Innovative technologies as ultrasound-assisted extraction (UAE) (550 W, 60% amplitude, 50°C) or subcritical water extraction (SWE) (200°C, 11.7 MPa) were more effective than hot water extractions to obtain β-glucan-enriched fractions from shiitake mushrooms. UAE required longer extraction time (60 min) than SWE (15 min). Combination of UAE + SWE or pre-treatment of the raw material with supercritical CO2 (SFE) (40°C, 35 MPa, 3 h) before both extractions yielded extracts containing larger β-glucan concentrations Fluorimetric/colorimetric determinations indicated that obtained fractions contained (1 → 3)- and (1 → 3),(1 → 6)-β-glucans. NMR confirmed their presence as well as (1 → 3)-α-glucans and heteropolymers including mannose and galactose. SWE (15 min), SFE + SWE or UAE + SWE extracts showed larger glucose levels and lower mannose and galactose residues than the other extractions suggesting certain extraction specificity towards β-glucans. They also included more chitin-derivatives than UAE. The extracts obtained after combination of technologies partially retained their immunomodulatory properties but they showed high hypocholesterolemic activities according to in vitro studies.

Lugana and Verdicchio are two Italian white wines with a Protected Designation of Origin (PDO) label. These two wine types are produced in different regions using the same grape variety. The aim of this work is to investigate the existence of volatile chem. markers that could help to elucidate differences between Lugana and Verdicchio wines both at chem. and sensory levels. Thirteen com. wine samples were analyzed by Gas Chromatog.-Mass Spectrometry (GC-MS), and 76 volatile compounds were identified and quantified. Verdicchio and Lugana had been differentiated on the basis of 19 free and glycosidically bound compounds belonging to the chem. classes of terpenes, benzenoids, higher alcs., C6 alcs. and norisoprenoids. Samples were assessed by means of a sorting task sensory anal., resulting in two clusters formed. These results suggested the existence of 2 product types with specific sensory spaces that can be related, to a good extend, to Verdicchio and Lugana wines. Cluster 1 was composed of six wines, 4 of which were Lugana, while Cluster 2 was formed of 7 wines, 5 of which were Verdicchio. The first cluster was described as “”fruity””, and “”fresh/minty””, while the second as “”fermentative”” and “”spicy””. An attempt was made to relate anal. and sensory data, the results showed that damascenone and the sum of 3 of esters the Et hexanoate, Et octanoate and isoamyl acetate, was characterizing Cluster 1. These results highlighted the primary importance of geog. origin to the volatile composition and perceived aroma of Lugana and Verdicchio wines.

Aliphatic polyamides containing pendent allyl groups were prepared by anionic ring-opening copolymerization of ε-caprolactam with substituted lactam 3-(3-propenyl)-2-azepanone (1). Copolymerization experiments revealed that up to 11 mol % of 1 was integrated successfully into these novel polyamides, which ranged in mol. weight from 27 to 72 kDa. Relative to the well-known com. polyamide-6 (PA-6), the degree of crystallinity of the copolymers decreased with incorporation of functional monomer. Moreover, the pendent allyl groups afforded rapid access to numerous functional aliphatic polyamides, using photoinitiated thiol-ene chem., providing a pathway to cross-linked polyamide films and gels.

Wine production processes still rely on post-production evaluation and off-site laboratory analyses to ensure the quality of the final product. We report the selective hydroxylation of small alkanes with H2O2 catalyzed by an artificial P 450 peroxygenase system generated from engineered cytochrome P450BM3 variants in assistance with dual-functional small mol. (DFSM), in which DFSM acts as a general acid-base co-catalyst for activating H2O2. This peroxygenase system exhibited comparable catalytic turnover number (TON) to the fungal peroxygenase AaeUPO, the only known H2O2-dependent natural alkane hydroxylase. Moreover, when compared with evolved/engineered NADPH-dependent P 450 variants, the current system yielded similar or even better product formation rates (PFRs) but lower total TONs. The substitution of the highly conserved T268 with amino acids having hydrophobic side chains was identified to play critical roles in improving the hydroxylation activity of the DFSM-facilitated P450BM3 peroxygenase system, which is distinct from NADPH-dependent P 450 enzymes. These results offer useful insights into how to tune the catalytic functions and chem. of P 450 peroxygenases.

The effects of the carbonic maceration and conventional winemaking on the volatile composition and aromatic sensory characteristics of Tempranillo Blanco wines were studied for the first time, during three consecutive vintages. Relationships between instrumental (volatiles) and sensory variables were analyzed applying partial least squares regression (PLS). Carbonic macerated wines had higher contents of alcs. and carbonyl compounds, yet lower concentrations of C6 alcs. and volatile acids than wines conventionally produced. The Odor Activity Values (OAV) exhibited an increase in wines when carbonic maceration was applied. According to the geometric mean (% GM) obtained from aroma descriptors the effect of the winemaking process was significant for seed fruit, ripe fruit and floral notes. When subjected to PLS the data from the instrumental anal. yielded a satisfactory model for the prediction of aroma descriptors in this set of wines.

E-cigarette or vaping product use-associated lung injury (EVALI) is a serious pulmonary condition that is associated with the extended use of certain vaping products. EVALI was first characterized in the summer of 2019 and has since been reported in all 50 U.S. states. From August 2019 through June 2021, the New York State Department of Health has reported more than 197 confirmed cases emanating from all regions of the state. The Wadsworth Center at the New York State Department of Heath received vaping cartridges recovered from EVALI patients for chem. anal. of their contents. Untargeted anal. methods using gas chromatog.-mass spectrometry and liquid chromatog.-high-resolution mass spectrometry as well as targeted analyses for a variety of analytes including cannabinoids, pesticides, vitamin E acetate (VEA) and mycotoxins were used to characterize the composition of the vaping fluids and several com. vaping fluid additives. From the analyses of the 284 e-cigarette devices recovered from patients, 82 were found to be nicotine-containing pods, and 202 devices containing cannabis oil, apparently from unauthorized or black-market dealers. The fluids from the cannabis-oil cartridges tended to have lower levels of THCs (Δ9-tetrahydrocannabinol + Δ8-tetrahydrocannabinol) and total cannabinoids compared with those of com. produced formulations and contained significant levels of diluents including VEA, medium-chain triglycerides, polyethylene glycol, and castor oil. VEA was the diluent most frequently detected, which was present in 132 (65.3%) of the vaping fluids that contained cannabis oil. When present, VEA ranged from 2.0 to 67.8% of the total mass of the oil with a mean content of 37.0%. In some cases, two or three diluents were detected in the same sample. The ratio of VEA to THCs varied widely, from 0.07 to 5.34. VEA and specifically the high ratios of VEA to THCs in black-market vaping fluids may be causative in EVALI. The safety of addnl. components and additives that are present in vaping fluids are likewise of concern.

The cryoprotective effects of lactitol, maltodextrin, palatinit, polydextrose, trehalose on the protein denaturation of threadfin bream surimi during six months of frozen storage were investigated. The characteristics analyzed were Ca2+-ATPase activity, protein solubility, sulfhydryl (SH) content, electrophoresis, surface morphol., and thermal stability. The effectiveness of all cryoprotectants generally decreased as the storage time increased. Polydextrose was able to maintain Ca2+-ATPase activity, protein solubility, and SH content of 33.5%, 71.3%, 71.8%, resp. Sucrose was able to maintain Ca2+-ATPase activity, protein solubility, and SH content of 32.4%, 68.1%, 75.6%, resp. Electrophoresis showed that the myosin heavy chain (MHC) and actin band intensity almost disappeared by the end of storage. Anal. by SEM (SEM) demonstrated that the number of pores formed increased after storage. Thermal stability anal. showed a decrease in myosin but no significant change in actin content. This study suggests that polydextrose as a cryoprotectant is able to maintain protein from denaturation as the same as sucrose.
