The extraction, purification, immunobiol. activities, and structure of Agaricus bisporus polysaccharides (ABP) were investigated. Especially we purified and identified the polysaccharides with the highest in vitro immunobiol. activity. The extraction conditions of ABP were optimized using single factor and orthogonal experiment ABP Ia was screened after double purification with DEAE-52 and Sephadex G-200 and showed the best immunoregulatory activity. UV spectra anal. and high-performance gel permeation chromatog. results indicated that the ABP Ia fraction did not contain any proteins or nucleotides and was a homogeneous polysaccharide with a relative mol. weight of 784 kDa. Gas chromatog. mass spectroscopy results showed that ABP Ia was a heteropolysaccharide consisting of ribose, rhamnose, arabinose, xylose, mannose, glucose, and galactose at a molar ratio of 2.08:4.61:2.45:22.25:36.45:89.22:1.55. FT-IR and periodic acid oxidation anal. indicated that ABP Ia was an α-pyran polysaccharide composed of 1 → 2 and 1 → 4 glycosidic bonds, as well as a possible 1 → 3 glycosidic bond. Furthermore, at. force microscopy revealed that ABP Ia polysaccharide chains twisted to form a rod-like architecture and, at a 5% concentration, aggregated into a tight structure similar to the shape of a stone forest. These findings identify ABP Ia as a potential functional food ingredient or pharmaceutical for immunoregulation.

In the current study, fucoidans from brown alga Nizamuddinia zanardinii were isolated with conventional and non-conventional extraction procedures to evaluate the effects of recently introduced technologies on biochem. characteristics and saccharide composition of the extracts, along with their antibacterial, antiviral and cytotoxic properties. The results demonstrated that subcritical water extraction showed the highest fucoidans yield (13.15%), while the lowest yield was obtained using ultrasound extraction method (3.6%). The polysaccharide chains consisted of fucose, galactose, glucose, mannose and xylose, whose molar percentages differed according to the extraction method used. The weight mean average mol. weight of fucoidans varied between 444 and 1184 kDa. The FT-IR spectroscopy confirmed the presence of sulfate esters by bending vibration of C-O-S and stretching vibration of S=O peaks at 818 and 1250 cm-1, resp. Antibacterial assays showed that microwave- and subcritical water-extracted fucoidans inhibited the growth of E.coli and that enzyme-ultrasound, ultrasound-microwave and subcritical water extracted fucoidans exhibited inhibitory effects against P. aeruginosa at 2 mg/mL. Antiviral studies revealed that all the extracted fucoidans exerted strong antiviral activity against HSV-2 infection, with EC50 values in the 0.027-0.123 μg/mL range; indeed the viscozyme-extracted macromols. displayed the best selectivity index.

Botanical biopesticides have potential for use in mosquito control because they exhibit low mammalian toxicity, are readily biodegraded, show target specificity and insecticidal activity. As populations of mosquito species grow more resistant to currently used organic insecticides, a need for new and effective insecticides for vector control becomes more urgent. This study reports the effects of synergistic and antagonistic essential oils (EOs) from Cymbopogon schoenanthus, Lantana camara, Lippia chevalieri and Lippia multiflora and their binary combinations against Anopheles gambiae s. l. larvae and adults. EOs insecticidal properties were tested with third to fourth-instar larvae and, non-blood-fed 3-5-day old field-collected An. gambiae using WHO and CDC bottle bioassays, resp. Many compounds were found in the EOs mixtures All EOs showed larvicidal and adulticidal activities with mortality from 0 to 100% against An. gambiae which were concentration dependent and EOs specific. EO mixture from C. schoenanthus and L. multiflora showed synergistic effect with LC50 of 38 ppm and EOs from L. chevalieri and C. schoenanthus showed antagonistic effect against larvae with LC50 of 100.84 ppm. On the other hand, EOs combinations showed additive effect on all adults. This study describes the potential of using EOs and their synergistic mixtures as potential insecticide(s) as a safer alternative to synthetic insecticides.

Four extraction methods of hot-water (HWE), ultrasound-assisted (UAE), microwave-assisted (MAE), and ultrasound-microwave assisted (UMAE) were utilized to determine the best technique to extract pectin from the common fig (Ficus carica L.) skin. UMAE (11.71%) significantly showed a more extraction yield than MAE (9.26%), UAE (8.74%), and HWE (6.05%). A significant dose-dependent antioxidant capacity for pectins 1,1-diphenyl-2-picryl-hydrazyl (DPPH), and 2,2′-azinobis (3-ethylbenzothiazoline-6-sulfonicacid) (ABTS) radical scavenging activity was found. The UMAE-pectin with the maximum galacturonic acid content (76.85%) and mol. weight (6.91 × 103 kDa) presented the highest emulsifying activity (61.2-61.3%) and emulsion stability (94.3-95.2%) with a monomodal droplet size distribution at cold and ambient storage temperatures A non-Newtonian shear-thinning behavior was detected at 1.5-3.0% pectic-solutions X-ray diffraction (XRD) anal. showed a non-crystalline nature for pectins extracted by UMAE. FTIR spectroscopy and HPLC photodiode array detector revealed that conventional and novel extraction technologies do not change the chem. structure and monosaccharide composition of pectin significantly.

A novel GH36 α-galactosidase gene (LrAgal36A) from Lichtheimia ramosa was synthesized and highly expressed in Pichia pastoris. The enzyme titer and protein yield for high-d. fermentation in a 5 L fermentor were up to 953.6 U mL-1 and 4.36 g L-1. Purified recombinant LrAgal36A showed the maximum activity at pH 6.0 and 65°C and was thermostable with a half-life of 70 min at 60°C. LrAgal36A displayed the highest specific activity (353.17 ± 4.19 U mg-1) toward p-nitrophenyl-α-D-galactopyranoside (pNPGal) followed by galacto-oligosaccharides and could act slightly on galactomannans. The Km and catalytic efficiency (kcat/Km) of LrAgal36A for pNPGal were 0.33 mM and 1569.50 mM-1 s-1, resp. LrAgal36A and GH5 β-mannanase from L. ramosa showed a significant synergistic effect on the degradation of locust bean gum (LBG), resulting in release more reducing sugars (1.56 folds) and galactose (7.6 folds) by simultaneous or sequential reactions. Due to its hydrolysis properties, LrAgal36A might have potential applications in the area of pulp biobleaching, feed and food processing.

Pectin is one of high-value functional food ingredients. Better knowledge of properties of chayote pectin would help to promote the application of the chayote as a pectin source. In this study, ultrasound-assisted extraction (UAE) was used to extract chayote pectin. The extraction parameters were optimized by a Box-Behnken response surface design. The highest yield (6.19%) was obtained at a liquid/solid ratio of 50 mL/g, ultrasonic temperature of 70°C and ultrasonic time of 40 min as optimized extraction conditions. The chem. properties, spectral information and rheol. properties of pectin extracted with UAE under the optimum conditions (PEUO) were measured and the results indicated that the PEUO exhibited a low degree of esterification, high molar mass (2.47 × 106 g/mol) and suitable foaming capacity. The PEUO featured a stronger antioxidant capacity compared to com. apple pectin and the PEUO solutions (1%, 2%, 3% and 5%) showed a non-Newtonian behavior. Hence, PEUO may provide beneficial characteristics to find use in food industry.

Camellia Oleifera Shell (COS) is a kind of renewable lignocellulose resource and contains abundant hemicelluloses. In this work, the hemicelluloses in COS were extracted by alkali treatment and precipitated by ethanol with different concentration Thermal pyrolysis kinetics of COS hemicelluloses were investigated using a thermogravimetric analyzer at the heating rates of 5, 10, and 20 °C/min based on Coats-Redfern, Flynn-Wall-Ozawa (FWO), and Kissinger-Akahira-Sunose (KAS) model. The results showed that the best fitting thermal pyrolysis mechanism of COS hemicelluloses was one-dimensional diffusion reaction analyzed by Coats-Redfern model. The activation energies of COS hemicelluloses ranged from 175.07 to 247.87 kJ·mol-1 and from 174.74 to 252.50 kJ·mol-1 calculated by FWO and KAS, resp. The thermal stabilities of COS hemicelluloses were enhanced with the precipitated ethanol concentration increasing, and reflected by thermodn. parameters ΔH, ΔG and ΔS. This study may provide basic theor. supports for the thermochem. conversion of COS hemicelluloses.

A neutral ginger polysaccharide fraction (NGP) was isolated from ginger, and its primary structures and immunomodulatory activity were investigated. The results showed that NGP had a low mol. weight of 6305 Da. NGP principally consisted of glucose (93.88%) together with minor levels of galactose (3.27%) and arabinose (1.67%). Besides, results of methylation anal. and 1D/2D NMR spectroscopy demonstrated that NGP was α-glucan which had the main chain of 1,4-linked α-D-Glcp and α-D-Glcp residues branched at C-6 position which was different from the common triple helical β-glucans. NGP also displayed a remarkable immunol. activity on the RAW264.7 cells in vitro. It could significantly enhance the proliferation of macrophages without cytotoxicity and increase the production of immune substances (NO, TNF-α, IL-1β and IL-6). The secretion at the concentration of 200μg/mL was 29.41μM, 1496.71, 44.30 and 1889.83 pg/mL for each substance, resp. The results indicated that NGP could be a potential immune agent and might provide meaningful information for further chain conformation and immune mechanism research.

Polysaccharides were extracted from Schizophyllum commune (a common mushroom) and their structural and immune-enhancing properties were investigated. Crude and fractions (F1 and F2) were composed of sugars (50.3-82.8%), proteins (1.46-20.1%), and sulfates (1.33-7.01%). Monosaccharide compositions of Cr and F1 were mainly composed of glucose (75.5% and 88.2%) with small amounts of mannose, galactose and xylose whereas the F2 was mainly composed of manose (55.2%) with minor amounts of galactose, glucose, and xylose. Their immune-enhancing activities were tested using RAW264.7 cells. Proliferation activity of RAW264.7 cells was over 100% after treatment with these polysaccharides. In addition, RAW264.7 cells produced large amounts of nitric oxide and various cytokines by up-regulating mRNA expression levels and the activation of nuclear factor-kappa (NF-κB) and mitogen-activated protein kinase (MAPK) after treatment with these polysaccharides. In addition, RAW264.7 cells were activated mainly through CR-3 and TLR-4 receptors. The backbone of F2 with excellent immune-enhancing activity was mainly linked by (1→3)-linked-mannopyranosyl and (1→2,3)-linked-mannopyranosyl residues.

The polysaccharide obtained from edible fungi has been regarded as the major bioactive component related to the nutritional and human’s health. In the present study, three purified polysaccharides (LCP-1, LCP-2, and LCP-3) were obtained from Leccinum crocipodium (Letellier.) Watliag. The characterizations of LCP-1, LCP-2, and LCP-3 were determined by high-performance liquid chromatog. (HPLC), UV, FT-IR spectrometrys and 1H NMR spectrum. LCP-1 had a mol. weight of 2.303 × 105 Da and 7.519 × 103 Da, and was composed of mannose (Man), ribose (Rib), rhamnose (Rha), glucuronic acid (GluA), galacturonic acid (GalA), glucose (Glu), galactose (Gal), xylose (Xyl), arabinose (Ara) and fucuronic (Fuc). The mol. weight of LCP-2 was 2.655 × 105 Da, and its monosaccharide constituents were Man, Rib, Rha, GluA, Glu, Gal, Xyl, Ara and Fuc. The mol. weight of LCP-3 was 3.783 × 105 Da, and its monosaccharide constituents were Man, Rib, Rha, GluA, GalA, Glu, Gal, Xyl, Ara and Fuc. For the in vitro immunomodulatory experiments demonstrated that three purified polysaccharides could enhance immunomodulatory activities on macrophage RAW 264.7 cells, moreover, LCP-2 and LCP-3 showed stronger immunomodulatory activity than LCP-1. The results indicated that the LCP-1, LCP-2, and LCP-3 could be further developed as functional food or medicine.
