2-Aryl-2,3-dihydroquinolin-4(1H)-ones have recently been identified as important structures with potent biological activities such as antitumor and antidiabetic effect. Herein, a total of 25 novel N-Tf 2-aryl-2,3-dihydroquinolin-4(1H)-ones were expediently synthesized via the oxidative aza-Michael cyclization of N-Tf-2 ‘-aminodihydrochalcones by ligand-free palladium(II) catalysis. This study presents a new synthetic approach to yield N-Tf 2-aryl-2,3-dihydroquinolin-4(1H)-ones, which can be easily transformed into pharmacologically interesting aza-flavanones and other N-heterocycles, such as quinolines and tetrahydroquinolines, in yields up to 84 %. This methodology has various advantages, which includes short reaction times under mild conditions and suitable functional group tolerance. Furthermore, a plausible mechanism was proposed and demonstrated by kinetic analysis.

Selective conversion of biomass-derived alcohols into carbonyl compounds via visible-light photocatalysis is realized over hierarchical hollow WO3 microspheres with tailored surface oxygen vacancies, which presents the remarkably boosted photoactivity in terms of selectivity and activity, intrinsically attributing to the strong synergetic effect of hierarchical spherical cavity and surface oxygen vacancies simultaneously. The hierarchical spherical cavity, substantially constructed by the self-interconnected nanosheets, enhances the light-harvesting ability via multiple light reflections not only in spherical cavity but also among the self-interconnected nanosheets. Surface oxygen vacancies favor the energy band gap narrowing via forming a miniband just below the conduction band and then extend the photoresponse region, further boosting the light-harvesting ability. Importantly, surface oxygen vacancies function as the electron sinks to capture photoelectrons and thus restrict their recombination probability with holes, finally improving the photoelectron-hole separation efficiency. Meanwhile, this photocatalyst presents excellent reusability, showing its promising potential in practical applications. This work sheds light on a new application of hierarchical WO3 microspheres with tailored surface oxygen vacancies and its strong synergetic effect of hierarchical structures and surface oxygen vacancies on photocatalytic performance, delivering new insights for rationally designing highly active photocatalysts applied in future green and sustainable organic transformation reactions.

A new magnetically recoverable catalyst consisting of phenanthroline Cu(II) complex supported on nickel ferrite nanoparticles was prepared. The synthesized catalyst was characterized by Fourier transform in-frared spectroscopy, X-ray diffraction, transmission and scanning electron microscopes, thermogravimetry, energy dispersive X-ray spectroscopy, vibrating sample magnetometry and inductively coupled plasma. Supported copper complex used for solvent free oxidation of 1-phenyl ethanol as a model. Influence of the reaction parameters (kind of oxidant, amount of the catalyst, reaction time, solvent and reaction temperature) were studied. Because of the immobilized complex has been shown to be an efficient het-erogeneous catalyst for the selective oxidation of 1-phenyl ethanol to acetophenone (94% yield) by hydro-gen peroxide so this green approach extended to other benzylic alcohols. The catalyst had been reused 10 times with no significant loss of catalytic activity. SEM, EDX, XRD, and ICP analysis of reused catalyst indicated that the catalyst was stable after the reaction. A thermal O-to-C [1,3]-rearrangement of alpha-hydroxy acid derived enol ethers was achieved under mild conditions. The 2-aminothiophenol protection of carboxylic acids facilitates formation of the [1,3] precursor and its thermal rearrangement via stabilization of a radical intermediate. Experimental and theoretical evidence for dissociative radical pair formation, its captodative stability via aminothiophenol, and a unique solvent effect are presented. The aminothiophenol was deprotected from rearrangement products as well as after derivatization to useful synthons.

A novel late-stage solubilization of peptides using hydrazides is described. A solubilizing tag was attached through a selective N-alkylation at a hydrazide moiety with the aid of a 2-picoline-borane complex in 50% acetic acid-hexafluoro-2-propanol. The tag, which tolerates ligation and desulfurization conditions, can be detached by a Cu-mediated selective oxidative hydrolysis of the N-alkyl hydrazide. This new method was validated through the synthesis of HIV-1 protease.

A convenient protocol for the one-pot synthesis of quinazolin-4(3H)-ones using palladium(II) complexes via dehydrogenative coupling of readily available benzyl alcohols and 2-aminobenzamide has been described. New structurally related Pd(II) N boolean AND O chelating complexes of general configuration [Pd(L)Cl(PPh3)] (where L = dimethylamino benzoylhydrazone ligands) have been designed and synthesized. The formation of the complexes has been recognized by analytical and spectral methods (FT-IR, NMR, HR-MS). The presence of a square-planar geometry around the palladium(II) ion was confirmed by single crystal X-ray diffraction study. A wide range of substituted quinazolinones have been successfully achieved from a diverse range of benzyl alcohols in good to excellent yields using 1.0 mol % of catalyst loading under aerobic conditions. Furthermore, control experiments reveal that the dehydrogenative coupling reaction involves initially the formation of an aldehyde intermediate and subsequent formation of a cyclic aminal intermediate.

Damage to the blood-brain barrier (BBB) during the process of cerebral ischemic injury is a key factor that affects the treatment of this condition. The present study aimed to assess the potential effects of 4-methoxybenzyl alcohol (4-MA) on brain microvascular endothelial cells (bEnd.3) against oxygen-glucose deprivation/reperfusion (OGD/Rep) using an in vitro model that mimics in vivo ischemia/reperfusion injury. In addition, the present study aimed to explore whether this underlying mechanism was associated with the inhibition of pro-inflammatory factors and the activation status of the PI3K/Akt signaling pathway. bEnd.3 cells were subjected to OGD/Rep-induced injury before being treated with 4-MA, following which cell viability, lactate dehydrogenase (LDH) release and levels of nitric oxidase (NO) were detected by colorimetry, pro-inflammatory factors including tumor necrosis factor-alpha (TNF-alpha), interleukin (IL)-1 beta and IL-6, were detected by ELISA. The expression levels of occluding and claudin-5were evaluated by immunofluorescence staining. The expression levels of AKT, phosphorylated (p)-Akt, endothelial nitric oxide synthase (eNOS) and p-eNOS were also measured by western blot analysis. After bEnd.3 cells were subjected to OGD/Rep-induced injury, cell viability and NO levels were significantly decreased, whilst LDH leakage and inflammatory factor (TNF-alpha, IL-1 beta and IL-6) levels were significantly increased. Treatment with 4-MA significantly ameliorated cell viability, LDH release and the levels of NO and pro-inflammatory factors TNF-alpha, IL-1 beta and IL-6 as a result of OGD/Rep. Furthermore, treatment with 4-MA upregulated the expression of occludin, claudin-5, Akt and eNOS, in addition to increasing eNOS and AKT phosphorylation in bEnd.3 cells. These results suggest that 4-MA can alleviate OGD/Rep-induced injury in bEnd.3 cells by inhibiting inflammation and by activating the PI3K/AKT signaling pathway as a possible mechanism. Therefore, 4-MA can serve as a potential candidate for treating OGD/Rep-induced injury.

Direct oxidative coupling of alcohols with amines using a non-precious metal oxide catalyst under mild conditions is highly desirable for imine synthesis. In this work, a mesoporous Mn1ZrxOy solid solution catalyst prepared by a co-precipitation method showed excellent catalytic performance in imine synthesis from primary alcohols and amines without base additives in an air atmosphere. XRD, N-2 physisorption, H-2-TPR, O-2-TPD, EPR and XPS were comprehensively used to unravel its structural, redox and amphoteric properties that closely depended on the interaction between MnOy and ZrO2 with a variable Zr ratio. The Mn1Zr0.5Oy catalyst presented the highest fractions of Mn3+ ions and reactive oxygen species on the surface, and the highest concentrations of acidic-basic sites, which were disclosed to play important roles in activating alcohols and molecular O-2 in the rate-determining step. In the model reaction of oxidative coupling of benzyl alcohol with aniline, such enhanced features of the Mn1Zr0.5Oy catalyst can promote the intrinsic catalytic activity (iTOF of 1.87 h(-1)) and boost benzylideneaniline formation (5.56 mmol g(cat).(-1) h(-1)) based on a >99% yield at 80 degrees C respectively at a fast response. It can also work effectively at a room temperature of 30 degrees C, as well as for the gram-grade synthesis. This is one of the best results among all the MnOy-based catalysts in the literature. Moreover, this catalyst showed good stability and a wide substrate scope with good to excellent yields of imines.
