There has been limited research done on the downstream processing of nanosuspensions into solid oral dosage forms. This paper demonstrates the bead layering process with a layering level at 150% and 240%, as well as the selection and justification of the outer phase excipients for tabletability and disintegrating properties. In a previous study, an itraconazole nanosuspension stabilized by SDS and HPMC E5 was layered onto sugar beads with coating polymer HPMC VLV. In the current study, compression studies with these layered beads utilizing the small bead size at 150% or 240% layering levels with outer phase cushioning excipients MCC, copovidone or isomalt were performed. Other excipients such as co-compressed crospovidone-PEG 4000; DCP functioning as a disintegrant; and HPC as a binder was also added. Target output variables were achieved with a balance between an adequate tensile strength and fast dissolution rate with a release of 99.0% (±1.0% SD) within 10 min, which is in accordance with the FDA guidance for dissolution testing. The results show that the compaction of nanosuspension-layered beads is a suitable process for processing an itraconazole nanosuspension into a solid dosage form such as a compacted tablet without compromising on drug release.

Fine size tuning and narrow size distributions are easily obtained by varying independently the flow rates of the injected fluids and the concentration of the dye stock solution The flash nanopptn. process employed herein is successfully applied to the fabrication of bicomposite FONs designed to allow energy transfer. Considerable enhancement of the emission signal of the energy acceptors is promoted and its origin is found to result from polarity rather than steric effects. Finally, we exploit the high spatial confinement encountered in FONs and their ability to encapsulate hydrophobic photosensitizers to induce photocrosslinking. An increase in the photocrosslinked FON stiffness is evidenced by measuring the elastic modulus at the nanoscale using at. force microscopy. These results pave the way toward the straightforward fabrication of multifunctional and mech. photoswitchable FONs, opening novel opportunities in sensing, multimodal imaging, and theranostics.

Transparent wood (TW) is an interesting polymer biocomposite with potential for buildings and photonics applications. TW materials need to be eco-friendly and readily processed with few defects, for high optical transmittance and low transmission scattering at wide angles (haze). Two wood templates with different lignin-content are impregnated with a new thiol-ene thermoset system. The more eco-friendly bleached wood template results in transparent wood with high optical transmission and much reduced transmission haze, due to strong reduction of interfacial air gaps. Characterization includes template composition, thiol-ene distribution, and polymerization in wood cell wall by EDX and confocal Raman microscopy, also NMR and DSC, tensile testing and FE-SEM fractog. for morphol. and wood/thiol-ene interface adhesion assessment. The wood template is a true nanocomposite with thiol-ene polymer located inside the nanoporous wood cell wall. Advanced TW applications require not only appropriate wood template modification and careful polymer matrix selection but also tailoring of the process to impregnation and polymerization mechanisms, in order to reduce optical defects.

Insect repellent textiles are currently being investigated as a complementary tool to control disease vectors, such as mosquitoes. The application of microencapsulated biopesticides to textiles is a promising low-toxicity alternative to other control methods, as microcapsules may prolong the release of the repellent agent. In this work, microcapsules containing two biopesticides, namely citronella essential oil and citriodiol were prepared and applied to cotton textiles using a variety of techniques. The aim of this work was to evaluate these functional textiles to select the most efficient, durable systems. Citriodiol-treated cotton presented a prolonged durability, and 100% repellency could be achieved for more than 30 days after padding onto cotton fabrics. It has to be pointed out that all these repellent textiles have been obtained by scalable, low-cost methods which would require no addnl. investment for textile finishing industries.

The aim of the present study is to evaluate the effect of a new formulation parameter on the characteristics of dosage form and manufacturing process. In the authors’ previous work the effects of isomalt and compression force on compaction and tablet properties were investigated. In this research paper the particles were agglomerated with purified water without using other binders. To set up the 33 type factorial design a new independent variable – mixing speed (X2) – was added to paracetamol:isomalt ratio in granules (X1) and to compression force during the tabletting process (X3). Each of these variables was examined at 3 levels to characterize their power and interactions. As granule’s properties mass flow, densities and particle size were investigated. From the aspect of compression process lubrication coefficient and friction work were the evaluated response variables. As tablet characteristics tablet resistance to crushing and tensile strength were studied. The results indicated that the agglomeration process did not need any other binders to obtain granules with good quality. Moreover, the addition of mixing speed as a new independent variable was appropriate, since it had significant effect among others on the particle size distribution of the granules and on tablet characteristics. According to the results of the tablet quality tests the optimal mixing speed at granulation is needed which offers not only sufficient homogeneity of the granules but also appropriate tablet characteristics. Furthermore, the enhancing effect of isomalt on the compaction of granules and on the tablet characteristics is observed

Isomalt, a sugar alc., was melt-extruded prior to compression in order to improve its tabletting properties. After fusion, crystalline isomalt was transformed into an amorphous form as shown by x-ray diffraction and DSC. The tabletting properties of amorphous isomalt were dramatically improved. Mixtures formulated with paracetamol (50%) and extruded isomalt yielded hard tablets. However, extruded isomalt powder showed agglomeration problems due to recrystallization of the amorphous phase into a stable crystalline form in the presence of atm. moisture. The evolution of the moisture content correlated well with the compressibility data. The tablets made of extruded isomalt powder had a lower friability in comparison to the tablets formulated with non-extruded isomalt powder. Their disintegration was fast and a rapid dissolution rate was recorded. Extruded isomalt displayed excellent tabletting properties; however, further experiments should be conducted to delay or even prevent recrystallization of amorphous isomalt.

It is well known in industrial applications involving powders and granular materials that the presence of electrostatic charges influences drastically the material flowing properties. The triboelec. charges are produced during flow at the contacts between the grains and at the contacts between the grains and the container. Unfortunately, the triboelec. effect is still poorly understood, even at the fundamental level. Therefore, the approach to solve practical problems is mostly empirical. Moreover, reproducible electrostatic measurements are difficult to perform. In the present study, the ability of a set of excipients and active pharmaceutical ingredients (APIs) to produce electrostatic charges during flow in contact with different materials is analyzed with a recently developed instrument called GranuCharge. While different excipients have almost the same triboelec. behavior and a low chargability, APIs show complex triboelec. properties. Some APIs charge a lot while other APIs charge less. Afterward, the electrostatic behavior of API/excipient blends is considered. We show that the net charge of the blend is a complex function of the relative quantity of API in the mixture Moreover, both the quantity and the sign of the charge are found to depend on the material in contact with the powder during the flow.

Optical diffusers with high transmission, high haze and excellent diffusing ability are widely needed in backlight unit (BLU) of liquid crystal displays (LCDs). However, there are still drawbacks in the particles-doped optical diffusers, including the distribution of the scattered particles due to agglomeration of particles and complex coating process. In this paper, polymer/liquid crystal composite films which are based on the epoxy resins, polyamine (PA) and varied thiol monomers were firstly prepared by thermally initiated polymerization The morphologies and optical properties of the composite films were systematically investigated. By controlling the functionality of thiol in the system of epoxy resins/thiol/PA/LCs, as the light scattering of anisotropic LCs and isotropic polymer framework is both achieved in the optical diffusers, good diffusing ability and high visible transmission (total transmission >96.0% and transmission haze >95.0%) are obtained.

Alzheimer′s disease is a neurodegenerative disorder whose pathol. mechanisms, despite recent advances, are not fully understood. However, the deposition of beta amyloid -peptide and neuroinflammation, which is probably aggravated by dysbiotic microbiota, seem to play a key role. Anti-Gal are the most abundant xenoreactive natural antibodies. They are supposed to stem from immunization against the gut microbiota and have been implicated in the pathogenesis of several diseases, including multiple sclerosis. These antibodies target the alpha-Gal epitope, expressed on the terminal sugar units of glycoprotein or glycolipid of all mammals except apes, Old World monkeys and humans. The alpha-Gal is constitutively expressed in several bacteria constituting the brain microbiota, and alpha-Gal-like epitopes have been detected in gray matter, amyloid plaque, neurofibrillary tangles and corpora amylacea of the human brain, suggesting a potential link between anti-Gal and Alzheimer′s disease etiopathogenesis. For the first time, our study searched for possible alterations of anti-Gal Ig levels in Alzheimer′s disease patients. IgG and IgM blood levels were significantly lower, and IgA significantly higher in patients than in healthy subjects. These results suggest that such Igs might be implicated in Alzheimer′s disease pathogenesis and open new scenarios in the research for new biomarkers and therapeutic strategies.

Renal fibrosis is the common pathol. feature in a variety of chronic kidney diseases. Aging is highly associated with the progression of renal fibrosis. Among several determinants, mitochondrial dysfunction plays an important role in aging. However, the underlying mechanisms of mitochondrial dysfunction in age-related renal fibrosis are not elucidated. Herein, we found that Wnt/β-catenin signaling and renin-angiotensin system (RAS) activity were upregulated in aging kidneys. Concomitantly, mitochondrial mass and functions were impaired with aging. Ectopic expression of Klotho, an antagonist of endogenous Wnt/β-catenin activity, abolished renal fibrosis in D-galactose (D-gal)-induced accelerated aging mouse model and significantly protected renal mitochondrial functions by preserving mass and diminishing the production of reactive oxygen species. In an established aging mouse model, dickkopf 1, a more specific Wnt inhibitor, and the mitochondria-targeted antioxidant mitoquinone restored mitochondrial mass and attenuated tubular senescence and renal fibrosis. In a human proximal tubular cell line (HKC-8), ectopic expression of Wnt1 decreased biogenesis and induced dysfunction of mitochondria, and triggered cellular senescence. Moreover, D-gal triggered the transduction of Wnt/β-catenin signaling, which further activated angiotensin type 1 receptor (AT1), and then decreased the mitochondrial mass and increased cellular senescence in HKC-8 cells and primary cultured renal tubular cells. These effects were inhibited by AT1 blocker of losartan. These results suggest inhibition of Wnt/β-catenin signaling and the RAS could slow the onset of age-related mitochondrial dysfunction and renal fibrosis. Taken together, our results indicate that Wnt/β-catenin/RAS signaling mediates age-related renal fibrosis and is associated with mitochondrial dysfunction.
