The study of bioactive mols. of natural origin is a focus of current research. Thymus algeriensis and Artemisia herba-alba are two medicinal plants widely used by the Moroccan population in the traditional treatment of several pathologies linked to inflammation. This study aimed to evaluate the single and combined antioxidant, anti-inflammatory and analgesic effects of the essential oils extracted from these two medicinal plants, and also their potential toxicity. Essential oils were extracted using hydro-distillation in a Clevenger-type apparatus The antioxidant activity was evaluated by two methods: the scavenging of the free radical DPPH, and the reduction in iron. Anti-inflammatory activity was evaluated by evaluating the edema development induced by carrageenan injecting, while the analgesic power was evaluated according to the number of abdominal contortions induced by the i.p. injection of acetic acid (0.7%). The acute oral toxicity was performed to assess the potential toxicity of the studied EOs, followed by an anal. of the blood biochem. parameters. The results of the two antioxidant tests indicated that our extract mixture exhibits good iron reduction capacity and very interesting DPPH free radical scavenging power, with an IC50 of around 4.38 ± 0.98 μg/mL higher than that of the benchmark antioxidant, BHT. The anti-inflammatory test demonstrated that the mixture administered orally at a dose of 150 mg/kg has a better activity, exceeding that of 1% Diclofenac, with a percentage of maximum inhibition of the edema of 89.99 ± 4.08. The number of cramps in the mice treated with the mixture at a dose of 150 mg/kg is significantly lower (29.80 ± 1.92) than those of the group treated with Tramadol (42.00 ± 2.70), resp. The toxicity results show no signs of toxicity with an LD50 greater than 150 mg/Kg. These interesting results show that the two plants′ EOs had an important anti-inflammatory, analgesic, and antioxidant activity, and also a powerful synergistic effect, which encourages further in-depth investigations on their pharmacol. proprieties.

As a class of promising green media, natural deep eutectic solvents (NADESs) have attracted more and more attention in recent years. In this paper, a novel method based on microwave-assisted natural deep eutectic solvents pretreatment coupled with microwave hydrodistillation (MA-NADES-MHD) was used for the extraction of essential oil from Cuminum cyminum L. seed. NADESs were used as pretreatment solvents for adsorbing microwaves and dissolving cellulose. The maximum yield of essential oil obtained with this three-stage microwave extraction technol. was 2.22% (weight/weight) when the natural deep eutectic solvent composed of choline chloride and L-lactic acid (with 1:3 of molar ratio) with 40% (weight/weight) of water. Compared with microwave hydrodistillation (MHD) and ultrasound-assisted natural deep eutectic solvents pretreatment combined with microwave hydrodistillation (UA-NADES-MHD) in which essential oil yield of 1.13% (weight/weight) and 1.57% (weight/weight) were obtained, resp., the MA-NADES-MHD naturally brought more essential oil. Notably, a total of 45, 48, and 58 volatile components were identified by using gas chromatog.-mass spectrometry with MHD, UA-NADES-MHD, and MA-NADES-MHD, resp. Hence, NADESs had a significant impact on the extraction of essential oil with higher yield, premium quality, and more quantity, especially when it was combined with microwave technol. Moreover, the MA-NADES-MHD as an economical and eco-friendly technique had the potential to be applied to other plant materials.

The physicochem. properties of Holothuria leucospilota polysaccharides (HLP) were measured, and the changes of mol. weight (Mw), antioxidant activity and microstructure in vitro were studied. The Mw of HLP was approx. 52.80 kDa, and the monosaccharide composition was primarily fucose, rhamnose and glucuronic acid. HLP showed good antioxidant activity at high concentration (≥2 mg/mL), and the DPPH (1,1-Diphenyl-2-picryl-hydrazil) free radical scavenging rate reached approx. 84%. It was observed that saliva did not have an effect on the Mw and antioxidant capability of HLP. Gastric and intestinal digestion significantly reduced the Mw of HLP (from 52.80 kDa to 14.67 kDa and 12.01 kDa) and maintained high antioxidant activity. In addition, the structure and surface morphol. were markedly altered. After simulated gastric digestion, the HLP changed from a smooth, irregular, and angular structure to a layered honeycomb with a large volume This result demonstrated that the decrease of Mw after gastric digestion was caused by the destruction of aggregates. Due to the decomposition of glycosidic bonds after digestion in intestine, HLP showed loose, small and blocky structures. Thus, HLP had antioxidant activity, and this activity was improved after gastric and intestinal digestion.

A polysaccharide was extracted in high yield from tamarind (Tamarindus indica L.) seed (TSP) by acidic hot water extraction and ethanol precipitation It was composed of 86.2% neutral polysaccharide, 5.4% uronic acid and 1.3% protein. The mol. weight of TSP was estimated to be about 1735 kDa, with glucose, xylose, and galactose in a molar ratio of 2.9:1.8:1.0 as the major monosaccharides. The steady shear and viscoelastic properties of TSP aqueous solutions were investigated by dynamic rheometry. Results revealed that TSP aqueous solution at a concentration above 0.5% (w/v) exhibited non-Newtonian shear-thinning behavior. Dynamic oscillatory anal. revealed that 10% (w/v) TSP showed as a “”weak gel”” structure. Apparent viscosities and viscoelastic parameters of TSP solutions decreased drastically in an alk. solution of pH > 10, but slightly influenced by acidic solution, high temperature and the presence of salt ions and sucrose. These results indicated that TSP possessed excellent pH-resistance and thermo-stability, which might be suitable for applications in acidic beverages and high-temperature processed foodstuffs.

Ultrasonic-assisted enzymic extraction (UAEE) technol. was employed to extract the polysaccharides from dandelion leaves (DLP). The extraction conditions were optimized by response surface method (RSM) and Box-Behnken design (BBD). The polysaccharide yield (14.05 ± 0.95%), which well matched with the predicted value of 14.27%, was obtained under the optimum parameters: ultrasound power 500 W, ultrasound time 42 min, cellulase amount 4.90%, and particle size 80 mesh. After purification by AB-8 Macroporous Resin and Sephadex G-100 column, a novel polysaccharide (DLP-I) was isolated and its mol. weight was estimated to be 87,000 g/mol by high performance liquid chromatog. (HPLC). Gas chromatog. (GC) anal. showed that DLP-I was composed of rhamnose, arabinose, mannose, galactose and glucose in the molar ratio of 1.14:1.00:1.05:4.76:1.52. 1D and 2D NMR spectra confirmed that the backbone of DLP-I was mainly consisted of →4)-α-D-Galp-(1→, →4)-β-D-Manp-(1→, →4)-α-D-Glcp-(1→, →2,4)-α-L-Rhap-(1→, and α-L-Araf-(1→ with branching at O-2 and O-4 of →2,4)-α-L-Rhap-(1→. In addition, the digestion results showed DLP-I could keep its stability during the simulated digestive juices.

Two polysaccharides, DGS1 and DGS2, were obtained by solid-state fermentation (SSF) of Fusarium solani DO7, an endophytic fungus isolated from the orchid Dendrobium officinale. Structural characterizations revealed that DGS1 consisted of arabinose, glucose, mannose and galactose with a molar ratio of 2.9:13.4:3.0:1, resp., and contained (1 → 5)-Araf, (1 → 4)-Glcp, (1 → 6)-Glcp, (1 → 3)-Manp, (1 → 2,6)-Manp and (1 → 6)-Galp glycosidic linkages, while DGS2 was composed of arabinose, glucose, mannose and galactose in a molar ratio of 3.5:8.1:2.1:1, resp., and contained (1 → 5)-Araf, (1 → 4)-Glcp, (1 → 6)-Glcp, (1 → 3)-Manp and (1 → 6)-Galp glycosidic linkages. Neither polysaccharide was toxic to human embryonic kidney cells or mouse RAW 264.7 macrophage cells. An immunomodulatory activity assay indicated that both polysaccharides could significantly enhance the levels of TNF-α, IL-6 and NO by activating TNF-α, IL-6 and iNOs gene expression, resp., especially DGS2. Interestingly, DGS2 also possessed relatively high antioxidant activity. These results illustrate that, due to its cost-effectiveness and environmentally friendly features, SSF has significant potential as a com. competitive source of natural products, including fungal polysaccharides, with immunomodulatory activity.

The fusion points and fusion enthalpies of borneol and isoborneol were determined by differential scanning calorimetry. The Apelblat equation, Wilson model, and NRTL model were employed to correlate the measured solubility data, and the NRTL model fits the measured data better. Mixing properties of each solute-solvent pair are calculated by the NRTL model.

A water-soluble acidic polysaccharide containing 3-O-Me galactose, named YB-PS4, was isolated from the twigs and leaves of Juniperus tibetica Kom. Its structure was characterized by monosaccharide composition anal., methylation, and NMR spectroscopy. It was concluded that YB-PS4 had a backbone composed of (2,4)-a-Rhap-(1, 3,5)-a-Araf-(1, 2,4)-a-Galp-(1 and 4)-a-GalpA-(1, with branches of 2)-a-Rhap-(1,3)-a-Araf-(1 and 2)-3-O-Me-a-Galp-(1). The possible repetitive units were speculated and further analyzed by oligosaccharide anal. YB-PS4 showed inhibitory effects on complement activation through the classical pathway (CH50 = 94.23 ± 8.90 mug/mL) and alternative pathway (AP50 = 194.76 ± 9.20 mug/mL). Preliminary mechanism study indicated that it interacted with C1q, C2, C3, C4 and C5. These studies pointed a way to understand the active constituents of J. tibetica and provided scientific bases for YB-PS4 as a complement inhibitor.

The anal. hierarchy process (AHP) combined with response surface methodol. (RSM) was used to optimize the purification conditions of crude polysaccharides from Carex meyeriana Kunth (CMKP) by macroporous resin. The optimal purification conditions as follows: elution volume of 2.74 BV, flow rate of 1.88 BV/h, and sample concentration of 2.10 mg/mL. Under the optimal conditions, the purification comprehensive score was 63.59 ± 1.56% (n = 3), which was agreed closely to the predicted value (64.02%). Two fractions of polysaccharides (CMKP-1 and CMKP-2) were obtained using a DEAE-52 column. Fourier Transform IR (FTIR) spectroscopy spectra revealed the general characteristic absorption peaks of CMKP-1 and CMKP-2. Congo-red test indicated that CMKP-1 and CMKP-2 have triple helix conformation. CMKP-1 and CMKP-2 were both composed of Man, Rib, Rha, GluA, GalA, Glu, Gal, Xyl, Ara and Fuc in a molar ratio of 8.93:1:4.62:1.41:1.78:10.7:24.69:4.51:15.92:1.27 and 7.85:1:5.05:1.99:3.15:12.47:15.41:1.54:6.91:3.15, resp. The major mol. weight of CMKP-1 and CMKP-2 was 22.30 kDa and 48.10 kDa, resp. Moreover, CMKP-1 and CMKP-2 had immunostimulatory effects on RAW264.7 cells in a certain concentration range. It could be used as potential immunomodulatory agents in medicine or functional food fields.

In this study, three strains of lactic acid bacteria isolated from Tibetan kefir grains, including two strains of Lactobacillus pentosus LZ-R-17 and L. helveticus LZ-R-5, and one strain of Lactococcus lactis subsp. lactis LZ-R-12. The ability of three strains to produce exopolysaccharide (EPS) was tested, and L. pentosus LZ-R-17 was found to have the highest EPS yield. One EPS (R-17-EPS) was isolated from the fermented milk by L. pentosus LZ-R-17 and purified by DEAE-52 anion exchange chromatog. Furthermore, R-17-EPS preliminary structure and macrophage immunomodulatory activity in vitro were investigated. On the basis of the anal. results of UV-visible spectrum, Fourier transform-IR spectrum, monosaccharide composition anal. and one-dimensional and two-dimensional NMR (NMR) spectra, R-17-EPS was found to have an average mol. weight of 1.20 x 106 Da and was composed of galactose and glucose residues with a molar ratio of 1.00:3.15. NMR anal. revealed that the R-17-EPS was a linear hetero-galactoglucan containing repeating units of →2)-α-D-Galp-(1 → 4)-β-D-Glcp-(1 → 4)-β-D-Glcp-(1 → 4)-β-D-Glcp-(1→. In addition, R-17-EPS could effectively enhanced the proliferation, phagocytosis, nitric oxide and cytokines production of RAW264.7 cells, suggesting that R-17-EPS had potent immunostimulatory activity and could be explored as immunomodulator in functional food and/or medicine fields.
