoniae species complex (KpSC); however, the dynamics of KpSC transmission and gene flow within flying fox colonies are poorly understood. High-resolution genome comparisons of 39 KpSC isolates from gray-headed flying foxes identified five putative strain transmission clusters (four intra- and one inter-colony). The instance of inter-colony strain transmission of K. africana was found between two flying fox populations within flying distance, indicating either direct or indirect transmission through a common food/water source. All 11 plasmids identified within the KpSC isolates showed 73% coverage (mean) and ≥95% identity to human-associated KpSC plasmids, indicating gene flow between human clin. and gray-headed flying fox isolates. Along with strain transmission, inter-species horizontal plasmid transmission between K. pneumoniae and Klebsiella africana was also identified within a flying fox colony. Finally, genome-scale metabolic models were generated to predict and compare substrate usage to previously published KpSC models, from human and environmental sources. These models indicated no distinction on the basis of metabolic capabilities. Instead, metabolic capabilities were consistent with population structure and ST/lineage.
