This study aimed to investigate the activity, structural characterization and present forms of selenium in two different artificial selenium-enriched tea polysaccharides (Se-TPS). The physicochem. anal. showed that both CSe-tps1 and ASe-tps1 were acidic heteropolysaccharides with different monosaccharide composition, molar ratio, and selenium content. Structural investigations including FT-IR, Raman spectra and 1D, 2D NMR anal. revealed that selenium in CSe-tps1 replaced the hydroxyl group at the C-6 position in the polysaccharide with the form of selenyl ester, while most of the selenium in ASe-tps1 replaced the hydroxyl group at the C-1 and C-6 position in the form of the Se-H bond on the branch of the polysaccharide. Besides, a series of studies on the structural characteristics of the Congo red test, I2-KI reaction, FESEM, DSC, and XRD anal. showed that the two artificial Se-TPSs had a triple helix structure and more branches. However, there were significant differences in crystal morphol., apparent morphol., heat release ability, and the α-glucosidase inhibition activity. These results indicated that the difference in artificial selenization methods not only made the polysaccharide exhibit different structural characteristics and the hypoglycemic activity, but also had a significant effect on the form of selenium in polysaccharides.

Bio-based nanoemulsions are part of green pest management for sustainable agriculture. This study assessed the physicochem. properties and the herbicidal activities of the peppermint essential oil nanoemulsions (PNs) in concentrations 1.0-10% stabilized by Eco-Polysorbate 80 on germinating seeds and young plants of maize and barnyard grass. Based on the design of experiment (DOE) results, the final nanoemulsion formulations were obtained with 1, 1.5, 2, and 5% of essential oil concentration Biol. analyses were conducted to select the most promising sample for selective control of barnyard grass in maize. Seedlings growing in the presence of PNs displayed an overall inhibition of metabolism, as expressed by the calorimetric analyses, which could result from significant differences in both content and composition of carbohydrates. Concentration-response sub estimation showed that leaf-sprayed concentration of PN causing 10% of maize damage is equal to 2.2%, whereas doses causing 50% and 90% of barnyard grass damage are 1.1% and 1.7%, resp. Plants sprayed with PN at 5% or 10% concentration caused significant drops in relative water content in leaves and Chlorophyll a fluorescence 72 h after spraying. In summary, peppermint nanoemulsion with Eco-Polysorbate 80 at 2% concentration is a perspective preparation for selective control of barnyard grass in maize. It should be analyzed further in controlled and field conditions.

The increased interest in functional materials of natural origin has resulted in a higher market demand for preservative-free, “”clean label””, or natural ingredients-based products. The gummy bear food supplements are more acceptable to consumers and have fewer limitations compared to other dosage forms. The aim of our study was to produce natural ingredients-based gummy bear composition, and evaluate the influence of the selected ingredients on the product’s textural properties, its acceptance in vivo, and the gummy bear’s quality. The optimal base composition was determined using a surface response design: gelatin 4.3 g and agave syrup 6.3 g. The investigated sweeteners did not affect the textural properties (p > 0.05). However, further studies demonstrated that a 100% increase of agave results in up to 27% higher flexibility (p < 0.05). The addition of calcium and cholecalciferol reduced firmness by 59.59 ± 1.45% (p < 0.05). On the other hand, acai berry extract had no significant effect. The presence of calcium resulted in a decreased smell and taste; however, the data indicated that exptl. texture anal. is a more accurate technique than in vivo evaluation. The acai berry extract did not improve all of the tested sensory properties. We can conclude that the suggested gummy bear base can be supplemented with various active ingredients and commercialized, though further studies are needed to investigate the other natural sources to mask the unpleasant taste of active ingredients and avoid water loss. Gleditsia sinensis, fenugreek and guar galactomannans (referred to as GSG, FG, and GG) were extracted from their gums and investigated using various techniques. Mannose to galactose ratios were 3.55, 1.11, and 1.65, resp. The intrinsic viscosity of GSG was very close to that of GG, while that of FG was the lowest one. This was attributed to the influence of high galactose substitution of FG on the mannan backbone, which induced a lower chain dimension due to intermol. entanglement. High degrees of substitution and high temperatures contributed to improving the solubility of galactomannan. Rheol. behavior indicated that GG had the highest apparent viscosity, yet the power-law model could well-fitted the flow curves of GSG and FG, but not GG. Through morphol. observations, the extracted galactomannans exhibited rod-like structure in deionized water and showed fibrous filament network structure after dehydration by freeze-drying. The thermal behavior was greatly influenced by the degree of side groups and Mw of galactomannans.

Stomatal responses to environmental signals differ substantially between ferns and angiosperms. However, the mechanisms that lead to such different responses remain unclear. Here we investigated the extent to which leaf metabolism contributes to coordinate the differential stomatal behavior among ferns and angiosperms. Stomata from all species were responsive to light and CO2 transitions. However, fern stomatal responses were slower and minor in both absolute and relative terms. Angiosperms have higher stomatal d., but this is not correlated with speed of stomatal closure. The metabolic responses throughout the diel course and under different CO2 conditions differ substantially among ferns and angiosperms. Higher sucrose content and an increased sucrose-to-malate ratio during high CO2-induced stomatal closure was observed in angiosperms compared to ferns. Furthermore, the speed of stomatal closure was pos. and neg. correlated with sugars and organic acids, resp., suggesting that the balance between sugars and organic acids aids in explaining the faster stomatal responses of angiosperms. Our results suggest that mesophyll-derived metabolic signals, especially those associated with sucrose and malate, may also be important to modulate the differential stomatal behavior between ferns and angiosperms, providing important new information that helps in understanding the metabolism-mediated mechanisms regulating stomatal movements across land plant evolution.

Oxidative phosphorylation is compromised in hypoxia, but many organisms live and exercise in low oxygen environments. Hypoxia-driven adaptations at the mitochondrial level are common and may enhance energetic efficiency or minimize deleterious reactive oxygen species (ROS) generation. Mitochondria from various hypoxia-tolerant animals exhibit robust functional changes following in vivo hypoxia and we hypothesized that similar plasticity would occur in naked mole-rat skeletal muscle. To test this, we exposed adult subordinate naked mole-rats to normoxia (21% O2) or acute (4 h, 7% O2) or chronic hypoxia (4-6 wk, 11% O2) and then isolated skeletal muscle mitochondria. Using high-resolution respirometry and a fluorescent indicator of ROS production, we then probed for changes in: (i) lipid- (palmitoylcarnitine-malate), (ii) carbohydrate- (pyruvate-malate), and (iii) succinate-fueled metabolism, and also (iv) complex IV electron transfer capacity, and (v) H2O2 production Compared to normoxic values, (a) lipid-fueled uncoupled respiration was reduced ∼15% during acute and chronic hypoxia, (b) complex I-II capacity and the rate of ROS efflux were both unaffected, and (c) complex II and IV uncoupled respiration were suppressed ∼16% following acute hypoxia. Notably, complex II-linked H2O2 efflux was 33% lower after acute hypoxia, which may reduce deleterious ROS bursts during reoxygenation. These mild changes in lipid- and carbohydrate-fueled respiratory capacity may reflect the need for this animal to exercise regularly in highly variable and intermittently hypoxic environments in which more robust plasticity may be energetically expensive.

The concentrations of different metabolites in olive (Olea europaea (L.)) fruit at harvest can be affected by water availability, with significant consequences on the composition and the quality of the resulting oil. The aim of the present study was to profile the metabolic composition of ripe olives (cv. Cipressino) grown under water-stress and irrigated conditions applied during the last part of the fruit developmental cycle (from pit hardening to com. harvest). The imposed conditions resulted in a moderate water stress (-3.5 MPa) at the end of the exptl. period. Samples (pulp + skin) of fruit collected at the stage of complete epicarp pigmentation were analyzed by means of GC-MS. In total, 176 metabolites were detected, of which 57 were identified. Principal component anal. (PCA) of stress and non-stress treatments resulted in clearly separated profiles with the first principal component (PC1) mostly correlated with the organic acid content. Of the 57 compounds identified, 19 metabolites (organic acids, fatty acids, soluble sugars, and terpens) accumulated differently in the two sets of samples. A reduction in soluble sugars and unsaturated fatty acids was detected in water stressed samples, suggesting an acceleration of the ripening process. These results highlighted the validity of metabolic profiling to study the effects of water stress in terms of both fruit composition and physiol.

Examination of antioxidant activity of Pleurotus ostreatus polysaccharide (POP) was directed by utilizing 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and 2, 2-azino-bis-3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) techniques. Cytotoxicity of POP was evaluated using an MTT assay. The exptl. data were fitted to a quadratic equation utilizing multiple regression investigations, and the ideal conditions were as per the following: water/crude material proportion, 26.04 mL/g; an extraction time of 62.08 min; and an extraction temperature 70.5°C. Under such conditions, the polysaccharide yield was 5.32 ± 0.12% with the anticipated yield.
