The effects of black garlic aged at different periods as a substitute for fresh garlic (FG) on the overall quality of pastirma cemen paste were evaluated during 90 days of storage. The addition of black garlic increased the phenolic content and antioxidant activities of the cemen pastes, and decreased the firmness, stickiness, pH and color values compared to CON (Control). Total aerobic mesophilic bacteria counts of cemen pastes containing black garlic aged for ≥21 days were similar to those of CON, but the counts of yeast and molds of black garlic added-groups were higher than CON during storage. Incorporation of black garlic to cemen paste increased the content of volatile compounds such as esters and terpenes with fruity and sweet notes, while the concentration of sulfur compounds decreased. In this way, the off-flavor of cemen paste was eliminated and its sensory acceptability improved. Considering the overall data, the best results were obtained in the CP3 group produced with black garlic aged for 21 days. Consequently, black garlic can be used as a substitute for FG to improve the consumer acceptance and bioactive properties of cemen paste. Novelty impact statement : Some consumers avoid pastirma consumption because of the unpleasant off-flavor from cemen paste. Overcoming this problem is very important for both the producer and the consumer. The results of this study showed that black garlic improved the sensory acceptability and bioactive properties of cemen paste by eliminating the unpleasant smell of fresh garlic and increasing the antioxidant capacity. In conclusion, the use of cemen paste with black garlic in pastirma production has promising potential to enhance consumer acceptance and retard oxidative reactions.

Phthalate and alternative plasticizers are semivolatile organic compounds (SVOCs) and among the most abundant indoor pollutants. Although ingestion of dust is one of the major exposure pathways to them, migration knowledge from source products to indoor dust is still limited. Systematic chamber measurements were conducted to investigate the direct transfer of these SVOCs between source products and dust in contact with the source. Substantial direct source-to-dust transfer of SVOCs was observed for all tests. The concentration of bis(2-ethylhexyl)phthalate in dust was 12 times higher than the pre-exptl. level after only two days of source-dust contact. A mechanistic model was developed to predict the direct transfer process, and a reasonable agreement between model predictions and measurements was achieved. The octanol/air partition coefficient (Koa) of SVOCs, the emission parameter of the source product (y0), and the characteristics of the dust layer (i.e., porosity and thickness) control the transfer, affecting the SVOC concentration in dust, the kinetics of direct transfer, or both. Dust mass loading has a significant influence on the transfer, while relative humidity only has a limited effect. The findings suggest that minimizing the use of SVOC-containing products and house vacuuming are effective intervention strategies to reduce young children’s exposure to SVOCs.

Recently, the aerobic biol. treatment of MSW has increased in rural areas of China. These facilities are usually open setup and close to nearby residents, and complaints tend to be received regarding odorous gases. A semi-in-vessel setup facility was built in recent years, and its impact on the environment and personnel is not clear. Ammonia was the predominant compound released from the windrow and its concentration was one order of magnitude higher than those of other compounds Terpenes and reduced sulfur compounds (RSCs) persisted throughout the entire active fermentation process, with only slight decreases. The biofilter pool had a greater odor impact on the surrounding neighborhood than the waste unloading and sorting operation. The concentrations of most compounds were reduced by the biofilter pool. Ten major substances that caused odors at the facility were analyzed, where most comprised RSCs. The odor impact of the facility was low, but it was associated with a carcinogenic risk to the operators, thereby indicating the need to improve personal protection. Large amounts of ammonia and volatile organic compounds were released from the composting units, but they had a low odor impact on the environment because of the semi-in-vessel setup. The odor impact of the facility was low but it was associated with a carcinogenic risk to the operators. This technol. can be popularized in rural areas.

Diesel exhaust particles (DEPs) are known pathogenic pollutants that constitute a significant quantity of air pollution. Given the ubiquitous presence of macrophages throughout the body, including the lungs, as well as their critical role in tissue and organismal metabolic function, we sought to determine the effect of DEP exposure on macrophage mitochondrial function. Following daily DEP exposure in mice, pulmonary macrophages were isolated for mitochondrial analyses, revealing reduced respiration rates and dramatically elevated H2O2 levels. Serum ceramides and inflammatory cytokines were increased. To determine the degree to which the changes in mitochondrial function in macrophages were not dependent on any cross-cell communication, primary pulmonary murine macrophages were used to replicate the DEP exposure in a cell culture model. We observed similar changes as seen in pulmonary macrophages, namely diminished mitochondrial respiration, but increased H2O2 production Interestingly, when treated with myriocin to inhibit ceramide biosynthesis, these DEP-induced mitochondrial changes were mitigated. Altogether, these data suggest that DEP exposure may compromise macrophage mitochondrial and whole-body function via pathol. alterations in macrophage ceramide metabolism

Biogas is an important and sustainable biofuel. Encouraged by renewable energy policies, the biogas industry has developed rapidly worldwide. Double membrane gasholders are widely used as biogas storage facilities. This work aimed to reveal the odorant permeation characteristics of double membrane gasholders, evaluate the related odor pollution risk, and identify the priority odorants for permeation prevention. Gas samples below the inner membrane, between the inner and outer membranes, and in the ambient environment were collected and analyzed. Odor potential and contribution were evaluated using the odor activity value (OAV). The relationship between the samples was explored by the Pearson correlation anal. The permeation characteristics of the major odor contributors were analyzed by comparing odorant concentrations in samples from both sides of the membrane. Terpenes were found to be the dominant compounds (>75%) inside the double membrane gasholder. The total OAVs of the gas below the inner membrane, between the inner and outer membranes and in the ambient environment were 47,500, 484, and less than 100, resp. Terpenes and sulfur compounds were the dominant odor contributors (>95% in total) for the gas inside the double membrane gasholder. The composition of the gas between the membranes was significantly correlated to that of the gas below the inner membrane (r2 = 0.98, p ≤ 0.001), but had a poor correlation with that of the gas in the ambient environment (r2 < 0.01). Odorant permeation occurred through both the inner and outer membranes. Hydrogen sulfide and limonene were selected as the main indicators for determining the permeability of the double membrane gasholder. Suggestions were made for the permeability test methods and material selection for double membrane gasholders. Lack of phys. activity can lower hormone levels and increase free radicals that can accelerate the aging process. Hence, phys. activity is very important to maintain functions of organs. This research was aimed to study the effects of low and moderate treadmill exercise on D-Galactose-exposed aging rat model by evaluating the degree of hepatic fibrosis, number of M1 and M2, and M1/M2 ratio. Twenty-four 3-mo-old male Wistar aging model rats were randomly divided into four groups, i.e., three treatment groups with daily 300 mg kgBW-1D-Galactose injection administrated i.p. for 4 wk and 1 control group with normal saline injection. Two of the D-Galactose treated groups were given low and moderate treadmill exercise for 4 wk. It was concluded that low intensity treadmill exercise significantly lowered the degree of D-Galactose-exposed hepatic fibrosis, and moderate treadmill exercise was able to restore the injured liver tissue back to the non-aging state. Administration of D-Galactose causes inflammation marked by the elevated number of M1 and M2 macrophages. Moderate treadmill exercise drove M1/M2 ratio back to the control condition.

Elderly population and age-related diseases are on the rise. On the contrary, aging studies are tech. hard to conduct, because they require elderly animals, the maintenance of which requires ample effort and is expensive. To tackle this problem, -galactose is used to hasten the aging process in various tissues in rodent models and it has been shown to successfully mimic the oxidative alterations that take place in the natural aging process in various tissues both by our group and others. In the present study, the validity of -galactose aging model in skeletal muscles was tested both on predominantly slow-twitch (soleus) and rather fast-twitch (gastrocnemius) muscle in male Sprague-Dawley rats and the results are compared with young littermate controls and naturally aged rats. Redox-related modifications in soleus and gastrocnemius were assessed by measurement of protein carbonyl groups, advanced oxidation protein products, lipid hydroperoxides, total thiol, and Cu, Zn-superoxide dismutase activities. In the present study, we provide biochem. evidence demonstrating that -galactose-induced mimetic aging does result in oxidative stress-related redox alterations that are comparable with the alterations that occur in natural aging in soleus. On the contrary, in the -galactose-induced mimetic aging of gastrocnemius, even though the oxidative stress markers were significantly increased, the endpoint redox homeostasis markers were not statistically comparable with the redox status of naturally aged group.

Sarcopenia has a significant neg. impact on healthspan in the elderly and effective pharmacol. interventions remain elusive. We have previously demonstrated that sarcopenia is associated with reduced activity of the sarcoplasmic reticulum Ca2+ ATPase (SERCA) pump. We asked whether restoring SERCA activity using pharmacol. activation in aging mice could mitigate the sarcopenia phenotype. We treated 16-mo male C57BL/6J mice with vehicle or CDN1163, an allosteric SERCA activator, for 10 mo. At 26 mo, maximal SERCA activity was reduced 41% in gastrocnemius muscle in vehicle-treated mice but maintained in old CDN1163 treated mice. Reductions in gastrocnemius mass (9%) and in vitro specific force generation in extensor digitorum longus muscle (11%) in 26 vs. 16-mo-old wild-type mice were also reversed by CDN1163. CDN1163 administered by intra-peritoneal injection also prevented the increase in mitochondrial ROS production in gastrocnemius muscles of aged mice. Transcriptomic anal. revealed that these effects are at least in part mediated by enhanced cellular energetics by activation of PGC1-α, UCP1, HSF1, and APMK and increased regenerative capacity by suppression of MEF2C and p38 MAPK signaling. Together, these exciting findings are the first to support that pharmacol. targeting of SERCA can be an effective therapy to counter age-related muscle dysfunction.

This study assessed whether isolated, permeabilized cardiomyocytes from male and female C57BL/6 mice differ in terms of their respiration with multiple substrates and overall intracellular diffusion restriction estimated by the apparent ADP-affinity of respiration. Using respirometry, we recorded 1) the activities of respiratory complexes I, II and IV, 2) the respiration rate with substrates fuelling either complex I, II, or I + II, and 3) the apparent ADP-affinity with substrates fuelling complex I and I + II. The respiration rates were normalized to protein content and citrate synthase (CS) activity. We found no sex differences in CS activity (a marker of mitochondrial content) normalized to protein content or in any of the respiration measurements. This suggests that cardiomyocytes from male and female mice do not differ in terms of mitochondrial respiratory capacity and apparent ADP-affinity. Pyruvate modestly lowered the respiration rate, when added to succinate, glutamate and malate. This may be explained by intramitochondrial compartmentalization caused by the formation of supercomplexes and their association with specific dehydrogenases. To our knowledge, we show for the first time that the apparent ADP-affinity was substrate-dependent. This suggests that substrates may change or regulate intracellular barriers in cardiomyocytes.
