In this study, response surface methodol. (RSM) was used to optimize the extraction parameters of dandelion root polysaccharide (DRP). Two novel polysaccharides (DRP-2b, DRP-3a) were isolated from dandelion root by graded ethanol precipitation and column chromatog. Structural analyses indicated that DRP-2b, with a mol. weight of 31.8 kDa, was composed of rhamnose, glucuronic acid, glucose, galactose and arabinose, whereas DRP-3a, with a mol. weight of 6.72 kDa, was composed of rhamnose, glucose, galactose and arabinose. The backbones of DRP-2b and DRP-3a were mainly composed of (1 �5)-linked-a-D-Ara and (1 �6)-linked-a-D-Glc, resp. In addition, the antioxidant activities and protective effects against H2O2-induced damage in hepatic L02 cells were evaluated. DRP-3a exhibited higher radical scavenging activity than DRP-2b against the DPPH, hydroxyl and superoxide anions. Furthermore, DRP-3a exhibited strong protective effects against H2O2-induced damage in hepatic L02 cells. Thus, DRP-3a could be used as a potential antioxidant in medicine or as a functional food.

In order to explore snow chrysanthemum polysaccharides (SCPs) as functional food ingredients and natural antioxidants for industrial applications, both microwave-assisted extraction (MAE) and ultrasonic-assisted extraction (UAE) were firstly optimized for the extraction of SCPs. Furthermore, the effects of conventional hot water extraction, UAE, and MAE on the chem. structures and antioxidant activities of SCPs were investigated. The maximum extraction yields of SCPs extracted by UAE (4.13 ± 0.24%) and MAE (4.26 ± 0.21%) were achieved at the optimized extraction parameters as follows: ultrasound amplitude (68%) and microwave power (500 W), ultrasound extraction time (21 min) and microwave extraction time (6.5 min), and ratio of liquid to raw material (42.0 mL/g for UAE and 59.0 mL/g for MAE). In addition, different extraction methods significantly affected the contents of uronic acids, the mol. weights, the molar ratio of constituent monosaccharides, and the degree of esterification of SCPs. SCPs exhibited remarkable DPPH (IC50 = 1.702 mg/mL), ABTS (IC50 = 1.121 mg/mL), and nitric oxide (IC50 = 0.277 mg/mL) radical scavenging activities, as well as reducing power (= 80.17 ± 4.8μg Trolox/mg), which suggested that SCPs might be one of the major contributors toward the antioxidant activities of snow chrysanthemum tea. The high antioxidant activities (DPPH, IC50 = 0.693 mg/mL; ABTS, IC50 = 0.299 mg/mL; nitric oxide, IC50 = 0.105 mg/mL; and reducing power, 127.79 ± 2.57μg Trolox/mg) observed in SCP-M extracted by the MAE method might be partially attributed to its low mol. weight and high content of unmethylated galacturonic acids. Results suggested that the MAE method could be an efficient technique for the extraction of SCPs with high antioxidant activity, and SCPs could be further explored as natural antioxidants for industrial application.

Intensified process based on subcritical water conditions (120°C and 1.5 atm, for 2 h) was utilized to prepare thyme oil in water (O/W) nanoemulsions. In this technique, water and xanthan gum, as green solvent and natural microbial emulsifier, were utilized. Results of gas chromatog. revealed that Thymol and Carvacrol were two main bioactive compounds of the extracted thyme oil. Effects of amounts of xanthan gum (0.05-0.25 g) and thyme essential oil (0.2-0.8 mL) on size of oil nanodroplets and polydispersity index (PDI) of the resulted nanoemulsions were evaluated using response surface methodol. Results demonstrated that the produced thyme O/W nanoemulsion by 0.242 mL thyme oil and 0.140 g xanthan gum had smallest average nanodroplet size (150 nm) and PDI (0.088). Furthermore, monodispersed and spherical in shape thyme oil nanodroplets were provided in the nanoemulsion using these optimal conditions with zeta potential value of -10.1 mV and antioxidant activity of 17.4%. Results also indicated that this prepared nanoemulsion had high fungicidal and bactericidal activities toward Penicillium digitatum and Escherichia coli, resp.

Besides, according to the GC-MS and LC-MS/MS results, this species was determined to contain considerable amounts of ferruginol (9292.9 μg analyte/g extract) and rosmarinic acid (15612.08 μg analyte/g extract). In parallel with the high total phenolic and rosmarinic content of the species, its ethanol extract has a high antioxidant potential. It was determined that the species was not active against acetylcholinesterase enzyme, however, the root ethanol extract showed good activity (Inhibition%: 63.01 ± 1.12) against butyrylcholinesterase enzyme. All extracts of the species were found to have fairly high anti-urease activity (Inhibition%: 89.67 ± 0.72), however, all extracts of S. poculata showed moderate and similar activities in anti-tyrosinase and antielastase activity assays.

Polysaccharides were extracted from jujube fruit using an efficient, optimized subcritical water extraction process. A maximum crude polysaccharide yield of 7.9% was obtained at an extraction temperature of 140°C and a time of 60 min. The crude polysaccharides extracted under these conditions were purified. Three polysaccharide fractions, ZP1, ZP2 and ZP3, were obtained and their structures were investigated. The three fractions were mainly composed of arabinose, galactose, glucose, mannose, rhamnose, and galacturonic acid. ZP3 was further investigated using methylation and NMR spectroscopy, and ZP3 comprised a main chain of (1 â†?4)-α-D-GalAp, (1 â†?2,4)-α-L-Rhap, with a (1 â†?4,6)-α-D-Galp branch. Assays of antioxidant activity showed that ZP3 exhibited a remarkable DPPH capacity and moderate hydroxyl radical scavenging ability, suggesting that it has potential applications as a natural antioxidant in foods and pharmaceuticals.

Major causatives for cutaneous leishmaniasis, the most common form of leishmaniasis, in the old world are Leishmania major and Leishmania tropica. Investigating the leishmanicidal effects of essential oils and preparing their nano/formulation is a promising approach for developing new green drugs. In this study, the ingredients of Cinnamomum zeylanicum essential oil were investigated using GC/MS anal., and its antioxidant effect was then evaluated using the DPPH assay. Moreover, the leishmanicidal effect of the essential oil against L. major and L. tropica was investigated; obtained half-maximal inhibitory concentrations (IC50)s were 16.53 and 7.56μg/mL. After that, a practical dosage form of C. zeylanicum, i.e., the nanoemulsion with a 52 ± 4 nm droplet size and a low droplet size distribution index (SPAN = 0.97), was prepared Interestingly, after treatment of both species with nanoemulsion, their viability was reduced to 0%. The prepared nanoformulation could be considered a potent anti-leishmanial agent for other species and in vivo studies.

In this study, a novel water-soluble polysaccharide (PVLP-1) was extracted and purified from Sacha inchi (Plukenetia volubilis L.) seeds and the structure, antioxidant and immunomodulatory activity of PVLP-1 were investigated. PVLP-1 (144 kDa) consisted of glucose (69.76%), mannose (14.86%), arabinose (10.53%), galactose (2.42%), ribose (1.23%), rhamnose (0.27%) and xylose (0.93%). PVLP-1 displayed characteristic polysaccharide bands in Fourier transform NMR spectra and IR. The primary structure of PVLP-1 was a heteropolysaccharide with a backbone of (1 â†?6)-linked glucose, sidechains of (1 â†?4)-linked mannose, (1 â†?4)-linked glucose and (1 â†?3, 6)-linked mannose and a residue unit of â†?)-linked arabinose as revealed the methylation anal. PVLP-1 possessed good water-holding capacity (WHC), oil-holding capacity (OHC) and antioxidant capacities. Besides, PVLP-1 induced the proliferation of RAW264.7 cell and enhanced the expression of inflammatory cytokines IL-6, TNF-alpha(TNF-α) and IL-1 beta (IL-1β). The present study indicated that PVLP-1 possessed immune-enhancing bioactivities and could be functional food or adjuvant drug to improve biol. immunity of immunodeficiency diseases and hypoimmunity.

Chinese water chestnut peels are a kind of vegetable processing waste containing many active components such as polysaccharides, the structure of which remains unknown. To elucidate the structure of polysaccharides from Chinese water chestnut peels, two polysaccharides named WVP-1 and WVP-2 were isolated. WVP-1 (3.16 kDa) consisted of mannose (1.75%), glucose (84.69%), galactose (6.32%), and arabinose (7.24%), while WVP-2 (56.97 kDa) was composed of mannose (3.18%), rhamnose (1.52%), glucuronic acid (1.42%), galacturonic acid (4.83%), glucose (11.51%), galactose (36.02%), and arabinose (41.53%). Linkage and NMR data indicated that WVP-1 was composed mainly of â†?)-α-D-Glcp(1â†?and a certain proportion of â†?)-β-D-Glcp-(1â†? including linear and branched polysaccharides simultaneously. WVP-2 was a pectin-like polysaccharide with â†?)-α-D-GalpA6Me-(1â†?units and the branch points of â†?,4)-α-L-Arap-(1â†? â†?,6)-β-D-Galp-(1â†? WVP-2 exhibited stronger potential antioxidant and immunomodulatory activities than WVP-1 in vitro. In particular, drought induced an increase in sesquiterpene production, a class of terpenoids that is important in the food, cosmetics, and pharmaceutical industries. Thus, controlled drought, in addition to water savings during cultivation, can be applied to improve the production of secondary metabolites in S. dolomitica.

Herein, we isolated and purified polysaccharides from Craterellus cornucopioides. Chem. composition anal. revealed that both polysaccharides (CCPs-1 amd CCPs-2) were composed of different molar ratios of rhamnose, fucose, arabinose, xylose, mannose, glucose, and galactose. The structure of polysaccharides was determined using FT-IR, periodate oxidation, smith degradation, methylation anal., and NMR, which demonstrated that the two polysaccharides were primarily connected by mannose with (1â†?)-linked. Notably, the configuration of the two polysaccharides exhibited random coil with pyranoid polysaccharide containing α or β glycosidic bond. Furthermore, to increase the antioxidant activity of polysaccharides (CCPs-1 amd CCPs-2), two carboxymethylated polysaccharides were obtained as cmCCPs-1 and cmCCPs-2 with the degree of substitution values of 0.34 and 0.52, resp. Structural anal. elucidated that nonselective carboxymethylation occurred in cmCCPs-1 and cmCCPs-2, where C-2, C-4 or C-6 were partially substituted. Of note, cmCCPs-2 exhibited the most significant antioxidant activity.
