In this study, different extraction methods of polysaccharides from Crataegus pinnatifida Bunge (CPP) were compared by studying the extraction yield, structural characteristics and antioxidant activities. Firstly, polysaccharides were obtained using hot water extraction (CPPh), ultrasound assisted extraction (CPPu), enzyme assisted extraction (CPPe) and enzyme-ultrasound assisted extraction (CPPc), resp. Meanwhile, the optimum extraction conditions of enzyme-ultrasonic assisted extraction were determined by response surface method (RSM). The extraction yields, structural characteristics and antioxidant activities were investigated and compared by visual photos, gas chromatog., UV-visible and Fourier-transform IR spectroscopy. The results clearly showed that enzyme-ultrasonic assisted extraction possessed the highest extraction yield (10.39 ± 0.04%). The mol. weight of CPPh was the highest while the other polysaccharides had no significant difference. Besides, the monosaccharide composition of CPPc, CPPh, CPPu and CPPe were similar but the molar percentages of monosaccharide were different. Finally, the results of antioxidant activities showed that CPPc exhibited the highest scavenging effect of superoxide radical and lipids inhibiting ability. In summary, enzyme-ultrasonic assisted extraction was a high-efficient and low-energy consumption method for CPP extraction

Traditional glutinous rice wine (TGRW) has been fermented in China for over 9000 years. Recently, an innovative regional variation of TGRW, chestnut rice wine, banli mijiu (BLMJ), was developed by adding Chinese chestnut (Castanea mollissima Blume) into the fermentation brew. The objective of this study was to characterize the effects of chestnut on the nutritional, aromatic, and antioxidant properties of TGRW. To compare the aromatic sensory profiles between TGRW and BLMJ, the free amino acids and Et carbamate, phenolic, and flavonoid contents were determined In addition, the antioxidant properties, including reducing power, metal chelation, and free radical scavenging activities, were also compared. A total of 98 distinct flavor components were identified in BLMJ, among which 38 were detected by sniffing instrument, compared to 77 distinct flavor components in TGRW. BLMJ thus contains a wider range of flavor components, but similar alc., acid and reducing sugar profiles compared with TGRW. Twenty-five free amino acids were detected in both wines, with lower contents of each in BLMJ compared with TGRW. BLMJ also exhibited stronger antioxidant properties than TGRW. The findings of this study suggest that chestnut can increase the diversity of aromatic components and improve antioxidant qualities of traditional rice wine.

Sparassis crispa polysaccharides have recently attracted considerable attention due to their excellent bioactivities. However, their extraction procedure is often tedious, time-consuming, and environmentally unfriendly; it even causes damage to their structures and reduces their bioactivities, all of which hinder their further development to some extent. Therefore, the ultrasonic-assisted enzymic extraction (UAEE) technol. was optimized to extract polysaccharides from Sparassis crispa (SCP) by the response surface methodol. The yields, physicochem. properties, and antioxidant activities of SCPs obtained from UAEE and conventional hot water extraction (HWE) were evaluated. According to the optimal parameters, the yield of SCPs extracted by UAEE reached up to 14.63%, which increased by 68.54% compared with that obtained from the conventional hot water extraction (HWE) method. Addnl., the UAEE methods affected the contents of the polysaccharides, mol. weights, and the molar percentage of the constituent monosaccharides of SCPs. SEM anal. indicated that the microstructures of the two SCPs were notably different. Antioxidant assays showed that both SCPs possessed good antioxidant activities against DPPH, ABTS +, and hydroxyl radicals in vitro. Addnl., the SCPs extracted by UAEE attenuated the HT22 cell neurotoxicity induced by H2O2 by the means of ameliorating cell viability, reducing extracellular LDH release, and decreasing the levels of intracellular ROS.

Abstract: Inflammatory bowel disease (IBD) is often associated with a decrease in energy-dependent nutrient uptake across the jejunum that serves as the main site for absorption in the small intestine. This association has prompted us to investigate the bioenergetics underlying the alterations in jejunal absorption in 2,4,6-trinitrobenzenesulfonic acid-induced colitis in rats. We have found that mitochondrial oxygen consumption did not change in state 2 and state 3 respirations but showed an increase in state 4 respiration indicating a decrease in the respiratory control ratio of jejunal mitochondria during the peak of inflammation. This decrease in the coupling state was found to be guanosine diphosphate-sensitive, hence, implicating the involvement of uncoupling protein-2 (UCP2). Furthermore, the study has reported that the production of reactive oxygen species (ROS), known to be activators of UCP2, correlated neg. with UCP2 activity. Thus, we suggest that ROS production in the jejunum might be activating UCP2 which has an antioxidant activity, and that uncoupling of the mitochondria decreases the efficiency of energy production, leading to a decrease in energy-dependent nutrient absorption. Hence, this study is the first to account for an involvement of energy production and a role for UCP2 in the absorptive abnormalities of the small intestine in animal models of colitis.

In this study, high-pressure ultrasonic-assisted extraction (HUE) technol. was used for polysaccharide extraction from Hovenia dulcis (HDPS). A maximal extraction yield (11.81 ± 0.26%) was obtained using the response surface method (RSM) with optimized HUE conditions (Ultrasound power 330 W, 68 °C, and 22 min). Results showed that HDPS mainly contained neutral sugar (47.81 ± 1.56%), uronic acid (12.52 ± 0.15%), and protein (3.64 ± 0.24%), specifically. HDPS was comprised of mannose, rhamnose, galacturonic acid, glucose, galactose and arabinose in a molar ratio of 3.20%, 20.40%, 4.81%, 11.14%, 27.37% and 33.08%, resp. And with an average mol. weight of 185.22 kDa for its major component (accounting for 90.13% for HDPS). The result obtained from the X-ray diffraction pattern confirmed a semi-crystalline nature of the sample. HUE-HDPS suggested typical shear thinning behavior and non-Newtonian properties. Addnl., HUE-HDPS exhibited better chem. antioxidant activities in DPPH, ABTS, hydroxyl radical scavenging and Fe2+ chelation activity. The in-vitro and in-vivo experiments indicated that HUE-HDPS had obviously hypoglycemic effects. Accordingly, the polysaccharides from Hovenia dulcis might serve as a natural antioxidant and hypoglycemic agent.

A new polysaccharide (pFSP) was first isolated from the originally discarded fibrous roots part of Chinese traditional herb, Bletilla striata. PFSP was composed of D-glucose, D-galactose and D-mannose in a molar ratio of 1: 2.03: 3.45 with mol. weight of 9.1 × 104 Da. It could effectively scavenge DPPH and superoxide radicals with inhibition rate of 64.47% and 72.27% at 5.0 mg/mL, higher than that of polysaccharide from Bletilla striata tuber. Structural investigations of the periodate oxidation studies and Smith-degradation as well as the FT-IR spectroscopy were performed, and combined with 1D and 2D NMR spectroscopy, the repeating unit of pFSP contained (1→4)-linked-α-D-Glcp, (1→4)-linked-β-D-Manp and (1→3,6)-linked-β-D-Manp units, together with the branches of (1→6)-linked-β-D-Galp and terminated with (1→)-linked-β-D-Manp residue.

Microwave-assisted extraction of the polysaccharide from blackcurrant (Ribes nigrum L.) fruits (MBP) was optimized using response surface methodol. (RSM). After purification using D4006 macroporous resin and Sepharose-6B column chromatog., a water-soluble acidic heteropolysaccharide (PMBP) was obtained with a mol. weight of 1.99 × 106 g/mol. PMBP was thermally stable and exhibited excellent rheol. properties. Fourier transform IR spectroscopy, gas chromatog. and NMR anal. confirmed the presence of α-, β-pyranose and six main glycosidic linkages in PMBP. SEM, at. force microscopy, and X-ray diffraction studies demonstrated that PMBP was amorphous aggregates with irregular shape, porous, and smooth surface. Bioactivity assays revealed the notable antioxidant and hypoglycemic activities of PMBP. Moreover, PMBP was kinetically characterized as a competitive inhibitor for α-amylase and α-glucosidase.

In the present study, the native polysaccharide (RNP) extracted from blackcurrant fruits was carboxymethylated. Physicochem. characteristics and antioxidant activities in vitro of RNP and three carboxymethylated polysaccharides (CRNPs) were determined GC anal. proved that RNP and CRNPs were composed of the same six monosaccharides (galacturonic acid, rhamnose, arabinose, mannose, glucose and galactose), but the molar ratios of monosaccharides were different. HPLC demonstrated that the mol. weights of CRNPs were improved. The assays of the antioxidant properties indicated that CRNPs possessed stronger scavenging activities on radicals (hydroxyl and superoxide radicals) and better anti-lipid peroxidation activities, as well as better protection effects on erythrocyte hemolyzes in vitro compared with RNP. The activities of CRNPs were significantly enhanced with the increase of the degree of substitution (DS). These results proved that the carboxymethylation could effectively increase the antioxidant activities of the polysaccharide.

Magnolol, a neolignan natural product with antioxidant properties, contains inherent, orthogonal, phenolic, and alkenyl reactive groups that were used in both direct thermoset synthesis, as well as the stepwise synthesis of a small library of monomers, followed by transformation into thermoset materials. Each monomer from the small library was prepared via a single step functionalization reaction of the phenolic groups of magnolol. Thermoset materials were realized through solvent-free, thiol-ene reactions, and the resulting crosslinked materials were each comprised of thioether and ester linkages, with one retaining the hydrophilic phenols from magnolol, another having the phenols protected as an acetonide, and two others incorporating the phenols into addnl. crosslinking sites via hydrolytically labile carbonates or stable ether linkages. With this diversity of chem. compositions and structures, the thermosets displayed a range of thermomech. properties including glass transition temperatures, Tg, 29-52 °C, onset of thermal degradation, Td, from about 290-360 °C, and ultimate strength up to 50 MPa. These tunable materials were studied in their degradation and biol. properties with the aim of exploiting the antioxidant properties of the natural product. Hydrolytic degradation occurred under basic conditions (pH = 11) in all thermosets, but with kinetics that were dependent upon their chem. structures and mech. properties: 20% mass loss was observed at 5, 7, 27, and 40 wk for the thermosets produced from magnolol directly, acetonide-protected magnolol, bis(allyl carbonate)-functionalized magnolol, and bis(allyl ether)-functionalized magnolol, resp. Isolated degradation products and model compounds displayed antioxidant properties similar to magnolol, as determined by both UV-vis and in vitro reactive oxygen species (ROS) assays. As these magnolol-based thermosets were found to also allow for extended cell culture, these materials may serve as promising degradable biomaterials.

Distichochlamys citrea M.F. Newman (commonly known as Black Ginger) is an endemic plant to Vietnam and has been extensively exploited by folk medication for treatments of infection-related diseases and diabetes. In this work, its rhizomes were subjected to fractionated extraction, phytochem. examination, evaluation of antioxidant effect by DDPH free radical neutralization, and inhibitory activity toward α-glucosidase. The compositional components were subjected to in silico screening, including d. functional theory calculation, mol. docking simulation, physicochem. anal., and pharmacokinetic regression. In the trials, EtOAc fraction is found as the bioactive part of most effectiveness, regarding both antioxidant effect (IC50 = 90.27 μg mL-1) and α-glucosidase inhibitory activity (IC50 = 115.75 μg mL-1). Chem. determination reveals there are 13 components of its composition DFT-based calculations find no abnormal constraints in their structures. Docking-based simulation provides order of inhibitory effectiveness: 3-P53341 > 12-P53341 > 7-P53341 > 4-P53341 > 11-P53341 > 10-P53341. QSARIS-based investigations implicate their biocompatibility. ADMET-based regressions indicate that all candidates are generally safe for medicinal applications. The findings would contribute to the basis for further studies on the chem. compositions of Distichochlamys citrea and their biol. activities.
