Although World Health Organization-approved emergency vaccines are available in many countries, the mortality rate from COVID-19 remains high due to the fourth or fifth wave and the delta variant of the coronavirus. Thus, an effective mechanistic investigation in treating this disease is urgently needed. In this work, we extracted phytochems. from two mangrove plants, Pistacia integerrima and Pandanus odorifer, assessing their potential actions against the receptor-binding domain (RBD) of the spike protein of SARS-CoV-2. The antioxidant activities of Pistacia integerrima leaves and fruits were 142.10 and 97.13 μg/mL, resp., whereas Pandanus odorifer leaves and fruits were 112.50 and 292.71 μg/mL, resp. Furthermore, leaf extracts from both plants had lower cytotoxicity against Artemia salina than fruit extracts Gas chromatog.-mass spectrometry anal. revealed a total of 145 potential phytochems. from these extracts Three phytochems., 28-demethyl-beta-amyrone, 24-Noroleana-3,12-diene, and stigmasterol, displayed binding free energy values of – 8.3, -7.5, and – 8.1 Kcal/mol, resp., in complexes with the spike protein of SARS-CoV-2. The root-mean-square deviation, solvent-accessible surface area, radius of gyration, root-mean-square fluctuations, and hydrogen bonds were used to ensure the binding stability of the docked complexes in the atomistic simulation. Thus, wet-lab validations are necessary to support these findings.

Kruk, Jerzy;Gabruk, Michal;Kryszczak, Marian;Wasik, Piotr research published 《 Protochlorophylls in Cucurbitaceae – Distribution, biosynthesis and phylogeny》, the research content is summarized as follows. Using high-resolution chromatog. we resolved monovinyl (MV)- and divinyl (DV)-protochlorophylls (Pchls) and detected all of their side-chain homologues in the inner seed coat of Cucurbita maxima, C. pepo and their varieties. Furthermore, we analyzed other less common representatives of the Cucurbitaceae family that were found to accumulate mostly MV-Pchls. All these species and varieties showed the characteristic composition of individual Pchls. Addnl., we also detected all of the corresponding protopheophytins, which accounted for between 1.1 and 35.5% of Pchls and are supposed to be degradation products of Pchls, formed during seed storage. A pigment composition anal. of C. maxima seedlings performed during deetiolation revealed that chlorophyll (Chl) a content increased gradually, while the levels of Pchl-GG and Chl-GG, a precursor of Chl a, were low and did not change significantly. However, when the seedlings were incubated with the precursor of tetrapyrrole biosynthesis (δ-aminolevulinic acid) before illumination, the Chl-GG content increased dramatically, while synthesis of Chl a was inhibited. These data indicate that in C. maxima seedlings, Chl a is not synthesized from geranylgeranyl-pyrophoshate via Chl-GG, but rather directly from phytyl-pyrophosphate. Phylogenetic anal. of Chl synthase genes revealed that many species, including those of the Cucurbitaceae family, have two or more Chl synthase genes. This suggests that these addnl. genes, at least in some species, might encode isoforms involved in Pchl synthesis.

Kont, Riin;Pihlajaniemi, Ville;Niemela, Klaus;Kuusk, Silja;Marjamaa, Kaisa;Valjamae, Priit research published 《 H2O2 in Liquid Fractions of Hydrothermally Pretreated Biomasses: Implications of Lytic Polysaccharide Monooxygenases》, the research content is summarized as follows. Lytic polysaccharide monooxygenases (LPMOs) are important players in enzyme-aided valorization of lignocellulose. However, the recently discovered dependency of LPMO catalysis on H2O2 along with H2O2-caused inactivation of the enzyme calls for an in-depth understanding of the levels and the dynamics of H2O2 in various streams of the processing of lignocellulosic biomass. Using an LPMO-based sensitive detection, we assessed the dynamics of H2O2 in the liquid fractions (LFs) of hydrothermally pretreated wheat straw (agricultural residue), birch (hardwood), and pine (softwood). Upon contact with air, H2O2 was formed in the LFs of all biomasses. The initially high rate of H2O2 formation decayed with the half-life around 1-2 h to a low but stable plateau value. The rates of H2O2 formation were much higher than the rates of its accumulation, suggesting that H2O2 is an intermediate in aerobic oxidation of the compounds in LFs. Although the general traits were similar, LFs of different biomasses had different rates of H2O2 formation and accumulation. LFs of different biomasses also differed by their effect on the enzymic degradation of cellulose. Compositional analyses revealed a number of different compounds that were formed and disappeared upon aerobic oxidation of LFs.

The liver plays critical roles to maintain homeostasis of living organisms and is also a major target organ of chem. toxicity. Meanwhile, nuclear receptors (NRs) are known to regulate major liver functions and also as a critical target for hepatotoxic compounds In this study, we established mammalian one-hybrid assay systems for five rat-derived NRs, namely PXR, PPARα, LXRα, FXR and RXRα, and evaluated a total of 326 compounds for their NR-activating profiles. Then, we assessed the association between their NR-activating profile and hepatotoxic endpoints in repeated-dose toxicity data of male rats from Hazard Evaluation Support System. In the in vitro cell-based assays, 68, 38, 20, 17 and 17 compounds were identified as positives for PXR, PPARα, LXRα, FXR and RXRα, resp. The association analyses demonstrated that the PXR-pos. compounds showed high frequency of endpoints related to liver hypertrophy, such as centrilobular hepatocellular hypertrophy, suggesting that PXR activation is involved in chem.-induced liver hypertrophy in rats. It is intriguing to note that the PXR-pos. compounds also showed statistically significant associations with both prolonged activated partial thromboplastin time and prolonged prothrombin time, suggesting a possible involvement of PXR in the regulation of blood clotting factors. Collectively, our approach may be useful for discovering new functions of NRs as well as understanding the complex mechanism for hepatotoxicity caused by chem. compounds

Due to a lack of economic incentive to develop new drugs, current treatments have severe limitations in terms of safety, efficacy, and ease of administration. In an effort to develop new HAT therapeutics, author report the structure-activity relationships around T. brucei for a series of benzoxazepinoindazoles previously identified through a high-throughput screen of human kinase inhibitors, and the subsequent in vivo experiments for HAT. Author identified compound I (R1 = H; R2 = Me; R3 = NH2) , which showed an improved kinase selectivity profile and acceptable pharmacokinetic parameters, as a promising lead. Although treatment with I (R1 = H; R2 = Me; R3 = NH2) cured 60% of mice in a systemic model of HAT, the compound was unable to clear parasitemia in a CNS model of the disease. Two implementations are employed, the direct and indirect methods. Reference of 16545-68-9

Kleinpeter, Erich;Krueger, Stefanie;Koch, Andreas research published 《 Anisotropy Effect of Three-Membered Rings in 1H NMR Spectra: Quantification by TSNMRS and Assignment of the Stereochemistry》, the research content is summarized as follows. The spatial magnetic properties (through space NMR shieldings, TSNMRSs) of cyclopropane; of the heteroanalogous oxirane, thiirane, and aziridine; and of various substituted mono-, dis-, and tris-cyclic analogs were computed by the GIAO perturbation method employing the nucleus independent chem. shift (NICS) concept and visualized as iso-chem.-shielding surfaces (ICSSs) of various size and direction. The TSNMRS values, thus obtained, can be employed to visualize the anisotropy (ring current) effect of the cyclopropane ring moiety. This approach was employed to qualify and quantify substituent influences and contributions of appropriate ring heteroatoms O, NH, and S on the anisotropy (ring current) effect of three-membered ring moieties, and to assign the stereochem. of mono-, bis-, and tris-cyclic structures containing cyclopropane as a structural element. Characteristic examples are included.

72824-04-5, Allylboronic acid pinacol ester is a useful research compound. Its molecular formula is C9H17BO2 and its molecular weight is 168.04 g/mol. The purity is usually 95%.
Allylboronic acid pinacol ester is an allylation reagent that is used to produce aldehydes from ketones. It reacts with water, yielding the desired product and formaldehyde as a byproduct. The reaction proceeds through a sequence of steps, in which the boronate ester first reacts with water to form an allylboronate ion and hydrogen gas. This intermediate then reacts with potassium t-butoxide to produce the desired allyl alcohol and potassium borohydride. Finally, the palladium complex catalyst reduces the carbonyl group of the starting material, converting it into an aldehyde. Allylboronic acid pinacol ester is commercially available as a white solid, but can also be synthesized from 2-chloro-5-pinacolylborane (pinacol) in high yield using catalytic cross coupling reactions., Related Products of 72824-04-5

With respect to acute toxicity, simple alcohols have low acute toxicities. Doses of several milliliters are tolerated. 72824-04-5, formula is C9H17BO2, For pentanols, hexanols, octanols and longer alcohols, LD50 range from 2–5 g/kg (rats, oral). Ethanol is less acutely toxic. All alcohols are mild skin irritants. Related Products of 72824-04-5

Kitson, Russell R. A.;Moody, Christopher J. research published 《 Synthesis of novel geldanamycin derivatives》, the research content is summarized as follows. The toxicity associated with the geldanamycin (I) family of benzoquinone ansamycins when used as heat shock protein-90 inhibitor mol. therapeutics is ameliorated by substitution at the 19-position. The resulting 19-substituted derivatives have greater potential for success in oncol. clin. trials and for other medicinal purposes such as the treatment of neurodegenerative conditions. Having overcome hurdles associated with the sensitivity and complexity of these mols., through a variety of synthetic approaches, the synthesis of a series of 19-substituted geldanamycin derivatives is reported herein using optimized Stille and Suzuki coupling reactions. Further compounds were accessible via copper-mediated coupling and nucleophilic addition reactions. He new compounds are of significant medicinal interest, in view of their significantly reduced toxicity previously observed for this class of substrate compared to their 19-unsubstituted counterparts that have been evaluated in the clinic.
