Previously, linalool was found to be the most abundant component among the cocktail of volatiles released from flwers of Freesia hybrida. Linalool formation is catalyzed by monoterpene synthase TPS1. However, the regulatory network developmentally modulating the expression of the TPS1 gene in Freesia hybrida remains unexplored. In this study, three regulatory genes, FhMYB21L1, FhMYB21L2, and FhMYC2, were screened from 52 candidates. Two MYB transcription factor genes were synchronously expressed with FhTPS1 and could activate its expression significantly when overexpressed, and the binding of FhMYB21L2 to the MYBCORE sites in the FhTPS1 promoter was further con firmed, indicating a direct role in activation. FhMYC2 showed an inverse expression pattern compared with FhTPS1; its expression led to a decreased binding of FhMYB21 to the FhTPS1 promoter to reduce its activation capacity when co-expressed, suggesting a role for an MYB-bHLH complex in the regulation of the FhTPS1 gene. In Arabidopsis, both MYB21 and MYC2 regulators were shown to activate the expression of sesquiterpene synthase genes, and the regu latory roles of AtMYB21 and AtMYC2 in the expression of the linalool synthase gene were also confirmed, implying conserved functions of the MYB-bHLH complex in these two evolutionarily divergent plants. Moreover, the expression ratio between MYB21 and MYC2 orthologues might be a determinant factor in floral linalool emission.

The SN2-type stereoselective Mitsunobu glycosylation is generally applicable to various unprotected pyranoses as glycosyl donors in combination with a wide range of acidic glycosyl acceptors such as carboxylic acids, phenols, and imides, retaining its high stereoselectivity (33 examples). Glycosylation of a carboxylic acid with unprotected α-D-mannose proceeded also in an SN2 manner to directly afford a usually less accessible 1,2-cis-mannoside. One- or two-step total syntheses of five simple natural glycosides were performed using the glycosylation strategy presented here using unprotected α-D-glucose.

This work presented the synthesis of Cu (I) immobilized on modified boehmite nanoparticles as an environmentally friendly heterogeneous catalyst for the synthesis of sulfides and their oxidation to the corresponding sulfoxides in good to excellent yields. The structure of the catalyst was studied by SEM, X-ray diffraction, thermogravimetric, inductively coupled plasma at. emission spectroscopy, and Brunauer-Emmett-Teller techniques. The use of this heterogeneous nanocatalyst in mentioned organic reactions allowed achieving good results such as the excellent stability, high efficiency, low cost, easy recovery, and reusability of catalyst for four continuous cycles.

(+)-Bicyclogermacrene (I) is a strained bicyclic and common sesquiterpene found in several essential oils. A short and good yielding synthesis of bicyclogermacrene proceeding in seven steps is reported. This terpene is used as key platform intermediate for a biomimetic access to several aromadendrene sesquiterpenoids, such as ledene, viridiflorol, palustrol, and spathulenol. Furthermore, bicyclogermacrene is shown to be the terpene component in the synthesis of the meroterpenoids psiguadial A, C, and D.

Abstract: Background: Prostate cancer (PC) can display very heterogeneous phenotypes ranging from indolent asymptomatic to aggressive lethal forms. Understanding how these PC subtypes vary in their striving for energy and anabolic mols. is of fundamental importance for developing more effective therapies and diagnostics. Here, we carried out an extensive anal. of prostate tissue samples to reveal metabolic alterations during PC development and disease progression and furthermore between TMPRSS2-ERG rearrangement-pos. and -neg. PC subclasses. Methods: Comprehensive metabolomics anal. of prostate tissue samples was performed by non-destructive high-resolution magic angle spinning NMR (1H HR MAS NMR). Subsequently, samples underwent moderate extraction, leaving tissue morphol. intact for histopathol. characterization. Metabolites in tissue extracts were identified by 1H/31P NMR and liquid chromatog.-mass spectrometry (LC-MS). These metabolomics profiles were analyzed by chemometric tools and the outcome was further validated using proteomic data from a sep. sample cohort. Results: The obtained metabolite patterns significantly differed between PC and benign tissue and between samples with high and low Gleason score (GS). Five key metabolites (phosphocholine, glutamate, hypoxanthine, arginine and α-glucose) were identified, who were sufficient to differentiate between cancer and benign tissue and between high to low GS. In ERG-pos. PC, the anal. revealed several acylcarnitines among the increased metabolites together with decreased levels of proteins involved in β-oxidation; indicating decreased acyl-CoAs oxidation in ERG-pos. tumors. The ERG-pos. group also showed increased levels of metabolites and proteins involved in purine catabolism; a potential sign of increased DNA damage and oxidative stress. Conclusions: Our comprehensive metabolomic anal. strongly indicates that ERG-pos. PC and ERG-neg. PC should be considered as different subtypes of PC; a fact requiring different, sub-type specific treatment strategies for affected patients.

Abstract: In this study, the immobilization technol. was used to improve the LA yield and shorten the fermentation time. The optimum conditions to immobilize Lactobacillus pentosus ATCC 8041 cell were determined by Taguchi design L16 (45). The immobilized L. pentosus ATCC 8041 cells prepared by 2% sodium alginate (SA) and 6% polyvinyl alc. (PVA) with the immobilization process by 0.10 M calcium chloride (CaCl2) and 2.5% boric acid (H3BO3) had the best performance of LA yield at the temperature of 35°C, which is significantly higher than that of L. pentosus ATCC 8041 free cells. These cells maintained the stable and efficient performance in 15 repeated batch fermentation, and they also have excellent mech. strength to keep from breakage caused by cell growth and agitation.

The polysaccharide preparation from Pimpinella anisum seeds (PAP) was isolated and characterized to evaluate its laser burn wound-healing and anti-inflammatory activities in mice. The structure characterization of PAP by Infra-red spectrometry (IR), NMR (NMR), Gas chromatogram-Mass spectrometer (GC-MS) and colorimetric methods revealed an optimum yield of 8.84%, a high quantity of carbohydrate (64.75%) and low levels of lipids, protein and sulfate. Galactose (33.47%), β-D-Glucose (26.71%) and a-D-Mannose (18.21%) were the major monosaccharides components presenting in PAP, and a smaller amounts of β-D-Galactose, D-Fructose, a-D-Glucose, a-L-Galactose and arabinose were detected. PAP showed noticeable antioxidant and antibacterial properties. The anti-inflammatory activity of PAP in the carrageenan-induced paw edema model in mice, demonstrated by reduced edema and cellular infiltration, and oxidative stress markers in muscle tissue. A beneficial wound healing effect was also revealed. The topical application of PAP based gel on laser burn lesions accelerates wound contraction, the re-epithelization and remodeling phases after seven days of treatment. The results demonstrated that PAP is a novel promising source of natural wound healing and anti-inflammatory drugs. The high content and varied PAP monosaccharides seem to be responsible for the observed biol. activities.

Food authenticity concerning the geog. origin becomes increasingly important for consumers, food industries, and food authorities. In this study, nontargeted1H NMR metabolomics combined with machine learning methodologies was applied to successfully distinguish the geog. origin of 237 samples of white asparagus from Germany, Poland, The Netherlands, Spain, Greece, and Peru. Support vector classification of the geog. origin achieved an accuracy of 91.5% for the entire sample set and 87.8% after undersampling the majority class. Important regions of the spectra could be identified and assigned to potential chem. markers. A subset of samples was compared to isotope-ratio mass spectrometry (IRMS), an established method for the determination of origin of white asparagus in Germany. Here, SVM classification led to accuracies of 79.4% for NMR and 70.9% for IRMS. Finally, the classification of asparagus from different German regions was evaluated, and the influence of year and variety was analyzed.

The physicochem. properties, structural features and immunomodulatory effects of the white asparagus (Asparagus officinalis L.) skin polysaccharides (WASP) were systematically studied. To conclude, the pectic-like polysaccharides from white asparagus (Asparagus officinalis L.) skin could be potentially used as an immunomodulatory agent in functional food.
