Xylem sap is the major transport route for nutrients from roots to shoots. In the present study, we investigated how variations in nitrogen (N) nutrition affected the metabolome and proteome of xylem sap and the growth of the xylem endophyte Brennaria salicis, and we also report transcriptional re-wiring of leaf defenses in poplar (Populus 3 canescens). We supplied poplars with high, intermediate or low concentrations of ammonium or nitrate. We identified 288 unique proteins in xylem sap. Approx. 85% of the xylem sap proteins were shared among ammonium- and nitrate-supplied plants. The number of proteins increased with increasing N supply but the major functional categories (catabolic processes, cell wallrelated enzymes, defense) were unaffected. Ammonium nutrition caused higher abundances of amino acids and carbohydrates, whereas nitrate caused higher malate levels in xylem sap. Pipecolic acid and N-hydroxypipecolic acid increased, whereas salicylic acid and jasmonoyl-isoleucine decreased, with increasing N nutrition. Untargeted metabolome analyses revealed 2179 features in xylem sap, of which 863 were differentially affected by N treatments. We identified 124 metabolites, mainly from specialized metabolism of the groups of salicinoids, phenylpropanoids, phenolics, flavonoids, and benzoates. Their abundances increased with decreasing N, except coumarins. Brennaria salicis growth was reduced in nutrient-supplemented xylem sap of low- and high- NO3–fed plants compared to that of NH4+-fed plants. The drastic changes in xylem sap composition caused massive changes in the transcriptional landscape of leaves and recruited defenses related to systemic acquired and induced systemic resistance. Our study uncovers unexpected complexity and variability of xylem composition with consequences for plant defenses. Abstract: Green synthesis of nanoparticles is an important tool to reduce the harmful effects associated with traditional methods. In the present investigation, we have synthesized gold nanoparticles (AuNPs) using aqueous extract prepared from fresh aerial parts (leaf and stem) of Vernonia cinerea as bioreducing agent. The visual indication of change in color from pale yellow to brown to ruby-red indicated the successful formation of the AuNPs. Characterization of nanoparticles was carried out by UV-visible spectroscopy, X-ray crystallog. (XRD), Transmission electron microscopy (TEM) and Energy dispersive X-ray anal. (EDX). UV-Vis spectra showed a specific peak at 546 nm which was the initial confirmation of the biosynthesized AuNPs. TEM images showed spherical and triangular shape of AuNPs with an average size of 25 nm. From FTIR spectrum, different functional groups were identified that could be responsible for the formation, stabilization, and capping of biosynthesized AuNPs. Aqueous plant extract and biosynthesised AuNPs were sep. tested for their antimicrobial activity against six bacterial strains and four fungal strains. Biosynthesised AuNPs (2 mg/mL) showed significantly high zone of inhibition against the selected bacterial strains as compared to the aqueous plant extract Maximum zone of inhibition (18.2 mm) was observed with AuNPs against Streptococcus pyogenes whereas comparatively less value (12.5 mm) was recorded with the plant extract Interestingly, the inhibitory activity observed against bacterial strains was even better than ampicillin. Antifungal activity recorded with AuNPs (5 mg/mL) was maximum (17.4 mm) against R. oryzae and it was higher than pos. control (17.00 mm) and plant extract (13.2 mm). The present study clearly showed that AuNPs coated with Vernonia cinerea extract were as good as pos. control in inhibiting bacterial and fungal growth. In this work, poly(urethane-imide)s (PUIs) with Lewis bases were synthesized in two steps only from various functional diols (N-methyldiethanolamine, N-tert-butyldiethanolamine, N-phenyldiethanolamine and 1,4-diethanolpiperazine) containing one or two tertiary amine groups with different chem. structures. The synthesis led to high yields, high mol. weights and a good control of the PUI amine content. These good film-forming polymers were then used as SO2 sensitive coatings on Love surface acoustic wave (L-SAW) micro-sensors. The phys. and mech. properties required for an optimal micro-sensor design were determined by High Resolution Brillouin Spectroscopy of PUI thin layers. Three-layer micro-sensors were developed by respecting the conditions for the L-SAW generation. The three-layer structure included Quartz ST-90 as the piezoelec. substrate, a ZnO guiding layer and a PUI sensitive layer. These micro-sensors were weakly sensitive to temperature and thus compatible with the targeted application. The exptl. results for SO2 detection showed that all the PUI coatings greatly improved the sensitivity compared to a micro-sensor without polymer coating. The basicity of the amine groups was not determining for the micro-sensor sensitivity to the SO2 acid gas and the key factor was their steric hindrance. Finally, the micro-sensor sensitivity increased with the accessibility of the amine groups in the order Piperazine-diol < tBu-DEA ≃ Ph-DEA ≪ MDEA. The final goal of this work is to provide biomarkers that can help to better understand how the COVID-19 illness evolves and to predict how a patient could progress based on the metabolites profile of plasma obtained at an early stage of the infection. In the present work, several metabolites were found as potential biomarkers to distinguish between the end-stage and the early-stage (or non-COVID) disease groups. These metabolites are mainly involved in the metabolism of carnitines, ketone bodies, fatty acids, lysophosphatidylcholines/phosphatidylcholines, tryptophan, bile acids and purines, but also omeprazole. In the present study, the activation of antioxidant machinery was observed at 8 and 24 h after 250 mM NaCl stress treatment. Further, GC-MS/MS based metabolomics was performed, independently in root (NR8 and NR24) and shoot (NS8 and NS24). A total of 53 and 62 metabolites were detected in roots and shoots, resp., that mainly included sugars, amino acids, organic acids and polyols. On the basis of statistical significance, sucrose and ethanolamine were identified as differentially altered metabolites (DAMs) shared under all the treatment conditions. Heat-map based clustering anal. substantiated the role of sugars and amino acids in NR8; while, sugars in NS8/NS24 treatments. For most of the DAMs, extent of change at NR24 was significantly dampened than NR8, which also coincided with reduced correlation strength between different metabolite groups, indicating partial metabolite restoration. The enrichment anal. identified sugars, amino acid and TCA cycle metabolism as major pathways altered under salt stress conditions. A total of 30 metabolomic datasets from 7 different halophytes, including the present study, were utilized to perform meta-anal. On the basis of cumulative pathway score (CPS) of both root and shoot, aspartate-alanine-glutamate (290.62) was identified as the most significant pathway followed by arginine biosynthesis (0.62) and glyoxylate-dicarboxylate metabolism (0.46) that significantly contributed to salt-adaptation in halophytes.
