In European countries, silage-free feeding is an ancient tradition and has a particularly pos. reputation among consumers. In the present study, we compared grass-based forages from the same plot conserved as hay or silage or fed fresh either on pasture or indoors, and we evaluated the differences in sensory properties of milk and uncooked pressed cheese. All herbage from the first cut of a grassland dominated by perennial ryegrass was harvested on the same day and preserved either as hay or silage. The first regrowth of the same plot was used for strip grazing or green feeding indoors. Balanced by breed, 24 Montbeliarde and 24 Holstein cows were allocated to the 4 treatments. Apart from the forages, the late-lactation cows received 3 kg/d of dry matter from concentrate After 2 wk of dietary adaptation, the bulk milk of 3 subgroups, each with 4 cows, was collected. Part of the milk was pasteurized, and part was left raw and partly transformed to small-sized Cantal-type cheese ripened for 9 wk. Milk and cheese underwent descriptive sensory anal. by a trained sensory panel, as well as analyses of physicochem. traits. Volatile organic compounds of the cheeses were also analyzed. Raw and pasteurized milk from hay-fed cows had less intense odors of cooked milk, cream, and barnyard than milk from grazing cows, whereby the effect of pasteurization did not differ between herbage utilization methods. Cheeses obtained from cows fed fresh herbage (grazing and indoors) were clearly yellower than cheeses from silage- and hay-fed cows, which coincided with the color intensity perceived by the panelists. Moreover, cheeses from cows fed fresh herbage had more intense barnyard and dry fruit flavors, were perceived as creamier and having less lactic odor, and exhibited more fat exudation than those from cows fed conserved herbage. Only a few differences were observed in milk and cheeses from hay-fed compared with silage-fed cows, and those differences were far less pronounced than those of milk and cheeses from cows fed fresh herbage. In conclusion, the present study did not substantiate assumptions of clear sensory differences of milk and uncooked pressed cheese from hay-fed compared with silage-fed cows. For the first time, this study reports that the global flavor intensity of cheeses from indoor green-fed cows is similar to that of cheeses derived from cows fed conserved forages, whereas cheeses from grazing cows have the greatest global flavor intensity.

Citrus grandis ‘Tomentosa’ (CGT) which is a region-famous medicinal and edible plant contains plentiful bioactive polysaccharides, however, its chem. structures and specific bioactivities still need to be further explored. In the present study, an acidic polysaccharide (CGTP-AP) was extracted and purified from the pulps of CGT. The structure elucidation and anti-cancer activity of CGTP-AP were investigated. Structure characterization indicated that CGTP-AP was a homogeneous heteropolysaccharide composed of arabinose, galactose and galacturonic acid in a molar ratio of 2.45:1:2.77, with an average mol. weight of 2721.68 kDa. Partial acid hydrolysis, methylation and NMR spectrometry revealed that the backbone of CGTP-AP mainly composed of (1 → 4)-α-D-galacturonan, while the branch principally consisted of (1 → 5)-α-L-Araf. In addition, CGTP-AP exhibited effective anti-proliferation against colon cancer cells LOVO and SW620 cells in dose- and time-dependent manners, with IC50 values of 5.55, 4.35 and 3.52 mg mL-1 after 24, 48 and 72 h, and 5.33, 3.63 and 2.97 mg mL-1 after 24, 48 and 72 h, resp. This study indicated that CGTP-AP might be utilized as a promising food supplement for the patients of colon disorders.

An acidic polysaccharide (SAZMP3) was isolated from jujube fruit residues by alk. extraction and purified with DEAE-Sepharose Fast Flow and Sephacryl S-300 column chromatog. The structural characterizations of SAZMP3 were determined by high-performance gelpermeation chromatog., gas chromatog., Fourier transform IR spectroscopy, methylation anal., NMR spectroscopy, SEM, and at. force microscopy. The results determined SAZMP3 was an acidic polysaccharide mainly contained rhamnose, galactose, and galacturonic acid with a mol. weight of 9.37 kDa and its backbone might mainly be composed of 1,4-α-D-GalAp with side chains of 1,3-β-D-Galp, 1,3,5-linked Araf, 1,2,4-α-L-Rhap and terminals of 1-linked Araf, 1-linked Rhap, 1-linked Galp by methylation and NMR anal. Besides, SAZMP3 has shown potential antioxidant activity in scavenging free radicals suggesting that it is an antioxidant agent, which might contribute to the functional food qualities of jujube fruit.

Plants secrete malate from guard cells to apoplast under stress conditions and exogenous malate induces stomatal closure. Malate is considered an extracellular chem. signal of stomatal closure. However, the mol. mechanism of malate-induced stomatal closure is not fully elucidated. We investigated responses of stomatal aperture, ion channels, and cytosolic Ca2+ to malate. A treatment with malate induced stomatal closure in Arabidopsis thaliana wild-type plants, but not in the mutants deficient in the slow (S-type) anion channel gene SLOW ANION CHANNEL-Associated 1 (SLAC1). The treatment with malate increased S-type anion currents in guard-cell protoplasts of wild-type plants but not in the slac1 mutant. In addition, extracellular rather than intracellular application of malate increased the S-type currents of constitutively active mutants of SLAC1, which have kinase-independent activities, in a heterologous expression system using Xenopus oocytes. The treatment with malate transiently increased cytosolic Ca2+ concentration in the wild-type Arabidopsis guard cells and the malate-induced stomatal closure was inhibited by the Ca2+ channel blocker and the Ca2+ chelator. These results indicate that extracellular malate directly activates SLAC1 and simultaneously stimulates Ca2+ signalling in guard cells, resulting in steady and solid activation of SLAC1 for stomatal closure.

Elevated-temperature cured epoxy resins are widely used for high-performance applications especially for structural adhesive and as a matrix for structural composites. This is due to their superior chem. and mech. properties. The thermosetting nature of epoxy produces a highly crosslinked polymer network during the curing process where the resulting material exhibited excellent properties. However, due to this crosslinked mol. structure, epoxies are also known to be brittle, and once a crack initiated in the material, it is difficult to arrest the crack propagation. Earlier research found that the inclusion of encapsulated healing agents is able to introduce self-healing ability to the room-temperature cured epoxies. The current study investigated the self-healing behavior of an elevated-temperature cured epoxy, which incorporated the dual-capsule system loaded with bisphenol A diglycidyl ether (DGEBA) resin and mercaptan. The microcapsules were prepared by the in-situ polymerization method while the fracture toughness and the self-healing capability of the tapered-double-cantilever-beam (TDCB) epoxy specimens were measured under Mode-I fracture toughness testing. We investigated the effect of temperature on viscosity of the healing agents and how these values influence the formation of uniform healing on the fracture surfaces. It was found that incorporation of the dual-capsule self-healing onto an elevated-temperature cured epoxy slightly changed the fracture toughness of the epoxy as indicated by the Mode-I testing.

The lipid content of eight samples of aeolian particles collected on the Frioul Islands (north-western Mediterranean Sea) during the year 2010 was investigated. Lipid analyses indicated a significant contribution of terrestrial higher plant debris to some of the samples, while meat cooking residues were also found. Specific degradation products of sterols and monounsaturated fatty acids were employed to assess the importance of biotic and abiotic degradation processes on land and during aeolian transport. Degradation of higher plant lipids on land appears to involve Type II (i.e., singlet oxygen-mediated) photooxidation during senescence and bacterial degradation processes in soils, while during aeolian transport this terrestrial material mainly undergoes autoxidation and/or Type I (i.e., radical-mediated) photooxidation and is relatively well preserved towards bacterial degradation In contrast, bacteria play a significant role in the degradation of meat cooking residues in these particles.

Analogs of the tumor-associated gangliosides GM3 and GM2 containing terminal S-linked neuraminic acid residues and an amino terminated, truncated ceramide homolog have been synthesized and conjugated to a protein. The synthesis involved coupling of a S-linked sialyl α(2→3) galactose disaccharide with a glucosyl sphingosine analog, followed by elaboration and deprotection to give amino-terminated glycosyl ceramide. Glycosyltransferase-catalyzed extension of the trisaccharide provided access to the modified GM2 tetrasaccharide or sulfur-containing GD3 analog. Owing to their potentially enhanced resistance to endogenous exo-glycoside hydrolases and their inherent non-self character, carbohydrate antigens containing non-reducing terminal thioglycosidic linkages may be more immunogenic than O-linked antigens and may stimulate the production of antibodies capable of recognizing naturally occurring oligosaccharides. Our initial results suggest that in fact these antigens are viable immunogens and furthermore, that immune sera cross reacts with O-gangliosides in the context of a heterologous glycoprotein conjugate.

A new type of glycocluster based on polyhedral oligo-silsesquioxanes (POSS) has been efficiently prepared from unprotected mannoside and lactoside employing a convergent approach of thiol-radical addition reaction. Preliminary lectin binding study indicated that these novel glycoclusters showed strong inhibition of the binding of asialo-oligosaccharide mixture derived from human 1-acid glycoprotein by RCA120.

Thirty one terpenes were analyzed by selected reaction monitoring via GC-MS/MS and isotope dilution using β-myrcene-d6 as a surrogate. All quant. analyses can be accomplished using less than 1 g of material, with minimal solvent and consumable use, on low resolution instruments in less than 30 min of instrument time. Of important note is this method’s power of selectivity, working ranges, and lack of need for extraction consumables such as SPE or QuEChERS, thereby minimising anal. costs and time.
