Purpose: As a third-generation EGFR TKI has been taken orally, Osimertinib effectively inhibits mutant EGFR, including T790M EGFR resistance mutations. Here, we examined real-world efficacy and tolerability of Osimertinib among Chinese patients with advanced EGFR T790M-mutant NSCLC. Patients and Methods: A total of 106 advanced NSCLC patients who were taking Osimertinib following disease progression after EGFR-TKIs or other treatments were retro-spectively recruited in this study. The PFS and OS after Osimertinib treatment were analyzed as the primary endpoints. Results: Osimertinib was used as a second line and >= 3rd line treatment in 22.6% and 77.4% of the patients, respectively. DCR and ORR were 93.4% and 57.5%, respectively. Median PFS was 12.4 12 (95% CI, 10.5-13.5) months. The PFS was 11 (8.0, 14.0) and 12 (10.3,13.7) months (p = 0.373), in patients with and without CNS metastasis, respectively. PFS in 2nd and >= 3rd line treatment was 11 (9.0, 13.0) and 12.4 12 (8.9, 15.1) months (p = 0.799), respectively. In patients with EGFR exon 19 deletion and exon 21 L858 mutation, the median PFS was 11 (9.2, 12.8) and 12 (9.2, 14.8) months, respectively (p = 0.833). Median PFS in the monotherapy group and combined anti-angiogenesis group was 11 (9.9,12.1) and 14 (11.2,16.8) months, respectively. Median OS after Osimertinib initiation was 27 (19.6, 34.4) months: 15 (6.9, 23.1) and 27 (22, 32) months in patients with and without CNS metastasis (p=0.027), 27 (20.3,33.7) months and (undefined) as second line or >= 3rd line of treatment (p = 0.421), respectively. In patients with exon 19 deletion, the median OS was not reached, and in patients with exon 21 L858 mutations, the median OS was 23 (19.1,29.9) months (p=0.027). Median OS in the monotherapy group was 27 (21.7,32.3) months, and in combined anti-angiogenesis group was not reached (p=0.68). Conclusion: Osimertinib can effectively treat advanced NSCLC with T790M mutations independently of previous treatment lines.

Herein, we designed and synthesized an SBA-15 supported 1-methyl-2-azaadamanane N-oxyl (1-Me-AZADO) and investigated its catalytic performance for selective oxidation of alcohols under Anelli’s conditions. The first example of immobilization of 1-Me-AZADO was very important to advance the oxgenation effectively because this supported N-oxyl has excellent catalytic activity for oxidation of alcohols to carbonyl compounds, and more importantly, it can be conveniently recovered and reused at least 6 times without significant effect on its catalytic efficiency.

A novel and effective photochemical approach for access to 2-substituted pyrido[2,3-d]pyrimidines is described starting from the corresponding 2-(2-aminopyridin-3-yl)ethenol through a palladium-catalyzed reaction. The recombination of photogenerated carriers seriously restricts their utilization efficiency in photocatalysis. Herein, surface oxygen vacancies (SOVs) were constructed in Pd-Bi2MoO6 interface to bridge ultra-low loading Pd cluster and Bi2MoO6 semiconductor (Pd/BMO-SOVs). It was found SOVs in Pd/Bi2MoO6-x serve as Electron Bridge to bridge ultra-low loading Pd cluster and Bi2MoO6-x, thus tremendously enhance utilization efficiency of photoexcited carriers and ultra-low loading Pd active sites for blue LED driven selective oxidation reaction. The Pd(0.05)/Bi2MoO6-SOVs exhibited 57.8 % conversion for selection oxidation of benzyl which are 6.5, 3.3 and 2.1 times higher than pristine Bi2MoO6, Bi2MoO6-x and Pd(0.05)/Bi2MoO6. Combined with theoretical calculations, SOVs was proposed as Electron Bridge to transfer photogenerated electrons from Bi2MoO6-x to ultra-low loading Pd clusters, thus greatly boosting separation and utilization efficiency of photogenerated electron-hole pairs.

A novel method was used to prepare a magnetic phenylene-based periodic mesoporous organosilica nanocomposite with yolk-shell structure (Fe3O4@YSPMO). The physiochemical properties of the designed Fe3O4@YSPMO@Ru nanocomposite were studied using PXRD, FT-IR, TGA, SEM, TEM, ICP, VSM and XPS analyses. Fe3O4@YSPMO@Ru was effectively employed as a highly recoverable nanocatalyst in the selective aerobic oxidation of alcohols.

alpha-Alkylation of methyldiarylphosphine oxides with (hetero)arylmethyl alcohols was performed under nickel catalysis. Various arylmethyl and heteroarylmethyl alcohols can be used in this transformation. A series of methyldiarylphosphine oxides were alkylated with 30-90% yields. Functional groups on the aromatic rings of methyldiarylphosphine oxides or arylmethyl alcohols including OMe, NMe2, SMe, CF3, Cl, and F groups can be tolerated. Chiral Lewis acid-catalyzed asymmetric alcohol addition reactions to cyclic carbonyl ylides generated from N-(alpha-diazocarbonyl)-2-oxazolidinones featuring a dual catalytic system are reported. Construction of a chiral quaternary heteroatom-substituted carbon center was accomplished in which the unique heterobicycles were obtained in good yields with high stereoselection. The alcohol adducts were successfully converted to optically active oxazolidine-2,4-diones by hydrolysis. Mechanistic studies by DFT calculations revealed that alcohols could be activated by Lewis acids, enabling enantioselective protonation of the carbonyl ylides.

A convenient protocol for the one-pot synthesis of quinazolin-4(3H)-ones using palladium(II) complexes via dehydrogenative coupling of readily available benzyl alcohols and 2-aminobenzamide has been described. New structurally related Pd(II) N boolean AND O chelating complexes of general configuration [Pd(L)Cl(PPh3)] (where L = dimethylamino benzoylhydrazone ligands) have been designed and synthesized. The formation of the complexes has been recognized by analytical and spectral methods (FT-IR, NMR, HR-MS). The presence of a square-planar geometry around the palladium(II) ion was confirmed by single crystal X-ray diffraction study. A wide range of substituted quinazolinones have been successfully achieved from a diverse range of benzyl alcohols in good to excellent yields using 1.0 mol % of catalyst loading under aerobic conditions. Furthermore, control experiments reveal that the dehydrogenative coupling reaction involves initially the formation of an aldehyde intermediate and subsequent formation of a cyclic aminal intermediate.

Noble-metal-free photocatalysts with high and stable performance provide an environmentally-friendly and cost-efficient route for green organic synthesis. In this work, CdS nanoparticles with small particle size and different amount were successfully deposited on the surface of covalent organic frameworks (COFs). The deposition of suitable content of CdS on COFs could not only modify the light adsorption ability and the intrinsic electronic properties, but also enhance the photocatalytic activity and cycling performance of CdS for the selective oxidation of aromatic alcohols under visible light. Especially, COF/CdS-3 exhibited the highest yield (97.1%) of benzaldehyde which is approximately 2.5 and 15.9 times as that of parental CdS and COF, respectively. The results show that the combination of CdS and COF can improve the utilization of visible light and the separation of photo-generated charge carriers, and COF with the pi-conjugated system as supports for CdS nanoparticles could provide efficient electron transport channels and improve the photocatalytic performance. Therefore, this kind of COF-supported photocatalysts with accelerated photo-induced electrons and charge-carrier separation between semiconductors possesses great potentials in future green organic synthesis.
