Here, we report the nitric oxide dioxygenation (NOD) reactions of NiIII-peroxo, [(12-TMC)NiIII(O22-)]+ (NiIII-O22-, 1), and CoIII-peroxo, [(12-TMC)CoIII(O22-)]+ (CoIII-O22-, 2), complexes bearing N-tetramethylated cyclam (TMC) ligands. The reaction of complex 1 with nitric oxide (NO) generates NiII-nitrato complex [(12-TMC)NiII(NO3-)]+ (NiII-NO3-, 3). However, in contrast to the NOD reaction of 1, compound 2 produces CoII-nitrito complex, [(12-TMC)CoII(NO2-)]+ (CoII-NO2-, 4), with O2 gas formation. Results suggested the formation of a presumptive Ni-peroxynitrite ([Ni-PN]+, [Ni-ONOO-]+) intermediate, in the NOD reaction of 1, prior to the generation of NiII-NO3- (3). Although the end product of the reaction of 2 with NO is different from that of 1, the mechanistic study explored by using 2,4-di-tert-butylphenol (2,4-DTBP) suggested that the reaction of 2 with NO also occurs via a presumed Co-peroxynitrite ([Co-PN]+, [Co-ONOO-]+) intermediate. Furthermore, exploring these NOD reactions and tracking the oxygen atom using 18O-labeled hydrogen peroxide (H218O2) revealed that two oxygen atoms of 3 (NiII-18O2N16O-) and one oxygen atom of 4 (CoII-18ON16O-) are derived from the peroxo (O22-) moieties of 1 and 2, resp. This work is the first-ever report which showed the exact NO dioxygenase (NOD) enzyme catalytic cycle. Furthermore, we have also explored the NO activation of 3 and 4, and surprisingly, we observed the formation of Co-nitrosyl (5) from 4 upon reaction with NO; in contrast, 3 was unreactive towards NO. Immersion of the mixed SAM in 0.1M HClO4 at open circuit potential or oxidation in 0.1M H2SO4 did not produce this surface-bound redox couple. The monolayers were thoroughly characterized by XPS and polarization modulation IR reflection absorption spectroscopy (PM IRRAS) after the different preparation steps indicating only minor changes in the overall composition of the monolayer, in particular, the preservation of the heteroatoms. The new redox couple is likely a diimine, in agreement with its ability to bind nucleophiles such as anilines by conjugate addition that could be followed by multicycle voltammetry and XPS. Although current antiretroviral therapy can control HIV-1 replication and prevent disease progression, it is not curative. Identifying mechanisms that can lead to eradication of persistent viral reservoirs in people living with HIV-1 (PLWH) remains an outstanding challenge to achieving cure. Utilizing a phenotypic screen, we identified a novel chem. class capable of killing HIV-1 infected peripheral blood mononuclear cells. Tool compounds ICeD-1 and ICeD-2 (“inducer of cell death-1 and 2”), optimized for potency and selectivity from screening hits, were used to deconvolute the mechanism of action using a combination of chemoproteomic, biochem., pharmacol., and genetic approaches. We determined that these compounds function by modulating dipeptidyl peptidase 9 (DPP9) and activating the caspase recruitment domain family member 8 (CARD8) inflammasome. Efficacy of ICeD-1 and ICeD-2 was dependent on HIV-1 protease activity and synergistic with efavirenz, which promotes premature activation of HIV-1 protease at high concentrations in infected cells. This in vitro synergy lowers the efficacious cell kill concentration of efavirenz to a clin. relevant dose at concentrations of ICeD-1 or ICeD-2 that do not result in complete DPP9 inhibition. These results suggest engagement of the pyroptotic pathway as a potential approach to eliminate HIV-1 infected cells. Hesperidin and narirutin are the major flavanones present in orange juice, and they are associated with a reduction in risk of cardiometabolic disease. However, there is heterogeneity in their biol. responses, which is partly due to the large interindividual variation in these flavonoids bioavailability. We investigated the relation between interindividual variability in the excretion of phase II conjugates and gut-derived phenolic acids, and cardiometabolic biomarkers response. Seventy-four subjects, both men and women, were included in a single-arm study. Over the 60 days, volunteers consumed 500 mL of orange juice daily. All measurements and blood collections were performed before and after the intervention period. Moreover, 24 h urine collection was performed after first consumption. Individuals were stratified according to the excretion of phase II conjugates and, for the first time, according to phenolic acids in high, medium, and low excretors. Furthermore, for the first time, the ratio between phenolic acids and flavanones-phase II conjugates has shown groups with different metabolization patterns. Groups with a low or intermediate ratio, corresponding to a higher amount of phase II conjugates excreted, showed a significant reduction in body fat % and blood pressure. This finding suggests that these improvements could be associated in a major way to flavanones-phase II conjugates, as well as to phenolic acids and stratification of volunteers according to metabolite excretions could be a good strategy to better understand the effects of orange juice on metabolism and health.
