Purpose By increasing circulating free fatty acids and the rate of fatty acid oxidation, obesity decreases glucose oxidation and myocardial tolerance to ischemia. Partial inhibition of fatty acid oxidation may improve myocardial tolerance to ischemia/reperfusion (I/R) in obesity. We assessed the effects of oxfenicine treatment on post ischemic cardiac function and myocardial infarct size in obese rats. Methods Male Wistar rats were fed a control diet or a high calorie diet which resulted in diet induced obesity (DIO) for 16 wk. Oxfenicine (200 mg/kg/day) was administered to control and DIO rats for the last 8 wk. Isolated hearts were perfused and infarct size and post ischemic cardiac function was assessed after regional or global ischemia and reperfusion. Cardiac mitochondrial function was assessed and myocardial expression and activity of CPT-1 (carnitine palmitoyl transferase-1) and IRS-1 (insulin receptor substrate-1) was assessed using Western blot anal. Results In the DIO rats, chronic oxfenicine treatment improved post ischemic cardiac function and reduced myocardial infarct size after I/R but had no effect on the cardiac mitochondrial respiration. Chronic oxfenicine treatment worsened post ischemic cardiac function, myocardial infarct size and basal mitochondrial respiration in control rat hearts. Basal respiratory control index (RCI) values, state 2 and state 4 respiration rates and ADP phosphorylation rates were compromised by oxfenicine treatment. Conclusion Chronic oxfenicine treatment improved myocardial tolerance to I/R in the obese rat hearts but decreased myocardial tolerance to I/R in control rat hearts. This decreased tolerance to ischemia of oxfenicine treated controls was associated with adverse changes in basal and reoxygenation mitochondrial function. Oxidative damage was estimated by measuring thiobarbituric acid reactive substances (TBARS). IPOC improved contractile recovery and cell viability in the fed but attenuated them in the fasted hearts. In both groups ischemia lowered glycogen. IPOC further reduced it. Triacylglycerol remained unchanged during ischemia-reperfusion in both groups, but triacylglycerol mobilization was activated by IPOC in the fasted group. ATP was increased by IPOC in the fed hearts, but lowered in the fasted ones, which appeared to be associated with the rates of ATP synthesis in isolated mitochondria. In the fed hearts IPOC raised glucose-6-phosphate dehydrogenase activity and GSH/GSSG, and lowered TBARS. These results suggest that IPOC effects are associated with changes in the ATP supply, mobilization of energy sources and glutathione antioxidant ratio. Tertiary amines are being considered as absorption solvents for post-ombustion CO2 capture, but their potential to form harmful byproducts nitrosamines is yet to be evaluated. This study examined the factors affecting the formation of nitrosamines from tertiary alkanolamines under simulated desorber conditions and the effects of amine structural characteristics. Total nitrosamine formation from tertiary alkanolamine was determined to be 1st-order with respect to nitrite concentration and the absorbed CO2, but was 0-order with respect to amine concentration in the range of 0.5-2.5M. Tertiary alkanolamines formed less nitrosamine than their secondary amine analogs. For tertiary alkanolamines with the same number of 2-hydroxyethyl groups, smaller steric hindrance resulted in more nitrosamine formation and higher yields based on nitrite consumption. The anal. of specific nitrosamines revealed that the cleavage of 2-hydroxyethyl group was preferred over demethylation, but comparable to de-ethylation. Side chain structure and number of OH- groups in primary, secondary, and tertiary amines effect on CO2 absorption and desorption kinetics, equilibrium loads, heat duty, cyclic capacity, heat of absorption, and pKa were assessed and used to develop rational criteria to select components to formulate an optimum amine blend. Based on these criteria, amines which had a combination of high absorption and desorption parameters were selected. Their mixing ratios and concentrations were varied to obtain best overall performance. Results showed that in comparison with their straight-chain analogs, steric hindrance present in branched-chain alkanolamines resulted in much faster desorption rate, higher CO2 solubility and cyclic capacity, and much lower heat duty for solvent regeneration, but just a slight decrease in CO2 absorption rate. Developed criteria resulted in formulating an excellent bi-solvent aqueous amine blend (comprised of 2 M BEA + 2 M AMP), shown to have outstanding desorption characteristics/heat duty and very good absorption characteristics. Also, this work developed a non-trial-and-error procedure to determine heat of CO2 absorption based on Gibbs-Helmholtz equation. It showed the CO2 absorption rate and heat of CO2 absorption may not necessarily be proportional to the heat of absorption and heat duty, resp. Significantly dysregulated expressions of several metabolites were observed in the intergroup comparisons of the PCOS, POF, and healthy control groups. Furthermore, 6 DEMs were most frequently observed among the three groups. The expressions of these DEMs were not only directly correlated but also exhibited potential significance in patients with PCOS and POF. Novel metabolites with up/downregulated expressions can be discovered in patients with PCOS and POF using serum-based metabolomics; these metabolites show good diagnostic performance and can act as effective biomarkers for the early detection of PCOS and POF. We studied 112 subjects with a high CV risk profile, subdividing them according to the presence (CAD/High-RFs) or absence of CAD (No-CAD/High-RFs), assessed by computed tomog. angiog. The metabolic differences between the two groups were identified by gas chromatog.-mass spectrometry. Characteristic patterns and specific metabolites emerged for each of the two phenotypic groups: high concentrations of pyruvic acid, pipecolic acid, p-cresol, 3-aminoisobutyric acid, isoleucine, glyceric acid, lactic acid, sucrose, phosphoric acid, trimethylamine-N-oxide, 3-hydroxy-3-methylglutaric acid, erythritol, 3-hydroxybutyric acid, glucose, leucine, and glutamic acid; and low concentrations of cholesterol, hypoxanthine, glycerol-3-P, and cysteine in the CAD/High-RFs group vs the No-CAD/High-RFs group. Our results show the existence of different metabolic profiles between patients who develop CAD and those who do not, despite comparable high CV risk profiles.
