Site-specific incorporation of deuterium into drug molecules to study and improve their biological properties is crucial for drug discovery and development. Herein, we describe a palladium-catalyzed room-temperature deuterogenolysis of carbon-oxygen bonds in alcohols and ketones with D-2 balloon for practical synthesis of deuterated pharmaceuticals and chemicals with benzyl-site (sp(3) C-H) D-incorporation. The highlights of this deoxygenative deuteration strategy are mild conditions, broad scope, practicability and high chemoselectivity. To enable the direct use of D2O, electrocatalytic D2O-splitting is adapted to in situ supply D-2 on demand. With this system, the precise incorporation of deuterium in the metabolic position (benzyl-site) of ibuprofen is demonstrated in a sustainable and practical way with D2O.

A thermal O-to-C [1,3]-rearrangement of alpha-hydroxy acid derived enol ethers was achieved under mild conditions. The 2-aminothiophenol protection of carboxylic acids facilitates formation of the [1,3] precursor and its thermal rearrangement via stabilization of a radical intermediate. Experimental and theoretical evidence for dissociative radical pair formation, its captodative stability via aminothiophenol, and a unique solvent effect are presented. The aminothiophenol was deprotected from rearrangement products as well as after derivatization to useful synthons.

The dendritic ligand 2, 4, 6-tris (di-4-chlorobenzamido)-1, 3, 5-triazine3supported Zn/Cu (1:1) 4a, 2, 4, 6-tris (di-4-chlorobenzamido)-1, 3, 5-triazine3supported Zn/Cu (1:2) 4b, and 2, 4, 6-tris (di-4-chlorobenzamido)-1, 3, 5-triazine3supported Zn/Cu (2:1) 4cbimetallic nanoparticles (NPs) were synthesized successfully by following the co-complexation route in which the desired molar ratio of Zn and Cu was confirmed by the obtained results of electron diffraction X-ray and X-ray fluorescence spectroscopy analysis. The average particle size of these NPs was detected as 15-20 nm from transmission electron microscopy investigations and agglomerate spherical surface morphology was found by scanning electron microscopy, whereas the face-centered cubic structure of these bimetallic NPs was observed by X-ray diffraction assessment. Also, the formation of the ligand was proven by IR,(HNMR)-H-1,(CNMR)-C-13, and elemental analysis. Remarkably, the chemoselective oxidation of aromatic alcohols to the corresponding aldehydes or ketones at 25 min and reduction of aromatic nitro substituents to the corresponding aniline at 20 min in aqueous medium at room temperature have been studied by the most effective catalyst Zn/Cu (2:1) 4cNPs among other molar ratios of Zn/Cu (1:1)4aand Zn/Cu (1:2) 4bNPs under atmospheric air (O-2) conditions with good to excellent yields. This green catalytic approach of Zn/Cu (2:1) 4cNPs catalytic was easily recovered by simple filtration and recycled at least five consecutive runs without a noticeable loss of its catalytic effectiveness.

The development of eco-friendly and low cost catalysts is challenging. Here, heterostructures of Bi2S3 quantum dots (QDs) doped onto ultrathin BiOCl nanosheets were synthesized by a facile hydrothermal method to exploit efficient photosensitizers with appropriate electronic states to enhance the transfer of electrons. The obtained Bi2S3-BiOCl showed highly efficient photocatalytic ability for selective oxidation of aromatic alcohols to aldehydes. More oxygen vacancies are formed on the exposed 001 facet of the ultrathin BiOCl, which can effectively trap electrons and form O-.(2)- radicals. The cooperation between the BiOCl and the Bi2S3 QDs effectively separates photogenerated electron-hole pairs at the heterointerface and facilitates the cooperative actions of O-.(2)- radicals and holes, which brings about a desirable photocatalytic eciency for the selective oxidation of aromatic alcohols. This work highlights the synergistic effect of semiconductor QDs and two-dimensional materials on selective conversion processes and provides a new design paradigm using noble metal-free heteromaterials with high photocatalytic activity. This opens new possibilities for photocatalyst design using heteromaterials with high photocatalytic activity.

Damage to the blood-brain barrier (BBB) during the process of cerebral ischemic injury is a key factor that affects the treatment of this condition. The present study aimed to assess the potential effects of 4-methoxybenzyl alcohol (4-MA) on brain microvascular endothelial cells (bEnd.3) against oxygen-glucose deprivation/reperfusion (OGD/Rep) using an in vitro model that mimics in vivo ischemia/reperfusion injury. In addition, the present study aimed to explore whether this underlying mechanism was associated with the inhibition of pro-inflammatory factors and the activation status of the PI3K/Akt signaling pathway. bEnd.3 cells were subjected to OGD/Rep-induced injury before being treated with 4-MA, following which cell viability, lactate dehydrogenase (LDH) release and levels of nitric oxidase (NO) were detected by colorimetry, pro-inflammatory factors including tumor necrosis factor-alpha (TNF-alpha), interleukin (IL)-1 beta and IL-6, were detected by ELISA. The expression levels of occluding and claudin-5were evaluated by immunofluorescence staining. The expression levels of AKT, phosphorylated (p)-Akt, endothelial nitric oxide synthase (eNOS) and p-eNOS were also measured by western blot analysis. After bEnd.3 cells were subjected to OGD/Rep-induced injury, cell viability and NO levels were significantly decreased, whilst LDH leakage and inflammatory factor (TNF-alpha, IL-1 beta and IL-6) levels were significantly increased. Treatment with 4-MA significantly ameliorated cell viability, LDH release and the levels of NO and pro-inflammatory factors TNF-alpha, IL-1 beta and IL-6 as a result of OGD/Rep. Furthermore, treatment with 4-MA upregulated the expression of occludin, claudin-5, Akt and eNOS, in addition to increasing eNOS and AKT phosphorylation in bEnd.3 cells. These results suggest that 4-MA can alleviate OGD/Rep-induced injury in bEnd.3 cells by inhibiting inflammation and by activating the PI3K/AKT signaling pathway as a possible mechanism. Therefore, 4-MA can serve as a potential candidate for treating OGD/Rep-induced injury.

The influence of pH on the photocatalytic partial oxidation of 4-methoxybenzyl alcohol (MBA) and vanillyl alcohol (VA) to their corresponding aldehydes in aqueous suspension under UVA irradiation was investigated by using poorly crystalline home-prepared and crystalline commercial TiO2 (BDH, Merck and Degussa P25) photocatalysts. The results clearly show as tuning pH can strongly impart selectivity and activity to photocatalytic processes which are often quite unselective in aqueous suspensions. It was found that pH effect on reaction rate and product selectivity strongly depended on TiO2 crystallinity and substrate type. In the case of MBA oxidation, photoreactivity and selectivity were very high at low pH values for all of TiO2 catalysts, and the crystalline samples showed to be more active than the poorly crystalline ones. At pH= 1 the photoactivity of Degussa P25 was the highest one, and 88% selectivity at 50% conversion was determined. At acidic pH values, selectivity and activity were higher in the presence of HCl than H2SO4 or H3PO4. For VA oxidation, high selectivity values were obtained at high pH’s for all of the samples, and the crystalline samples showed higher activity at the alkaline pH values with respect to that observed at the acidic ones. Experiments starting from the obtained products, that are p-anisaldehyde and vanillin, showed that the selectivity depends on the resistance of those compounds to be subjected to further oxidation under the experimental conditions used.

An efficient magnetically recyclable bimetallic catalyst by anchoring copper and manganese complexes on the Fe(3)O(4)NPs was prepared and named as Fe3O4@Cu-Mn. It was founded as a powerful catalyst for the domino one-pot oxidative benzimidazole and Biginelli reactions from benzyl alcohols as a green protocol in the presence of air, under solvent-free and mild conditions. Fe3O4@Cu-Mn NPs were well characterized by FT-IR, XRD, FE-SEM, TEM, VSM, TGA, EDX, DLS, and ICP analyses. The optimum range of parameters such as time, temperature, amount of catalyst, and solvent were investigated for the domino one-pot benzimidazole and Biginelli reactions to find the optimum reaction conditions. The catalyst was compatible with a variety of benzyl alcohols, which provides favorable products with good to high yields for all of derivatives. Hot filtration and Hg poisoning tests from the nanocatalyst revealed the stability, low metal leaching and heterogeneous nature of the catalyst. To prove the synergistic and cooperative effect of the catalytic system, the various homologues of the catalyst were prepared and then applied to a model reaction separately. Finally, the catalyst could be filtered from the reaction mixture simply, and reused for five consecutive cycles with a minimum loss in catalytic activity and performance. Graphic A new magnetically recyclable Cu/Mn bimetallic catalyst has been developed for domino one-pot oxidation-condensation of benzimidazole and Biginelli reactions from alcohols.

Deuterium labeling is an interesting process that leads to compounds of use in different fields. We describe the transfer hydrogenation of aldehydes and the selective C-1 deuteration of the obtained alcohols in D2O, as the only deuterium source. Different aromatic, alkylic and alpha,beta-unsaturated aldehydes were reduced in the presence of [RuCl(p-cymene)(dmbpy)]BF4, (dmbpy=4,4 ‘-dimethyl-2,2 ‘-bipyridine) as the pre-catalyst and HCO2Na/HCO2H as the hydrogen source. Moreover, furfural and glucose, were selectively reduced to the valuable alcohols, furfuryl alcohol and sorbitol. The processes were carried out in neat water or in a biphasic water/toluene system. The biphasic system allowed easy recycling, higher yields, and higher selective D incorporation (using D2O/toluene). The deuteration took place due to an efficient effective M-H/D+ exchange from D2O that allows the inversion of polarity of D+ (umpolung). DFT calculations that explain the catalytic behavior in water are also included.

alpha-Alkylation of methyldiarylphosphine oxides with (hetero)arylmethyl alcohols was performed under nickel catalysis. Various arylmethyl and heteroarylmethyl alcohols can be used in this transformation. A series of methyldiarylphosphine oxides were alkylated with 30-90% yields. Bark from the Handroanthus impetiginosus (Mart. ex DC.) Mattos (Bignoniaceae) tree has long been used in traditional South American healing practises to treat inflammation. However, its anti-inflammatory activity has not been closely examined. Here we use chemical extraction, qualitative phytochemical examination, toxicity testing and quantitative examination of anti-inflammatory activity on human cells ex vivo. All extracts were found to be nontoxic. We found different extracts exhibited unique cytokine profiles with some extracts outperforming a positive control used in the clinic. These results verify the immunomodulatory activity of Handroanthus impetiginosus (Mart. ex DC.) Mattos (Bignoniaceae) tree bark-derived compounds. Collectively, combining a lack of toxicity and potency in human immune cells supports further fractionation and research.
