Abstract: The use of non-conventional yeast species to obtain interesting flavors and aromas has become a new trend in the fermented beverages industry. Among such species, Brettanomyces bruxellensis (B. bruxellensis) has been reported as capable of producing desirable or at least singular aromas in fermented beverages like beer and wine. However, this yeast can also produce an aromatic defect by producing high concentrations of phenolic compounds like, 4-ethylguaiacol and particularly 4-ethylphenol (4-EP). In the present study, we designed a mutant screening method to isolate B. bruxellensis mutants with reduced 4-EP production More than 1000 mutants were screened with our olfactory screening method, and after further sensory and chem. anal. we were able to select a B. bruxellensis mutant strain with a significant reduction of 4-EP production (more than threefold) and less phenolic perception. Notably, the selected strain also showed higher diversity and concentration of Et esters, the most important group of odor active compounds produced by yeasts. Based on these results, we consider that our selected mutant strain is a good candidate to be tested as a non-conventional yeast starter (pure or in co-inoculation) to obtain wines and beers with novel aromatic properties. Doenjang is a traditional Korean fermented soybean-based food that is now produced industrially as traditional Doenjang and modernized Doenjang, depending on the conditions of the manufacturing process. In this study, the effect of the production process on Doenjang metabolites was analyzed using untargeted capillary electrophoresis time-of-flight mass spectrometry (CE-TOF/MS). Of the 247 metabolites detected in 19 traditional and three modernized Doenjang samples, the level of 55 metabolites were significantly different between the two production groups. The S-plot of the orthogonal projection to latent structures-discriminant anal. (OPLS-DA) revealed that nine compounds (tryptamine, 2-phenylethylamine, citrulline, gamma-aminobutyric acid, putrescine, tyramine, 2-aminoisobutyric acid, stachydrine, and N5-ethylglutamine) were highly distributed in the traditional group, and six compounds (arginine, citric acid, choline, cytidine, hypoxanthine, and glucaric acid) were considered distinguishable metabolites of the modernized group. Microbial community anal. indicated that the levels of these metabolites were significantly altered by the presence of Bacillus spp., Enterococcus faecium, Tetragenococcus halophilus, Aspergillus oryzae, Penicillium spp., and Saccharomyces cerevisiae. These findings will give better understanding of the influence of the manufacturing process on Doenjang production in terms of both microbial activity and metabolite profiles. Improving hydrophilicity is a key factor for enhancing the biocompatibility of polymer surfaces. Nevertheless, previous studies have reported that poly(2-methoxyethyl acrylate) (PMEA) surfaces demonstrate markedly better biocompatibility than more hydrophilic poly(2-hydroxyethyl methacrylate) (PHEMA) surfaces. In this work, the origins of the excellent biocompatibility of the PMEA surface are investigated using mol. dynamics (MD) simulations of simplified binary mixtures of acrylate/methacrylate trimers and organic solvents, with n-nonane, 1,5-pentanediol, or 1-octanol serving as the probe organic foulants. The interactions between the acrylate/methacrylate trimers and solvent mols. were evaluated by calculating the radial distribution function (RDF), with the resulting curves indicating that the 2-methoxyethyl acrylate (MEA) trimer has a lower affinity for n-nonane mols. than the 2-hydroxyethyl methacrylate (HEMA) trimer. This result agrees with the exptl. consensus that the biocompatibility of PMEA surfaces is better than that of PHEMA surfaces, supporting the hypothesis that the affinity between an acrylate/methacrylate trimer and a foulant mol. in a simplified binary mixture is a significant factor in determining a surfaces antifouling properties. The RDF curves obtained for the other two solvent systems exhibited behavior that further highlighted the advantages of the PMEA surfaces as biocompatible polymers. In addition, the validity of employing the second virial coefficient (B2) as a predictor of antifouling properties was explored. The order of the B2 values of different binary mixtures indicated that the MEA trimers have the lowest affinities with n-nonane mols., which confirms that although PMEA is more hydrophobic than PHEMA, it exhibits better biocompatibility. This anal. demonstrates that the MEAs weaker miscibility with nonpolar foulants contributes to the excellent biocompatibility of PMEA. Thus, B2 is a promising criterion for assessing the miscibility between acrylate/methacrylate materials and nonpolar organic foulants, which indicates the potential for predicting the antifouling properties of acrylate/methacrylate polymer materials by evaluating the value of B2. Meat production performance is one of the most important factors in determining the economic value of poultry. Peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PPARGC1A) as a transcriptional coactivator has been found to be widely involved in a series of biol. processes. However, PPARGC1A is still poorly understood in chickens. In this manuscript, we reported that PPARGC1A was highly expressed in slow-twitch myofibers. PPARGC1A facilitated mitochondrial biogenesis and regulated skeletal muscle metabolism by mediating the flux of glycolysis and the TCA cycle. Gain- and loss-of-function analyses revealed that PPARGC1A promoted i.m. fatty acid oxidation, drove the transformation of fast-twitch to slow-twitch myofibers, and increased chicken skeletal muscle mass. Mechanistically, the expression level of PPARGC1A is regulated by miR-193b-3p. Our findings help to understand the genetic regulation of skeletal muscle development and provide a mol. basis for further research on the antagonism of skeletal muscle development and fat deposition in chickens. Present study assessed antioxidant and anti-inflammatory properties of Samsum ant venom (SAV) extract in carbon tetrachloride (CCL4)-induced spleen toxicity. Toxicity and oxidative stress were measured in four exptl. groups: a neg. control group without any treatment, a pos. control group (CCl4-treated rats; a single dose of 1 mL/kg CCL4), an exptl. group of CCl4-treated rats co-treated daily with SAV (100 μl), and a group to determine safe use with rats treated only with SAV (100 μl) daily for 3 wk. CCl4-treatment led to an elevation in toxicity and oxidative stress. CCl4 significantly elevated malondialdehyde (MDA) levels, as well as expression of inhibitor of κB (IκB) and tumor necrosis factor-α (TNF-α) proteins. On the other hand, a decrease in glutathione (GSH) and catalase (CAT) levels were detected in CCl4-treated rats. Co-treatment with SAV was found to reduce these inflammatory and oxidative parameters. SAV elucidated a significant recovery of MDA concentration as well as a significant restoration in GSH levels compared to CCl4-treated rats; however, SAV increased CAT levels compared to normal rats. Hence, SAV was found to restore splenomegaly induced in CCl4-treated rats. Histopathol. anal. also favored the biochem. anal. showing improvement in splenic architecture in CCl4 and SAV co-treated rats. The antioxidant properties of SAV may potentially enhance anti-inflammatory actions and improve spleen structure and function in CCl4-challenged rats. In the present study, the lipid profile from the fat body and eggs of Rhipicephalus microplus was evaluated after exposure of engorged females to (E)-cinnamaldehyde and α-bisabolol, substances which have acaricide potential according to the literature. Engorged females collected from artificially infested cattle were immersed in a concentration of 10.0 mg/mL of each substance. Dissection of the female fat bodies was performed at different times (72 h and 120 h), for subsequent lipid extraction In addition, on the fifth day of oviposition, were collected 50.0 mL50.0 mL aliquots of the egg mass of each treatment to perform the same lipid extraction procedure. To assess the lipid profiles, the samples were submitted to the thin layer chromatog. (TLC) and gas chromatog.-mass spectrometry (GC-MS) anal. Furthermore, an in silico anal. was performed using PASS online software to predict the possible mol. targets of (E)-cinnamaldehyde and α-bisabolol. As result, the main lipids identified from the fat body were triacylglycerides, fatty acids, and cholesterol, whereas, triacylglycerides (TAG), fatty acids (FA), and cholesterol (CHO) and cholesterol esters (CHOE), were identified in the eggs. The results also showed a significant increase (p < 0.05) of CHO in the fat body in the group exposed to (E)-cinnamaldehyde at 72 h (0.12 μg/fat body) and 120 h (0.46 μg/fat body), in the eggs from females treated with this same substance, there was a significant reduction (p < 0.05) in the amount of CHO (0.21 μg), compared to the water control group (0.45 μg). In the GC-MS technique, 5 chem. classes were found, and variations were observed between these substances, mainly hydrocarbons and steroids, in the different groups, and (E)-cinnamaldehyde promoted the greatest changes. From the predictions of the in silico study, 38 and 20 targets were selected, resp., which are mainly related to alterations in lipid metabolism, immune system and nervous system. This study provides the first report of changes in lipid metabolism of R. microplus exposed to (E)-cinnamaldehyde and α-bisabolol, as well as presenting possible activity on the mol. targets of these substances, expanding knowledge for the potential use of these compounds in the development of botanical acaricides. Tuo-Min-Ding-Chuan decoction (TMDCT) is a Traditional Chinese Medicine (TCM) formula consisting of twelve herbs that can relieve the symptoms and treat allergic asthma. Yet, the underlying mechanism of action is still unclear. In this study, we investigated the effect of TMDCT in regulating Treg/Th17 cells immune balance and explored potential metabolic and gut biomarkers associated with Treg and Th17 cells in eosinophilic asthma mice treated by TMDCT. We found that TMDCT increases Treg cells percentage and decreases Th17 cells percentage in the ovalbumin (OVA) -induced eosinophilic asthma mice model. Furthermore, Imidazoleacetic acid, DL-glutamine, L-pyroglutamic acid, 2-deoxy-D-glucose were preliminary identified as biomarkers in plasma metabolites treated by TMDCT, meanwhile genus Desulfovibrio, genus Butyricimonas and genus Prevotella 9 were preliminary identified as gut microbiota biomarkers after TMDCT treatment.
