The aim of this study is to investigate hypolipidemic and antioxidant effects of Pine needle polysaccharide (PNP) from Pinus massoniana in high-fat diet (HFD)-induced mice. PNP could significantly improve the serum lipid levels (total cholesterol, triacylglycerols, high-d. lipoprotein cholesterol, low-d. lipoprotein cholesterol), enhance the antioxidant enzymes levels (total antioxidant capability, superoxide dismutase, glutathione peroxidase, catalase), and decrease malondialdehyde (MDA) content in HFD-induced mice. PNP exhibited distinct antioxidant ability on the superoxide anions, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and ferric ion reducing antioxidant power (FRAP) in vitro. The average mol. weight (Mw) of PNP was 6.17 × 105 Da, and mainly of fucose, arabinose, galactose, glucose, galacturonic acid. These results suggested that PNP might be used as functional foods and natural drugs in enhancing antioxidant ability and alleviating the hyperlipidemia.

Plant-parasitic nematodes and arbuscular mycorrhizal fungi (AMF) have been reported to alter the yield and chem. composition of basil (Ocimum basilicum) essential oil. The aim of this study was to evaluate the effectiveness of AMF to control the root-knot nematode Meloidogyne javanica in basil and to investigate the effects of nematode-AMF interactions on plant growth, phosphorus (P) absorption, and essential oil composition The experiment was conducted under greenhouse conditions following a completely randomized 3 x 2 factorial (two fungal species and an uninoculated control x inoculated and uninoculated seedlings) arrangement with 10 replicates. Substrates were inoculated with Claroideoglomus etunicatum, Rhizophagus clarus, or no fungi (control) and sown with basil seeds. After 20 days, half of the seedlings were inoculated with 4,000 M. javanica eggs. After 60 days, the vegetative parameters, P absorption, essential oil composition, nematode population d., AMF root-colonization efficiency, and AMF spore d. were determined The presence of AMF increased the basil’s fresh weight and ability to absorb P, while reducing the M. javanica reproduction In total, 21 compounds were identified in basil essential oil, the concentrations of which varied according to the treatments. The major components were eucalyptol, linalool, eugenol, β-elemene, trans-α-bergamotene, and τ-cadinol. Inoculation with AMF decreased the linalool levels but increased the amount of eucalyptol. Mycorrhizal plants showed increased shoot height, P uptake, and essential oil yield and a decreased nematode population d. in their roots.

The hydrodistd. essential oils from eighteen samples of Salvia rosmarinus Spenn. collected in six localities of Elba Island (Tuscany, Italy) during three different seasonal periods were analyzed by GC-MS for the first time. Fifty-five components were identified, representing 96.8-99.6% of the total chem. composition All the tested essential oils were characterized by a high content of monoterpenes (oxygenated 49.2-80.3%; hydrocarbons 18.7-48.3%). Among them, 1,8-cineole (26.4-49.1%), α-pinene (4.5-34.8%), camphor (1.1-18.8%) and borneol (1.7-16.2%) were the main constituents. The high amount of 1,8-cineole/α-pinene/camphor/borneol may suggest the presence of an intermediate rosemary chemotype. Ganoderma leucocontextum is a new species of Ganoderma discovered in 2014. Up to now, the structural characteristics and immunoregulatory activity of its polysaccharides remain virtually unknown. In this study, a water-soluble polysaccharide termed, GLP-3, was purified from G. leucocontextum by ultrafiltration and column chromatog. The results revealed that GLP-3 mainly consisted of glucose (92.7%) and its weight average mol. weight was 159.7 kDa. The structural anal. indicated that the backbone of GLP-3 was →4-α-D-Glcp-(1→4,6)-β-D-Glcp-1→ with a β-Glcp-1→ branch. Atomic force microscopy and Congo red experiments revealed that GLP-3 might possess a globular structure with triple-helix conformation in water. Moreover, GLP-3 was recognized by toll-like receptor 2 (TLR2) and exerted immunomodulatory effects via activating mitogen-activated protein kinases (MAPKs), phosphatidylinositol-3-kinase (PI3K)/Akt and nuclear factor-κB (NF-κB) signaling pathways in RAW 264.7 macrophages. Collectively, these results suggested that GLP-3 could be developed as a potential functional food ingredient for immunomodulation.

A water-soluble polysaccharide, named GPN, with mol. mass 38.7 kDa was isolated from Glycyrrhiza glabra with hot water extraction, ethanol precipitation, and purified by column chromatog. Monosaccharide composition anal. confirmed the presence of predominant glucose (98.03%) and trace amount of mannose, arabinose, and galactose. Methylation and GC-MS anal. revealed that the main glycosidic bonds in GPN comprised 1,4-linked Glcp, T-linked Glcp, 1,4,6-linked Glcp, and 1,6-linked Glcp. Based on these results and 1D/2D NMR spectroscopy, GPN has a linear backbone of 1,4-linked α-D-Glcp and 1,6-linked α-D-Glcp which substituted at C-4 of glucose. The side chain probably composed from 1,4-linked to main side α-D-Glcp and terminal 1-linked β-D-Glcp to the C-3 of D residue. Congo red assay confirmed the existence of triple helix structure. Moreover, SEM and XRD anal. revealed that the GPN had irregular fibrous, filaments like surface; and both crystalline and amorphous structure. GPN also displayed favorable thermal stability. Moreover, G.glabra polysaccharide showed good antioxidant activity.

A heteropolysaccharide, LP5, was purified from Lycium barbarum L. leaves. It was identified as a calcium-rich polysaccharide (8.6 mg calcium/g) with a mol. weight of 2.50 × 105 Da. The polysaccharide was composed of six kinds of monosaccharides, of which mannose and xylose are the main components. And it contains 16.37% glucuronic acid. Studies on RAW264.7 cells demonstrated that this polysaccharide exhibited potent immunomodulatory activity, including an increase in phagocytic activity, as well as the release of both nitric oxide and cytokines. However, after the depletion of calcium, the polysaccharide exhibited greater immunomodulatory activity (p < 0.05). Further conformation anal. confirmed that the polysaccharide changed from a compact spherical conformation to a random coil structure in aqueous solution after the depletion of calcium, which resulted in increased immunomodulatory activity by LP5. To characterize the immuno-stimulating ingredient from the Korean citrus, Cheongkyool, a crude polysaccharide (CCE-0) was isolated from the pectinase digests of Cheongkyool peels, from which the complex polysaccharide CCE-I was purified to homogeneity by gel filtration. CCE-I highly enhanced the production of IL-6, TNF-α, and NO in RAW 264.7 cell lines. It augmented the mRNA expression of IL-6, TNF-α, and iNOS in a dose-dependent manner. Moreover, CCE-I dose-dependently induced phosphorylation of MAPKs and NF-κB related proteins and led to the nuclear translocation of p65. The effect of CCE-I on NO and IL-6 production was suppressed by treatment with specific antibodies for TLR2, TLR4, and scavenger receptors. Conversely, the primary structure of CCE-I that exhibited potent immunostimulatory activity was characterized by sugar composition, linkage anal., and oligosaccharide anal. after β-elimination. The results suggested that CCE-I may be a rhamnogalacturonan-I type, highly branched polysaccharide with short arabinan and galactan side chains.

Essential oils have been widely used in traditional medicine for the eradication of lice, including head lice, but due to the variability of their constitution the effects may not be reproducible. In an attempt to assess the contribution of their component monoterpenoids, a range of common individual compounds were tested in in vitro toxicity model against both human lice (Pediculus humanus, an accepted model of head lice lethality) and their eggs, at different concentrations No detailed study into the relative potencies of their constituent terpenoids has so far been published. Adult lice were observed for lack of response to stimuli over 3 h and the LT50 calculated, and the percentage of eggs failing to hatch was used to generate ovicidal activity data. A ranking was compiled for adult lice and partially for eggs, enabling structure-activity relationships to be assessed for lethality to both, and showed that, for activity in both life-cycle stages, different structural criteria were required. (+)-Terpinen-4-ol was the most effective compound against adult lice, followed by other mono-oxygenated monocyclic compounds, whereas nerolidol was particularly lethal to eggs, but ineffective against adult lice.
