The aim of this study was to define optimal conditions by using Simplex lattice mixture design for the formulation of sucrose-free chocolate produced in a ball mill. Milk chocolate mass were made using three different polyols including maltitol, xylitol and isomalt along with Stevia as high potency sweetener. The influences of polyols mixtures as sucrose substitutes on rheol. properties and main phys. quality parameters were investigated. According to the results, the fitted models demonstrated a high coefficient of determination (≥93%). The optimization of the variables illustrated that utilizing 11.16% weight/weight maltitol, 8.9% weight/weight xylitol and 12.93% weight/weight isomalt produced the optimum milk chocolate with the highest desirability (1.00) without unwanted variations in the rheol. and quality characteristics. Also the optimum formulation was produced to validate the optimum model. The sensory anal. of the optimized formulation satisfied the consumer demand.

The β-D-galactosyl-(1,3)-α-N-acetyl-D-galactosamine disaccharide is present in antifreeze glycoproteins (AFGPs). Analogs of this disaccharide including the β-linked (1,3)-, (1,4)-, and (1,6)-galactosyl-N-acetyl galactosamine and the β-(1,3)-galactosyl-galactoside were synthesized and evaluated for ice recrystallization inhibition (IRI) activity. The results from this study demonstrate that the β-linked-(1,4) disaccharide exhibits more potent IRI activity than the native β-linked-(1,3) disaccharide. The C2 N-acetyl group of the disaccharide does not affect IRI activity but in monosaccharides, the presence of the C2 N-acetyl group decreases IRI activity. The current study will facilitate the design of potent small-mol. ice recrystallization inhibitors.

In this research, bacterial cellulose (BC), one of the most promising biopolymers of the recent years, was saturated with thyme, eucalyptus and clove essential oils (EOs) and applied against staphylococcal and pseudomonal biofilms formed on hydroxyapatite (HA). BC dressings were thoroughly analyzed with regard to their phys. properties. Moreover, the exact composition and ability of particular EO mols. to adhere to HA was assessed. Addnl., cytotoxicity of oil-containing, cellulose-based dressings towards osteoblasts and fibroblasts as well as their impact on reactive oxygen species (ROS) production by macrophages was assessed. The results revealed the high ability of BC dressings to absorb and subsequently release EOs from within their microstructure; the highest number of compounds able to adhere to HA was found in the thyme EO. The eucalyptus EO displayed low, while thyme and clove EOs displayed high cytotoxicity towards fibroblast and osteoblast cell lines. The clove EO displayed the highest eradication ability toward staphylococcal, while the thyme EO against pseudomonal biofilm. Taken together, the results obtained indicate the suitability of EO-saturated BC dressings to eradicate pseudomonal and staphylococcal biofilm on HA surface and moreover, to not trigger reactive oxygen species production by immune system effector cells. However, due to cytotoxic effects of thyme and clove EOs towards cell lines in vitro, the eucalyptus EO-saturated BC dressing is of highest potential to be further applied.

Liquid crystal elastomers (LCEs) are shape morphing materials promising for many applications including soft robotics, actuators, and biomedical devices, but current LCE synthesis techniques lack a simple method to program new and arbitrary shape changes. Here, we demonstrate a straightforward method to directly program complex, reversible, non-planar shape changes in nematic LCEs. We utilize a double network synthesis process that results in a competitive double network LCE. By optimizing the crosslink densities of the first and second network we can mech. program non-planar shapes with strains between 4-100%. This enables us to directly program LCEs using mech. deformations that impart low or high strains in the LCE including stamping, curling, stretching and embossing methods. The resulting LCEs reversibly shape-shift between the initial and programed shape. This work widens the potential application of LCEs in biomedical devices, soft-robotics and micro-fluidics where arbitrary and easily programed shapes are needed.

In Brazil, humane slaughter regulations do not include fish, and hypothermia or immersion in ice water is used in most fish slaughterhouses for pre-slaughter handling. Such a method is not considered humanitarian as it does not induce an immediate loss of consciousness and fish show aversive behavior when immersed in a water and ice mixture This study aimed to assess the times for induction and recovery using Ocimum americanum and Lippia alba essential oils as anesthetics in Nile tilapia, and their influence on the flavor and aroma of fillets. For the anesthetic evaluation, Nile tilapia (224.05 ± 67.56 g, 22.10 ± 2.41 cm) were divided into four groups (40 fish/group): ethanol (ET), 2-phenoxyethanol (PE), Ocimum americanum (OA) and Lippia alba (LA), all at 500μL/L. The times taken to reach total loss of equilibrium, anesthesia and consciousness recovery were recorded using a stopwatch. Blood was collected from the caudal vein after 0, 1, 4, 8 and 24 h of anesthesia for the anal. of glucose, lactate and cortisol. For the sensory anal., fillets from fish exposed to essentials oils and hypothermia at pre-slaughter were evaluated by 122 untrained assessors to determine differences in the aroma and flavor attributes. The induction of deep anesthesia (stage 4) was faster in the OA group (mean ± standard error of mean SEM) (126.8 ± 4.9 s), than the PE (152.3 ± 6.6 s) and LA groups (169.7 ± 10.9 s) (p < .01). A total of 91.7% (110/120) of the fish showed VOR (Vestibulo-Ocular Reflex) as the first behavioral response during consciousness recovery after anesthesia. The levels of glucose (mmol/L) were similar amongst the groups at each time. The lactate levels (mmol/L) were higher at 0 h of sampling in LA (2.0 ± 0.2) than in ET (1.4 ± 0.8) (p < .05) and the cortisol levels (ng/mL) at 0 h were higher in ET (150.3 ± 55.3) than in OA (33.9 ± 9.7). Fillets prepared from Nile tilapia anesthetized using OA essential oil received higher sensory scores (1.5 ± 0.1) than fillets from both the Lippia alba (1.0 ± 0.1) and control groups (0.7 ± 0.1). OA and LA essential oils at 500μL/L were effective in inducing deep anesthesia with unconsciousness duration of 126 s, allowing time for bleeding. Anesthesia with Ocimum americanum and Lippia alba essential oils may be indicated as an alternative to hypothermia for the pre-slaughter handling of fish in Brazil. A robust computational approach may guarantee a reduction in costs in the initial stages of drug discovery, novelty and accurate results. In this work, a virtual screening workflow of a library consisting of ∼6 million mols. was set up, with the aim to find potential lead compounds that could manifest activity on the KOR. This in silico study provides a significant contribution in the identification of compounds capable of interacting with a specific mol. target. The main computational techniques adopted in this exptl. work include: (i) virtual screening; (ii) drug design and leads optimization; (iii) mol. dynamics. The best hits are tripeptides prepared via solution phase peptide synthesis. These were tested in vivo, revealing a good antinociceptive effect after s.c. administration. However, further work is due to delineate their full pharmacol. profile, in order to verify the features predicted by the in silico outcomes.

Polyurethane foams currently dominate the global foam market. Nevertheless, the main drawback of these foams remains the use of toxic isocyanate reagents. To overcome this issue, several ways have been explored. The most promising route appears to be the aminolysis of 5-membered cyclic carbonates leading to formation of polyhydroxyurethanes (PHUs). However, the lack of reactivity of systems using C5-cyclic carbonates currently limits the application range of PHUs. Hence, this study reports the first synthesis of PHUs foams using a new tetrafunctional 6-membered cyclic carbonate (TC6). The TC6 was reacted with different diamines under catalyst-free and mild temperature conditions in presence of a chem. blowing agent. Three different amines were employed, allowing access to various foams properties. Finally, promising flexible foams have been obtained, with interesting properties that could make them possible new candidates for applications as seating or insulation mattresses.

In this research, the effects of replacement of 30% of wheat flour in a control biscuit formulation with different whole grain flours (barley, buckwheat, oat, amaranth or soy) or 15% of wheat flour with apple- or oat dietary fiber (DF) were investigated in terms of their impact on the nutritional quality, rates of starch digestibility and sensory characteristics of the final product. Modifications of control formulation resulted in improved macronutrient distribution ranges and increase of dietary fiber (DF) and protein content. Soy- and amaranth enriched samples contained significantly lower rapidly available glucose (RAG) and total starch (TS) content in comparison to control sample. The anal. of starch digestibility showed that the most significant increase of RAG content in the starch fraction was observed in oat- and apple dietary fiber enriched samples defining them as samples with the highest expected glycemic index (GI). All modifications of control sample resulted in formulations with acceptable sensory characteristics; addition of pure fiber can be considered as significantly less favorable in comparison to whole grain flours especially regarding the taste and texture of the final product.

Fusarium oxysporum is the primary pathogen causing root rot disease that severely affects cultivation of jujube fruit in the Xinjiang province of China. The aim of this study was to identify endophytic bacteria in healthy jujube organs that could effectively suppress F. oxysporum growth. Different plant organs (leaves, twigs and roots) were collected from healthy Chinese jujube cultivated in southern Xinjiang province of China. The endophytic bacterium Brevibacterium halotolerans JZ7 was selected for its strong antagonistic activity and growth-promoting characteristics. Gas-chromatog. mass-spectrometry anal. showed that acetoin, 2,3-butanediol and fenretinide were the three dominant volatile organic compounds produced by strain JZ7. Fenretinide strongly suppressed spore germination of F. oxysporum in vitro. Pot experiments showed that strain JZ7 colonized both the roots and rhizosphere soil of Chinese jujube and significantly reduced F. oxysporum level in jujube rhizosphere soil. We demonstrated that B. halotolerans JZ7 can be developed into a biol. control agent to combat root rot disease of Chinese jujube in the Xinjiang province of China. Significance and Impact of the Study : The suggested strategy for biol. control of jujube root rot disease is fully in accordance with the current principles of sustainability.

The reaction process was studied by calorimetry, and the materials obtained were characterized by thermogravimetry, thermomech. studies and mech. testing. Thiol-epoxy materials showed a better mech. performance than thiol-ene photocured thermosets. This new epoxy monomer of renewable origin allowed to prepare materials with final properties better than those obtained from the diglycidylether of bisphenol A, owing to the high functionality of bis-eugenol and its rigid and compact structure.
