A novel polysaccharide (PFP1) with an average mol. weight of 2.02 × 105 g/mol was isolated from Passiflora foetida fruits through hot water extraction, ethanol precipitation and column chromatog. The structure of PFP1 was determined by GPC-MALS-RI, IC, FT-IR, GC-MS and NMR. The structural anal. showed that PFP1 was a heteropolysaccharide and composed of mannose (48.83%), galactose (32.46%), glucose (6.21%), arabinose (5.88%), fructose (2.24%), galacturonic acid (2.20%), xylose (1.17%), fucose (0.17%), ribose (0.05%), and glucuronic acid (0.78%), with a backbone structure of →1)-α-D-Manp→1,2)-β-D-Manp-linked 1,2,6)-β-D-Manp residues and side chains consisted of →1)-β-D-Galp, →1,4)-α-D-Manp, →1, 4)-β-D-Glcp, →1,3)-α-D-Galp, →1,6)-β-D-Manp, →1,6)-β-D-Galp, →1,2,3)-β-D-Manp and →1,3,6)-β-D-Galp residues. The results of immune-enhancing assays revealed that PFP1 could obviously promote the production of NO and secretion of cytokines (TNF-α and IL-6) of macrophage RAW264.7. These findings demonstrate that P. foetida fruit polysaccharides can be utilized as a potential immune-enhancing functional food.

Boehmite (BO) nanoparticles (NPs) were prepared via the injection of aqueous NaOH solution to aqueous aluminum nitrate solution at room temperature Afterwards, a new complex of copper was immobilized on BO-NPs (Cu-Guanidine@BO-NPs). This heterogeneous nanocatalyst was used as a practical, recyclable, chemo and homoselective nanocatalyst in the organic processes, i.e. the preparation of tetrazole five-membered heterocycles and chemoselective sulfoxidation of sulfides using H2O2 as oxidant. In this sense, the prepared nanocatalyst was characterized by AAS, N2 adsorption-desorption isotherms, WDX, EDS, SEM, and TGA techniques. The reusability of this catalyst was investigated in the described organic reactions for several runs without notable loss of its catalytic activity. Moreover, all of the tetrazole and sulfoxide derivatives were isolated in high Turn Over Number (TON) and Turn Over Frequency (TOF) numbers indicating the high activity and selectivity of Cu-Guanidine@BO-NPs in the described reactions.

Linalool is a natural product present in fruits and aromatic plants with biol. activities. Researchers have reported that the inhalation of linalool exerts anti-inflammatory activities. In this study, we examined the therapeutic effects of linalool on airway inflammation and mucus overproduction in mice with allergic asthma. Oral administration of linalool significantly inhibited the levels of eosinophil numbers, Th2 cytokines and IgE (IgE) caused by ovalbumin (OVA) exposure. Linalool exerted preventive effects against the influx of inflammatory cells and mucus hypersecretion in the lung tissues. Linalool also dose-dependently decreased the levels of inducible nitric oxide synthase (iNOS) expression and protein kinase B (AKT) activation in the lung tissues. Linalool effectively downregulated the activation of mitogen-activated protein kinases (MAPKs) and nuclear factor-kappaB caused by OVA exposure. Furthermore, linalool exerted inhibitory effect on OVA-induced airway hyperresponsiveness (AHR). In the in vitro study, the increased secretion of MCP-1 was attenuated with linalool treatment in lipopolysaccharide (LPS)-stimulated H292 airway epithelial cells. In conclusion, linalool effectively exerts a protective role in OVA-induced airway inflammation and mucus hypersecretion, and its protective effects are closely related to the downregulation of inflammatory mediators and MAPKs/NF-kappaB signaling.

Herein, we report a Pd(II)-catalyzed highly enantioselective (up to 99.5% ee) arylation of cyclopropyl C-H bonds with aryl iodides using mono-N-protected amino acid (MPAA) ligands, providing a new route for the preparation of chiral cis-aryl-cyclopropylmethylamines, e.g., I [R1 = H, Me, CH2Ph, Ph, C6H3F2-2,6, CH2OSiMe2CMe3, CO2Me]. 2,2,2-Trifluoroethanol (TFE) is used more and more widely in industry, but there are few reports on its separation at present. The purpose of this work is to explore the feasibility of separating 2,2,2-trifluoroethanol from aqueous solution using different extractants, as isopentanol, 2-Ethyl-1-hexanol, n-Pr acetate, and dichloromethane. The liquid-liquid equilibrium data of TFE + water +{isopentanol or 2-ethyl-1-hexanol or Pr acetate or dichloromethane} at standard pressure and 303.2 K were measured. The estimated partition coefficient (D) and separation factor (S) of TFE are higher than 1. Meanwhile, Othmer-Tobias and Hand’s empirical equations verified the exptl. data’s consistency, and the squares of its linear correlation are all greater than 0.99. The NRTL and UNIQUAC activity coefficient models were used to correlate the exptl. data, and different model parameters were obtained by regression. After comparing the calculated values of the model with the exptl. data, it was found that the root means square deviation (RMSD%) was less than 0.34%, which indicate that both models have a reasonable correlation with the exptl. data of the ternary system and can accurately predict the exptl. data.

Auxin plays a key role in different plant growth and development processes, including flower opening and development. The perception and signaling of auxin depend on the cooperative action of various components, among which auxin/indole-3-acetic acid (Aux/IAA) proteins play an imperative role. In a recent study, the entire Aux/IAA gene family was identified and comprehensively analyzed in Hedychium coronarium, a scented species used as an ornamental plant for cut flowers. Phylogenetic anal. showed that the Aux/IAA gene family in H. coronarium is slightly contracted compared to Arabidopsis, with low levels of non-canonical proteins. Sequence anal. of promoters showed numerous cis-regulatory elements related to various phytohormones. HcIAA genes showed distinct expression patterns in different tissues and flower developmental stages, and some HcIAA genes showed significant responses to auxin and ethylene, indicating that Aux/IAAs may play an important role in linking hormone signaling pathways. Based on the expression profiles, HcIAA2, HcIAA4, HcIAA6 and HcIAA12, were selected as candidate genes and HcIAA2 and HcIAA4 were screened for further characterization. Downregulation of HcIAA2 and HcIAA4 by virus-induced gene silencing in Hedychium coronarium flowers modified the total volatile compound content, suggesting that HcIAA2 and HcIAA4 play important roles in Hedychium coronarium floral scent formation. The results presented here will provide insights into the putative roles of HcIAA genes and will assist the elucidation of their precise roles during floral scent formation.

Azodisalicylate, used to treat ulcerative colitis, causes diarrhea in ≤12.5% of patients. The in vitro effects of azodisalicylate, sulfasalazine, and 5-aminosalicylic acid on rabbit intestinal electrolyte transport were studied. Distal ileal mucosae mounted in Ussing chambers were exposed to varying concentrations of the drugs. Mucosal addition of azodisalicylate (>5 mM) caused the greatest anion-dependent increase in short-circuit current of 83 μA/cm2. Isotope flux measurements suggest that azodisalicylate may stimulate predominantly electrogenic HCO3- secretion and induce net NaCl secretion. In contrast, serosal addition of azodisalicylate and sulfasalazine decreased short-circuit current, and 5-aminosalicylic acid had no effect. Azodisalicylate had no effect on ion transport in distal colon. The effects of azodisalicylate in ileum were not inhibited with piroxicam (an inhibitor of cyclooxygenase). Mucosal cAMP and cGMP levels were unchanged after ileal exposure to azodisalicylate. Azodisalicylate appears to be a mechanistically unusual secretagogue, possibly explaining the increased incidence of diarrhea seen in patients taking the drug.

Abstract: Background: Deracemization, the transformation of the racemate into a single stereoisomeric product in 100% theor. yield, is an appealing but challenging option for the asym. synthesis of optically pure chiral compounds as important pharmaceutical intermediates. To enhance the synthesis of (R)-1,3-butanediol from the corresponding low-cost racemate with minimal substrate waste, we designed a stereoinverting cascade deracemization route and constructed the cascade reaction for the total conversion of racemic 1,3-butanediol into its (R)-enantiomer. This cascade reaction consisted of the absolutely enantioselective oxidation of (S)-1,3-butanediol by Candida parapsilosis QC-76 and the subsequent asym. reduction of the intermediate 4-hydroxy-2-butanone to (R)-1,3-butanediol by Pichia kudriavzevii QC-1. Results: The key reaction conditions including choice of cosubstrate, pH, temperature, and rotation speed were optimized systematically and determined as follows: adding acetone as the cosubstrate at pH 8.0, a temperature of 30°C, and rotation speed of 250 rpm for the first oxidation process; in the next reduction process, the optimal conditions were: adding glucose as the cosubstrate at pH 8.0, a temperature of 35°C, and rotation speed of 200 rpm. Conclusions: The step-by-step cascade reaction efficiently produced (R)-1,3-butanediol from the racemate by biosynthesis and shows promising application prospects.

Total soluble solid (TSS) in tomatoes is an important trait that affects consumer’s preference. Previous studies have observed high TSS content in parthenocarpic tomato fruits induced by a parthenocarpic gene, pat-k. However, the mechanisms regulating high TSS content remain unknown. Hence, we have investigated the water content, metabolites involved with high TSS content, and gene expression related to starch synthesis in tomato fruits to elucidate the mechanism regulating high TSS content. We have found that independent of fruit water content, parthenocarpic fruits induced by pat-k had higher TSS content than pollinated fruits on Pat-k homozygous plants. Addnl., metabolome anal. of mature fruits has revealed that fructose influenced the difference in TSS content between pollinated fruits on Pat-k homozygous plants and the parthenocarpic fruits on pat-k homozygous plants. Furthermore, high performance liquid chromatog. (HPLC) anal. showed that not only fructose contents but also glucose contents were higher in the parthenocarpic fruits on pat-k homozygous plants than pollinated fruits on Pat-k homozygous plants, suggesting that increases in these sugar contents influenced the high TSS content in the pat-k-induced parthenocarpic fruits. We also have found that parthenocarpic fruits had high starch content and upregulated Sus4, which is involved in starch synthesis, during the early stages of fruit development. Since previous reports have suggested that high starch content during the early stages of fruit development attributed to high sugar content in mature tomato fruits, our results suggest that high accumulation of starch in young fruits could be involved with high TSS content in parthenocarpic fruits induced by pat-k.

This paper summarized 16 controlled studies and evaluated the correlation of resveratrol supplementation with metabolic parameters such as the body weight, waist circumference (WC), systolic blood pressure (sbp), HDL, total cholesterol, triglyceride and glucose levels. This meta-anal. was carried out to determine the association between the resveratrol intake with metabolic parameters in metabolic syndrome patients. PubMed, Scopus, Cochrane and Google Scholar were searched from inception to Dec. 2018 using relevant keywords. All articles were independently reviewed by two authors using predetermined selection criteria. We have selected the studies that investigated the effects of resveratrol on metabolic parameters. Of 16 studies, 10 were performed on human subjects, and in 6 studies animal models were used. Standard mean difference (SMD) with 95% confidence interval were determined using Der Simonian and Laird random-effects modeling, when there was a significant heterogeneity between studies. Funnel plot and Egger′s test were conducted to examine the risk of publication bias. Pooled effect sizes in human studies indicated a significant impact of resveratrol supplementation on glucose level [-1.73 (-2.99, -0.47); p = 0.007] and WC [-1.73 (-2.79, -0.67); p = 0.001] compared with the control group. Also combining the results of studies on rat samples (n = 6), indicated significant effect of resveratrol on decreasing weight [-22.95 (-44.74, -1.17); p = 0.04], TGs [-6.76 (-11.10, -2.42); p = 0.001], sbp [-7.30 (-12.48, -2.13); p = 0.006], and it can influence significantly on increasing HDL level (4.75 (1.87, 7.63); p = 0.001). However, resveratrol was not significantly effective on total cholesterol in both samples. The results of subgroup anal. of human studies showed that resveratrol has significant effect on metabolic parameters (glucose level and WC) at the dosage of > 500 mg and with long-term interventions ≥ 10 wk. Administration of resveratrol can meaningfully reduce the BW, WC, TGs, and glucose level, also it can increase HDL, but not total cholesterol.
