Plants emit a specific blend of volatiles in response to herbivory and these volatiles, which often attract predators and parasitoids function as an indirect plant defense. The impact of plant volatiles in shaping herbivore defenses is unclear. Here, we report that specific plant volatiles induce immune responses in the polyphagous herbivore, Spodoptera litura. We characterized the hemocyte profile and established their functional significance with respect to ontogeny and exposure to specific plant volatiles. Fifth instar larvae showed the highest number and hemocytes diversity. We characterized seven different types of hemocytes, of which granulocytes performed phagocytosis, oenocytoids showed melanization activity, and plasmatocytes along with granulocytes and oenocytoids were found to be involved in encapsulation. Among the six volatiles tested, exposure to (E)-β-ocimene caused the highest increase in total hemocytes number (THC) followed by linalool and (Z)-3-hexenyl acetate exposure. Although THC did not differ between these three volatile treatments, circulating hemocytes diversity varied significantly. (E)-β-ocimene exposure showed higher number of plasmatocytes and phenol oxidase activity. The interaction of the parasitic wasp Bracon brevicornis with (E)-β-ocimene exposed larvae was poor in terms of delayed paralysis and lower egg deposition. In choice assays, the wasp showed clear preference towards control larvae indicating (E)-β-ocimene treatment renders the host unattractive. Hemocyte profiles post-parasitoid exposure and (E)-β-ocimene treatment were similar indicating cue-based priming. When challenged with Bacillus thuringiensis, linalool exposure resulted in the highest survival as compared to other volatiles. Our results show that specific HIPVs can modulate cellular immunity of S. litura, revealing a new role for HIPVs in tri-trophic interactions.

Two magnetic nano catalysts of nickel and copper, Fe3O4@SiO2@DOP-BenPyr-M(II), (M=Ni and Cu) have been synthesized. These catalysts were applied as recoverable, efficient and new heterogeneous catalysts for the high yielding and room temperature one-pot procedure of selective oxidation of sulfides to sulfoxides and oxidative coupling of thiols to disulfides. In addition, the catalytic activity of Fe3O4@SiO2@DOP-BenPyr-Ni(II) was investigated as heterogeneous nanocatalyst for synthesis of 2,3-dihydroquinazolin-4(1H)-ones, 5-substituted 1H-tetrazoles and polyhydroquinolines. The synthesized catalysts were characterized by FT-IR, TGA, XRD, VSM, EDX, ICP and SEM techniques. These catalysts were recovered by an external magnet and reused several times without significant loss of catalytic efficiency.

This is the first computational study that maps the conformational change to the preferential hydrogen bond donating capacity of one of the secondary hydroxyl groups of CD, in consensus with an NMR experiment We have developed a simple and novel geometry-based technique to identify water mols. occupying the nonspherical CD cavity, and the computed water occupancies are in close agreement with the exptl. and d. functional theory studies. Our anal. reveals that a water mol. in CD cavity loses out about two hydrogen bonds and remains energetically frustrated but possesses higher orientational degree of freedom compared to bulk water. In the context of CD-drug complexation, these imply a nonclassical, i.e., enthalpically driven hydrophobic association of a drug in CD cavity.

The purpose of this study was to investigate antibacterial activity of essential oil from Cinnamomum camphora var. linaloofera Fujita (EOL) at vapor phase and its mechanism of bactericidal action against Escherichia coli. Results showed that the vapor-phase EOL had significant antibacterial activity with a min. inhibitory concentration (MIC) and min. bactericidal concentration (MBC) of 200μl/L. Further analyses showed that treatment of E. coli with vapor-phase EOL resulted in partial degradation of cell membrane, increased membrane permeability, leakage of cytoplasm materials, and prominent distortion and shrinkage of bacterial cells. FTIR showed that EOL altered bacterial protein secondary and tertiary structures. GC/MS anal. showed that the components of vapor-phase EOL included linalool (69.94%), camphor (10.90%), nerolidol (10.92%), and safrole (8.24%), of which linalool had bactericidal activity. Quantum chem. anal. suggested that the antibacterial reactive center of linalool was oxygen atom (O10) which transferred electrons during antibacterial action by the donation of electrons.

Various industries highly regard the functionalities and bioactivities of bacterial polysaccharides. We aimed to characterize the exopolysaccharide (EPS) produced by novel probiotic Lactobacillus plantarum C70 (accession number KX881779) isolated from camel milk and to investigate its bioactivities and rheol. properties. EPS-C70 had a weight-average mol. weight (Mw) of 3.8 × 105 Da. Arabinose (13.3%), mannose (7.1%), glucose (74.6%), and galactose (5.0%) were the major monosaccharides constituents. EPS-C70 had two endothermic peaks at 76.95°C and 158.76°C corresponding to glass transition (Tg) and m.p. (Tm), resp. Zeta potential and particle size of EPS-C70 were -330.71 mV and 525.5 nm, resp. DPPH and ABTS of EPS-C70 were 75.91% and 49.42% at 10 mg/mL concentration, resp. The cytotoxic activities against colon cancer and breast cancer lines were 88.1% and 73.1% at concentration 10 mg/mL, resp. EPS-C70 exhibited shear-thinning behavior. Salts and pH values had a significant impact on the rheol. properties of EPS-C70.

The endothelial-mesenchymal transition (EndMT) participates in the progression of diabetic retinopathy (DR), but cell-intrinsic factors modulating this process remain elusive. In this study, we explored the role of lysophosphatidic acid (LPA) – producing enzyme, acylglycerol kinase (AGK), in the EndMT of human retinal microvascular endothelial cells (HRECs) under high-glucose (HG) conditions. We found that AGK was significantly elevated in HG-treated cells. In addition, AGK knockdown reversed the HG-induced EndMT in HRECs, which was evidenced by the increased endothelial markers (CD31 and VE-cadherin) and decreased mesenchymal markers (FSP1 and β-SMA). Furthermore, downregulation of AGK inhibited the HG-induced activation of transforming growth factor β (TGF-β)/Notch pathways, whereas exogenous TGF-β1 (10 ng/mL) impeded the inhibitory effects of AGK knockdown on HG-induced EndMT in HRECs. Addnl., the silencing of AGK abolished the HG-induced upregulation of LPA and its receptor, LPA receptor 1 (LPAR1), and overexpression of LPAR1 further rescued the AGK knockdown-mediated inhibition of the EndMT process. In conclusion, we demonstrate that downregulation of AGK suppresses HG-induced EndMT in HRECs through regulating the LPAR1/TGF-β/Notch signaling pathway, indicating that AGK might be a potential therapeutic target for the treatment of DR.

This study aims to determine the antitumor potential of cashew gum in vitro and in vivo. The cashew gum structure is similar to already showed in literature. The cytotoxicity effect of CG was performed by MTT assay, and B16-F10 melanoma model was used to evaluate antitumor effect. The tumor inhibition was calculated based on tumor weight Hematol., histopathol., FTIR, oxidative stress and Western Blot anal. were performed to elucidate the mechanism of inhibition and toxic effects. As results, CG did not demonstrate cytotoxicity in vitro, however showed a significant tumor inhibition in vivo, with about 36.9 to 43% of reduction in tumor mass, with no toxicity to organs. Animals treated with CG did not show toxicity in normal tissues, FTIR spectrum and oxidative stress anal. of the tumor tissue indicated that CG cause tumor inhibition with the presence of apoptosis morphotype cells, without alterations in the levels of antioxidants components. In addition, it was observed that CG reduced the expression of γH2AX without changing the expression of caspase-3. With this, we can suggest that this polymer can assist in the anticancer activity and/or decrease the side effects of standard drugs used in treatment of cancer.

Though studies in bioaerosols are being conducted with increasing frequency over the past decade, the total breadth of knowledge on bioaerosols and their role in atm. processes is still minimal. In order to better characterize the chem. composition of fresh biol. aerosol for purposes of source apportionment and tracing in the atm., several plant pollen species were selected for detailed chem. analyses. For this purpose, different pollen species were purchased and collected around Reno, Nevada, USA, for further extraction and detailed chem. anal. These species included aspen, corn, pecan, ragweed, eastern cottonwood, paper mulberry, rabbitbrush, bitterbrush, lodgepole pine, and Jeffrey pine. Saccharides, free amino acids, and various other polar compounds (e.g., anhydrosugars and resin acids) were quant. analyzed using gas chromatog. and ultra-high performance liquid chromatog. coupled with mass spectrometry techniques (GC-MS and UPLC-MS), with the purpose to identify differences and nuances in chem. composition of specific pollen species. The saccharides β-D-fructose, α-D-glucose, and β-D-glucose were ubiquitously found across all pollen samples (10), and sucrose was found in five samples. D-galactose was also found in pine species. Total saccharides were 4.0 to 29% of total dry weight across all samples. Total free amino acids were 0.29% to 15% of total dry weight across all samples, with the most common amino acid being proline. Chem. profiles (including both saccharides and amino acids) of surface-deposited aerosol in the Lake Tahoe area correlated most closely with pine pollen than other analyzed pollen species, indicating that chem. profiles of pollen can be used to infer its contribution to local aerosols.

In this study, uridine-diphosphate-dependent glycosyltransferase (UGT) candidate genes were selected by correlation anal. and their response to airborne (Z)-3-hexenol, which has been shown to be taken up by the tea plant. The allelic proteins UGT85A53-1 and UGT85A53-2 showed the highest activity towards (Z)-3-hexenol and are distinct from UGT85A53-3, which displayed a similar catalytic efficiency for (Z)-3-hexenol and nerol. A single amino acid exchange E59D enhanced the activity towards (Z)-3-hexenol, whereas a L445M mutation reduced the catalytic activity towards all substrates tested. Transient overexpression of CsUGT85A53-1 in tobacco significantly increased the level of (Z)-3-hexenyl glucoside. The functional characterization of CsUGT85A53 as a (Z)-3-hexenol UGT not only provides the foundation for the biotechnol. production of (Z)-3-hexenyl glucoside but also delivers insights for the development of novel insect pest control strategies in tea plant and might be generally applicable to other plants.
