In this study, high-pressure ultrasonic-assisted extraction (HUE) technol. was used for polysaccharide extraction from Hovenia dulcis (HDPS). A maximal extraction yield (11.81 ± 0.26%) was obtained using the response surface method (RSM) with optimized HUE conditions (Ultrasound power 330 W, 68 °C, and 22 min). Results showed that HDPS mainly contained neutral sugar (47.81 ± 1.56%), uronic acid (12.52 ± 0.15%), and protein (3.64 ± 0.24%), specifically. HDPS was comprised of mannose, rhamnose, galacturonic acid, glucose, galactose and arabinose in a molar ratio of 3.20%, 20.40%, 4.81%, 11.14%, 27.37% and 33.08%, resp. And with an average mol. weight of 185.22 kDa for its major component (accounting for 90.13% for HDPS). The result obtained from the X-ray diffraction pattern confirmed a semi-crystalline nature of the sample. HUE-HDPS suggested typical shear thinning behavior and non-Newtonian properties. Addnl., HUE-HDPS exhibited better chem. antioxidant activities in DPPH, ABTS, hydroxyl radical scavenging and Fe2+ chelation activity. The in-vitro and in-vivo experiments indicated that HUE-HDPS had obviously hypoglycemic effects. Accordingly, the polysaccharides from Hovenia dulcis might serve as a natural antioxidant and hypoglycemic agent.

Sparassis crispa polysaccharides have recently attracted considerable attention due to their excellent bioactivities. However, their extraction procedure is often tedious, time-consuming, and environmentally unfriendly; it even causes damage to their structures and reduces their bioactivities, all of which hinder their further development to some extent. Therefore, the ultrasonic-assisted enzymic extraction (UAEE) technol. was optimized to extract polysaccharides from Sparassis crispa (SCP) by the response surface methodol. The yields, physicochem. properties, and antioxidant activities of SCPs obtained from UAEE and conventional hot water extraction (HWE) were evaluated. According to the optimal parameters, the yield of SCPs extracted by UAEE reached up to 14.63%, which increased by 68.54% compared with that obtained from the conventional hot water extraction (HWE) method. Addnl., the UAEE methods affected the contents of the polysaccharides, mol. weights, and the molar percentage of the constituent monosaccharides of SCPs. SEM anal. indicated that the microstructures of the two SCPs were notably different. Antioxidant assays showed that both SCPs possessed good antioxidant activities against DPPH, ABTS +, and hydroxyl radicals in vitro. Addnl., the SCPs extracted by UAEE attenuated the HT22 cell neurotoxicity induced by H2O2 by the means of ameliorating cell viability, reducing extracellular LDH release, and decreasing the levels of intracellular ROS.

In this study, different extraction methods of polysaccharides from Crataegus pinnatifida Bunge (CPP) were compared by studying the extraction yield, structural characteristics and antioxidant activities. Firstly, polysaccharides were obtained using hot water extraction (CPPh), ultrasound assisted extraction (CPPu), enzyme assisted extraction (CPPe) and enzyme-ultrasound assisted extraction (CPPc), resp. Meanwhile, the optimum extraction conditions of enzyme-ultrasonic assisted extraction were determined by response surface method (RSM). The extraction yields, structural characteristics and antioxidant activities were investigated and compared by visual photos, gas chromatog., UV-visible and Fourier-transform IR spectroscopy. The results clearly showed that enzyme-ultrasonic assisted extraction possessed the highest extraction yield (10.39 ± 0.04%). The mol. weight of CPPh was the highest while the other polysaccharides had no significant difference. Besides, the monosaccharide composition of CPPc, CPPh, CPPu and CPPe were similar but the molar percentages of monosaccharide were different. Finally, the results of antioxidant activities showed that CPPc exhibited the highest scavenging effect of superoxide radical and lipids inhibiting ability. In summary, enzyme-ultrasonic assisted extraction was a high-efficient and low-energy consumption method for CPP extraction

The present study aimed to determine the essential oil compositions of four different types of Juniperus species including Juniperus oxycedrus ssp oxycedrus L., Juniperus foetidissima Willd., Juniperus excelsa M. Bieb, and Juniperus communis L. with their antioxidant and antiproliferative activities. The antiproliferative activity of the essential oils was determined against HeLa (human cervix carcinoma) and C6 (rat brain tumor cell) cell lines by BrdU cell proliferation assay. The essential oils were tested for their total reducing power activity, DPPH (2,2′-diphenyl-1-picrylhydrazyl) free radical scavenging activities, and hydrogen peroxide scavenging activities. The overall results of the study showed that the needles of Juniperus species have strong antiproliferative activities as cell selective against C6 cells.

In vitro provision of biol. activities through their antioxidant power (ABTS.+), ability to inhibit 15-lipoxygenase (15-LOX), acetylcholinesterase (AChE), xanthine oxidase (XOD), as well as, cytotoxic potency were undertaken. Chem. profiling showed that the methanolic extracts of both species were the richest in total phenolic compounds GC-MS anal. of the two Eucalyptus species revealed that spathulenol, globulol, epiglobulol, and trans-Z-alpha-bisabolene-epoxide were the main volatile compounds detected in the different extracts In addition, the derivatization procedure allowed the identification of ethylene glycol, and D-fructofuranose as very abundant mols. The HPLC-DAD has enabled the identification of eight new mols. among them icariin (1.1 mg/g dr), and 5-hydroxy-3prime-methoxyflavone (7.5 mg/g dr) in the E. stricklandii MeOH and EtOAc extracts Aqueous E. brevifolia extract possessed the highest anti-AChE with an IC50 (10 mug/mL). Whereas, the MeOH E. stricklandii extract possessed the highest anti 15-LOX with an IC50 (9.7 mug/mL), as well as, the most important percentage of XOD inhibition 51.1%. All extracts revealed an antiproliferative potency against human cancer cell lines HCT-116, MCF-7 and OVCAR, with an inhibition percentage up to 96%. Taken together, both Eucalyptus species have a broad range of potential applications in several fields, such pharmaceutical, medical and food industries as nutraceutical supplements.

In this study, a high Me ester pectin polysaccharide, AER-A3 (Mw, 1.12 x 105 g/mol; O-Me ester groups (wt%), 3.81%), was isolated and purified from the root bark of Aralia elata by ion exchange and gel-filtration chromatog. Its planar structure was investigated in combination with UPLC-ESI+-MS, FT-IR, HILIC-UPLC-ESI–HCD-MS/MS, GC-MS and NMR techniques. The main chain of AER-A3 was unambiguously determined to be smooth region and hairy region with a chain length ratio of 1:1, characterized by occurrence of (1 → 2)-α-Rhap, (1 → 2,3)-α-Rhap, (1 → 2,4)-α-Rhap, (1 → 2,3,4)-α-Rhap, and (1 → 4)-α-GalpA, whereas the branched chain included T-α-Rhap, T-α-Araf, (1 → 5)-α-Araf, (1 → 3,5)-α-Araf, T-β-Galp, (1 → 3)-β-Galp, (1 → 3,4,6)-β-Galp, (1 → 4)-Glcp, (1 → 3)-Glcp, and (1 → 3)-Manp. Meanwhile, SEC-MALLS-RID, CD and Congo red assays showed that AER-A3 had no helical conformations but existed as a globular shape with branching. In addition, AER-A3 had good scavenging activities against DPPH, hydroxyl, and superoxide radicals. Anti-tumor assay investigated the effects of AER-A3 on human A549 and HepG2 cancer cell lines in vitro. These results provided a scientific basis for the use of the polysaccharides in A. elata root barks in pharmaceuticals.

The purpose of the present research was to compare the effects of four extraction methods on the extraction yields, physicochem. characteristics, and antioxidant activities of polysaccharides from the cup plant (Silphium perfoliatum L.) (CPPs). The extraction methods included the hot water extraction method (HEM), ultrasonic-assisted extraction method (UEM), enzyme-assisted extraction method (EEM), and the ultrasonic-enzyme-assisted extraction method (UEEM). Four corresponding CPP extracts, HEM-CPPs, UEM-CPPs, EEM-CPPs, and UEEM-CPPs, were obtained. The results indicated that there were notable differences between the extraction methods regarding the yield, average mol. weight, uronic acid content, monosaccharide proportions, and the antioxidant activities of the CPPs. HEM-CPPs were extracted with the lowest yield (6.44%). EEM-CPPs had the largest extraction yield (9.87%) and the most notable antioxidant abilities of ferric reducing power and free radical scavenging activity. According to correlation anal., the higher antioxidant abilities of EEM-CPPs might be due to their smaller average mol. weight and a higher content of uronic acid than those of the other extracted CPPs. Therefore, EEM could serve as a promising technique for extracting CPPs.

Addnl., they exhibited obvious radical scavenging activities on DPPH and hydroxyl radicals, which reached to 59.6-71.5% and 44.5-70.4%, resp. Moreover, the antitumor effects were studied using human colon cancer cell lines HCT8. The results showed that they had significant antitumor effects considering the inhibitory effects on cell viabilities (28.3-18.0% on HCT8, resp.). Thus, these exopolysaccharides from microalgae species are worth further investigating as alternative potential antitumor agents.

The mol. weights were 1.94 × 105, 7.39 × 103 and 3.24 × 104 g/mol, resp. Monosaccharide anal. indicating that these exopolysaccharides consisted of ten different kinds of sugars and derivatives, resp. Furthermore, they exhibited obvious free radicals scavenging abilities, which reached to 55.2-71.4% and 63.1-77.5% on DPPH and hydroxyl radicals, resp. Moreover, the antitumor effects were investigated using human colon cancer cell lines HCT116 and HCT8. The results showed that they had significant antitumor effects considering the inhibitory effects on cell viabilities (17.2-19.2% on HCT116, and 22.9-38.6 on HCT8, resp.), and could decrease the colony amounts (21-42.8% on HCT116, and 11.5-35.2% on HCT8, resp.) at the tested dosage. Thus, these exopolysaccharides may be worth further investigating as effective potential antitumor agents.

Chronic liver damage has serious medical consequences. To investigate the hepatoprotective effect of dry Zingiber officinale (ginger) and its essential (volatile) oil against diethylnitrosamine (DEN) toxicity in rats. Phenols and flavonoids components were characterized in dry ginger using HPLC-UV instrument while ginger essential oil (E.O.) was investigated via GC-MS technique. Antioxidant activity was determined in-vitro. In rat model, ginger was administrated for 2 mo. Lipid profile, antioxidant biomarkers, liver functions and histopathol. were assessed. Chlorogenic acid (63.85ppm) and hesperidin (156.91ppm) are among the major phenolic and flavonoid constituents in dry ginger. Curcumene (15.21%) and linalool (13.47%) represent the main E.O. constituents. In rats treated with ginger E.O., a significant elevation in serum HDL (31.14%) was accompanied by a decrease in LDL (55.14%). A significant decrease in serum ALT and ALP was reported (56.85% and 53.84%, resp.). Serum GSH-Px activity has significantly increased 75.06%. Meanwhile, E.O. showed anticancer potential against HepG2 cell line (IC50 = 40μg/mL). Liver histopathol. examinations confirmed the protective effect against abnormalities. Ginger was able to reduce the severity of DEN-cytotoxicity in rats, which suggests a novel antioxidant role originating from this medicinal plant.
