A novel polysaccharide (MFP4P) was isolated from mulberry fruits (Morus alba L.) pulp using a combination of DEAE-Sepharose fast-flow and Sephadex G-100 chromatog. columns. Structural characterization revealed that MFP4P was composed of arabinose, rhamnose, galactose, glucose and galacturonic acid with a mol. weight of 198.2 kDa. The linkage types of MFP4P were proved to be (1 â†?6)-linked α-D-Glc, (1 â†?2)-linked α-L-Rha, (1 â†?3)-linked α-D-Gal, (1 â†?3)-linked β-L-Rha and (1 â†?)-linked α-L-Ara. The MFP4P showed good antioxidant activity with an oxygen radical absorbance capacity value of 121.8 ± 16.5 μM trolox equivalent/g. In addition, the MFP4P promoted pancreatic cell proliferation, increased insulin secretion, and enhanced glucose consumption. In the non-toxic range, the polysaccharide and zinc complexes (MFP4P-Zn) exhibited higher antioxidant and hypoglycemic activities than MFP4P at the same concentration In vivo hypoglycemic study, MFP4P and MFP4P-Zn both showed decreased blood glucose level and glucose tolerance compared with the model control group.

Fucoidan is a complex sulfated polysaccharide and an active component found in the cell wall of brown seaweeds. In the present study, fucoidans were obtained from Sargassum fusiforme using different extraction methods, including hot water (prepared fucoidan was named as WSFF), dilute hydrochloric acid (ASFF), and calcium chloride solution (CSFF). The assessments were performed on S. fusiforme fucoidans based on their chem. composition, mol. conformations, and in vitro antioxidant activities. ASFF showed the maximum extraction yield (11.24%), whereas CSFF exhibited the min. yield (3.94%). The monosaccharide composition of WSFF, ASFF, and CSFF was similar, but the molar ratio of monosaccharide was quite different. Moreover, their mol. weight, Fourier transform IR (FT-IR) spectrum, surface morphol., uronic acid content and degree of sulfation were distinct. The Congo red test and CD spectroscopy anal. displayed some differences in solution conformation of these samples. Furthermore, WSFF, ASFF, and CSFF showed distinct in vitro antioxidant activities evaluated by DPPH and hydroxyl radical scavenging assays. The present study provides scientific evidence on the influences of extraction methods on the physicochem. characteristics, conformation behaviors and antioxidant activities of S. fusiforme fucoidans.

In the current study, sunflower byproduct pectin (SFBP) was obtained using ultrasound-assisted extraction (UAE) technique. The UAE variables were successfully optimized, and the optimal condition (irradiation time 30 min, temperature 33°C and ultrasound power 400 W) yielded 11.15%. Under this condition, SFBP was rich in low-esterified galacturonic acid content (72.94%), and had long side branches of galactan, arabinogalactan, and arabinan, with an average mol. weight of 175.353 kDa. The structural properties were characterized using FTIR, NMR, XRD spectroscopies. The thermal anal. by DSC and TGA suggested suitable thermal stability for SFBP. The solubility, water and oil holding capacities, emulsifying capacity, emulsifying stability (vs various conditions), foam capacity, foam stability, DPPH and ABTS inhibitions, and reducing power assay were measured to evaluate the potential of SFBP to be considered as a food ingredient.

Potato peel was used for the extraction of three types of polysaccharides (PW, PAL, and PAC) using water, alk., and acid treatments, resp. The structure of the PP polysaccharides was examined by means of Fourier transform IR spectroscopy (FT-IR) anal. and Gas chromatog.-mass spectrometry (GC-MS). The results suggest that the extracted polysaccharides constituted essentially three functional groups: C=O, C-H, and O-H. The polysaccharides were comprised of low proportions of proteins, 17-23% uronic acids, and approx. 70% carbohydrates. PAL, PW, and PAC with mol. weights of 2.25 × 103, 2.18 × 103, and 1.92 × 103 kDa, resp., were composed of rhamnose, xylose, mannose, arabinose, glucose, and galactose. Functional properties (solubility, oil holding capacity, foaming, and emulsion properties) of these polysaccharides were evaluated. Among three, PAL showed the highest fat-binding capacity which was 7.50 g/g with the solubility of 95.06%. The three polysaccharides possessed appreciable in vitro anti-oxidant (scavenging DPPH and ABTS+ radicals, chelating ferrous ions, and reducing power) potential. PAL exhibited the strongest reducing power, scavenging activity on DPPH radicals and chelating capability on ferrous ions. PP polysaccharides can be used as promising natural antioxidants in food, pharmaceutical, and cosmetic preparations

The structure of polysaccharides was determined using FT-IR, periodate oxidation, smith degradation, methylation anal., and NMR, which demonstrated that the two polysaccharides were primarily connected by mannose with (1â†?)-linked. Notably, the configuration of the two polysaccharides exhibited random coil with pyranoid polysaccharide containing α or β glycosidic bond. Furthermore, to increase the antioxidant activity of polysaccharides (CCPs-1 amd CCPs-2), two carboxymethylated polysaccharides were obtained as cmCCPs-1 and cmCCPs-2 with the degree of substitution values of 0.34 and 0.52, resp. Structural anal. elucidated that nonselective carboxymethylation occurred in cmCCPs-1 and cmCCPs-2, where C-2, C-4 or C-6 were partially substituted. Of note, cmCCPs-2 exhibited the most significant antioxidant activity.

Chinese water chestnut peels are a kind of vegetable processing waste containing many active components such as polysaccharides, the structure of which remains unknown. To elucidate the structure of polysaccharides from Chinese water chestnut peels, two polysaccharides named WVP-1 and WVP-2 were isolated. WVP-1 (3.16 kDa) consisted of mannose (1.75%), glucose (84.69%), galactose (6.32%), and arabinose (7.24%), while WVP-2 (56.97 kDa) was composed of mannose (3.18%), rhamnose (1.52%), glucuronic acid (1.42%), galacturonic acid (4.83%), glucose (11.51%), galactose (36.02%), and arabinose (41.53%). Linkage and NMR data indicated that WVP-1 was composed mainly of â†?)-α-D-Glcp(1â†?and a certain proportion of â†?)-β-D-Glcp-(1â†? including linear and branched polysaccharides simultaneously. WVP-2 was a pectin-like polysaccharide with â†?)-α-D-GalpA6Me-(1â†?units and the branch points of â†?,4)-α-L-Arap-(1â†? â†?,6)-β-D-Galp-(1â†? In this study, a novel water-soluble polysaccharide (PVLP-1) was extracted and purified from Sacha inchi (Plukenetia volubilis L.) seeds and the structure, antioxidant and immunomodulatory activity of PVLP-1 were investigated. PVLP-1 (144 kDa) consisted of glucose (69.76%), mannose (14.86%), arabinose (10.53%), galactose (2.42%), ribose (1.23%), rhamnose (0.27%) and xylose (0.93%). PVLP-1 displayed characteristic polysaccharide bands in Fourier transform NMR spectra and IR. The primary structure of PVLP-1 was a heteropolysaccharide with a backbone of (1 â†?6)-linked glucose, sidechains of (1 â†?4)-linked mannose, (1 â†?4)-linked glucose and (1 â†?3, 6)-linked mannose and a residue unit of â†?)-linked arabinose as revealed the methylation anal. PVLP-1 possessed good water-holding capacity (WHC), oil-holding capacity (OHC) and antioxidant capacities. Besides, PVLP-1 induced the proliferation of RAW264.7 cell and enhanced the expression of inflammatory cytokines IL-6, TNF-alpha(TNF-α) and IL-1 beta (IL-1β). The present study indicated that PVLP-1 possessed immune-enhancing bioactivities and could be functional food or adjuvant drug to improve biol. immunity of immunodeficiency diseases and hypoimmunity.

Investigating the leishmanicidal effects of essential oils and preparing their nano/formulation is a promising approach for developing new green drugs. In this study, the ingredients of Cinnamomum zeylanicum essential oil were investigated using GC/MS anal., and its antioxidant effect was then evaluated using the DPPH assay. Moreover, the leishmanicidal effect of the essential oil against L. major and L. tropica was investigated; obtained half-maximal inhibitory concentrations (IC50)s were 16.53 and 7.56μg/mL. After that, a practical dosage form of C. zeylanicum, i.e., the nanoemulsion with a 52 ± 4 nm droplet size and a low droplet size distribution index (SPAN = 0.97), was prepared Interestingly, after treatment of both species with nanoemulsion, their viability was reduced to 0%. The prepared nanoformulation could be considered a potent anti-leishmanial agent for other species and in vivo studies.

Three polysaccharide fractions, ZP1, ZP2 and ZP3, were obtained and their structures were investigated. ZP3 was further investigated using methylation and NMR spectroscopy, and ZP3 comprised a main chain of (1 â†?4)-α-D-GalAp, (1 â†?2,4)-α-L-Rhap, with a (1 â†?4,6)-α-D-Galp branch. Assays of antioxidant activity showed that ZP3 exhibited a remarkable DPPH capacity and moderate hydroxyl radical scavenging ability, suggesting that it has potential applications as a natural antioxidant in foods and pharmaceuticals.
