M. pumilum has been claimed to protect the bone against the adverse effect of estrogen deficiency. Addnl., it also exhibits anti-diabetic activity. In view of these, this study aims to identify the mechanisms underlying the bone protective effect of M. pumilum in the presence of both estrogen deficiency and diabetes mellitus (DM). Ovariectomized, diabetic female rats were given M. pumilum leave aqueous extract (MPLA) (50 and 100 mg/kg/day), estrogen, glibenclamide and estrogen plus glibenclamide for 28 consecutive days. At the end of the treatment, fasting blood glucose (FBG), serum insulin, Ca2+, PO3-4 and bone alk. phosphatase (BALP) levels were measured. Rats were sacrificed and femur bones were harvested for determination of expression level and distribution of RANK, RANKL, OPG and oxidative stress and inflammatory proteins by mol. biol. techniques.100 mg/kg/day MPLA treatment decreased the FBG and BALP levels but increased the serum insulin, Ca2+ and PO3-4 levels in estrogen deficient, diabetic rats. Expression and distribution of RANKL, NF-κB p65, IKKβ, IL-6, IL-1β and Keap-1 decreased however expression and distribution of RANK, OPG, BMP-2, Type-1 collagen, Runx2, TRAF6, Nrf2, NQO-1, HO-1, SOD and CAT increased in the bone of estrogen deficient, diabetic rats which received 100 mg/kg/day MPLA with greater effects than estrogen-only, glibenclamide-only and estrogen plus glibenclamide treatments. MPLA helps to overcome the adverse effect of estrogen deficiency and DM on the bone and thus this herb could potentially be used for the treatment and prevention of osteoporosis in postmenopausal women with diabetes. Monitoring amino acids and vitamins is important for understanding human health, food nutrition and the culture of mammalian cells used to produce therapeutic proteins in biotechnol. A method including ion pairing reversed-phase liquid chromatog. with tandem mass spectrometry was developed and optimized to quantify 21 amino acids and 9 water-soluble vitamins in Chinese hamster ovary (CHO) cells and culture media. By optimizing the chromatog. separation, scan time, monitoring time window, and sample preparation procedure, and using isotopically labeled 13C, 15N and 2H internal standards, low limits of quantitation (≤0.054 mg/L), good precision (<10%) and good accuracy (100 ± 10%) were achieved for nearly all the 30 compounds Applying this method to CHO cell extracts, statistically significant differences in the metabolite levels were measured between two cell lines originated from the same host, indicating differences in genetic makeup or metabolic activities and nutrient supply levels in the culture media. In a fed-batch process of manufacturing scale bioreactors, two distinguished trends for changes in amino acid concentrations were identified in response to feeding. Ten essential amino acids showed a zigzag pattern with maxima at the feeding days, and 9 non-essential amino acids displayed a smoothly changing profile as they were mainly products of cellular metabolism Five of 9 vitamins accumulated continuously during the culture period, suggesting that they were fed in access. The method serves as an effective tool for the development and optimization of mammalian cell cultures. Streptokinase (SK) is a potent plasminogen activator naturally produced by beta-hemolytic streptococcus bacteria and used as a thrombolytic drug. Optimize high yield production of recombinant streptokinase (rSK) in Escherichia coli and evaluate its thrombolytic activity. Synthetic gene encoding mature SK protein with optimization for rare codons and mRNA secondary structure was cloned into the expression vector pET-3a and transformed into Escherichia coli BL21 (DE3). Seed banks were established for high rSK expression clones. The native rSK protein expression was optimized using IPTG induction. The nonsol. rSK inclusion bodies were purified, denatured in 6 M guanidinium chloride, and refolded using the rapid dilution method. The refolded rSK protein was purified using anion exchange chromatog. and evaluated with ELISA. The activity of rSK was evaluated using the casein digestion method and in vitro blood clot lysis assay with reference drug Sedonase as standard Seed banks with high stable expression of native rSk (MW 47 kDa) were established. High rSK expression was optimized using 1 mM IPTG at bacterial OD600 0.6. The refolded rSK was prepared and purified successfully with high productivity (494 mg purified rsk/L culture). Using ELISA, the purified rSK mol. identity and conservation of native SK epitopes were confirmed. The enzymic activity of the purified rSK was 1.945×106 IU/mg with 62.94 ± 2.3% clot lysis efficiency. A high yield production of proper rSK protein with in vitro thrombolytic activity similar to com. Gastrointestinal co-digestion of wine polyphenols with glucose/whey proteins affects their bioaccessibility and impact on colonic microbiota was written by Tamargo, Alba;Cueva, Carolina;Silva, Mariana;Molinero, Natalia;Miralles, Beatriz;Bartolome, Begona;Moreno-Arribas, M. Interactions between food components during their gastrointestinal digestion are constant and could affect compounds digestibility and bioaccessibility. These interactions could have a key role in the bioactivity of dietary polyphenols. This study aimed to investigate the food matrix effects during the co-digestion of red wine with glucose and whey proteins using the gastrointestinal dynamic simulator simgi. Bioaccessibility of wine polyphenols and nutrients and the effect of co-digestion on colonic microbiota composition and metabolism were evaluated. Co-digestion with red wine led to a reduction of over 50% of glucose bioaccessibility and lowered α-lactalbumin gastric degradation Still, co-digestion with the food matrixes modified polyphenols profiles, including their bioaccessible and non-bioaccessible fractions. For instance, the (-)-epicatechin bioaccessible fraction increased 70% when the wine was co-digested with glucose. Hence, the combined feeding of wine and each food matrix affected microbiota composition and functionality at colonic level. Glucose and whey proteins reduced bacterial diversity, but homogenization of beta-diversity by wine was observed Moreover, wine presence favored intestinal health-related taxa as Akkermansia or Bifidobacterium, and the co-digestion of wine and food matrixes significantly increased total short- and medium-chain fatty acids production, especially butyric acid. Overall, this study provides evidence of the convenience of the simgi system to evaluate the effects of co-digestion and highlights the importance of food matrix effects on our understanding of polyphenol bioactivity. A review. Chemotherapy is the main treatment used for cancer patients failing surgery. Doxorubicin (DOX) is a well-known chemotherapeutic agent capable of suppressing proliferation in cancer cells and triggering apoptosis via inhibiting topoisomerase II activity and producing DNA breaks. This activity of DOX restrains mitosis and cell cycle progression. However, frequent application of DOX results in the emergence of resistance in the cancer cells. It seems that genetic and epigenetic factors can provide DOX resistance of cancer cells. Long non-coding RNAs (lncRNAs) are a subcategory of non-coding RNAs with role in the regulation of several cellular processes such as proliferation, migration, differentiation and apoptosis. LncRNA dysregulation has been associated with chemoresistance, and this profile occurs upon DOX treatment of cancer. In the present review, we focus on the role of lncRNAs in mediating DOX resistance and discuss the mol. pathways and mechanisms. LncRNAs can drive DOX resistance via activating pathways such as NF-κB, PI3K/Akt, Wnt, and FOXC2. Some lncRNAs can activate protective autophagy in response to the stress caused by DOX, which mediates resistance. In contrast, there are other lncRNAs involved in the sensitivity of cancer cells to DOX, such as GAS5, PTCSC3 and FENDRR. Some anti-tumor agents such as polydatin can regulate the expression of lncRNAs, enhancing DOX sensitivity. Overall, lncRNAs are potential players in DOX resistance, and their identification and targeting are of importance in chemosensitivity. Furthermore, these findings can be translated into clin. for treatment of cancer patients. In order to differentiate the extra virgin olive oils (EVOO) of different origin of purchase, such as monovarietal Italian EVOO with protected denomination of origin (PDO) and com.-blended EVOO purchased in supermarkets, a number of samples was subjected to the anal. of volatile aroma compounds by both targeted gas chromatog./mass spectrometry (GC-MS) and untargeted profiling by comprehensive two-dimensional gas chromatog./mass spectrometry (GC×GC-TOF-MS), anal. of phenols by targeted high-performance liquid chromatog./mass spectrometry (HPLC-DAD-ESI/MS), and quant. descriptive sensory anal. Monovarietal PDO EVOOs were characterized by notably higher amounts of pos. LOX-derived C6 and C5 volatile compounds, which corresponded to the higher intensities of all the assessed pos. fruity and green odor sensory attributes. Com.-blended EVOOs had larger quantities of generally undesirable esters, alcs., acids, and aldehydes, which coincided with the occurrence of sensory defects in many samples. Many minor volatile compounds that were identified by GC×GC-TOF-MS were found to differentiate each of the two investigated groups. The differences between the groups with respect to phenols and taste characteristics were evident, but less pronounced. The results that were obtained in this study have undoubtedly confirmed the existence of the large heterogeneity of oils that are sold declared as EVOO. It was shown that GC-MS, GC×GC-TOF-MS, and HPLC-DAD-ESI/MS analyses have complementary outputs, and that their use in combination has advantages in supporting the results of sensory anal. and objectively differentiating these groups of EVOO. Serodiagnosis of surra, caused by Trypanosoma evansi, is still based on native antigens purified from bloodstream form of T. evansi grown in rodents. In order to investigate prospective diagnostic possibilities as an alternative for native antigens, we cloned, expressed 26 kDa calflagin protein containing 218 amino acids from T. evansi (Indian Strain) in Escherichia coli. The potential of recombinant calflagin (rCLF) protein as diagnostic antigen was evaluated in immunoblot and indirect ELISA using exptl. infected equine serum samples from 0 to 84 days post infection. The antibodies against T. evansi were detected with rCLF antigen in serum samples of exptl. infected equines as early as 10 days and 14 days post infection, using immunoblot and ELISA resp. No cross-reactivity was observed with rCLF antigen in ELISA with different serum samples of equines pos. for Equine herpesvirus 1, Burkholderia mallei, and Theileria equi infections. Several immunoreactive regions ranging from 10 to 28 kDa were detected using distinct T. evansi isolates (pony, cattle, donkey and camel origin) indicating presence of multiple calflagin family members in a single trypanosome. Indirect immunofluorescence antibody test with anti-CLF rabbit hyperimmune serum showed localisation of native immunogenic protein near attachment of flagellum. The rCLF protein was found to be a potential diagnostic candidate for distinguishing T. evansi pos. and neg. equine serum sample, suggesting that it could be used for serol. surveys in animals for surra. Phospholipid liposomes are the most popular kind of vesicles used in drug delivery due to their high biocompatibility with biomembranes. However, this type of liposomes show some limitations related with their short half-life in bloodstream. This problem can be resolved by the coating of their surface with the application of PEG mols. which form steric and entropic barrier, avoiding recognition of encapsulated liposomes by the reticuloendothelial system (RES). Such modification of liposomes significantly affects the physicochem. characteristics of PEGylated liposomes, which is related to the intermol. interactions between the lipopolymer mols. and the phospholipid that builds the liposome membrane. In our work we investigated the influence of the PEG-ylated lipid (DSPE-PEG750) on physicochem. properties of phospholipid monolayers and bilayers with different structure of the polar group (DPPC, DPPE, DPPS). The phospholipid monolayers were examined with the application of Langmuir monolayer technique and Brewster angle microscopy (BAM) as well as grazing incidence x-ray diffraction (GIXD). The studies performed on phospholipid bilayer systems were related to dynamic light scattering and ζ potential measurements and the experiments with the calcein release and steady-state fluorescence anisotropy of DPH. The obtained results showed that the type of polar group of phospholipid as well as the addition of PEG-ylated lipid significantly changes the mol. organization of phospholipid monolayers. Moreover, these parameters also influence the properties of phospholipid bilayers such as size, surface charge, stability and permeability. In this study, phenolic compounds were extracted from three varieties of sesame seeds (black, brown, and white) to determine the antioxidant activities and bioaccessibility of selected phenolic compounds during in vitro digestion and colonic fermentation The SCFAs production was also estimated Black sesame seeds performed the highest total phenolic content (TPC) (2.69 mg GAE/g) and antioxidant capacities during gastrointestinal digestion. During colonic fermentation, black and brown sesame seeds exhibited relatively higher TPC (4.13 mg GAE/g) and antioxidant activities (DPPH: 8.6 mg TE/g; FRAP: 4.1 mg TE/g). Kaempferol was the lowest bioaccessible phenolic compound presented in all three sesame seeds, which relied more on the action of gut microbiota. White sesame seeds displayed higher production of individual and total SCFAs followed by black sesame seeds, which could be beneficial to gut health. Novelty impact statement : Black and brown sesame seeds showed relatively higher content of phenolic compounds and remarkable antioxidant potential during in vitro gastrointestinal digestion and colonic fermentation The bioaccessibility of phenolic compounds present in brown and white sesame seeds was relatively higher than in black sesame seeds. Most of the detected phenolic compounds in selected sesame seeds were fully bioaccessible after 24 h of colonic fermentation, except kaempferol. White and black sesame seeds showed higher production of SCFAs, which would be more beneficial to gut health. This study involved multiple reactions and reactants, such as (2S,3R,4S,5S,6R)-2-(3-Hydroxy-5-((E)-4-hydroxystyryl)phenoxy)-6-(hydroxymethyl)tetrahydro-2H-pyran-3,4,5-triol (cas: 27208-80-6Application of 27208-80-6).

(2S,3R,4S,5S,6R)-2-(3-Hydroxy-5-((E)-4-hydroxystyryl)phenoxy)-6-(hydroxymethyl)tetrahydro-2H-pyran-3,4,5-triol (cas: 27208-80-6) belongs to alcohols. Similar to water, an alcohol can be pictured as having an sp3 hybridized tetrahedral oxygen atom with nonbonding pairs of electrons occupying two of the four sp3 hybrid orbitals. A multistep synthesis may use Grignard-like reactions to form an alcohol with the desired carbon structure, followed by reactions to convert the hydroxyl group of the alcohol to the desired functionality.Application of 27208-80-6

(2R)-3-(((2-Aminoethoxy)(hydroxy)phosphoryl)oxy)propane-1,2-diyl dipalmitate (cas: 923-61-5) belongs to alcohols. Alcohols are among the most common organic compounds. They are used as sweeteners and in making perfumes, are valuable intermediates in the synthesis of other compounds, and are among the most abundantly produced organic chemicals in industry. Converting an alcohol to an alkene requires removal of the hydroxyl group and a hydrogen atom on the neighbouring carbon atom. Dehydrations are most commonly carried out by warming the alcohol in the presence of a strong dehydrating acid, such as concentrated sulfuric acid.Formula: C37H74NO8P

Activity of Na+/K+-ATPase in model lipid membrane at air-water interface was written by Matyszewska, Dorota;Zatloukalova, Martina;Bilewicz, Renata. And the article was included in Electrochimica Acta in 2019.Formula: C37H74NO8P The following contents are mentioned in the article:

The Na+/K+-ATPase pump was reconstituted in proteoliposomes composed of DPPC:DPPE, which were then spread at the air-H2O interface using Langmuir technique. In the presence of the enzyme in the liposomes the resulting lipidic layer at the air-H2O interface became more liquid The effect of Na+/K+-ATPase on the morphol. of Langmuir lipidic layers was monitored by Brewster angle microscopy, which showed the formation of lattice-like structure in between round-shaped lipid domains. The presence of protein stabilized the DPPC:DPPE mixed monolayer at the air-H2O interface, which was revealed by surface pressure measurements in time and ascribed to H bond network formation between the protein and the lipids. The activity of the protein embedded in the lipidic Langmuir layer was measured using spectroscopy and voltammetry. Free phosphate released from ATP reacted with ammonium molybdate and the blue α-Keggin anion formed was detected spectrophotometrically at a wavelength of 710 nm. The results based on spectroscopic assay were complemented with electrochem. methods. The activity of the enzyme could by switched off using the inhibitor – ouabain. The Na+/K+-ATPase activity (2.62 nmol mg-1 min-1) was similar to the activity of the protein solubilized using detergents (3.21 nmol mg-1 min-1). The slightly lower activity was ascribed to the defined orientation of the embedded protein mols. with respect to the air-H2O interface needed for its activity at the air-H2O interface.
