The purpose of this study was to optimize the extraction method for polysaccharide from the fruiting bodies of Pleurotus ostreatus (Jacq.) P.Kumm and to assess the antioxidant and cytotoxic potentials of polysaccharide. In this investigation, polysaccharides from Pleurotus ostreatus (Jacq.) P.Kumm were extricated by utilizing the hot water. One-single factor and response surface methodol. was established to optimize the extraction conditions for polysaccharide from Pleurotus ostreatus (Jacq.) P.Kumm. Examination of antioxidant activity of Pleurotus ostreatus polysaccharide (POP) was directed by utilizing 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and 2, 2-azino-bis-3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) techniques. Cytotoxicity of POP was evaluated using an MTT assay. The exptl. data were fitted to a quadratic equation utilizing multiple regression investigations, and the ideal conditions were as per the following: water/crude material proportion, 26.04 mL/g; an extraction time of 62.08 min; and an extraction temperature 70.5°C. Under such conditions, the polysaccharide yield was 5.32 ± 0.12% with the anticipated yield. POP showed good scavenging activity against DPPH radical (p<0.001, EC50 = 1036.38 μg/mL, R2 = 0.8313) and ABTS radicals (p<0.001, EC50 = 824.37 μg/mL, R2 = 0.8223), with a dose (p<0.001)-and-time (p<0.001) dependent cytotoxic potential on Ehrlich ascites carcinoma cell line in vitro. This demonstrated that polysaccharides (POP) had certain cancer prevention agent exercises. In this manner, these examinations give reference to addnl. research and reasonable improvement of Pleurotus ostreatus (Jacq.) P. Kumm polysaccharide and POP may prove a useful therapeutic agent, due to its robust antioxidant and cytotoxic activity. An immunol. active polysaccharide named as UPP-2 (1035.52 kDa) was isolated from Undaria pinnatifida using traditional water extraction followed by DEAE Sepharose fast flow chromatog. UPP-2 was proven to be a low sulfated polysaccharide with relatively abundant uronic acid (13.08 ± 0.67%). UPP-2 mainly consisted of xylose (64.55%), glucose (23.81%), arabinose (5.90%) and mannose (4.26%), and its main glycosidic linkage types included →2-α-D-Xylp-(1→, →4)-α-D-Glcp-1→, α-D-Xylp-(1→ and →2,4)-β-D-Xylp-1→. Results indicated that UPP-2 significantly promoted the proliferation and pinocytic capacity of RAW264.7 cells, and upregulated the mRNA expressions of iNOS, TNF-α, IL-6, and IL-1β at 100-600μg/mL with a maximum of 195, 42, 768, and 539 times those of the neg. control, resp. Moreover, UPP-2 significantly increased the secretions of nitric oxide, TNF-α and IL-6 at 100-600μg/mL (8.0, 73.1, and 188.7 times compared to those of the neg. control, resp.), as well as promoted the production of IL-1β obviously at 600μg/mL. Overall, UPP-2 could be served as a potential dietary supplement or functional food based on its immunostimulatory activity.

Gleditsia sinensis, fenugreek and guar galactomannans (referred to as GSG, FG, and GG) were extracted from their gums and investigated using various techniques. Mannose to galactose ratios were 3.55, 1.11, and 1.65, resp. The intrinsic viscosity of GSG was very close to that of GG, while that of FG was the lowest one. This was attributed to the influence of high galactose substitution of FG on the mannan backbone, which induced a lower chain dimension due to intermol. entanglement. High degrees of substitution and high temperatures contributed to improving the solubility of galactomannan. Rheol. behavior indicated that GG had the highest apparent viscosity, yet the power-law model could well-fitted the flow curves of GSG and FG, but not GG. Through morphol. observations, the extracted galactomannans exhibited rod-like structure in deionized water and showed fibrous filament network structure after dehydration by freeze-drying. The thermal behavior was greatly influenced by the degree of side groups and Mw of galactomannans.

Structural analyses indicated that both polysaccharide fractions were heteropolysaccharides, their average mol. weights were 193 kDa and 107 kDa, resp. The backbone of WXA-1 was → 4-β-D-Xylp-1→, which was substituted at O-3 positions by arabinose, glucose and galactose residues, while the backbone of AXA-1 was → 4-β-D-Xylp-1→, which was mainly substituted at O-3 positions by arabinose. AXA-1 exerted a stronger inhibitory effect on the activities of α-amylase and α-glucosidase compared with WXA-1. Moreover, AXA-1 exhibited a competitive inhibition of α-amylase and a mixed-type noncompetitive inhibition of α-glucosidase. These results suggest that AXA-1 can be used as α-amylase and α-glucosidase inhibitors.

Lavandula dentata L. and Origanum syriacum L. essential oils have numerous health benefits and properties, such as possessing common components with a variant degree of depressive actions in the central nervous system. We investigated the depressive property of these oils on AMPA receptors, which are responsible for most of the fast-excitatory neurotransmission in the CNS and play a critical role in synaptic plasticity. Since excessive activation of AMPARs has been linked to neurotoxicity leading to various pathologies, we hypothesize that these oils have a neuroprotective role by acting directly on the kinetics of AMPARs. The current study results showed that the biophys. properties of AMPA receptor subunits showed a decrease in desensitization rate of GluA1 and GluA2 homomers, using O. syriacum, while administering L. dentata oil decreased the desensitization rate of GluA1 and GluA2 homomers, as well as GluA1/2 heteromers. As for the deactivation rate, both oils slowed the deactivation kinetics of all AMPA receptor subunits. Intriguingly, between the two oils, the effect of desensitization and deactivation was of a greater significance for L. dentata oil than O. syriacum. Our data suggest that the two oils contain components that are essential to identify, as those active components underlie the oils’ neuronal depressive properties reported, and to extract them to synthesize a potent neuroprotective drug to treat neurol. diseases potentially.

Alzheimer’s disease (AD) is a progressive neurodegenerative disorder more prevalent among the elderly population. AD is characterised clin. by a progressive decline in cognitive functions and pathol. by the presence of neurofibrillary tangles (NFTs), deposition of beta-amyloid (Aβ) plaque and synaptic dysfunction in the brain. Centella asiatica (CA) is a valuable herb being used widely in African, Ayurvedic, and Chinese traditional medicine to reverse cognitive impairment and to enhance cognitive functions. This study aimed to evaluate the effectiveness of CA in preventing D-galactose/aluminum chloride (D-gal/AlCl3) induced AD-like pathologies and the underlying mechanisms of action were further investigated for the first time. Results showed that co-administration of CA to D-gal/AlCl3 induced AD-like rat models significantly increased the levels of protein phosphatase 2 (PP2A) and decreased the levels of glycogen synthase kinase-3 beta (GSK-3β). It was further observed that, CA increased the expression of mRNA of Bcl-2, while there was minimal effect on the expression of caspase 3 mRNA. The results also showed that, CA prevented morphol. aberrations in the connus ammonis 3 (CA 3) sub-region of the rat’s hippocampus. The results clearly demonstrated for the first time that CA could alleviate d-gal/AlCl3 induced AD-like pathologies in rats via inhibition of hyperphosphorylated tau (P-tau) bio-synthetic proteins, anti-apoptosis and maintenance of cytoarchitecture.

The inhibiting effect of 2-propanol extract of apricot pomace (APE) in respect to mild steel corrosion in 0.5 M NaCl is studied by gravimetric and potentiodynamic polarization methods. The characterization of APE is carried out by Fourier transform IR spectroscopy (FTIR) and gas chromatog.-mass spectrometry (GC-MS) anal. It is found that APE acts as a corrosion inhibitor in chloride-containing media. The maximum inhibiting efficiency (about 98%) is obtained in APE presence of 500 ppm after 40 h immersion. The results of the polarization test reveal that APE acts as a mixed type inhibitor decreasing both the anodic and the cathodic reactions rates. The adsorption of APE on the mild steel surface is investigated by AFM and SEM. DFT simulation calculations support the exptl. results obtained.

We investigated the chem. composition and antioxidant activity of Eucalyptus citriodora (lemon-scented eucalyptus) leaf oil in terms of total antioxidant activity, ferric reducing antioxidant power (FRAP) assay, ferrous ion chelating activity, and scavenging of hydrogen peroxide (H2O2) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals and inhibition of lipid peroxidation GC-MS anal. of essential oil revealed the presence of 43 components constituting 99.2% of oil. The oil was monoterpenoid (94.35% of oil) with citronellal (60.66%), β-citronellol (12.58%) and isopulegol (8.19%) as the major monoterpenoids. Oil and its major monoterpenes exhibited moderate to strong antioxidant activity in terms of TAA, FRAP and Fe+2 chelating, DPPH and H2O2 scavenging, and lipid peroxidation inhibition. The study concludes that E. citriodora leaves contain monoterpenoid rich oil exhibiting antioxidant activity.
